Laboratory and field performance of an imidacloprid gel bait against German cockroaches (Dictyoptera: Blattellidae).

An experimental 2.15% imidacloprid gel bait containing approximately 44% water was evaluated in laboratory and field studies against the German cockroach, Blattella germanica (L). In continuous exposure tests, toxicity and presumably bait consumption varied with cockroach stage, deprivation of competitive food, and temperature. The LT50 values for cockroaches provided with competitive food ranged from approximately 0.9 h for adult females to 190 h for small nymphs. The LT50s for cockroaches not provided competitive food ranged from approximately 1.7 h for adult females to approximately 31 h for adult males. The LT50s decreased exponentially with temperature between 10 and 30 degrees C. Even though the bait was significantly more repellent (approximately 38%) than an untreated control (approximately 14%) when tested in Ebeling choice boxes, performance index values were positive and increased to nearly 100 (indicating high mortality and low repellency) after 14 d. When applied at 15-45 g per kitchen, the bait significantly reduced German cockroach trap catch in infested homes during a 4-wk period. There was a approximately 50% reduction after 1 wk and approximately 80% reduction 4 wk after treatment.

Oviposition preference and egg eclosion in different salt concentrations in the coastal malaria vector Anopheles aquasalis Curry.

Anopheles aquasalis is the main malaria vector in Sucre State, Venezuela. The larvae of this species are saltwater tolerant. The effects of different concentrations of salt on oviposition preference and egg survival were studied under laboratory conditions. Choice experiments with salt concentrations of 0, 10, 20, 30, and 40% in bottled water were set up for individual adult females and the number of eggs laid in each salt concentration was noted. Egg survival, as inferred by the number of hatched larvae also was determined for each salt concentration. Females preferred to oviposit in freshwater and rejected water salt concentrations of 40%, but they were neither attracted nor repelled by water with 10-30% of salt. Eggs hatched more quickly in the lower salt concentrations, but egg survival was not affected by salt concentrations of up to 20%. Thus, female oviposition preference in An. aquasalis determines egg survival.

Use of residue profile analysis to identify modes of insecticide activity contributing to control of plum curculio in apples.

Residue profile analysis techniques were developed, along with laboratory and field-based bioassays to describe the modes of insecticidal activity responsible for the control of the plum curculio, Conotrachelus nenuphar (Herbst), in apples (Malus spp.). Adult plum curculios were treated in laboratory topical bioassays to determine acute contact activity and lethal time for five insecticides. Azinphosmethyl had the highest levels of toxicity and shortest lethal time values, followed by the neonicotinoids thiacloprid, thiamethoxam, and imidacloprid, whereas indoxacarb had the highest LD50 and LT50 values for topical exposure. Field-based residual activity bioassays assessed adult mortality, and fruit and leaf injury from plum curculio exposed to 4 h, 7 d, and 14 d field-aged residues. All compounds caused significant levels of mortality to plum curculio when adults were exposed to fruit clusters 4 h post-application. Thiacloprid, thiamethoxam, and imidacloprid showed oviposition deterrence, antifeedant, and repellency effects in the 7- and/or 14 d residual bioassays and protected fruit in the absence of significant lethal activity. Indoxacarb maintained lethal activity throughout the study intervals, with the incidence of plum curculio feeding, suggesting that ingestion is an important mode of entry. For the neonicotinoids thiacloprid, thiamethoxam and imidacloprid plum curculio mortality was highly correlated with fruit and leaf surface residues. As surface residues declined, sublethal effects such as oviposition deterrence and antifeedant effect remained. The value of the plant-insect-chemistry triad model for describing the temporal dimensions of insecticidal modes of activity and understanding a compound's critical performance characteristics is discussed.

Influence of nutritional and physiological status on behavioral responses of Aedes aegypti (Diptera: Culicidae) to deltamethrin and cypermethrin.

