Effect of peroral glycerol administration on inner ear fluid electrolytes of guinea pigs.

After the oral administration of 50% glycerol (12 mL/kg), serum, CSF and inner ear fluids from scala tympani perilymph, scala vestibuli perilymph, and scala media endolymph were collected from normal guinea pigs under sodium pentobarbital anesthesia (25-35 mg/kg). The sodium and potassium concentrations were determined by microflame photometry. Increases in sodium concentrations were found in CSF, scala tympani perilymph, scala vestibuli perilymph, and cochlear endolymph. No significant change was observed in the serum. These sodium increases were considered to be due to the dehydration caused by the osmotic action of glycerol. Potassium concentration was increased only in scala tympani perilymph. Oral administration of glycerol was found to be more gradual and effective in dehydration compared to intravenous injection.

[Effect of cocoa on excretion of oxalate, citrate, magnesium and calcium in the urine of children].

Cocoa is a strong carrier of oxalic acid (average: 400 mg per 100 g). In three calcium oxalate stone formers clinical observation had been suggestive of excessive intake of cocoa products contributing to calculus formation. We studied the effect on renal oxalate excretion of an oral cocoa load (30 g per m2 body surface given on 2 consecutive days) in 12 former stone formers (group 1), 14 children with isolated microscopic haematuria (group 2), 13 healthy boys (group 3), and 12 healthy girls (group 4). A new enzymatic method was used to measure oxalic acid in cocoa products as well as in urine samples by a two step reaction: 1. Oxalate decarboxylase, 2. formiate dehydrogenase with photometry of NADH. In addition, the daily excretion of the following substances was measured: Citrate, magnesium, and calcium. There was a significant increase of urinary oxalate excretion from an average of 14.5 mg/24 hours before to an average of 22.2 mg/24 hours after the load in healthy children, and a similar increase in stone formers, but not in children with microscopic haematuria. The excretion of citrate and magnesium did not change following cocoa intake. The calcium excretion was higher in stone formers than in the other groups, but the difference was significant only compared to group 2. It is concluded that the risk of calculus formation may increase following continuous and excessive intake of cocoa products in children with a tendency toward hypercalciuria. Counselling of the stone formers resulted in a marked drop of the daily oxalate excretion, and there was no recurrence of calculus formation over a period of 6 years.(ABSTRACT TRUNCATED AT 250 WORDS)

Relationship between magnesium, potassium and sodium concentrations in lymphocytes and erythrocytes from normal subjects.

Mg, K and Na were measured in erythrocytes, lymphocytes and serum obtained from 20 normal subjects. The cells of 20 ml of blood were separated in Ficoll gradient, washed with a choline buffer and digested with H2SO4. The cations were measured by atomic absorption photometry, protein was determined by the Lowry method and the results were expressed as nmol/mg of protein. Intracellular lymphocyte concentrations (mean +/- SD, n = 20) were: Mg: 57.6 +/- 5.1, Na: 30.0 +/- 7.9, K: 614.3 +/- 69. In the red cells, ion concentrations were: Mg: 7.97 +/- 0.8, Na: 20.9 +/- 4.8, K: 343.0 +/- 42.0. There was a highly significant correlation between Mg and K concentration in erythrocytes (p less than 0.001) and lymphocytes (p less than 0.001). A negative correlation (p less than 0.05) was also found between lymphocyte and erythrocyte K concentration and K serum concentration.

[Introduction to the use of selective electrodes in clinical biology].

The relatively recent distribution of apparati which allow cation measurement by electrometry presents the biologist with a certain number of difficulties inherent to this methodology. A review of the principles of electrochemistry, limited to the facts necessary to master use of these apparati and avoiding the dissipated effort inevitably incurred by consulting treatises, is of an evident practicality. The points concerning, in particular, application in clinical biology have been treated in detail, whether they involved the definition of direct and indirect potentiometries, or the concepts of activity and of concentration, or the explanation of the actual problems posed by the plasma/plasmatic water difference and by electrometry/flame photometry comparison measurement. A survey of the different types of standard electrodes in biology completes this works.

[Characteristics of the course of hypertension in relation to on plasma renin activity].

