[Lymph, plasma and urinary hydrolases during animal feeding with different types of food].

In acute and chronic tests with dogs the effect of feeding the animals on meat, bread, or butter upon the pancreatic enzymes incretion was studied. The activity of the amylase, lipase, trypsin and of its inhibitor was determined during an acute test in the thoracic duct lymph, in the blood plasma of the portal and jugular veins, the amylase and lipase being also determined in the urine. The incretion of the pancreatic enzymes is adapted to the type of the food stimulant, viz. after feeding on bread it is the amylase incretion that increases to the most, on meat--it is that of trypsin and its inhibitor and on butter - of lipase. Most informative as to the level of the hydrolases increation adaptability is separation of corresponding enzymes forming part of the lymph, less so - their content in the plasma of the portal and still less - in the peripheral venous blood and the least - the content and segregation of the hydrolases from the urine.

Construction of a lactose-assimilating strain of baker's yeast.

A recombinant strain of baker's yeast has been constructed which can assimilate lactose efficiently. This strain has been designed to allow its propagation in whey, the byproduct resulting from cheese-making. The ability to metabolize lactose is conferred by the functional expression of two genes from Kluyveromyces lactis, LAC12 and LAC4, which encode a lactose permease and a beta-galactosidase, respectively. To make the recombinant strain more acceptable for its use in bread-making, the genetic transformation of the host baker's yeast was carried out with linear fragments of DNA of defined sequence, carrying as the only heterologous material the coding regions of the two K. lactis genes. Growth of the new strain on cheese whey affected neither the quality of bread nor the yeast gassing power. The significance of the newly developed strain is two-fold: it affords a cheap alternative to the procedure generally used for the propagation of baker's yeast, and it offers a profitable use for cheese whey.

Comparison of ethanol-soluble proteins from different rye (Secale cereale) varieties by two-dimensional electrophoresis.

The major storage proteins from six rye varieties, grown under the same conditions in 1997 and 1998 in Rønhave, Denmark, were analyzed by two-dimensional (2-D) polyacrylamide gel electrophoresis. The proteins were extracted from ground rye kernels with 70% ethanol and separated by 2-D electrophoresis. The gels were scanned, compared using ImageMaster software and the data sets were analyzed by principal component analysis (PCA) using THE UNSCRAMBLER software. Afterwards MATLAB was used to make a cluster analysis of the varieties based on PCA. The analysis of the gels showed, that the protein patterns (number of different proteins and their isoelectric points and molecular weights) from the six rye varieties were different. Based on the presence of unique cultivar-specific spots it was possible to differentiate between all six varieties if the two harvest years were investigated separately. When the results were combined from the two years five varieties could be differentiated. The results from the PCA confirmed the finding of the unique spots and cluster analysis was made in order to illustrate the results. The combination of the results from 2-D electrophoresis and other grain characteristics showed that one protein spot was located close to the parameters bread volume and bread height.

Assessment of the agronomic and technological characteristics of Triticum turgidum ssp. dicoccum Schrank and T. spelta L.

Recently, the peoples interest in natural and organic products led to a "rediscovery" of hulled wheat, particularly T. turgidum ssp. dicoccum, commonly known as "farro". Compared with durum wheat (T. turgidum ssp. durum), farro is characterized by low productivity and low quality semolina and flour, which makes it unsuitable for the production of bread and pasta. In this paper, the agronomic, technological and nutritional characteristics of three new varieties of farro obtained by crossing the T. turgidum ssp. dicoccum c.v. Molise with T. turgidum ssp. durum c.v. Simeto, were analysed. Data were then compared with those concerning older populations of T. turgidum and of two varieties of T. spelta. The aim of this work is to highlight the productive advantages of the new varieties and to emphasize their prospective use in the production of bread and pasta with considerable health properties due to nutritional characteristics of hulled cereals.

Occurrence of certain mycotoxins in corn and corn-based products and thermostability of fumonisin B1 during processing.

A total of 57 samples of corn and corn-based products collected from various districts of Egypt were analyzed for Fusarium mycotoxins (T-2, diacetoxyscripenol (DAS), deoxynivalenol (DON) and fumonisin B1 (FB1) and aflatoxins. FB1 was detected in about 80%, 53.85%, 33.3%, and 28.57% of yellow corn, corn meal, white corn, and popcorn samples, respectively. The levels of FB1 ranged from 10 to 780 microg/kg. T-2 and DAS were detected in 5% and 10% of yellow corn samples, respectively, and DON was detected in white corn and popcorn samples at levels of 28.8 and 10.1 microg/kg, respectively. Starch samples were found to be free from Fusarium mycotoxins. Baking balady bread at 450 degrees C/min reduced FB1 to 72.4% while baking Franco bread at 250 degrees C/20 min reduced FB1 to 57.4%. Boiling of macaroni and corn in water completely removed FB1 from contaminated samples. On the other side, corn flakes samples were found to be contaminated with aflatoxins B1 and G1 at levels ranging from 6 to 10 ppm, whereas 2.9% of samples were contaminated with aflatoxin B1 > 35 ppm and G1 > 16 ppm.

