The maximum attainable body size of herbivorous mammals: morphophysiological constraints on foregut, and adaptations of hindgut fermenters.

An oft-cited nutritional advantage of large body size is that larger animals have lower relative energy requirements and that, due to their increased gastrointestinal tract (GIT) capacity, they achieve longer ingesta passage rates, which allows them to use forage of lower quality. However, the fermentation of plant material cannot be optimized endlessly; there is a time when plant fibre is totally fermented, and another when energy losses due to methanogenic bacteria become punitive. Therefore, very large herbivores would need to evolve adaptations for a comparative acceleration of ingesta passage. To our knowledge, this phenomenon has not been emphasized in the literature to date. We propose that, among the extant herbivores, elephants, with their comparatively fast passage rate and low digestibility coefficients, are indicators of a trend that allowed even larger hindgut fermenting mammals to exist. The limited existing anatomical data on large hindgut fermenters suggests that both a relative shortening of the GIT, an increase in GIT diameter, and a reduced caecum might contribute to relatively faster ingesta passage; however, more anatomical data is needed to verify these hypotheses. The digestive physiology of large foregut fermenters presents a unique problem: ruminant-and nonruminant-forestomachs were designed to delay ingesta passage, and they limit food intake as a side effect. Therefore, with increasing body size and increasing absolute energy requirements, their relative capacity has to increase in order to compensate for this intake limitation. It seems that the foregut fermenting ungulates did not evolve species in which the intake-limiting effect of the foregut could be reduced, e.g. by special bypass structures, and hence this digestive model imposed an intrinsic body size limit. This limit will be lower the more the natural diet enhances the ingesta retention and hence the intake-limiting effect. Therefore, due to the mechanical characteristics of grass, grazing ruminants cannot become as big as the largest browsing ruminant. Ruminants are not absent from the very large body size classes because their digestive physiology offers no particular advantage, but because their digestive physiology itself intrinsically imposes a body size limit. We suggest that the decreasing ability for colonic water absorption in large grazing ruminants and the largest extant foregut fermenter, the hippopotamus, are an indication of this limit, and are the outcome of the competition of organs for the available space within the abdominal cavity. Our hypotheses are supported by the fossil record on extinct ruminant/tylopod species which did not, with the possible exception of the Sivatheriinae, surpass extant species in maximum body size. In contrast to foregut fermentation, the GIT design of hindgut fermenters allows adaptations for relative passage acceleration, which explains why very large extinct mammalian herbivores are thought to have been hindgut fermenters.

[Balkan endemic nephropathy in Bosnia and Herzegovina--renal registry report].

INTRODUCTION: Balkan Endemic Nephropathy (BEN) is still dominant cause of the end stage renal disease (ESRD) in North-Eastern Bosnia. The aim of this paper was to analyze the patients with BEN diagnosis on chronic dialysis treatment in Bosnia and Herzegovina. METHODS: In this study we used data from individual questionnaires which we collected for Renal Registry. Individual questionnaires include: sex, age, place of birth and address, primary renal disease, data of the first dialysis treatment, type of dialysis, kidney transplantation, co-morbid diseases, erythropoietin therapy and outcome. For patients with BEN diagnosis we gathered additional data: history of urothelial tumor and family history of similar kidney diseases and renal replacement therapy. We compared these data with data about others dialysis patients in Bosnia and Herzegovina. STATISTICAL ANALYSIS: descriptive statistical analysis. RESULTS: Prevalence of the chronic dialysis patients in Bosnia and Herzegovina in 2003 was 474 pmp, 70 pmp for patients with BEN and 54 pmp for patients with diabetic nephropathy. In North-Eastern Bosnia prevalence of chronic dialysis patients was 844 and of patients with BEN 520 pmp. Incidence of the new chronic dialysis patients in Bosnia and Herzegovina in 2003 was 113 pmp, 11 pmp for BEN, and 19 pmp for diabetic nephropathy. Mortality of the chronic dialysis patients in Bosnia and Herzegovina in 2003 was 11.24 %, and mortality of the BEN patients 10.75 %. CONCLUSION: From the total number of the chronic dialysis patients in Bosnia and Herzegovina 14.7 % are BEN patients and 11.3 % are patients with diabetes. BEN is still big medical and social problem in Bosnia and Herzegovina, especially in the North-Eastern Bosnia. There are certain indicators that the incidence of the BEN patients is in decrease such as decreased difference between the prevalence of the patients with BEN and diabetic nephropathy; as well as increase of average age of patients with BEN.