In 94 patients with hypertension the plasma renin activity was determined by the Yu. A. Serebrovskaya and I.A. Uchitel technique, the glomerular filtration--after Rehberg, potassium and sodium content in blood serum and 24-hour urine samples--by flame photometry. Hypertensive disease was found to be accompanied by an elevated plasma renin activity in 47.9% of the patients, by a normal level--in 26.9%, and by a reduced--in 25.6%, as compared with control group data. Cardiovascular complicatins were found in 1/5 of those with an elevated plasma renin activity, and no such complications were observed in those with a normal or reduced renin activity. An elevated plasma renin activity is a risk factor. The highest mean age and the longest duration of the disease history are typical for patients with low plasma renin activity. To choose the most rational therapy regimen it may be useful to calculate the renin-sodium index that reflects the relationship between the plasma renin activity and the urine excretion of sodium.

[Comparative study of bone mineral densities in the human skeleton].

The different mineral densities and their distribution within human bone of the iliac crest, the femoral and the humeral head are analysed by microradiography with computerized photometry. 10 male and 11 female corpses between the age of 17 to 80, show that the results obtained depended upon age, sex and upon the site of the bone biopsy. Bcause of the small number of cases in this preliminary investigation, careful interpretation of these differences is mandatory. The advantages of this method are discussed.

Physiological studies of exocrine pancreatic secretion in conscious rats. 3. Communication. Separation and determination of enzymes by gel-electrophoresis.

1. A method is described for the electrophoretic separation and following quantitative determination of the different protein fractions of pancreatic juice. 2. A polyacrylamide (PAA) gel-electrophoresis is performed in two systems: one anionic at pH = 9.0 with 7.5% PAA and a cationic one at pH = 5.0 with 10% PAA. 3. After about two hours of separation using a current of 3 and 4 mA respectively, the gels are stained with Amido-black 10B after fixation of the proteins with acetic acid. 4. The colour in excess is removed with acetic acid. 5. The coloured bands are isolated by cutting. 6. Evaluation of the colour by NaOH and determination of the colour intensity by photometry. 7. Evaluation of the colour extinction as relative percentages of the different protein and enzyme fractions. (Extinction of total dye = 100%) 8. There are at least 13 bands in the cationic system and about 7 additional ones in the anionic system distinctly separated, 12 of them could be identified using enzymatic methods. 9. Evidence is given that the colour intensity has a linear relationship to enzyme activity. 10. The validity of the presented method is discussed and compared with other electrophoretic methods.

[Function of the posterior lobe of the hypophysis in rats under long-term thyroliberin administration].

Thyroliberin injected intraperitoneally to rats in doses of 0.5 microgram/100 g body weight two times a day markedly increased the blood plasma thyrostimulating hormone (TSH) content by the 6th day of using, followed by its decrease down to the control level by the 9th day of the experiment. The blood plasma thyroid hormone levels of these rats were lower than those of the controls by the 6th and 9th days of the test. Photometry of the concentration of the Gomori-positive neurosecretory matter in the posterior hypophysis has shown a significant rise of its amount by the 9th day of the trial. However, changes in the neurosecretory matter did not correlate with the alteration of the TSH- and thyroid hormone levels. Simultaneous administration of thyroliberin and diiodothyrozine (7.5 micrograms/animal) led to the inhibition of thyroliberin TSH-stimulating effect. The above increase in the neurosecretory matter concentration testifies indirectly that long-term thyroliberin injection produces less intensive hormone secretion from the posterior hypophysis into the blood cannel.

Clinical chronopharmacology of ACTH 1-17. II. Effects on plasma testosterone, plasma aldosterone, plasma and urinary electrolytes (K, Na, Ca and Mg).