Excito-repellency responses of Aedes aegypti (L.) exposed to deltamethrin and cypermethrin were assessed using an excito-repellency test system. Contact irritancy and non-contact repellency assays compared non-bloodfed (unfed) parous (post-gravid), nulliparous, early blood-fed, late blood-fed, sugar-fed, and unmated female mosquitoes for behavioral responses based on nutritional and physiological conditions at the time of testing. Rates of escape during contact exposure with either compound were most pronounced in parous mosquitoes, followed by unmated mosquitoes, when compared to other conditional states. Significantly higher numbers of parous females also escaped from control chambers compared to other cohorts (P <0.05). Irritability of blood- and sugar-fed mosquitoes was noticeably suppressed. We conclude that nutritional and physiological conditions (including age) of mosquitoes at the time of testing can significantly influence behavioral responses (excito-repellency) to insecticides. The findings indicate that whether due to chronological age, nutrition, physiological state, or innate (circadian) activity patterns, careful consideration must be given to the selection of appropriate conditioned mosquitoes for testing.

A paradoxical gradient of a basal lamina-associated repellent is essential for pathfinding by the Ti1 pioneer axons in cockroach embryos.

Perturbations of in situ axon growth with proteolytic enzymes and monoclonal antibodies were used to determine the role of gradient guidance cues in the formation of the Ti1 pioneer axon trajectory in cultured cockroach embryos. Treatment with enzymes that degrade the basal lamina indicated that this substrate contains both an elastase-sensitive proximal directing cue and a collagenase-sensitive distal directing cue. The latter is shown to be a repellent of axon growth and is identical to the PROD-2 antigen that is distributed in a gradient along the proximal-distal axis of the leg with high levels in proximal regions. This means that throughout the course of their growth the axons extend in the direction of increasing levels of repellent. At a critical decision point in the trajectory the axons change both the direction of growth and the substrate to which their growth cones adhere. PROD-2 plays an essential role in both of these processes.

[Evaluation of cocoa oil for individual protection against Simulium damnosum s.i].

Thanks to larviciding and distribution of ivermectine, transmission of onchocerciasis was virtually halted in all countries covered by the WHO Onchocerciasis Control Program (OCP). However as a result of this success vector control has been stopped in most areas covered by the program and a recrudescence of files is an increasing problem. In some rural communities fly biting hinders normal agricultural activity. Since local farmers cannot afford ground larviciding, many rely on topical repellents. For this reason, development of an inexpensive method of individual protection is a major priority. The purpose of this study carried out in the savannah area of the Côte d'Ivoire was to assess the repellent properties of cocoa oil, which is frequently used in some rural areas of the Côte d'Ivoire to prevent fly biting. Results showed that cocoa oil provides excellent protection against Simulium damnosum s.l.

Prevention of the transmission of Chagas' disease with pyrethroid-impregnated materials.

Interrupting the transmission of Chagas' disease using insecticide-treated materials could be a cost-effective option, particularly for sylvatic vectors, which enter houses at night. A randomized trial was undertaken that included all houses in two communities in regions endemic for this disease, one in Venezuela (50 houses) and one in Colombia (47 houses). After a baseline study (including a short questionnaire survey, entomologic assessment, and Chagas' disease serology), each household was randomly allocated to either the intervention group, which used pyrethroid-impregnated bed nets, or the control group, which used unimpregnated bed nets. Serologic analysis of children in the baseline study showed active transmission of Chagas' disease in the Venezuelan community (10.7% of 103 children were positive), but none in the Colombian community (0% of 100 children were positive). Vectors were sylvatic and bugs entered the houses at night in both communities. The efficacy of pyrethroids against triatomine vectors was tested in Venezuela by exposing Rhodnius prolixus to lambdacyhalothrin-impregnated fabrics and in Colombia by residual house spraying with deltamethrin. This randomized trial showed that in both countries users of impregnated bed nets were well protected from vector bites (immediate benefit). The long-term effect on the community was high vector mortality. In Venezuela, all 62 vectors detected (mainly R. robustus) died within 72 hours of contact with impregnated bed nets. In houses that used unimpregnated nets, only 24.5% (13 of 53) of the vectors died (P < 0.001). The vectors most likely came from infested palm trees and they maintained transmission of the disease in this community (28.1% of 629 R. robustus were positive for Trypanosoma cruzi). Bioassays showed that the mortality rate of R. prolixus was 100% on lambda-cyhalothrin-impregnated materials. In addition, in Colombia (the main vector was R. prolixus), the effect of repellent on vectors (driving them away from impregnated nets) was significant. Thus, users of impregnated bed nets are well protected from transmission of Chagas' disease, and vector reduction or elimination can potentially be achieved in areas infested with R. prolixus and R. robustus.

