Glucagon: a stress hormone in the domestic fowl.

Chicks were injected intraperitoneally with glucagon (50 micrograms/kg) once daily for five days, beginning on the first day after hatching. At seven days of age the chicks were weighed and tissues sampled. Growth rate was unaffected, relative adrenal weight was increased and there were significant increases in adrenal cholesterol stores. The birds were hypoglycaemic, hypercholesteraemic but had a decreased concentration of corticosterone in the plasma. The results are consistent with the hypothesis that glucagon is a stress hormone and that it is responsible for activating the hypothalamo-hypophyseal-adrenocortical axis.

The efffect of sustained +Gz acceleration on extravascular lung water content in domestic fowl.

In order to examine the effects of sustained acceleration on pulmonary fluid balance, chickens were centrifuged at low sustained +G acceleration (LSG) intensities of +3Gz or +4Gz or high sustained +G acceleration (HSG) at +6Gz for varying periods. Animals were exposed to acceleration on a hydraulic-driven centrifuge. Controls (Group I) were not centrifuged. The vascular lungs were rapidly excised after centrifugation, and the extravascular lung water content (EVLW) was measured. EVLW in Group I animals = 2.19 +/- 0.38 g/g. Results indicate that positive acceleration elevated in animals exposed to +4Gz for 60 min (Group III). In this group, EVLW = 3.01 +/- 1.07 g/g. This increase is largely attributable to an elevated EVLW in those animals which died as a result of centrifugation, in which EVLW = 3.90 +/- 1.04 g/g. Exposure to +3Gz for 120 min (Group II) or +6Gz for periods up to 50 min (Group IV) did not produce significant elevations in EVLW in either group as a whole, but EVLW in those Group II animals which died as a result of centrifugation was significantly elevated. EVLW in these animals = 2.87 +/- 0.21 g/g. We conclude that pulmonary edema caused by centrifugation is dependent on both magnitude and duration of G force.

Right Müllerian duct in the domestic fowl during postnatal ontogenesis.

In this study the postembryonal changes of the right Müllerian duct are investigated. This structure was found in all female broilers of the hybrid combination Ross 1 as well as in hens of the female line of parent stock for the production of this hybrid. A thready structure of the length of several millimeters observed in one-day-old females develops in chicks into a long, thin-walled ampula filled up with clear liquid. In mature hens this rudiment is markedly long and its shape is rather different. In some cases the Müllerian duct was divided into several parts which might be compared with individual parts of the left oviduct.

On the structure of the vascular body in the domestic fowl.

The microscopic structure of the vascular body has been described. Three cell types have been identified. The arrangement of blood and lymph vessels has been demonstrated by means of injection techniques, and these vessels have been investigated at an ultrastructural level. The function of the vascular body is discussed.

Epidemiological studies of coccidiosis in the domesticated fowl (Gallus gallus): II. Physical condition and survival of Eimeria acervulina oocysts in poultry-house litter.

The survival of coccidial oocysts in poultry-house litter has been examined. The experimental design ensured that viable oocysts were not continuously replenished by multiplication in chicks. Hence, reliable oocyst survival data were obtained, which is not possible in a commercial house where there is a continuously changing population of coccidia. Litter was seeded directly from infected chicks with unsporulated oocysts of Eimeria acervulina. Whilst sporulation proceeded rapidly, oocysts simultaneously began to deteriorate after only 24 h. After five days, most oocysts were disintegrating. Viable oocysts were, however, detected (using sentinel birds) up to day 23, although subsequently the viability of the oocyst population began to wane. Few viable oocysts were detectable after 38 days, despite some intact sporulated oocysts being evident up to 54 days. The physical condition of oocysts is therefore not a reliable indication of their viability. The surface temperature of the litter was 25-28 degrees C throughout and the moisture content varied from 31.0 to 62.1%; these conditions were not limiting factors for oocyst sporulation.

Specific antibody production against a soluble antigen in the Harderian gland of the domestic fowl.

Three groups of chickens were immunized with bovine serum albumen (BSA). One group received the antigen i.v., the second had BSA dropped into the orbit and the third group was immunized both systemically and by topical application of BSA. Agar-gel diffusion studies showed antibodies to BSA to be present in the tears and sera of the first and third groups but only in the tears of the second group. Immunofluorescence revealed positive anti-BSA staining in lymphoid cells of the spleens of the first and third groups and in plasma cells of Harderian glands of all three groups. It is concluded that systemic and/or topical applications of BSA affect the tears and Harderian gland whereas only systemic injections produce an effect in the spleen and serum. The Harderian gland may have an important part to play in the local immunologic mechanism of the eye and upper respiratory tract of the fowl.