The aim of the investigation was to study the effects of ACTH 1-17 on plasma testosterone, plasma aldosterone as well as on both plasma and urinary electrolytes (K, Na, Mg and Ca) in healthy young adult males with regard to the time (clock hours) at which this polypeptide was injected. Eight healthy adults (males from 28 to 30 years) volunteered for the study. The were synchronized with a diurnal activity from 0700 to midnight and a nocturnal rest. Each week, during 6 consecutive weeks (January 19 to February 25, 1980) a 3-day test was performed on Saturday, Sunday and Monday. On Sundays 3 control-tests and the 3 ACTH-tests were programmed during which either saline or 100 microgram ACTH 1-17 were injected i.m. at respectively 0700, 1400 and 2100. During each 3 day-test period (72 h) the urinary excretion of K, Na, Mg and Ca was determined every 4 h at fixed clock hours. In addition, on Sundays, venous blood was sampled prior to control or ACTH injections at respectively 0700, 1400 and 2100 and 20, 40, 60, 90, 120, 150 and 180 min thereafter. Plasma testosterone, aldosterone (radioimmunoassays) K, Na (flame photometry), Mg and Ca (photocolorimetric methods) were determined in the collected samples. Both conventional and cosinor methods were used for statistical analyses. The injection of ACTH at 0700 was followed by a clear and statistically significant rise of plasma testosterone. No change with regard to control occurred when ACTH was injected at either 1400 or at 2100. A statistically significant rise of plasma aldosterone was observed after each of the ACTH injections. However, the highest plasma aldosterone level was reached when ACTH was administered at 1400 and the lowest level at 2100. ACTH-induced changes in plasma electrolytes were either nil (for Na and Ca) or small (for K and Mg). A more or less important increase of urinary K occurred after the ACTH injection at each of the 3 considered times. The highest values of excreted K occurred after the injection of ACTH at 0700, without shift of the acrophase. In contrast, injections of ACTH at 1400 and 2100 induced a dramatic alteration of the K rhythms. ACTH induced an important fall in the Na urinary excretion. This fall was the greatest when ACTH was injected at 1400. Na rhythm alterations also occurred, particularly after ACTH injections at 2100. However, this effect was less pronounced after ACTH injection at 0700 than at other considered time points. The urinary amount of excreted Ca did not seem to be affected by ACTH. Rhythm alterations occurred after ACTH injections at 1400 and 2100. Peaks of plasma testosterone, plasma aldosterone as well as plasma cortisol (reported in a previous paper) resulting from ACTH stimulation coincided in time with the acrophase of the physiological circadian rhythm in plasma levels of these hormones...

[Disturbed sodium and calcium exchange in erythrocytes of spontaneously hypertensive rats (author's transl)].

In red blood cells of spontaneously hypertensive and of normotensive rats intracellular NA+ activity (a(i)Na), Na+ concentration (]Na+]i) and Ca2+ activity (a(i)Ca) were measured by ion-selective electrodes and flame photometry, respectively. In spontaneously hypertensive rats a(i)Na and a(i)Ca were significantly elevated as compared with the normotensive controls, whereas [Na+]i showed no significant difference. In hypertensive animals a(i)Na, exceeded [Na+]i, in normotensive rats a(i)Na was lower than [Naf+]i. From these results it can be concluded: (1) In spontaneously hypertensive rats the transmembraneous distribution of free Na+ is altered, (2) an elevation of intracellular free Ca2+ may contribute to increased vascular contractility in hypertensive animals, (3) the described abnormalities may be caused by a lowered binding-capacity of intracellular macromolecules for Na+ and Ca2+.

The megakaryocyte DNA content and platelet formation after the sublethal whole body irradiation of rats.

The DNA content of rat bone marrow megakaryocytes (MK) was studied by Feulgen photometry, following whole body irradiation with 2 Gy. The DNA measurements were preceded by acetylcholinesterase staining to avoid missing the smaller 2N-8N MK. The number of 2N-8N MK declined immediately following irradiation, whereas the number of 16N-64N MK remained normal for 4 days before decreasing. The number of 2N-8N and 16N-64N MK reached minimum around days 7 and 10, respectively, and thereafter increased to supranormal values at days 14 and 20, respectively. Platelet production, measured by 35S incorporation into platelets, increased during the first 4 days, then decreased to minimum about day 10. A rise to supranormal values was present at day 20. All values were about normal 30 days after exposure. The observed pattern may be explained as follows: Most of the 16N-64N MK survive the applied dose and maintain their ability to produce platelets. Some of the 2N-4N and 8N MK survive irradiation and transform into platelet-producing MK. No influx of cells from the MK stem cell compartment into the MK compartment can be observed before day 7 after irradiation. One explanation for this time lag may be that thrombocytopenia, which does not occur before then, is an essential stimulus for MK stem cell activation.