A comparison study of house entering and exiting behavior of Anopheles vestitipennis (Diptera: Culicidae) using experimental huts sprayed with DDT or deltamethrin in the southern district of Toledo, Belize, C.A.

An investigation of the house entering and exiting behavior of Anopheles vestitipennis Dyar and Knab was undertaken in the Toledo District of Belize, Central America, between March and December of 1998. Three untreated experimental huts were either fitted with exit or entrance interception traps or used as a control for human landing collections. Human landing collections showed that An. vestitipennis exhibited a high level of biting activity shortly after sunset and continued biting at high levels throughout the night. Under unsprayed conditions, the use of exit and entrance interception traps demonstrated that doors, windows, and eaves were the primary mode of entry; whereas, cracks in the walls served a secondary role. The peak entrance time for An. vestitipennis occurred between 6:45 P.M. and 9:45 P.M. and a peak exit time occurred between 11:45 P.M. and 4:45 A.M. Additional trials were conducted after spraying one of the huts with DDT and another with deltamethrin. The excito-repellent properties of deltamethrin did not affect entrance times but did result in a peak exiting behavior that was five hours earlier than under pre-spray conditions. Deltamethrin also exhibited a repellency effect, showing 66% fewer An. vestitipennis entering the hut two weeks post-spray. DDT had an even more powerful repellency effect resulting in a 97% post-spray reduction of An. vestitipennis females entering the hut up to two weeks post-spray. The control hut showed only a 37% reduction in An. vestitipennis as compared to pre-spray conditions.

Perceived nuisance of mosquitoes on the Isle of Sheppey, Kent, UK.

Little is known about the biting nuisance of mosquitoes in the UK, despite the high numbers found in some locations. A telephone questionnaire survey was used to determine the perceived nuisance of biting insects on the Isle of Sheppey, Kent, a place notorious for mosquitoes. Two hundred randomly selected individuals were interviewed and asked if they suffered from mosquito bites. If they answered yes, they were asked to describe where and when they were bitten, and what measures they took against mosquitoes. Forty-six percent of respondents completed the questionnaire. Of those, 50% reported being bitten by mosquitoes, mostly outside during the summer. Seventy percent said that most biting occurred during the evening and night. Of those respondents who protected themselves against biting (27), most used repellents (70%), with the remainder changing their behaviour to avoid mosquitoes, including closing or screening windows (33%), wearing thicker clothes (7%) and spraying insecticide (4%). One person slept under a bednet in summer (4%). This study provides evidence that on the Isle of Sheppey mosquitoes are considered a major nuisance by a sizeable proportion of the population. Since there is growing interest in the threat posed by new and emerging diseases in the UK, health authorities will need to make substantial efforts to inform and reassure the public about the threats posed by mosquitoes in areas where they are common.

Effect of the combined treatment of insecticides and an attractant for the control of Phloeotribus scarabaeoides, a pest of Olea europea.