Isospora sp from ducks. Infectivity for the goose, four anatids and the domestic fowl.

A coccidium was isolated from mule duck farms, the sporulated oocysts of which most closely resemble that of Isospora anseris, a goose coccidium. By experimental infection, these oocysts were proved to be able to develop in the goose and in several duck species, but unable to develop in the chicken. No pathogenicity was noted in any species.

Appearance of intraportally infused [15N]urea in blood and urine of domestic fowl.

The time course of the occurrence of intraportally infused [15N]urea in blood and urine was investigated in chickens. The infused urea appeared in ureteral urine, mostly in the form of urea, as early as 30 min after the start of infusion and the excretion further increased up to the end of 2 hr infusion. Blood urea concentration rapidly increased and reached about three times the initial level at the end of the experiment (P < 0.05 after 20 min), but no significant effects were observed on uric acid, ammonia and glutamine concentrations. Fifty-seven percent of blood urea N and 3% of blood glutamine-amide N and 1% of blood ammonia N, which were determined at the end of experiment, were derived from the infused urea N. It is concluded that urea, which is rapidly increased in blood and urine after feeding urea to chickens, is mostly derived from dietary urea.

The gastro-intestinal helminth infections of domestic fowl in Dschang, western Cameroon.

Three hundred and fifty one chickens purchased from the Dschang animal market were examined for gastro-intestinal helminths. Ten species were found with the following prevalences: Heterakis brevispiculum (59.3%), Ascaridia galli (51.6%), Hymenolepis carioca (48.4%), Dispharynx spiralis (20.8%), Tetrameres americana (17.1%), Amoebotaenia cuneata (15.1%), Raillietina tetragona (14.5%), Syngamus trachea (13.7%), Hymenolepis cantaniana (5.7%) and Capillaria contorta (2.0%). Infections were predominantly mixed (93.5%). The infection rates were not influenced by host sex except for A. galli which was more prevalent in cocks. Older chickens showed some resistance to A. cuneata and S. trachea. Parasite prevalence and/or worm burdens were generally higher during the rainy season (April to October).

Phylogenetic relationships among eight Eimeria species infecting domestic fowl inferred using complete small subunit ribosomal DNA sequences.

Complete 18S ribosomal RNA gene sequences were determined for 8 Eimeria species of chickens and for Eimeria bovis of cattle. Sequences were aligned with each other and with sequences from 2 Sarcocystis spp., Toxoplasma gondii, Neospora caninum, and 4 Cryptosporidium spp. Aligned sequences were analyzed by maximum parsimony to infer evolutionary relationships among the avian Eimeria species. Eimecia bovis was found to be the sister taxon to the 8 Eimeria species infecting chickens. Within the avian Eimeria species, E. necatrix and E. tenella were sister taxa: this clade attached basally to the other chicken coccidia. The remaining Eimeria spp. formed 3 clades that correlated with similarities based on oocyst size and shape. Eimeria mitis and Eimeria mivati (small, near spherical oocysts) formed the next most basal clade followed by a clade comprising Eimeria praecox. Eimeria maxima, and Eimeria brumetti (large, oval oocysts), which was the sister group to Eimeria acervulina (small, oval oocysts). The 4 clades of avian Eimeria species were strongly supported in a bootstrap analysis. Basal rooting of E. necatrix and E. tenella between E. bovis and the remaining Eimeria species and the apparent absence of coccidia that infect the ceca of jungle fowl all suggest that E. necatrix and E. tenella may have arisen from a host switch, perhaps from the North American turkey, Meleagris gallopavo.

The effect of noradrenaline and acetylcholine on blood arginine vasotocin level in the domestic fowl.

The effect of naredrenaline and acetylcholine on the blood arginine vasotocin (AVT) level in hens as well as the effect of reserpine and atropine on the release of this hormone following the intravenous injection of hypertonic NaCl solution were investigated. It was dound that noradrenaline causes a decrease in blood AVT level in contrast to acetylcholine. Reserpine itself caused a rise in the blood AVT level; the effect was also found after administration of hypertonic solution. Atropine had no effect on the blood AVT level, but inhibited the AVT release following the adminstration of hypertonic Nacl solution.

[Proventriculitis in domestic fowl (Gallus gallus var. dom. L., 1758) caused by megabacteria].

Three cases of a disease similar to going light syndrome in budgerigars in a flock of young laying hens in Turkey are reported. In all cases megabacteria in association with E. coli could be isolated. The causing agent appeared to be large, rod-shaped, gram-positive or gram-labil, and catalase test positive bacteria with cytochromoxydase activity. Subcultivation of these bacteria could not be achieved.

