Asymmetric somatic cell hybridization in plants. II. Electrophoretic analysis of radiation-induced DNA damage and repair following the exposure of sugarbeet (Beta vulgaris L.) protoplasts to UV and gamma rays.

As part of an investigation into whether it would be possible to use UV radiation as a suitable pretreatment of the donor cells in asymmetric hybridization experiments, the effects of this treatment on sugarbeet (Beta vulgaris L.) protoplast DNA have been determined and compared with those of gamma radiation. Both nuclear and mitochondrial DNAs have been examined. The dose ranges chosen had previously been determined to be potentially applicable for fusion experiments. Pulsed field gel electrophoresis and standard agarose gel electrophoresis have been used in combination with laser scanning densitometry to gain an insight into the precise nature and degree of DNA damage resulting from irradiation. It was observed that UV radiation introduced substantial modifications to sugarbeet DNA. Double-strand breaks were detected, the number of which was found to be directly proportional to the dose applied. Such breaks indicate that UV radiation results in substantial chromosome/chromatid fragmentation in these cells. Chemical modifications to the DNA structure could be revealed by a significant reduction in DNA hybridization to specific mitochondrial and nuclear DNA probes. Following gamma irradiation at equivalent biological doses (i.e. those just sufficient to prevent colony formation) much less damage was detected. Fewer DNA fragments were produced indicating the presence of fewer double-strand breaks in the DNA structure. In comparison to UV treatments, DNA hybridization to specific probes following gamma radiation was inhibited less. For both treatments, mitochondrial DNA appeared more sensitive to damage than nuclear DNA. The possibility that DNA repair processes might account for these differences has also been investigated. Results indicate either that repair processes are not involved in the effects observed or that DNA repair occurs so fast that it was not possible to demonstrate such involvement with the experimental system used. The general relevance of such processes to asymmetric cell hybridization is discussed.

[In vitro regeneration of Nicotiana africana plants from explants of different type and mesophyll protoplasts].

Methods of in vitro cultivation of an African endemic species Nicotiana africana (Solanaceae) possessing some valuable traits have been elaborated. Influence of different concentrations of auxin (alpha-naphtaleneacetic acid) and cytokinins (6-benzylaminopurine, zeatin, thidiazuron) on morphogenesis and plant regeneration has been analysed using leaves and internodes as explants. The optimum method of regeneration of N. africana shoots in leaf explants is cultivation in the presence of 0.1 mg/l NAA and 1 mg/l BA; in internode explants--cultivation on the medium containing 0.5 mg/l NAA and 1 mg/l zeatin. The use of thidiazuron was the most effective at the concentration of 0.4 mg/l with subsequent transfer of explants to hormone-free medium. Method of isolation and culture of mesophyll protoplasts enabling to regenerate N. africana plants during 3-4 months has been proposed.

[Breeding of high-yield strain of taxol by mutagensis of protoplast and primary discussion of genetic differences between mutants and their parent strain].

The breeding of high-yield strain of taxol was performed by protoplast mutagenesis of strain NCEU-1 using ultraviolet radiation and combined treatment of UV and LiCl. The mutants UV40-19 and UL50-6 were obtained, which raised the taxol yield from 314.07 microg/L to 376.38 microg/L and 392.63 microg/L respectively. Genetic differences between the mutants UV40-19, UL50-6 and their parent strain were primarily compared through random amplified polymorphic DNA (RAPD) and isozyme technique. The results showed that the genetic differences were very obviously between the parent strain and its mutants and between the two mutants, which laid foundation of molecular mechanism for the study of genes related to the taxol biosynthesis and mutants for raising the taxol yield.

[Study on breeding up high-yield strain of taxol by protoplast mutagensis].

In order to obtain resistant mutants to nystatin, ultraviolet radiation and LiCl were used to mutagenize the protoplasts of taxol-producing fungi NCEU-1, and four positive mutants with high yield of taxol were screened out on nystatin flat. After further screening experiments on fermentation, a mutant strain--UL04-5 which was able to produce taxol with high yield and could be stably passed on in genetics was eventually found, it's ability to produce taxol was improved from 314.07 microg/L (strain NCEU-1) to 418.24 microg/L (strain U04-5).

A comparison of the phenotypic and genetic stability of recombinant Trichoderma spp. generated by protoplast- and Agrobacterium-mediated transformation.

Four different Trichoderma strains, T. harzianum CECT 2413, T. asperellum T53, T. atroviride T11 and T. longibrachiatum T52, which represent three of the four sections contained in this genus, were transformed by two different techniques: a protocol based on the isolation of protoplasts and a protocol based on Agrobacterium-mediated transformation. Both methods were set up using hygromycin B or phleomycin resistance as the selection markers. Using these techniques, we obtained phenotypically stable transformants of these four different strains. The highest transformation efficiencies were obtained with the T. longibrachiatum T52 strain: 65-70 transformants/microg DNA when transformed with the plasmid pAN7-1 (hygromycin B resistance) and 280 transformants/107 spores when the Agrobacterium-mediated transformation was performed with the plasmid pUR5750 (hygromycin B resistance). Overall, the genetic analysis of the transformants showed that some of the strains integrated and maintained the transforming DNA in their genome throughout the entire transformation and selection process. In other cases, the integrated DNA was lost.

