Acid phosphatase activity of cerebrospinal fluid cells in leptomeningeal haemorrhage.

One hundred and twenty samples of haemorrhagic spinal fluid were examined by acid phosphatase staining. This enzyme activity starts to appear in mono-histiocytic cells 2 days after bleeding and increases up to the 5th day. After 1 week the activity decreases rapidly. Similar results are found in mixtures of incubated clear spinal fluid, to which blood is added. Acid phosphatase staining is a useful additional method for determination of the age of a leptomeningeal bleed.

Effects of tunicamycin on the pH-activity pattern of acid phosphatase in Pseudomonas pseudomallei.

The liquid culture of Pseudomonas pseudomallei shows a complex feature in in the pH-activity pattern of acid phosphatase, not a single peak curve. There was an evident tendency that the higher activity shifted to the higher pH range with the growth of culture. The culture in the presence of tunicamycin (20 micrograms/ml) showed a decreased activity selectively in the higher pH range, while the activity in the lower pH was more heat-labile. The bacterial cells grown on agar plates containing tunicamycin were more heat-labile than the untreated control cells. The glucosidase-treatment reduced the enzymatic activity (of the phosphatase-active fractions from the living cells) with the shift of the optimum pH to lower pH. These observations together with some collateral findings suggest that the pH-activity pattern of acid phosphatase in P. pseudomallei is associated with the development of precursor enzyme proteins to mature glycoproteins.

Acid phosphatase elevation with symptoms following transurethral prostatic resection.

Symptoms following transurethral prostatic resection for benign prostatic hypertrophy were evaluated in 100 patients. The occurrence of post-transurethral resection symptoms was monitored postoperatively and was found to be associated with an increase in serum acid phosphatase. The transient elevation of serum acid phosphatase after transurethral prostatic resection indicates that intraoperative absorption of prostatic tissue substances occurs. The intraoperative absorption of prostatic tissue substances may have a significant role in producing symptoms of post-transurethral resection syndromes.

Comparative investigation of the mixed aggregation immunocytochemical technique and the indirect peroxidase technique for the detection of prostate specific acid phosphatase in paraffin or paraplast sections.

The results obtained with the indirect peroxidase technique for the identification of prostate specific acid phosphatase in formalin fixed, paraffin or paraplast embedded autopsy material are compared with the results obtained with the mixed aggregation immuno-cytochemical technique. When using a monospecific antiserum the former technique is prefered. However, when a monospecific antiserum is not available, one has to balance the advantages of the mixed aggregation immuno-cytochemical technique against the disadvantages of having to prepare a monospecific antiserum, necessary for the indirect peroxidase technique. Both methods appeared positive in 20 prostatic carcinomas and in 36 metastases of prostatic carcinomas. In the epithelium of the seminal vesicles and in osteoclasts no acid phosphatase could be detected with the antiserum. A comparison of both techniques, as well as different types of preincubation to diminish nonspecific background staining are discussed.

Histiocytic medullary reticulosis with parallel increases in serum acid phosphatase and disease activity.

Two months after presenting with autoimmune hemolytic anemia, a 57-year-old man developed increased serum acid phosphatase activity. Subsequently, abnormal histiocytes were found in the bone marrow and blood, and 8 months after the onset of his illness lymph node biopsy led to the diagnosis of histiocytic medullary reticulosis. At that time the level of serum acid phosphatase activity was more than 60 times the upper limit of normal and then paralleled the activity of his disease during temporary responses to chemotherapy, rising to over 90 times normal at the time of his death 1 year after presentation. No prostatic malignancy was demonstrated by biopsies or at autopsy, and electrophoretic studies of the serum enzyme suggested that it was of extraprostatic origin. It is postulated that the acid phosphate present in the serum at levels heretofore described only in metastatic carcinoma of the prostate may have arisen from the abnormal histiocytes.

Serum tartrate resistant acid phosphatase as a potential marker of bone metastasis from breast cancer.

BACKGROUND: Tartrate resistant acid phosphatase (TRACP) is exclusively localized in osteoclasts and has been suggested to be a unique marker of bone metastasis. PATIENTS AND METHODS: TRACP activity using an improved spectrophotometric assay was measured in 56 healthy volunteers and 113 breast cancer patients, including 35 with bone metastases (BM). RESULTS: TRACP activities (IU/l, mean +/- SD) of normal subjects in pre- and post-menopausal women were 5.7 +/- 1.3 and 6.6 +/- 1.2, respectively (p < 0.02). The specificity, sensitivity, and accuracy of TRACP in patients, were 91.0%, 65.7%, and 83.2%, respectively. TRACP was significantly higher in patients with BM than in patients without BM (p < 0.01). In patients with BM, TRACP increased in proportion to the number of BM. Regarding clinical assessment of treatment of BM, TRACP was significantly higher in patients with progressive disease. CONCLUSION: TRACP is a useful marker of metastatic bone disease and response to treatment in breast cancer patients.