Isolation of adenoviruses and reoviruses from avian species other than domestic fowl.

Adenoviruses and reoviruses were isolated from pigeons and mallard ducks. In addition, adenoviruses were isolated from budgerigars and a bantam and a reovirus was isolated from a turkey. Primary identification of these viruses was by electron-microscope examination. It was further possible to assign the 4 adenoviruses to recognized fowl serotypes, and the reoviruses shared a common antigen with fowl reoviruses. These viruses were isolated from a variety of clinical conditions.

Peptidergic pathways in the avian brain.

The results obtained by topographical studies on the immunoreactive peptide systems in the embryonic and adult avian brain (domestic fowl, domestic mallard, pigeon, Japanese quail, and zebra finch) can be realized only by means of phylogenetical comparisons. The comparative studies mainly demonstrate a fascinating constancy of the immunological properties and the spatial distribution of the neuropeptides. Independent of the development of the neopallium, and the increasing cerebral complexity, the spatial distribution of the neuropeptides, the location of their main perikaryal accumulations which are interconnected by immunoreactive fiber projections (and thereby forming widespread but continuous peptide systems) remain nearly unchanged during vertebrate evolution. The recognition of the neuropeptides as integral parts of the central nervous system is demonstrated by the fact that neuropeptidergic structures connect sensory inputs with central nervous areas as well as with the peripheral endocrinium.

Morphological changes in the efferent ducts during the main phases of the reproductive cycle of birds.

The changes that take place in the efferent ducts during the major phases of the reproductive cycle of birds were studied morphologically using standard histological, morphometric, and ultrastructural methods in prepuberal, sexually mature and sexually active, and sexually mature but sexually inactive domestic fowl (Gallus domesticus), drake (Anas platyrhynchos), and guinea fowl (Numida meleagris). Profound structural and dimensional changes occurred in both segments (proximal and distal) of the efferent ducts and, in particular, in the nonciliated (Type I) cell of the proximal duct of sexually mature but inactive birds. The subapical tubulovacuolar system was markedly atrophic in nonciliated (Types I and II) cells and the numerous round dense globules of Type I cells that normally occurred in sexually active birds were replaced by fewer and more pleomorphic bodies containing lipofuscin granules in sexually resting birds. Lipid droplets, few and extremely large in inactive drakes but numerous and smaller in size in guinea fowls and domestic fowls, occurred in the Type I cell at both infra- and supranuclear levels of resting but not in prepuberal or sexually active birds. Ciliated cells in both segments of the ducts exhibited fewer and less profound phase-dependent changes ultrastructurally. Generally, the Type I cells of the proximal efferent duct appeared to be more sensitive to androgen deprivation than the Type II cell of the distal efferent duct or ciliated cells in both ducts. These morphologically phase-dependent features of the efferent ducts of birds may be used, together with or independent of testicular changes, in the determination of the status of the testis and epididymis of a male bird with regard to the reproductive cycle, especially in seasonally breeding species.

[Epizootiology and pathogenesis of avian mycobacteriosis in the laughing gull (Larus ridibundus)].

The occurrence of avian mycobacteriosis and sensitivity to M. avium were investigated in the laughing gull in this paper. The patho-morphological and culture examination of 35 individuals of the laughing gull caught at four different localities of the Czech Republic did not reveal any tuberculous lesions nor did it enable to isolate mycobacteria. After experimental intramuscular infection by suspension of M. avium macroscopic tuberculous lesions were found at the spot of puncture in 35 days, in liver and spleen in 70 days since the beginning of infection. In the simultaneously infected domestic fowl the lesions were determined at the spot of puncture, in spleen and liver in 35 days. Mycobacteria were isolated from several organs and tissues in both species in 35 days after infection. Tuberculous lesions were found in liver, spleen and intestines of the laughing gull in 118 days since peroral infection by feed contaminated with the liver of tuberculous fowl. This finding was identical in spleen of the domestic fowl only in 118 days. Mycobacteria were demonstrated in both species in 118 days, in liver, spleen, intestine and/or brain in the laughing gull, while in spleen only in the domestic fowl. Intestinal tuberculosis was determined in the laughing gull in 118 days after infection. The results of experimental infections show that the sensitivity of the laughing gull to M. avium is the same as in the domestic fowl. The occurrence of tuberculosis in the laughing gull is apparently depending upon the local epizootological situation, particularly upon the occurrence of M. avium sources and their accessibility.(ABSTRACT TRUNCATED AT 250 WORDS)
