[The characteristics of the ultrastructural changes in yeast-like fungi of the genus Candida exposed to therapeutic-range ultrasound and nystatin].

The electron microscopic study of the ultrastructure of yeast-like fungi of the genus Candida under the action of nystatin and sonication, as well as the action of these two factors used in combination, was carried out. In this study the degree of preservation of the structure of fungal cells was evaluated and the percentage of cells with different ultrastructural changes was determined. Observations made 3 and 24 hours after sonication and treatment with nystatin, used in combination, showed that ultrastructural changes in fungal cells were more pronounced than in the cells subjected to the action of nystatin alone. 24 hours after the combined physico-medicinal treatment of Candida the percentage of nonviable blastospores among them was 5-fold higher than that of such cells among Candida subjected to treatment with nystatin alone and more than 20-fold higher than among Candida subjected only to sonication. These data on the increased fungicidal effect of nystatin used in combination with sonication substantiate the expediency of the above-mentioned physico-medicinal treatment in cases of candidal lesions of the skin and its appendages, which is one of the ways for increasing the effectiveness of the treatment of this mycosis.

Fluorescence and ultrastructural localization of actin distribution patterns in the nucleus of HL-60 and K-562 cell lines treated with cytostatic drugs.

Actin has been studied extensively for decades, but so far its well-defined role has been found localized to the cytoplasm. The present study characterizes the distribution pattern of actin in the nucleus of HL-60 and K-562 cell lines treated with etoposide and doxorubicin. F-actin labeled with TRITC-phalloidin was found in the nucleus in both cell lines independently of the cytostatic drugs used. Using confocal microscopy F-actin is seen in the center of the nucleus showed labeling of the nucleoli by phalloidin. There are also distinct tracts of F-actin seen from nucleoli to the nuclear envelope in some sections. HL-60 cells treated with 200 micro M etoposide and 5, 10 micro M doxorubicin showed cells with characteristic features of apoptosis at the ultrastructural level. Intense immunogold labeling for actin was seen in nucleus of HL-60 cells with compaction and margination of chromatin. In K-562 cells more intense labeling was often found in the cytoplasm of the cells. Positive labeling for actin was not found after control incubations. F-actin is present in the nucleus and there is labeling of nucleoli by phalloidin. The observation at the ultrastructural level suggests that actin can be involved in chromatin reorganization during the process of apoptosis. Increase of labeling at the ultrastructural level in the nucleus of HL-60 cells in relation with compaction and margination of nuclear chromatin might also suggest translocation of actin from cytoplasm to the nucleus in connection with chromatin reorganization.

[Effect of microwave irradiation on neurocyte mitochondrial ultrastructure and mtTFA mRNA expression in rats cerebral cortex and hippocampus].

OBJECTIVE: To explore molecular controlling mechanism of mitochondrial injury induced by different density of microwave irradiation. METHODS: Rats were exposed to microwave irradiation for 1 hour at average power density of 3 mW/cm(2) or 30 mW/cm(2). After microwave irradiation, the changes of pathological ultrastructure of rat cerebral cortex and hippocampus were observed by electron microscope, and mitochondrial transcription factor A (mtTFA) mRNA expression level were determined by RT-PCR. RESULTS: After 3 mW/cm(2) microwave irradiation for 0, 3, 24 h, mitochondrial ultrastructure and mtTFA mRNA expression level didn't significantly change in rat cerebral cortex and hippocampus. After 30 mW/cm(2) microwave irradiation for 0, 3, 24 h, mitochondrial ultrastructure obviously changed, mtTFA mRNA expression in rat hippocampus significantly increased by 67.00%, 80.00%, 30.00% respectively, and in rat cerebral cortex by 133.00%, 86.00%, 233.00% respectively. There were significant differences between the corresponding groups of hippocampus and cerebral cortex (P < 0.01). CONCLUSION: No obvious change in mitochondria was found after 3 mW/cm(2) microwave irradiation, but it was found after 30 mW/cm(2) microwave irradiation. Mitochondria injury in cerebral cortex was more severe than that in hippocampus. mtTFA mRNA may have certain regulation in mitochondrial energy metabolism.

[Ultrastructural changes in muscular tissues of dystrophin/utrophin double-knockout mice after bone marrow-derived mesenchymal stem cell transplantation].