Effect of target gene CpG content on spontaneous mutation in299 transgenic mice.

Transgenic mutation assays utilizing bacterial target genes display a high frequency of spontaneous mutation at CpG sequences. This is believed to result from the fact that: (1) the prokaryotic genes currently being used as transgenic mutation targets have a high CpG content and (2) these sequences are methylated by mammalian cells to produce 5-methylcytosine (5MC), a known promutagenic base. To study the effect of CpG content on the frequency and type of spontaneous mutation, we have synthesized an analogue of the bacterial lacI target gene (mrkII) that contains a reduced number of CpG sequences. This gene was inserted into a lambda vector and used to construct transgenic mice that undergo vector rescue from genomic DNA upon in vitro packaging. Results on spontaneous mutation frequency and spectrum have been collected and compared to those observed at the lacI gene in Big Blue transgenic mice. Spontaneous mutations at the mrkII gene occurred at a frequency in the mid-10-5 range and were predominantly base pair substitutions, similar to results seen in Big Blue. However, mrkII mutations were distributed toward the carboxyl end of the gene instead of the bias toward the amino terminus seen in lacI. Unexpectedly, 23% of the spontaneous mrkII mutations were GC-->AT transitions at CpG sequences (compared to 32% in lacI), despite the reduction in CpG number from 95 in lacI to only 13 in mrkII. Nine of the CpG bases undergoing transition mutations in mrkII have not been recorded previously as spontaneous sites in Big Blue. Therefore, substantial reduction of the number of CpG sequences in the lacI transgene did not significantly reduce the rate of spontaneous mutation or alter the contribution of CpG-related events. This suggests that other factors are also operating to establish frequency and composition of spontaneous mutations in transgenic targets.

Cancer patterns in three African populations compared with the United States black population.

Cancer incidence rates and patterns in three African populations in the Gambia, Mali and South Africa, have been compared with corresponding data on the Black population in Connecticut, USA. In the African populations, total rates for cancer are much lower than that of US Blacks, even allowing for under-reporting. Chief cancers are those of the oesophagus, liver and cervix. In Mali, stomach cancer is very common. As to trends, among South African Blacks, a population in transition, rates are rising, albeit slowly, of cancers of prosperity--prostate, lung, breast and colon-rectum. Salient questions are: can the number of cancers of underprivilege be lessened, and can cancers linked with rising socioeconomic states be restrained? Discussions of common risk factors, including diet, reproduction, smoking and drinking practices, indicate that for Africans as a whole, continuing poverty will prevent major changes in cancer pattern and rises in occurrence of the disease. However, should prosperity increase for Africans in big cities, rates are ultimately likely to attain those prevailing in the US Black population. Significant avoiding action seems almost impossible.

[A functional assessment of the masticatory muscles in mammals and humans].

We have amended and added to Fabian's tables giving a functional assessment of individual masticatory muscles. The data in our tables refer only to the temporalis, masseter, pterygoideus medialis, pterygoideus lateralis and digastricus muscles. The weights of these muscles were determined in three fixed human cadavers and the mean values compared with those of apes, carnivores, herbivores and rodents. In humans, the most powerful masticatory muscle is the M. temporalis, followed by the M. massater, as in apes and carnivores. The M. pterygoideus is also one of the most important. This is remarkable, since in the other groups this muscle occupies the last place. This relative strengthening of the M. pterygoideus lateralis is an important characteristic of the human masticatory apparatus. In humans, the difference between the relative weights of the individual masticatory muscles is not nearly so great as in other mammalian groups. The M. pterygoideus lateralis does lie close behind the other two big adductors (Mm temporalis and masseter) but, as regards power and weight, it hardly differs from the M. pterygoideus medialis and the M. digastricus. In humans the strengthening affects not only the M. pterygoideus lateralis but also the M. digastricus. It would seem that these two masticatory muscles could become the key to the understanding of the specific changes in human mastication.

