Mobilization of plasmid-borne drug resistance determinants for transfer from Pseudomonas aeruginosa to Escherichia coli.

RSU2, a plasmid transmissible between strains of P. aeruginosa but not to Escherichia coli can be mobilized by R751. Conjugatants receive a single plasmid composed of DNA from both R751 and RSU2 which has the compatibility properties of a member of group P (like R751). Study of this fusion plasmid suggests that the failure of RSU2 to transfer into enterobacteria is due to an inability to replicate in these bacteria. The fusion plasmid replicates using the genes of R751.

In vivo transfer of the ti-plasmid of Agrobacterium tumefaciens to Escherichia coli.

The Ti-plasmids are naturally self-transmissible from their normal host Agrobacterium to E. coli. They are however unable to stably establish themselves as a replicon in E. coli. It is nevertheless possible to study the Ti-plasmids in E. coli with the help of Ti::RP4 cointegrate plasmids that transfer and maintain themselves very efficiently in E. coli. An E. coli harbouring such a Ti::RP4 plasmid is unable to catabolize octopine and unable to induce crown-gall tumours on plants.

In vitro susceptibility of Helicobacter pylori to trospectomycin, pirlimycin (U-57930E), mirincamycin (U-24729A) and N-demethyl clindamycin (U-26767A).

The in vitro activity of trospectomycin, pirlimycin, mirincamycin and N-demethyl clindamycin was measured against 46 clinical isolates of Helicobacter pylori using an agar dilution technique. The MIC50 and MIC90 were 4 and 64 micrograms/ml for pirlimycin and N-demethyl clindamycin, and 32 and 128 micrograms/ml for mirincamycin, respectively. All 46 strains were sensitive to trospectomycin with an MIC50 of 8 micrograms/ml and an MIC90 of 16 micrograms/ml. Of seven strains with the highest trospectomycin MICs (8 or 16 micrograms/ml) 100% were found to be resistant to metronidazole. Among ten strains with low trospectomycin MICs (2 micrograms/ml or less) 100% were sensitive to metronidazole. Possible explantations for the apparent correlation between the MICs of the two drugs are discussed. Since all metronidazole resistant strains were sensitive to trospectomycin, this drug may be useful in treating infection with metronidazole resistant Helicobacter pylori.

Dissemination of streptomycin and sulfonamide resistance by plasmid pBP1 in Escherichia coli.

About one third of streptomycin resistance in Escherichia coli is mediated by APH-(3''). This enzyme is encoded by the plasmid pBP1 in 80% of all streptomycin resistant strains tested. pBP1, which in addition mediates sulfonamide resistance, has been found to be disseminated in Escherichia coli strains all over the world. It has a molecular weight of 4.0 megadalton and does not seem to be disadvantageous for the metabolism of the bacterial cell. The reason for the slow decrease of resistance to streptomycin and sulfonamide in clinical isolates, despite the restricted use of these drugs, is presumably the survival of bacteria harbouring pBP1 which have been selected by streptomycin and sulfonamides in the early days of chemotherapy.

Effect of an acidic environment on the susceptibility of Helicobacter pylori to trospectomycin and other antimicrobial agents.

The susceptibility of 30 clinical isolates of Helicobacter pylori to trospectomycin, ampicillin, metronidazole, clarithromycin, azithromycin and clindamycin under varying pH conditions was evaluated. An acidic environment was shown to affect unfavourably the activity of all the antimicrobial agents tested. This pH effect was most marked for the two macrolides and for clindamycin.

Plasmid shuttle vector with two insertionally inactivable markers for coryneform bacteria.

A new shuttle vector pCEM500 replicating in Escherichia coli and in Brevibacterium flavum was constructed. It carries two antibiotic resistance determinants (Kmr/Gmr from plasmid pSa of Gram-negative bacteria and Smr/Spr from plasmid pCG4 of Corynebacterium glutamicum) which are efficiently expressed in both hosts and can be inactivated by insertion of DNA fragments into the unique restriction endonuclease sites located within them. This vector was found to be stably maintained in B. flavum and can be used for transfer of the cloned genes into this amino-acid-producing coryneform bacterium.

Comparison of Neisseria gonorrhoeae isolates from the genital tract and pharynx of two gonorrhea patients.

Neisseria gonorrhoeae was isolated simultaneously from urethral and pharyngeal specimens of two gonorrhea patients. The pair of isolates from one of the two patients were identical in auxotype, pulsed-field gel electrophoresis pattern, and antimicrobial susceptibility, which indicated that both sites in that patient were infected with the same strain of N. gonorrhoeae. However, the two isolates from the other patient had different properties, indicating that the two sites in the second patient were infected with different strains. The gonococcal infections in these patients failed to respond to initial treatment with sparfloxacin or sulbactam/ampicillin, because the causative strains of N. gonorrhoeae were resistant to the respective antibiotics. Variable patterns and routes of gonococcal infection have recently been discovered in individual patients, suggesting that specimens for bacterial isolation should be taken not just from one site but from various sites that might be infected. This method may contribute to the successful treatment and epidemiological investigation of gonococcal infections.

Reduction in myocardial neutrophil accumulation and infarct size following administration of thromboxane inhibitor U-63,557A.

We examined the effects of a new selective thromboxane A2 (TXA2) synthetase inhibitor, U-63,557A, on myocardial infarct size 48 hours following left coronary ligation in rats. With a single 8 mg/kg dose of U-63,557A (furegrelate) administered prior to coronary ligation, platelet aggregation and serum TXA2 formation declined significantly (p less than 0.02) for up to 48 hours. Myocardial infarct size, as measured by planimetry of serial left ventricular sections, was decreased from 44 +/- 3% (saline-treated control rats) to 34 +/- 4% (p less than 0.05). Left ventricular creatine kinase (CK) following coronary ligation was also preserved in U-63,557A vs saline-treated control animals (p less than 0.05). These beneficial effects of U-63,557A were not accompanied by reduction in the indices of myocardial oxygen demand (heart rate and arterial pressure). Furthermore, neutrophil accumulation in the infarcted myocardium was significantly decreased by U-63,557A (26 +/- 6 vs 96 +/- 3/high-power field [p less than 0.01]). These data suggest that administration of a single dose of selective TXA2 synthetase inhibitor prior to coronary ligation modulates platelet function for up to 48 hours and reduces the extent of myocardial injury, which may, in part, relate to decrease in neutrophil accumulation.