OBJECTIVE: To observe the ultrastructural changes in the muscular tissues of Duchenne muscular dystrophy mouse models with dystrophin/utrophin double-knockout (DKO) after transplantation of bone marrow-derived mesenchymal stem cells (MSCs). METHODS: The fifth passage of in vitro cultured MSCs from SD rats were transplanted into the DKO mice via the tail vein, and the ultrastructural changes in the gastrocnemius of the recipient rats were observed using transmission electron microscope 15 weeks after the transplantation. RESULTS: The fifth passage of the in vitro cultured MSCs growing in colonies possessed good homogeneity with low immunogenic activity for transplantation via the tail vein. After transplantation, the DKO mice exhibited improved motor function and prolonged survival period. Ultrastructural observation of the gastrocnemius of the recipient DKO mice revealed much alleviated sarcolemmal disruption, dissociation and edema of the subsarcolemmal tissues, central nuclear shift, loosening of the local myofibrils, inflammatory cell infiltration and connective tissue hyperplasia in comparison with the tissues from the DKO mice without MSC transplantation. Fusion of several immature nuclei was observed in the subsarcolemmal region. CONCLUSIONS: MSCs possess strong plasticity in vitro and in vivo, and after transplantation, the MSCs may migrate to lesioned muscular tissues via the blood circulation to participate in the repair and regeneration the atrophied muscular tissues.

Qualitative and quantitative studies on the ultrastructure of ovine pinealocytes during postnatal development.

OBJECTIVE: The study was performed to analyze the ultrastructure of ovine pinealocytes during the period of postnatal development. MATERIAL AND METHODS: The pineals of newborn, 10-week and one-year old females of the domestic sheep were prepared for ultrastructural investigations. The point count analysis was used in quantitative studies of the pinealocyte substructures. RESULTS: The prominent feature of pinealocytes in the newborns was the presence of well developed rough endoplasmic reticulum and numerous polysomes. The pinealocyte cytoplasm contained also smooth endoplasmic reticulum, mitochondria, Golgi apparatus, lysosomes, dense core vesicles, multivesicular bodies and lipid droplets. Pinealocytes of the 10-week and 1-year old sheep were characterized by the occurrence of numerous vesicles and short cisterns of smooth endoplasmic reticulum, abundant microtubules and lipid droplets. Pinealocytes of the adult sheep were distinguished by well developed Golgi apparatus, numerous dense core vesicles and multivesicular bodies. The relative volume of rough endoplasmic reticulum in pinealocytes was significantly higher in the newborn sheep than in two other groups. The relative volume of mitochondria was significantly higher in pinealocytes of the 10-week old sheep than the newborns and one-year old animals. The relative volume of Golgi apparatus was significantly higher in the one-year old animals than in two other groups. No differences concern lysosomes. The relative volume of lipid droplets as well as the numerical density of dense core vesicles and multivesicular bodies increased significantly with age. CONCLUSION: The ultrastructure of ovine pinealocytes undergoes the marked changes during postnatal development. The changes concern mainly substructures involved in secretory activity.

Avian minor salivary glands: an ultrastructural study of the secretory granules in mucous and seromucous cells.

Ultrastructural descriptions in birds are scarce thus, in this study we have characterized the secretory granules of mucous and seromucous cells from the palatine and lingual salivary glands of birds with different diets. The samples were taken from the tongue and palatine mucosa of chicken (Gallus gallus), quail (Coturnix coturnix), chimango (Milvago chimango) and white heron (Egretta thula). The samples were processed for observation by transmission electron microscopy (TEM) employing 4% Karnovsky solution for fixation. The most noteworthy finding was the heterogeneous ultrastructural appearance of the secretory granules. Differences in substructure were found between the four species, between the palatine and lingual glands in the same species and even within the same acinus and the same cell. At variance with other authors, these differences cannot be attributed to the type of fixative solution used taking into account that all the samples were processed in the same way. Previous histochemical studies have shown the presence of sulfated and non sulfated glycoconjugates in these glands which can be associated to the maturation of the granules. These granules are probably representative of peculiar storage of the secretory products that would give rise to a heterogeneous and complex ultrastructural pattern of granules in the mucosa and seromucosa cells of these avian species.