Food consumption and cancer of the colon and rectum in north-eastern Italy.

The relation between dietary factors and the risk of colorectal cancer was investigated in a case-control study conducted in Pordenone province, North-eastern Italy, on 123 cases of colon cancer, 125 of rectal cancer and 699 controls admitted to hospital for acute, non-neoplastic or digestive disorders. Consistent positive associations were observed with more frequent consumption of bread (odds ratio, OR = 2.1 for colon and 2.2 for rectum for highest vs. lowest tertile), polenta (OR = 2.1 for colon, 1.9 for rectum), cheese (OR = 1.7 for colon, 1.8 for rectum) and eggs (2.5 for colon, 1.9 for rectum), whereas reduced ORs were observed in subjects reporting more frequent consumption of tomatoes (OR = 0.5 for colon and 0.4 for rectum). High consumption of margarine exerted a significant protection against cancer of the colon whereas high consumption of carrot spinach, whole-grain bread and pasta (favorably) and red meat (unfavorably) affected rectal cancer risk in particular. Thus the present study gives support for a protective effect associated with a fiber-rich or vegetable-rich diet, while it indicates that frequent consumption of refined starchy foods, eggs and fat-rich foods such as cheese and red meat is a risk factor for colo-rectal cancer.

Proteomic analysis of aneuploid lines in the homeologous group 1 of the hexaploid wheat cultivar Courtot.

Three monosomic lines (MSLs) and three nullisomic lines (NSLs) of the homeologous group 1 and one euploid line of the bread wheat Triticum aestivum cultivar Courtot were used in a proteomic approach to investigate the effects of zero, one or two doses of chromosomes 1A, 1B and 1D on the amount of endosperm proteins. Polypeptides whose amounts changed significantly between each aneuploid line and the euploid line were identified using image analyses of two-dimensional gel electrophoresis patterns resulting from specific endosperm protein extractions. Matrix-assisted laser desorption/ionization-time of flight mass spectrometry and electrospray ionization tandem mass spectrometry were also used for protein identification. Removing one chromosome or a chromosome pair allowed varying responses to be observed for the remaining endosperm protein genes. Compensation phenomena for the high molecular weight glutenin subunits (HMW-GS) were detected only in the MSLs. Subunits Bx7, By8 and Dy12 were the only HMW-GS overexpressed (from 152-737%) when chromosomes 1A or 1B or 1D were at hemizygous state. Thirteen new protein spots were detected only in the NSL1D, and seven were identified as HMW-GS analogs. These seven new spots may result from the expression of inactive genes. The HMW-GS were of significantly higher volume in MSLs, whereas the low molecular weight glutenin subunits and the gamma-gliadins were of lower volume in aneuploid lines. Most of the down-regulated proteins in the MSLs were storage proteins encoded at loci located on another chromosome pair. Complex regulations between chromosomes and loci of the homeologous groups 1 and 6 in bread wheat are discussed.

Towards royal jelly proteome.

The recent availability of the honey-bee Apis mellifera genome and trascriptome of both the female castes, has stimulated new efforts in investigating the protein composition of royal jelly (RJ), its role in caste differentiation and its quality and typicality by a proteomic approach. This study is aimed both to separate and identify proteins of royal jelly and to detect some of them in honey-bee pollen-bread by using two-dimensional gel electrophoresis, mass spectrometry and by de novo sequencing. All the identified proteins belonged to the Apis mellifera genome. Apalbumin 1 was also confirmed to be present in honey-bee pollen-bread where the presence of apalbumin 2 was also found. In addition several fragments of apalbumin 1 and apalbumin 3 were also found in RJ. These could be the result of protease activity other than that of serine-protease. This study is a contribution to the description of royal jelly proteome.

Glutenin and gliadin contents of flour derived from wheat infested with different aphid species.