Different insecticides have been tested for the control of the olive bark beetle, Phloeotribus scarabaeoides Bern. This scolytid can be managed at two points in its biological cycle: in pruned logs, where it excavates reproduction galleries, or in living trees, after emergence from the logs, where it digs feeding galleries. In mortality laboratory bioassays, the efficiency of organophosphorus insecticides has been ranked as follows: chlorpyrifos + dimethoate < formothion < methidathion. Formothion and methidathion, the two most efficient, were sprayed on olive logs together with a pyrethroid insecticide, deltamethrin, and a formulation which combined an organophosphorus (fenitrothion) and a pyrethroid (cypermethrin) insecticide. Deltamethrin inhibited the excavation of new reproduction galleries and induced a repellent effect on the olive pest. In contrast, none of the organophosphorus insecticides or the combination, fenitrothion + cypermethrin, were able to control the olive bark beetle. In olive trees, deltamethrin controlled this olive pest without showing the repellent effect observed for logs. Ethylene, a plant hormone, has been reported as an attractant for the olive bark beetle. The use of dispensers which released ethylene increased the number of P. scarabaeoides approaching the treated olive trees, thus favouring its use in a lure-and-trap control system.

Evaluation of the CatanDog's tag to prevent flea infestations, inhibit flea reproduction or repel existing flea infestations on cats.

To evaluate the ability of the CatanDog's tag to eliminate fleas, inhibit egg production and prevent flea infestations, six domestic shorthaired cats were randomly allocated to two treatment groups and housed individually in stainless steel metabolic cages. Three cats were each fitted with a CatanDog's tag; the other three cats were not fitted with tags and served as controls. Following a 42-day acclimation period, each of the six cats was infested with 100, 1-3 day post-emergence, adult Ctenocephalides felis felis (Bouché) on days 0, 7, 14, 21, and 27. Flea egg production was determined by collecting and enumerating eggs 2, 4 and 6 days after each infestation. Viability of eggs was determined by placing 100 eggs recovered from each cat in rearing media in an insect rearing chamber and determining adult emergence at 28 days. Adult fleas were recovered from cats 6 days post-infestation by thoroughly combing each cat to remove fleas. To determine if the tags provided protection from infestation, the six cats were placed into a 8.53mx4.36 m room with 400 cat fleas for 3h. Cats were then combed to remove and enumerate fleas. The CatanDog's tags had no significant effect upon egg production, egg viability, or adult fleas infesting cats. In addition there was no difference in the numbers of fleas recovered from the cats placed in the flea-infested room.

Effect of indoor residual spraying of DDT and bendiocarb on the feeding patterns of Anopheles pseudopunctipennis in Mexico.

Intense and persistent use of DDT for malaria control has increased resistance and induced exophilic behavior of Anopheles pseudopunctipennis. An evaluation of bendiocarb and DDT to control this species in Sinaloa, Mexico, showed that, in spite of DDT-resistance, both insecticides produced similar effects. Feeding patterns were analyzed to explain these results. Resting mosquitoes were collected over the dry and wet seasons. Anophelines were tested in an ELISA to determine the source of the meals. The human blood index (HBI) ranged from 3.3 to 6.8% in DDT- and from 12.7 to 26.9% in bendiocarb-sprayed houses. Irritability and repellency in DDT-sprayed houses could explain the reduced HBI. In contrast, bendiocarb produced higher mortality. These effects could have affected different components of the vectorial capacity and similarly reduced malaria.

Spatially restricted activity of a Drosophila lipid phosphatase guides migrating germ cells.

Temporal and spatial controls of cell migration are crucial during normal development and in disease. Our understanding, though, of the mechanisms that guide cells along a specific migratory path remains largely unclear. We have identified wunen 2 as a repellant for migrating primordial germ cells. We show that wunen 2 maps next to and acts redundantly with the previously characterized gene wunen, and that known wunen mutants affect both transcripts. Both genes encode Drosophila homologs of mammalian phosphatidic acid phosphatase. Our work demonstrates that the catalytic residues of Wunen 2 are necessary for its repellant effect and that it can affect germ cell survival. We propose that spatially restricted phospholipid hydrolysis creates a gradient of signal necessary and specific for the migration and survival of germ cells.

Lola regulates midline crossing of CNS axons in Drosophila.