[Ultrastructure of the cell wall in vegetative mycelia of Agaricus bisporus (Lange) Imbach].

A comparative description of the cell wall ultrastructure was made for vegetative mycelia in Agaricus bisporus, other species of Agaricus, and Pleurotus ostreatus. Apart from well known ultrastructural features of cell walls of the examined strains, inclusions of different kinds were observed. The cell wall thickness varied in different Agaricus strains. Maximum differences between the examined strains were 1.4 times. The cell wall thickness in P. ostreatus strains was by 3 to 4 times less than that in Agaricus strains. Using electron micrographs, a brief description of mycelia and A. bisporus basidia ultrastructure was composed.

[Localization of ATPase activity, respiration and ultrastructure of wheat root cells with modulated ion conductivity of plasma membrane].

Changes in the localization of ATPase activity, respiration and ultrastructure of wheat root cells with modulated ion conductivity of plasma membrane were studied. A 2 h treatment of excised root with valinomycin (20 microM), N,N-dicyclohexylcarbodiimid (100 microM), gramicidin S (20 microM) and chlorpromazine (100 microM) caused an increased loss of potassium by cells, a decreased respiration and changes in the localization of ATPase activity and in cell ultrastructure. Differences in the observed changes may be conditioned by different mechanisms of action of the membrane active compounds used. It is concluded that changes in the localization of ATPase activity and ultrastructure may indicate some early specific responses of root cells, whereas the increase in the ion conductivity and decrease in respiration under disruption of ion homeostasis caused by membrane active compounds indicate unspecific responses of cells.

[Macular corneal dystrophy including histologic and ultrastructural changes].

PURPOSE: The study reports the results of a histological and ultrastructural examination of the corneal button, obtained during penetrating keratoplasty from patient with clinically recognized macular corneal dystrophy. MATERIAL AND METHODS: 34-year-old male patient suffering from macular corneal dystrophy (MCD) has been treated on corneal epithelium defect and photophobia since his early childhood. Visual acuity was decreased on the Snellen test chart to 0.02. Slit-lamp examination, and ultrasonographical measurement of the cornea's thickness were performed. Removed during penetrating keratoplasty corneal button was divided into two pieces. One of them was prepared in standard procedure for histological examination in the light microscopy after having been stained with hematoxylin and eosin, alcian blue and paS-method. From the other part, slides for ultrastructural examination in the transmission electron microscopy were prepared with the use of standard method. The family history from the patient was also taken, and available relatives have undergone examination in search of typical MCD symptoms. RESULTS: Slit-lamp examination findings revealed diffuse, from limbus to limbus, stromal opacification. In measurement by pachymeter cornea's thickness was reduced. In the light microscopy, in typical stained slides, delaminations within stroma and deficit of endothelial cells were observed. After being stained with alcian blue, dark blue deposits in the places of delamination became visible. By transmission electron microscopic examination, intracellular and extracellular deposits were detected in the stroma, Descemet membrane and endothelium. Distended keratocytes with enormous vacuoles containing abnormal material were found. Pedigree was typical for autosomal recessive inherited disease. CONCLUSIONS: Histological and ultrastructural diagnosis is a basis of recognition of macular corneal dystrophy. Analysis of the pedigree as well as ultrasonographical measurement of the cornea's thickness is very helpful to establish the right diagnosis.

[Ultrastructure and clinicopathology of pleomorphic adenomas of the salivary gland. I. Methods and results].

The observations are a ultrastructural analysis of pleomorphic adenomas with one's own 14 cases. Pleomorphic adenoma is chief representative of the secretory tumour complex of salivary glands. Ultrastructure of three dominate secretory tumor cells and three nonsecretory celltypes are descripted. Ultramicroscopical differentiation of stroma elements an their transformations, henceforth regressive of tumour cells in the pleomorphic adenoma are discussed. The author give instructions for electromicroscopic diagnostic of this adenomas. Lastly there is tried to determine the "dignity" of pleomorphic adenomas by their ultrastructure for purpose of a adequate therapy.

[Ultrastructural basis of interactions between prokaryotes and eukaryotes in different symbiotic models].