Aphid feeding in cereals results in both quantitative and qualitative yield losses; moreover aphid-transmitted viruses are responsible for other quantitative and qualitative damage so that direct or indirect effects of aphid infection are always of interest. At the beginning of stem elongation, wheat plants were caged regardless of the presence or absence of indigenous cereal aphids. Half of the caged plants were artificially infected with Diuraphis noxia. The number of D noxia and Rhopalosiphum padi individuals on plants destined for flour preparation was approximately 150 aphids per tiller at peak population. The number of Sitobion avenae individuals was ca 25 aphids per tiller. Bread-making quality of wheat flour is determined primarily by the protein content, the gluten proteins (gliadins, glutenins) being the prime factors. The absolute amounts and/or the relative proportion of gliadins and glutenins are very important in dough making and in determining baking quality. Flour from grains originating from plants infected naturally with cereal aphids indigenous to Hungary, and flour infected artificially with D noxia, were analysed for glutenin and gliadin content using size exclusion HPLC. It was found that aphid infection had a significant effect on the gliadin/glutenin ratio, which was significantly lower in wheat flour prepared from aphid-infected plants than in that from uninfected control. The most significant decrease in gliadin/glutenin ratio was caused by D noxia infection, followed by R padi and then S avenae. As the gliadin/glutenin ratio was significantly lower in flour made from aphid-infected wheat seeds, it can be supposed that aphid feeding results in decreased bread making quality of wheat flour.

Dietary associates of serum total, LDL, and HDL cholesterol and triglycerides in patients with coronary heart disease.

BACKGROUND: Diet-lipid associations established in clinical trials have in general been weak or nonexistent in cross-sectional studies within a population. Our objective was to analyze the dietary associates of serum lipids in patients with coronary heart disease (CHD) not using lipid-lowering medication. METHODS: Patients with coronary bypass grafting (n = 49), balloon angioplasty (n = 46), acute myocardial infarction (n = 79), and acute myocardial ischemia (n = 79) participated in a survey (EUROASPIRE). Patients were selected from hospital records at least 6 months after hospitalization. Diet was assessed by a food record, a short questionnaire, and fatty acid composition of serum cholesteryl esters (CE). RESULTS: Neither the intake of total fat nor that of saturated, monounsaturated, or polyunsaturated fatty acids was associated with serum lipids. Use of soft margarine on bread (though not in cooking or baking) and high intake of fiber and cereal products were associated with low total cholesterol. Linoleic acid in CE was inversely associated with total cholesterol and triglycerides, and eicosapentaenoic acid was inversely associated with triglycerides and positively associated with HDL cholesterol. CONCLUSIONS: In the present study use of soft margarine on bread (though not in cooking or baking) and high intake of fiber and cereal products were associates of lowered serum cholesterol concentrations in CHD patients. Fatty acid composition of CE reflected dietary fatty acid intake involved in cholesterol lowering better than food records.

Bioavailability of cadmium in rats fed various diets.

Six-week-old female albino rats were fed rat diet or human foods: meat, bread or milk--3 days before and 6 days after a single oral dose of 115m Cd. All animals were killed 6 days after administration and the radioactivity in the whole body and in the gut-free carcass were determined in a double scintillation counter. Gut retentions were calculated as the difference: whole body minus carcass. All animals fed meat, bread or milk had much higher body retentions than animals fed rat diet (whole body 4-5 times, carcass about 3 times and gut 10-14 times). Our results point out the importance of nutritional factors in metal metabolism and toxicity.

Glycaemic responses to commonly ingested breakfasts in children with insulin-dependent diabetes mellitus.

UNLABELLED: The glycaemic response to commonly ingested breakfast with different glycaemic index (cornflakes, white bread, dark rye bread and muesli) was investigated in 14 children with insulin-dependent diabetes mellitus. The blood glucose response after cornflakes was significantly higher than after the other breakfasts, whereas the areas under the blood glucose curve were not statistically different. We observed no difference in blood glucose rise between the other breakfast types. CONCLUSION: Postprandial blood glucose measurements after meals with high glycaemic index may be useful in order to minimize the postprandial rise in blood glucose.

The nutritive value of new and traditional sorghum and millet foods from Sudan.

In the present study was examined the nutritive value of different sorghum and millet based baby foods. Different types of kisra, some fortified with chickpeas or peanuts, were also included in the study. Finally was compared the influence of replacing wheat bran with sorghum bran in bread. The nutritive value was assessed by chemical analyses and by rat balance studies. The addition of chickpea and peanuts improved the quality of kisra a staple food of Sudan. This type of kisra can be used as a well-balanced food for infants above the age of one year. The baby food developed from sorghum/millet malt, milk powder and processed by drum drier provides a safe level of protein for children above the age of one month. This formulation and way of processing is well suited for commercial production of sorghum/millet based baby food. Based on the results of the present investigation it is recommended that the use of brown bread for infants should not exceed ten percent wheat or sorghum bran as it affects the digestibility in a negative way.