Acid phosphatase activity in the rat neurohypophysis during increased levels of gonadothrophic hormones, in diabetes insipidus (Bratteboro strain) and after water loading.

The acid phos phatase activity of the rat neurohypophysis was measured during increased gonadotrophic hormone levels, in diabetes insipidus (DI) (Brattleboro strain) and after water loading, i.e. conditions that interfere with the function of the hypothalamo-neurohypophyseal system (HNS). In addition determination of tissue protein, lipid and DNA and of water metabolism were performed. Neurohypophyseal acid phosphatase activity expressed on a dry weight basis increased under all conditions. For gonadectomized females, Brattleboro rats and water loaded males an increased water metabolism was observed. The increased acid phosphatase activity is interpreted as being related to disposal of release residues during stimulation of the HNS (increased gonadotrophic hormone levels and DI) and to disposal of neurosecretory material during inhibition (water loading).

Isolation and characterization of recessive, constitutive mutations for repressible acid phosphatase synthesis in Saccharomyces cerevisiae.

Two new classes of mutants containing recessive constitutive mutations, phoT and phoU, that affect the repressible acid phosphatase (EC 3.1.3.2) in Saccharomyces cerevisiae were isolated along with many previously known phoR mutants. These loci segregated independently from each other, from the phoS gene, and from another regulatory gene, phoD, that exerts positive control for acid phosphatase synthesis. The phoR and phoU mutations showed the same genetic behavior in the double mutants, which also contained the phoS or phoD mutation. In contrast, the phoT mutation could not suppress the phoS mutation, which caused a loss of enzyme activity. Many mutant alleles of phoR and phoU were found to be temperature sensitive (ts), whereas those of phoT were not. These ts mutants were constitutive at 35 C but severely repressible at 25 C. These facts strongly suggest that both the phoR and phoU genes are cooperatively concerned with the production of the repressor, whereas the phoT gene might be involved in another mechanism distinct from that in which phoR and phoU are involved. No single mutation of phoR, phoT, or phoU result in an enzyme level comparable to that of fully derepressed enzyme activities, and the temperature sensitivity of the ts phoR and ts phoU mutations in such combinations almost disappeared. In addition to these observations, since the ts phoR phoS and ts phoU phoS double mutants showed some enzyme synthesis at 25 C under derepressing conditions, a defect in the ts mutant repressors was strongly suggested, even at 25 C.

Bone marrow acid phosphatase in staging prostatic carcinoma.

We evaluated bone marrow acid phosphatase in 30 patients as another parameter in staging prostatic carcinoma. This method is reliable and useful as part of the staging precedures. No falsely positive results were found and the procedure provided the only indication of metastatic diseases in 4 cases.

Acid phosphatase and zinc in semen of subjects with no clinical evidence of prostatic disease.

We evaluated the distribution of values for prostatic acid phosphatase (EC 3.1.3.2) activity and zinc concentration in semen of 80 men without clinical evidence of prostatic disease. Both substances have log normal distributions. Reference intervals were defined by parametric tests after logarithmic transformation of data and by nonparametric tests. There is a correlation between zinc concentration and prostatic acid phosphatase activity in semen.

Trypanosoma cruzi: experimental Chagas' disease in rhesus monkeys. II. Ultrastructural and cytochemical studies of peroxidase and acid phosphatase activities.

Ultrastructural and cytochemical studies of peroxidase and acid phosphatase were performed in skin, lymph node and heart muscle tissue of rhesus monkeys with experimental Chagas' disease. At the site of inoculation there was a proliferative reaction with the presence of immature macrophages revealed by peroxidase technique. At the lymph node a diffuse inflammatory exudate with mononuclear cells, fibroblasts and immature activated macrophages reproduces the human pattern of acute Chagas' disease inflammatory lesions. The heart muscle cells present different degrees of degenerative alterations and a striking increase in the number of lysosomal profiles that exhibit acid hydrolase reaction product. A strong inflammatory reaction was present due to lymphocytic infiltrate or due to eosinophil granulocytes associated to ruptured cells. The present study provides some experimental evidences that the monkey model could be used as a reliable model to characterize histopathological alterations of the human disease.

Acid phosphatase activity of the brain in SHR-- the first report of enzyme histochemical studies of spontaneously hypertensive rat brain.