The pattern and level of expression of axon guidance proteins must be choreographed with exquisite precision for the nervous system to develop its proper connectivity. Previous work has shown that the transcription factor Lola is required for central nervous system (CNS) axons of Drosophila to extend longitudinally. We show here that Lola is simultaneously required to repel these same longitudinal axons away from the midline, and that it acts, in part, by augmenting the expression both of the midline repellant, Slit, and of its axonal receptor, Robo. Lola is thus the examplar of a class of axon guidance molecules that control axon patterning by coordinating the regulation of multiple, independent guidance genes, ensuring that they are co-expressed at the correct time, place and relative level.

Laboratory and field performance of consumer bait products for German cockroach (Dictyoptera: Blattellidae) control.

Bait formulations containing boric acid, chlorpyrifos, or hydramethylnon were evaluated in field and laboratory experiments against the German cockroach, Blattella germanica (L.). In continuous exposure tests, chlorpyrifos formulations produced 100% mortality in less than 24 h, boric acid formulations had LT50's of approximately 5.5 d, and hydramethylnon had an LT50 of 0.86 d. LT50's estimated in Ebeling choice boxes were 1.6 to 10.8 times greater than those from continuous exposure tests for chlorpyrifos formulations and 2.5 times greater for the hydramethylnon formulation. The same LT50's were produced by choice box and continuous exposure tests for boric acid formulations. Relative repellency, measured as the mean percentage of live cockroaches in the light side of the choice box, was greatest for chlorpyrifos formulations, followed by boric acid formulations, then hydramethylnon. A performance index (PI) that combined choice box repellency and mortality data indicated that hydramethylnon (Combat) had the greatest potential for field effectiveness. Only hydramethylnon significantly reduced German cockroach populations in the field. Apartments treated with the IT WORKS formulation of boric acid bait had significantly greater cockroach trap catches. The PI is an accurate predictor of the field performance of toxic baits for German cockroach control.

U.S. Army soldiers' perceptions of arthropod pests and their effects on military missions.

A survey was conducted to determine the effects of biting and stinging arthropods on military personnel, operations, and training. Nearly 70% of respondents reported experiencing problems attributable to arthropods. Arthropods obstructed movement and field position, prevented concealment and cover, disrupted maneuvers, and caused panic. Twenty percent of respondents reported attendance at sick call for treatment of bites or stings, and 4% were hospitalized or assigned to quarters. Median lost time was 2 days. Bee, wasp, and ant stings and spider and chigger bites were the most frequent causes of lost time. Additional training on biting and stinging arthropods, use of repellents and other personal protective measures, first aid for bites and stings, and conditions requiring medical attention is needed in field units to enhance mission performance and reduce time lost because of arthropods. Materials for treatment of bites and stings should be included in first-aid kits issued for field use.

[Evaluation of a DEET-based repellent on 3 vectors of malaria in central Africa].

As part of the framework of studies on personal protection against disease-transmitting mosquitoes, a field evaluation was carried out in a forest area of Cameroon on human subjects to assess the efficacy of a common repellent: DEET (a 50% active ingredient formulation). One ml of DEET applied to the legs of resting persons gave an 85% reduction in Anopheles bites during the 5 hours following application. The repellent effect decreased gradually with time. The effect was more than 90% maximum after 3 hours, 50% by about the seventh hour and declined to 0% from the fifteenth hour onwards. No difference was found between the three Anopheles species: Anopheles gambiae ss, An. nili and An. funestus. The longevity and infectivity of Anopheles populations caught on humans with repellent and on untreated controls were similar. Thus the reduction of biting rate can be considered equivalent to the reduction in malaria transmission. The effect of sweating on the loss of the applied repellent was studied. Moderate sweating, corresponding to normal activity, did not reduce the efficacy of the repellent. More profuse sweating did not significantly reduce repellent efficacy. We conclude that the tested formulation of DEET has the same efficacy as commercial formulations already on the market. Protection did not last the whole night, which is the time required for protection against malarial infection. However, good protection levels lasted more than 5 hours.(ABSTRACT TRUNCATED AT 250 WORDS)