This paper reviews the Author's contribution to the knowledge of the ultrastructural basis of the prokaryote-eukaryote interactions in different models assessed by an ultrastructural approach. In agreement with the hypothesis of the origin of eukaryotic cells, which are chimeras of several prokaryotes with different morpho-functional specializations, symbiosis had major consequence for evolution of life. In Arthropods, one of the most successful lifestyles, the presence of endosymbiotic prokaryotes, plays an important role in their metabolism. In some cases, genome integration has occurred in the endosymbiotic relationships with the host, proving that intracellular symbiosis is not merely a nutritional supplement. Intracellular symbiotic bacteria are also described in nematodes. In particular, the presence of intracellular Wolbachia in filariae, even if its function is not yet completely known, influences positively the reproductive biology and the survival of the host, as proved by antibiotic treatment against this bacterium. The ultrastructural images reported in this review were obtained using different species of cockroaches, termites, ticks and filarial nematodes. The traditional methods of transmission (TEM), scansion (SEM) and immuno electron microscopy were used. In addition, also freeze-fracture and deep-etching techniques were employed. The cockroaches and the primitive termite Mastotermes darwiniensis host symbiotic bacteria in the ovary and in specialized cells (bacteriocytes) of the fat body. These bacteria have the typical cell boundary profile of gram-negative bacteria and are enveloped in a vacuolar membrane produced by the host cell. Molecular sequence data of 16S rDNA of endosymbionts of five species of cockroaches and M. darwiniensis indicate that they are members of the Flavobacteria-bacteroides group and that the infection occurred in an ancestor common to cockroaches and termites probably after the end of the Paleozoic (250 Ma BP). The symbiotic bacteria are transmitted transovarially and, during embryogenesis, they are integrated into the morphogenetic processes. In particular, we were able to demonstrate that the origin of the bacteriocyte should be looked for in the cells of the haemocyte line (embryonic plasmatocytes). The eggs are infected by the bacteria emerging from the bacteriocytes of the ovaric fat body and, at the end of the vitellogenesis, they are actively phagocytized by the egg membrane. In filarial nematodes, intracellular bacteria belonging to the genus Wolbachia have been described: they have evolved an obligatory mutualistic association with their host. In fact, antibiotic treatments lead to the clearance of bacteria and this loss produces a negative impact on reproduction and survival of the filarial host. We evidenced, by TEM, the degenerative events occurring during the embriogenesis of Brugia pahangi and Dirofilaria immitis after tetracycline treatment. The data suggest that the Wolbachia play a direct role in worm metabolism. Finally, a new additional model of the prokaryote-eukaryote interaction has been described: we have recently discovered a new intracellular alpha-proteobacterium, named Iric ES1, which resides in the ovarian tissues of the tick Ixodes ricinus. The intriguing characteristic of this bacterium is its ability to invade and consume the ovaric mitochondria. From an evolutionary perspective, it is interesting to note that Iric ES1 enters mitochondria in a similar way to that employed by the "predatory" bacterium Bdellovibrio bacteriovorus.

Tenon's capsule: ultrastructure of collagen fibrils in normals and infantile esotropia.

No detailed information about the ultrastructure of Tenon's capsule has been published. The purpose of the present study was to compare the ultrastructural features of collagen fibrils from Tenon's capsule in a nonstrabismic control group (seven children) to those in an infantile esotropic group (10 children). Small biopsy specimens from Tenon's capsule were taken during various operations to be examined by electron microscopy. On electron microscopy, the capsule was found to be composed of groups of collagen fibrils arranged irregularly in different orientations, forming a three-dimensional network that provides tissue resistance to stress. The cross-sectioned collagen fibrils were studied by an image analyzer. In both study groups, all fibrils had a round, regular contour. In the esotropic group, the Tenon's collagen fibrils were thicker, as reflected by their significantly greater mean diameter: 101 +/- 5 nm (mean +/- standard deviation) compared to 86 +/- 5 nm in the control group. Also, significantly greater heterogeneity was found in the collagen fibril thickness of each individual in the esotropic group compared to the control group. Moreover, the mean number of collagen fibrils per unit area was significantly higher in the esotropic group: 98 +/- 13 fibrils per 10(6) nm2 compared to 73 +/- 5 fibrils per 10(6) nm2 in the control group. These ultrastructural changes may be stress-induced secondary alterations of the Tenon's collagen fibrils resulting from prolonged deviation of the eye in infantile esotropia. The significantly denser collagen fibrils may cause a decrease in the elasticity of Tenon's capsule in infantile esotropia.