Polymorphism and inheritance of gliadin components controlled by chromosome 6A of spring durum wheat.

The gliadin composition of 78 spring durum wheat varieties has been studied by one-dimensional (Al-lactate, pH 3.1) and two-dimensional (first dimension, Al-lactate, pH 3.1; second dimension, sodium dodecyl sulfate-polyacrylamide gel) electrophoresis. Analysis of hybrids has shown that all components of the alpha zone of gliadin spectra are inherited together as blocks and are, probably, coded for by a cluster of tightly linked genes located on chromosome 6A. Fourteen variants of gliadin blocks have been identified, which can be classified into five families on the basis of component composition. All families but one have analogues among chromosome 6A-controlled blocks of bread wheat. The results indicate that some of the genome A diploid genotypes that were ancestors of durum wheats were also ancestors of bread wheats and that polyploid wheats were produced by repeated allopolyploidization events, as has been suggested earlier.

Diet and other risk factors for fissure-in-ano. Prospective case control study.

The findings of a partly prospective case control study of chronic fissure-in-ano conducted in two Danish outpatient clinics are reported. One hundred seventy-four patients with chronic fissure-in-ano were matched by age and sex to outpatients from the same community suffering from benign skin tumors. All subjects were interviewed regarding diet, beverage consumption, occupational exposures, and medical and surgical history. Significantly decreased risks were associated with frequent consumption of raw fruits, vegetables, and whole-grain bread, and significantly increased risks were associated with frequent consumption of white bread, sauces thickened with roux, and bacon or sausages. Risk ratios for consumption of coffee, tea, and alcohol were not significantly different. No statistical associations were found with particular occupational exposures. However, a history of previous anal surgery was reported significantly more often for cases than for controls. Current evidence indicates that anal fissure is likely to result at least partly from an inappropriate diet and that dietary manipulations might reduce the incidence of fissure-in-ano.

Supplementary selenium influences the response to fatty acid-induced oxidative stress in humans.

The mutual influences of wheat selenium (Se) and n-3 polyunsaturated fatty acids (n-3 PUFA) on plasma Se and indicators of increased oxidative stress were investigated in a randomized, double-blind study with 31 women (23.5 +/- 3.4 yr). Groups 1 and 2 ingested 5.4 g n-3 PUFA daily (as ethyl esters), whereas groups 3 and 4 received placebo capsules. Groups 2 and 3 received 3 slices of high-Se bread daily, providing 115 micrograms Se, in addition to the 77 +/- 26 micrograms Se in the diet. Groups 1 and 4 received placebo slices. Blood samples were drawn at baseline and at 3 and 6 wk. Serum Se concentrations increased in both groups given Se-enriched bread, but significantly less in subjects given n-3 PUFA (group 2). There were no changes in the plasma ratio alpha-tocopherol:mg cholesterol or plasma ascorbic acid levels. In group 1, plasma-conjugated dienes and thiobarbituric acid-reactive substances (TBARS) rose by 130% (p < 0.005) and 126% (p < 0.005), respectively. Two-way ANOVA showed significant interaction effects of Se and n-3 PUFA on changes in conjugated dienes (p = 0.03) and TBARS (p = 0.015), Se treatment apparently modifying the peroxidative effects of n-3 PUFA. In subjects receiving n-3 PUFA, changes in conjugated dienes and TBARS were negatively correlated with changes in serum Se. In summary, n-3 PUFA modified the effect of Se supplementation, whereas Se seemed to modify the peroxidative effects of n-3 PUFA.

A comparative study in rats of measures of the availability of dietary zinc and iron.

The purpose of this study was to compare three measures of the availability of dietary Zn and Fe in order to test their validity. Thirty-six 5-wk-old rats were fed deionized water and wheat crispbread made from endosperm flour, whole-grain flour, or endosperm flour supplemented with Zn and Fe to the whole-grain levels ad libitum for 14 d. The retention of 65Zn and 59Fe from test meals of the same breads after 1 wk and the sum of the excretion of endogenous Zn and Fe (injected 65Zn and 59Fe) with the Zn and Fe balances, respectively, were used as independent measures of Zn and Fe absorption. Measurements of Zn absorption, Zn balance, and serum Zn concentration gave quite different results with regard to the availability of Zn in the three breads, presumably because of the homeostatic regulation of the absorption and excretion of Zn when the Zn in the diet is in excess of the body's needs. Measurements of Fe absorption, Fe balance, and Fe concentrations in liver and serum were consistent in demonstrating overloading of Fe in the group given wheat-endosperm crispbread supplemented with Zn and Fe, but there was evidence that the isotope retention method overestimated iron absorption.