[Fusion of protoplasts of inactive variants of 2 producers of actinomycin C and the biosynthesis of an antibiotic of non- actinomycin nature].

Fusion of protoplasts of double auxotrophic mutants of spontaneous inactive variants of two cultures producing actinomycin C, i.e. Streptomyces chrysomalus 305 and Streptomyces sp. 26-115 induced by PEG-600 yielded a number of stable recombinants. One of the recombinants requiring proline for its growth was designated as recPro. Unlike its parent strains, it synthesized an antibiotic substance active against gram-positive bacteria and Saccharomyces cerevisiae. The nature of the substance is under investigation.

Promoter and genotype dependent transient expression of a reporter gene in plant protoplasts.

For analysis of expression of three different plant promoters such as CaMV 35S, rbc S and mas, compact plasmid vectors were constructed by use of beta-glucuronidase (GUS) gene and nos termination signal. The plasmid molecules were introduced into tobacco and tomato protoplasts by using the Mg2+/PEG transformation protocol described by Negrutiu et al. The transient assays revealed maximum expression two days after DNA uptake. The comparative studies show the following order of promoters mas, CaMV 35S, rbc S as far as the activity is concerned. We also detected genotype-dependent promoter activity in the case of tomato.

[Features of transformation of competent cells and Bacillus subtilis protoplasts by integrative vectors].

The effect of structural peculiarities of DNAs from integrative plasmids on the transformation activity was studied. Monomeric forms of the plasmids can only transform B. subtilis competent cells, when plasmid selective marker is inserted into chromosomal fragment within the plasmid. Polymeric forms are needed for efficient transformation. Both single- and double-stranded DNAs of integrative plasmids transform no B.subtilis protoplasts, this being irrespective of plasmid structure.

Oxytetracycline production in solid state and submerged fermentation by protoplast fusants of Streptomyces rimosus.

Streptomyces rimosus TM-55 was treated with 3% EMS, and 29 auxotrophic mutants (AM-1 - AM-29) were isolated from 5457 colonies at a survival rate between 6.6-66.7%. Three sets of the auxotrophic mutants, AM-3 and AM-27, AM-7 and AM-28, and AM-3 and AM-21, were chosen for protoplast fusion with 50% PEG 1000 for 30 minutes at 25 degrees C, and 25 fusants were isolated (f-1 - f-25). In solid substrate, oxytetracycline production of 20% fusants was higher than that of the wild strain, while in submerged fermentation, it was 44%. Oxytetracycline productions of fusants f-1, f-6, f-11, f-12, f-20 and f-21 were lower than that of the wild strain in solid substrate, but this was reversed in submerged fermentation. On the other hand, OTC production of fusant f-8 was higher than that of the wild strain in solid substrate, and this was reversed in submerged fermentation.

[Protoplast fragmentation as one of the possible mechanisms of L-transformation of bacteria (Clostridium perfringens)].

As revealed, the action of lysozyme on the cells of Clostridium perfringens BP-6K led to the formation of not only typical spheroplasts, but also of cells whose peripheral parts of the cytoplasm were fragmented by membrane component. Small bodies framed by the membrane proper and containing granular and fibrillar components were formed. They were polymorphic in osmium treatment, and had smooth contours in preliminary use of aldehyde fixation. In the latter case a dense lumpy material analogous to the one which fills the periplasmic zone and serves as a rigid wall component formed at the surface of the protoplasm and bodies-fragments. In case of escape of the bodies into the external environment through the perforations in the cell wall the principal mass of the protoplast remains intact. The morphology of the bodies-fragments indicated a principal possibility of their autonomic existence. It is supposed that the phenomenon described could serve as one of the mechanisms of L-cell formation.

Interspecific protoplast fusion in Streptomyces--selection of thermotolerant antibiotic-producing recombinant.

The thermotolerant fusants were obtained after interspecific protoplast fusion between S. qingfengmyceticus M15S (SMr, stop growth at 39 degrees C, producing qingfingmycin with wide antimicrobial spectrum) and S. hygroscopicus var. jinggangensis *75 (SMs, grow well at 42 degrees C, producing jingganmycin of antifungus) by directly selecting from the regeneration plates containing SM 100 micrograms/ml and incubated at 42 degrees C. The fusion frequency was about 10(-5) -10(-4). The stable thermotolerant recombinants with antimicrobial activity were obtained. The properties of their products were quite different from that of the parents (Qm, Jm). The antimicrobial substance produced by recombinant F6-6 consists of two components: one has acid-alkaline indicator property; the other is fluorescent under UV light. The antimicrobial products of F1-16, F1-38 and FM3-32 have absorption peaks at 274nm, which suggests that a cytosine moiety may be present in their molecules.