Diurnal rhythms of pinealocyte ultrastructure, pineal serotonin content and plasma melatonin level in the domestic pig.

The study was conducted to investigate diurnal changes in pinealocyte ultrastructure, pineal serotonin content and plasma melatonin concentration in the domestic pig. The immature pigs (n=24) were kept under a cycle of 12 h light : 12 h dark, with a photophase between 0800 and 2000. During the photophase the animals were exposed to direct sunlight. After four weeks the gilts were slaughtered at 0900, 1400, 2100 and 0200. The pineals were removed and divided into two parts - one for quantitative ultrastructural study (by a point count method) and one for serotonin assay. Simultaneously, blood samples were taken for melatonin assay. The relative volume of mitochondria in pinealocyte perikarya was significantly higher at 1400 than at 0200 and 0900 as well as at 2100 than at 0200. The relative volume of Golgi apparatus was higher at 0900 and 1400 than at 0200. The relative volume of dense bodies of the MBB-1 type in pinealocyte perikarya was significantly lower at 1400 and 2100 than at 0900. In contrast, the relative volume of MBB-2 was higher at 1400 than at 0900 and 0200. The numerical density of DCV in perikarya was significantly higher at 0200 than at 1400. No significant differences were found in rough endoplasmic reticulum, lysosomes and multivesicular bodies. The pineal serotonin content showed a prominent rhythm with the maximum at 1400. The plasma melatonin concentration was significantly higher at 0200 than at 0900, 1400 and 2100. The obtained results demonstrate that both pinealocyte ultrastructure and pineal biochemistry in the pig undergo significant changes in the course of the diurnal rhythm.

[Ultrastructure of resting cells of some non-spore-forming bacteria].

Using electron microscopy (ultrathin sections and freeze-fractures), we investigated the ultrastructure of the resting cells formed in the cultures of Micrococcus luteus, Arthrobacter globiformis, and Pseudomonas aurantiaca under conditions of prolonged incubation (up to 9 months). These resting cells included cyst-like forms that were characterized by complex cell structure and the following ultrastructural properties: (i) a thickened or multiprofiled cell wall (CW), typically made up of a layer of the preexisting CW and one to three de novo synthesized murein layers; (ii) a thick, structurally differentiated capsule; (iii) presence of large intramembrane particles (d = 180-270 A), occurring both on the PF and EF sides of the membrane fractures of M. luteus and A. globiformis; (iv) a peculiar structure of the cytoplasm, which was either fine-grained or lumpy (coarse-grained) in different parts of the cell population; and (v) a condensed nucleoid. Intense formation of cyst-like cells occurred in aged (2- to 9-month-old) bacterial cultures grown on diluted complex media or on nitrogen-, carbon-, and phosphorus-limited synthetic media, as well as in suspensions of cells incubated in media with sodium silicate. The general morphological properties, ultrastructural organization, and physiological features of cyst-like cells formed during the developmental cycle suggest that constitutive dormancy is characteristic of non-spore-forming bacteria.

Ultrastructural changes of pneumocytes of rat exposed to Arabian incense (Bakhour).

OBJECTIVE: Impacts of air pollution on the human health have been recognized over the last decades. Smokes, in particular, have deleterious effects on the respiratory system. According to a local tradition, incense "Bakhour" is burnt and the resultant heavy smokes are inhaled. The objective of the present study is to investigate the ultrastructural pulmonary changes which can be induced by Arabian incense, Bakhour, exposure. METHODS: The study was conducted from September through to December 2003, at the Animal House, College of Science, King Saud University, Riyadh, Kingdom of Saudi Arabia. Two groups of Wister albino rats, Rattus norvegicus, were used. One group (n=16) was exposed to 420 grams of Bakhour for 14-weeks at the rate of 4 grams/day in the exposure chamber. Additional group of rats, of equal number, was used as non-exposed control. At the end of the exposure period, lung tissues were removed from all experimental animals and processed for electron microscopy. RESULTS: Alveolar pneumocytes of exposed animals revealed significant ultrastructural changes which involved the cell organelles and surfactant material of type II cells. Hyperplasia of alveolar cells was a feature in the affected lung tissue. Neutrophils were recognized infiltrating pulmonary alveoli and accompanied with degenerative and necrotic changes of the alveolar cells. Deposition of collagen fibrils in the alveolar walls was also observed. CONCLUSION: Basing upon the results of electron microscopy, it was concluded that exposure to Bakhour can induce ultrastructural pulmonary changes which may imply compromised respiratory efficiency.

