So far classical prenatal detection of chromosome aberrations has been limited to the evaluation of metaphase by means of time-consuming cytogenetic techniques. The MultiVision PGT test enables a simultaneous detection of aneuploidies of chromosomes 13,18, 21, X, and Y, even 24 h after amniocentesis. In the presented case, this test detected prenatally a chromosomal mosaicism 69,XYY[35]/46,XY[65]. This result was not confirmed after birth, by the same test on blood smear. The discrepancy is difficult to explain.

It is considered that centromeric (CEN) regions play the leading role in the formation of chromocentres predominantly consisting of satellite DNA. Cloned mouse and human satellite DNA sequences from CEN and periGEN regions were used in order to trace their positions relative to chromocentres. Methods of fluorescent in situ hybridization (FISH) and immunoFISH with antibodies against CENP-B, known is a marker of prekinetochore, were employed. Peripheral position of the signals was observed at the chromocentres, but a combined signal of GEN and periGEN satellite DNAs never covered the whole brightly DAPI stained regions. A reasonable amount of the chromocentre body is not a satellite DNA. We suppose that the matrix-associated regions (MAR) of structural genes and, probably, the heavy methylated coding sequences of the genes, which are not expressed in the given cell type, are included in the chromocentres in addition to satellite DNAs.

Short-term cultures of fifty-two samples of fibroadenomas were cytogenetically analyzed. Thirty-three of the successfully karyotyped fibroadenomas were further investigated for the presence of amplifications in the CCND1, c-MYC and HER/2-neu genes by means of FISH analysis. Compared to carcinomas, fibroadenomas seem to have less complex cytogenetic rearrangements and limited alterations on HER-2/neu, CCND1 and c-MYC loci. A cytogenetic subgroup of fibroadenomas with hyperdiploid karyotypes and only numerical changes was observed. Amplification of CCND1 seems to play a more substantial role in benign tumor progression. These findings confirm that fibroadenomas do have genetic alterations and support the hypothesis that a fibroadenoma subset displays changes also found in carcinomas, thus indicating that patients belonging to this group might have an increased risk for subsequent breast cancer.
A study was made of the influence of cytosol and conditioned medium, obtained at different times of incubation of irradiated thymocytes, on native thymocytes, as well as of the influence of mixing the suspension and changing the medium and the enzyme inhibitors, involved in metabolism of arachidonic acid, on the radiation injury to thymocytes. Cytosol of thymocytes, incubated for 60 min after irradiation, was shown to exert an insignificant toxic effect on native thymocytes. The conditioned medium of irradiated thymocytes is virtually ineffective. The mixing of the suspension upon incubation and the changing of the medium reduced the damage to exposed thymocytes while inhibitors of phospholipase A2 and lipoxygenase prevented completely the radiation-induced pyknosis of nuclei, whereas a inhibitor of cyclooxygenase did not influence it. It is inferred from the data obtained that unstable lipoxygenase products may serve as mediators of intercellular interactions of exposed thymocytes.