[Study of the effect of protoplast formation on the antibiotic activity of erythromycin producers].

The influence of protoplasting and regeneration on the strains of the erythromycin-producing organism differing by their origin was studied with respect to changes in the antibiotic production property. 223 regenerates of the erythromycin-producing culture were tested in several generations and it was shown that there was a marked change in the range of the variation by that property. 40 to 70 per cent of the variants in the IInd generation increased their levels of erythromycin biosynthesis by 20 to 60 per cent as compared to the intact cultures. However, in the subcultures the antibiotic production level decreased and by the IVth generation only 3 to 6 per cent of the variants preserved its increase by 10 to 20 per cent over the control level because of the culture high instability.

[Isolation of mRNA from protoplasts of spore-forming bacteria and study of its translation products].

Total RNA was isolated from the protoplasts of Bacillus intermedius 7P which actively produced alkaline exocellular RNAse. A fraction of polyadenylated RNA was isolated from this RNA using chromatography on poly(U) Sepharose. The total RNA and poly(A) +RNA were translated in Xenopus laevis oocytes. Bacterial exocellular RNAse was identified among the products of translation by means of electrophoretic and immunological analyses. The mRNA of RNAse secreted by B. intermedius 7P was concluded to be polyadenylated.

Interspecific protoplast fusion of Streptomyces hygroscopicus var. yingchengenisis with Streptomyces qingfengmyceticus and biological characterization of their recombinants.

The frequency of recombination between Streptomyces hygroscopicus var. yingchengensis and Streptomyces qingfengmyceticus as a result of interspecific protoplast fusion was 4.5 x 10(-4). Characterization of biological properties revealed differences in spore mass color, drug resistance, antimicrobial activity, and antibiotic production capacity between the recombinants and their parents. Two recombinant strains, FL-42 and FL-48, exhibited production of the antibiotics originally elaborated by the two parents individually. They also produced two compounds with antimicrobial activity that their parents could not synthesize.

[Biosynthesis of oleandomycin by cultures of Streptomyces antibioticus obtained after regeneration and fusion of protoplasts].

The ability of Streptomyces antibioticus strains to synthesize oleandomycin is studied under the effect of regeneration and fusion of protoplasts. The production of strains-regenerants with an increased (by 30-50%) synthesis of oleandomycin is possible. Regenerants of mutants resistant to the proper antibiotic retain a high level of the oleandomycin synthesis more stably. Variations in the antibiotic-production ability are considered in regenerant populations of various generations.

[Use of a protoplasting method for altering the qualitative and quantitative composition of the anthracycline complex in Streptomyces peucetius var. caesius].

Conditions for efficient regeneration in mutant strains of the doxorubicin-producing organism Str. peucetius var. caesius were developed. The effect of the protoplast regeneration on changes in the proportion of the components of the anthracycline complex produced by these strains was shown. Variants with doxorubicin productivity 2 times higher than that of the parent strain were isolated.

[Fusion of protoplasts in Streptomyces antibioticus].

The analysis of Streptomyces antibioticus recombinants isolated under non-selective conditions for stability of inheritance of parental properties demonstrated the existence of non-stable diploids and stable haploid recombinants. Also, genetic analysis of heteroclones isolated on selective medium after plating spores of regenerants was conducted. The results of analysis of haploid recombinant and heteroclones pointed to linkage of genetic loci and the possibility to use the technique of protoplast fusion for establishment of genetic map of S. antibioticus.

[Fusion of Bacillus subtilis and Bacillus licheniformis protoplasts. The mapping of the mutations leading to the supersynthesis of riboflavin in interspecies hybrids].

As a result of fusion of the protoplasts of B. subtilis and B. licheniformis the majority of the prototrophic hybrids, as well as the auxotrophs with the Thi-Hom-phenotype or the Thi-phenotype acquired capacity for over-production of riboflavin lacking in the initial parent strains. When grown on the minimal Spizizen medium with aeration at 37 degrees C the auxotrophic recombinants accumulated 10-60 micrograms/ml of riboflavin for 2 days, while the prototrophic recombinants accumulated up to 90 micrograms/ml of riboflavin. The respective figures for their cultivation in the sucrose fermentation medium were 100-330 and 600 micrograms/ml. For mapping 3 random variants of them with different phenotypes, i.e. SL-7 (prototroph), SL-15 Thi-and SL-52 Thi-Hom-were used. Localization of the mutation on the chromosome of B. subtilis was based on transformation experiments with three marker crossing, where hybrid strains SL-7, S1-15 and SL-52 were used as the DNA donor and strain lys-rib-of B. subtilis was used as the recipient. The analysis showed that the required mutation designated as R1 was localized on the chromosome of B. subtilis in the regulatory region rib0 of the riboflavin operon.
