Sorbitol dehydrogenase inhibitors (SDIs): a new potent, enantiomeric SDI, 4-[2-1R-hydroxy-ethyl)-pyrimidin-4-yl]piperazine-1-sulfonic acid dimethylamide.

We report here on our medicinal chemistry and pharmacology efforts to provide a potent sorbitol dehydrogenase inhibitor (SDI) as a tool to probe a recently disclosed hypothesis centered on the role of sorbitol dehydrogenase (SDH) in the second step of the polyol pathway, under conditions of high glucose flux. Starting from a weak literature lead, 2, and through newly developed structure-activity relationships, we have designed and executed an unambiguous synthesis of enantiomeric SDI, 6, which is at least 10x more potent than 2. Also, 6 potently inhibits SDH in streptozotocin-diabetic rat sciatic nerve. We have described an expedient synthesis of a key building template, 33, for future research in the SDI area that may facilitate the discovery of even more potent SDIs with longer duration of action in vivo.

Development of human calcitonin gene-related peptide (CGRP) receptor antagonists. 1. Potent and selective small molecule CGRP antagonists. 1-[N2-[3,5-dibromo-N-[[4-(3,4-dihydro-2(1H)-oxoquinazolin-3-yl)-1-piperidinyl]carbonyl]-D-tyrosyl]-l-lysyl]-4-(4-pyridinyl)piperazine: the first CGRP antagonist for clinical trials in acute migraine.

Although the triptans have greatly improved the acute treatment of migraine headache, there are yet many shortcomings. Therefore, new strategies for the treatment of migraine are needed which offer advantages over current therapy, e.g. triptans. Our novel approach was based on the hypothesis that the release of calcitonin gene-related peptide (CGRP) could play a causative role in migraine headache. Thus we initiated a program aimed at the design and synthesis of small molecule CGRP receptor antagonists. High throughput screening led to the identification of (R)-Tyr-(S)-Lys dipeptide-like compounds that showed weak but unequivocal binding to the human CGRP receptor. Lead optimization afforded highly potent CGRP antagonists, the prototype being compound 19 (BIBN4096). This compound exhibiting a favorable biological profile was selected for initial clinical trials. A proof of concept study indicated that intravenous application of 19 was effective in the treatment of acute migraine headache. This finding strongly supports our initial working hypothesis that CGRP plays an important role in the pathophysiology of migraine.

Discovery of N-(2-chloro-6-methyl- phenyl)-2-(6-(4-(2-hydroxyethyl)- piperazin-1-yl)-2-methylpyrimidin-4- ylamino)thiazole-5-carboxamide (BMS-354825), a dual Src/Abl kinase inhibitor with potent antitumor activity in preclinical assays.

A series of substituted 2-(aminopyridyl)- and 2-(aminopyrimidinyl)thiazole-5-carboxamides was identified as potent Src/Abl kinase inhibitors with excellent antiproliferative activity against hematological and solid tumor cell lines. Compound 13 was orally active in a K562 xenograft model of chronic myelogenous leukemia (CML), demonstrating complete tumor regressions and low toxicity at multiple dose levels. On the basis of its robust in vivo activity and favorable pharmacokinetic profile, 13 was selected for additional characterization for oncology indications.

Effect of N-tris (Hydroxymethyl) Methylglycine and N-2-Hydroxyethyl-piperazine-N'-2-Ethanesulfonic Acid Buffers on Linolenic Acid as a Substrate for Flaxseed Lipoxidase.

The delay in, or loss of, flaxseed lipoxidase activity in N-tris (hydroxymethyl) methylglycine and N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid buffers with linolenic acid as a substrate appears due to an alteration of the lipid micelle. Flaxseed lipoxidase activity is dependent on the ionic strength of the assay solution. These effects are not observed with linoleic acid as substrate. The influence of these 2 buffers on linolenic acid micelles may have a direct bearing on recent reports of chloroplast structure and activity in these buffers.

Diethyl piperazine-1,4-diyldioxalate.

The ethyl oxamate group, N-C(O)-C(O)-OEt, in the title compound, alternatively called diethyl N,N':N,N'-bis(ethylene)dioxamate, C12H18N2O6, can be considered as being composed of two singly bonded amide and ester functionalities. The ethyl oxamate group is not planar. The two carbonyl groups are almost perpendicular, with an oxalyl O=C-C=O torsion angle of -111.34 (17) degrees. The molecule is located on an inversion centre. Infinite supramolecular tapes, propagating along the b axis, are formed through soft C-H...O interactions which form a centrosymmetric R(2)2(12) motif.

Comparative activity and beta-lactamase stability of cefoperazone, a piperazine cephalosporin.

The in vitro activity and beta-lactamase stability of 7-[d(-)-alpha-(4-ethyl-2,3-dioxopiperazino-carbonylamino) -p-hydroxyphenylacetamido]-3-[(1-methyl)-5-tetrazolylthiomethyl] -Delta(3)-cephem-4-carboxylic acid (cefoperazone), a cephalosporin analog of piperacillin, were compared with the activities and stabilities of other cephalosporins and cephamycins. The compound was less active than cephalothin or cefamandole in inhibiting Staphylococcus aureus; it was as active as cefamandole and cefoxitin against most of the Enterobacteriaceae but less active than cefotaxime. It was more active than carbenicillin or piperacillin against Pseudomonas aeruginosa. In general, the compound was not active against Bacteroides. It was hydrolyzed by the beta-lactamases of some Escherichia coli which hydrolyzed cefamandole, but was stable to most plasmid-mediated, chromosomally mediated, inducible beta-lactamases in the Enterobacteriaceae and Pseudomonas.

[Preparation and physico-chemical properties of alkyl esters of 2-, 3-, 4-(2-hydroxy-3-[4-diphenylmethyl) piperazine-1-yl] propoxy) phenylcarbamide].

The paper is devoted to the synthesis and study of some physico-chemical properties of a group of substances--potential antagonists of beta-adrenergic receptors. The substances are derivatives of aryloxyaminopropanol, where the basic part consists of diphenylmethylpiperazine and the aromatic part is modified with alkyl esters (methyl to butyl) of carbamic acid in positions 2-, 3-, 4-. The representation of the elements was confirmed by elemental analysis. The substances were characterized by melting point, IR and UV spectra. Their solubility and surface activity were determined. The purity of prepared substances was examined by means of TLC.