[Changes in antioxidative system and cell ultrastructure in the fruit peels of apple during sunburn development].

Fruits from 6-year-old apple trees (Malus domestica Borkh cv. Fuji) were used as materials to test the changes of antioxidative system and cell ultrastructure of fruit peels during sunburn development. Fruit sunburn appeared in mid July, 2002. The process of apple fruit sunburn was divided into three phases (0 degrees : control): 1 degrees : bleaching, 2 degrees : brownness, and 3 degrees : necrosis. Fuji apple fruits in different sunburn state were picked. Phenolic compounds, membrane protective enzymes (SOD, POD, PPO, CAT), cell membrane lipid peroxidation and cell ultrastructure in fruit peel were studied. The result showed that the degree of cell membrane lipid peroxidation enhanced along with development of sunburn. The activity of membrane protective enzymes also increased remarkably. However, the cell structure kept its integrity, except some organelles which partly disassembled, and cytoplasm and vacuoles became enriched with electron-dense substances while fruit peels became pale. As peel became brown, chlorogenic acid, quercetin, rutin and myricetin accumulated, and cells of outer layers of the epidermis collapsed correspondingly, cell wall became thicker. It is suggested that changes in both cell ultrastructure and antioxidative system confirm that physiological state of fruit peels becomes disordered during sunburn development.

Effects of endothelin-1 or of its receptor A a selective antagonist, on histological and ultrastructural patterns in experimental acute pancreatitis in rats.

The role of endothelin-1 (ET-1) and of its receptor A (ETA) blockade in oedematous acute pancreatitis (AP) remains unclear. In 40 male Wistar rats with i.p. cerulein-induced AP, lasting 4 hours, ET-1 2x0.5 nmol/kg and 2x1.0 nmol/kg or selective ETA antagonist LU 302146, 10 mg/kg and 20 mg/kg was given i.p. simultaneously with cerulein. Histological and ultrastructural studies of pancreatic specimens were done. ET-1 decreased the inflammatory infiltration, but not the mean scores of necrosis and vacuolization in AP. The ultrastructural damage of acinar cells was less evident after ET-1 than in untreated AP. Selective ETA antagonist slightly aggravated the vacuolization and necrosis of acinar cells and some ultrastructural alterations in AP. In conclusion, ET-1, in contrast to selective ETA antagonist, exerts some protective effect in the early course of oedematous cerulein-induced acute pancreatitis in rats.

Resin-dentin interfacial ultrastructure and microtensile dentin bond strength after five-year water storage.

OBJECTIVE: To evaluate a total-etch three-step adhesive system's resin-dentin interfacial ultrastructure and microtensile dentin bond strength (microTBS) after multi-year storage in water. METHODS: Resin composite crowns were formed on 600 grit SiC flattened extracted human molars using a total-etch three-step adhesive system (Optibond FL, Kerr) and a hybrid resin composite (Prodigy, Kerr). microTBS specimens were fabricated and placed in water with 0.5% chloramine T at 37 degrees C until respective static load to failure testing at one-month, six-months and five-year storage. Failure modes were determined by scanning electron microscopy. The interfacial ultrastructure of the resin-dentin interface was analyzed by transmission electron microscopy (TEM) at 48-hours and 44-months storage. microTBS was modeled with Weibull distribution for survival analysis and failure curve distributions were analyzed by the Wald chi-square statistic for significant differences at alpha=0.05. RESULTS: The characteristic tensile strength (sigma omicron) at one-month, six-months and five-year storage was 52.63, 14.77 and 23.57 Mpa, with a Weibull modulus of 3.04, 1.56 and 1.28, respectively. Failure distributions for all groups were significantly different (p<0.0001) with one-month > five-year > six-months. TEM interfacial morphology demonstrated hydrolytic degradation of hybrid layer components at 44-months storage. SIGNIFICANCE: The decrease in tensile strength and changes in ultrastructure may be caused by water sorption and resultant hydrolytic degradation of the adhesive joint.