Changes in acid phosphatase activity in the cerebrovascular system and brain parenchyma in SHR were investigated histochemically. An increased activity of the enzyme was demonstrated in the SHR endothelial and medial smooth muscle cells of the cerebral arterial system as compared to the control. The pericytes of intraparenchymal blood vessels also showed an intensified enzyme activity. The enzyme activity increased with advancing age. In SHR brain parenchyma, the enzyme activity was decreased in the cortical nerve cells. Glial cells with the enzyme activity were increased in number and showed an intensified activity. Causative factors of changes in acid phosphatase activity in SHR cerebral arteries and parenchyma were discussed.

[Electrophoretic polymorphism of lactate, malate and glutamate dehydrogenases, acid phosphatase and esterases of Providencia alcalifaciens, P. stuartii and P. rustigianii].

The polymorphism of glutamate, lactate and malate dehydrogenases, of acid phosphatase and of esterases of 27 strains of Providencia alcalifaciens, 35 strains of P. stuartii and 17 strains of P. rustigianii was investigated by conventional electrophoresis in polyacrylamide agarose gel and by isoelectric focusing in thin-layer polyacrylamide gel. For each enzyme analysed, the three species were characterized by a distinct electrophoretic pattern. The number of allozymes detected by conventional electrophoresis was greater than that detected by isoelectric focusing. The use of these two techniques in parallel led to improved detection of polymorphism of esterase alpha beta from P. alcalifaciens. A two-dimensional profile obtained by plotting isoelectric points against electrophoretic mobilities for malate dehydrogenase, acid phosphatase and beta-A esterase showed a molecular relationship between the diverse allozymes and demonstrated their taxonomic values. Polymorphism varied considerably according to the enzyme and species analysed and was correlated with DNA heterogeneity. The strains of P. alcalifaciens exhibited the greatest enzyme polymorphism and were classified into two main zymotypes reflecting genetic divergence within this species, whereas the strains of P. stuartii were electrophoretically less variable.

The forensic identification of semen by isoelectric focusing of seminal acid phosphatase.

An isoelectric focusing technique for the separation of acid phosphatases is described. The focusing patterns for semen and vaginal secretion are examined in detail and the technique is evaluated as a means of identifying and distinguishing between these body fluids and other materials of potential forensic interest. The method can reliably be used to diagnose the presence of semen in stains and in the post-coital vagina.

Acid phosphatase positional correlations in solid surface fungal cultivation: a fractal interpretation of biochemical differentiation.

Colour image analysis was used to measure the positional correlation between acid phosphatase intracellular concentration and hyphal cellular differentiation which leads to concentric circular zonal activity patterns. Acid phosphatase is strongly implicated in the biochemical control of hyphal branching, and exo-enzyme secretion, such as laccase in fungi, occurs predominately via the hyphal tips. Different concentrations of an organic dye were used to effect substrate induction of the enzyme response, which was shown to be statistically correlated according to a fractal power law (H approximately 0.39). A self-organized critical state for the molecular responsivity of dissipative enzyme expression is hypothesized as an efficient mechanism for hyphal adaptation, also accounting for the underlying biophysics of the observed pattern formations.

Influence of secretin and caerulein on acid phosphatase activity in human saliva.

Experimental studies have demonstrated a structural and functional correlation between the pancreas and salivary glands. Therefore, one may suppose that caerulein plus secretin, used in functional exploration of the exocrine pancreas, could exert an influence on acid phosphatase activity (AcP) and its isoenzymes (tartrate sensitive-TsAcP, tartrate resistant-TRAP acid phosphatases) in whole saliva in humans. The current study was conducted in 14 asymptomatic volunteers (six woman and eight men, mean age 34, range 27-45). Salivary secretions were collected under basal conditions (intravenous infusion of 0.15 mol l-1 NaCl) and during intravenous infusion of secretin plus caerulein at different doses. Stimulation with secretin (50 mKE kg-1 min-1) and caerulein (2.5 ng kg-1 min-1) resulted in a marked and rapid decrease of salivary flow rate. TRAP activity decreased during the first 20 min of hormonal infusion (1.09 +/- 0.19 mU ml-1 vs. 1.93 +/- 0.18 mU ml-1, 1.16 +/- 0.12 mU ml-1 vs. 1.86 +/- 0.17 mU ml-1, p < 0.05). AcP and TsAcP activity during intravenous infusion of secretin plus caerulein remained unchanged. These results point to a wider than expected participation of the digestive system in hormonal control of biochemical processes taking place inside the oral cavity.