The total and free concentrations of Ca2+ and Mg2+ inside platelet secretory granules. Measurements employing a novel double null point technique.

The inorganic ion contents of platelet alpha-granules were determined by a combination of neutron activation analysis and flame photometry. Total concentrations were estimated using intragranular water spaces measured by isotope dilution. To measure the free concentrations of Ca2+ and Mg2+ we developed a novel double null point titration technique. The method requires independent determinations of transmembrane delta pH and the availability of two divalent cation-H+ exchange ionophores with different Ca2+/Mg2+ selectivity ratios. A23187 and the halogenated analog 4-bromo-A23187 were used for this purpose. Absolute delta pH was measured by methylamine distribution, while relative pH changes induced by the ionophores were monitored with 9-aminoacridine. The free concentrations of Ca2+ and Mg2+ were found to be 12 and 326 microM, respectively. These values are markedly lower than the calculated total concentrations of these cations, i.e. 32 mM for Ca2+ and 172 mM for Mg2+.

[Change in the shape and volume of the secretory cells of the mammae in the course of the secretory cycle].

Changes occurring in the acini due to oxytocin were studied using the vital photometry and the morphometric analysis of materials obtained by biopsy. At the initial stages of the functional activation, the secretory cells become cylindric, their volumes simultaneously increasing. A study of intercellular interrelations using the atropine blockade of cholinoreceptors, revealed the lack of activation of the secretory cycle which suggests the functional dissociation of contractile and secretory functions of the cell alveolar complex.

Pseudoisochromatic plate design--Macbeth or tungsten illumination?

Three sets of pseudoisochromatic plates were evaluated by photometry and colorimetry. The luminance contrast between the figure and background was measured and compared with a contrast detection threshold. The chromaticity coordinates of the figure and background were evaluated on the basis of how closely they approached a dichromatic line of confusion. The separation of the coordinates of the figure and background are a measure of the severity of the defect for which the plate tests. The plates were evaluated under both Macbeth (C) and tungsten (A) illuminants; two sets of plates were found to be better designed for tungsten illumination.

Low ploidy megakaryocytes in steady-state rat bone marrow.

The amount of DNA in individual megakaryocytes (MK) was measured by Feulgen photometry performed on smears of steady-state rat bone marrow. The MK were identified by acetylcholinesterase (ACHE) staining. The method described is particularly suitable in the study of immature MK, hitherto difficult to examine for DNA content. The main MK ploidies were 8N, 16N, and 32N, comprising 33.4% +/- 1.4%, 40.8% +/- 1.4%, and 17.9% +/- 1.3%, respectively, of the total MK numbers. Only 6.7% +/- 1.7% had less than octoploid DNA content. Very few cells were found belonging to the 64N class. Among the immature MK, a relatively large fraction was found between the ploidy levels, indicating a rapid turnover within these ploidy classes. The demonstration of ACHE activity even in MK with a diploid DNA content indicates that the flow from the committed progenitors to differentiated MK occurs at the 2N level.

The effect of splenectomy on platelet formation and megakaryocyte DNA content in rats.

The DNA content of rat bone marrow megakaryocytes (MK) was studied by Feulgen photometry following splenectomy and sham operation, respectively. The DNA measurements were preceded by acetylcholinesterase staining for identification of the 2N-8N MK. The number of 2N-8N MK decreased to minimum values, while the number of 16N-64N MK increased to maximum values about 4 days following both splenectomy and sham operation. However, the changes were somewhat more pronounced following splenectomy than sham operation. The total MK number did not change significantly. Platelet production, measured by 35S incorporation into platelets, increased during the first 2 days and remained high for 6-7 days, increasing the platelet counts. All values were about normal 30 days after surgery, except for a minor thrombocytosis following splenectomy. The early, highly significant thrombocytosis, following both splenectomy and general surgery, is caused by increased production of platelets due to the surgical trauma. This is caused by a direct action on bone marrow MK by transforming 2N-8N MK into higher ploidy classes. Lack of splenic platelet pooling may influence the grade and duration of the early thrombocytosis after splenectomy. The late, long-lasting, minor thrombocytosis, which occurs after splenectomy but not after sham operation, can be explained by the removal of the splenic platelet pool.