Interaction between a DNA oligonucleotide and a dinuclear iron(II) supramolecular cylinder; an NMR and molecular dynamics study.

A tetracationic supramolecular cylinder, [Fe(2)L(3)](4+) (L=C(25)H(20)N(4)), with a triple-helical architecture, is just the right size to fit into the major groove of DNA but too big to fit into the minor groove. A detailed NMR spectroscopic analysis supported by molecular dynamics (MD) calculations shows unambiguously the close fit between the cylinder and a duplex oligonucleotide, [d(GACGGCCGTC)(2)]. Furthermore, only the left-handed enantiomer of the cylinder seems to fit the groove geometry. With both free and complexed species of [Fe(2)L(3)](4+) and DNA in solution, the NMR spectra are too complicated for a detailed structure determination. Based on differences in chemical shifts and extensive MD calculations, a realistic qualitative picture of the DNA-cylinder adduct is presented. Several sets of chemical shifts assigned to the protons of the three ligand strands in the cylinder indicate that the iron complex situated in the major groove exhibits restricted rotation on the NMR timescale around the cylindrical axis. The NMR NOE data support a model where the cylinder undergoes both a translational and rotational oscillation in the major groove. The results of an NOE restrained MD calculation indicates that the cylinder induces a 40 degrees bend of the double helix, in accordance with linear dichroism measurements. Other distinct features to be noticed are the very low value of the helical twist (16 degrees) induced at the G(4)C(5) step.

CGS 35601, a triple inhibitor of angiotensin converting enzyme, neutral endopeptidase and endothelin converting enzyme.

CGS 35601 (L-tryptophan, N-[[1-[[(2S)-2-mercapto-4-methyl-1-oxopentyl]amino]-cyclopentyl]carbonyl]) is one of a few single molecules capable of inhibiting the activities of angiotensin-converting enzyme (ACE), neutral endopeptidase (NEP) and endothelin converting enzyme (ECE) simultaneously, with IC(50) values of 22, 2, and 55 nM, respectively. Through the inhibition of ACE and ECE, it blocks the conversion of angiotensin I (AI) and big endothelin-1 (big ET-1) into the two most potent peptidic vasoconstrictors, angiotensin II (AII) and ET-1, respectively. By inhibiting NEP, CGS 35601 also prevents the degradation of peptidic vasodilators such as bradykinin (BK), natriuretic peptides (NPs) and adrenomedullin (ADM) and, hence, modulates the secondary release of other vasoactive mediators such as nitric oxide (NO) and prostaglandins. In chronic (30 days) experiments, CGS 35601 is well tolerated with a very good safety profile in healthy normotensive, hypertensive and type 2 diabetic rats. The antihypertensive efficacy of CGS 35601 was demonstrated in chronically instrumented, unrestrained and conscious rat models of hypertension (SHR and DSS) and type 2 diabetes (ZDF-fatty). It lowered blood pressure effectively as well as modulated plasma concentrations of a number of circulating vasoactive peptidic mediators that are keys to the regulation of the vascular tone. These data suggest that CGS 35601, a triple vasopeptidase inhibitor (VPI), may represent a novel class of antihypertensive drugs and may have the potential to reduce morbidity and mortality from cardiovascular disorders, diabetes and subsequent renal complications. Similar in vivo ACE, NEP, and ECE inhibitory activities were also observed with the orally active prodrug, CGS 37808 (L-tryptophan, N-[[1-[[(2S)-2-(acetylthio)-4-methyl-1-oxopentyl]amino]cyclopentyl]-carbonyl]-, methyl ester.

A 'big five' scoring system for the item pool of the adjective check list.

The item pool of the Adjective Check List (ACL; Gough & Heilbrun, 1980) is widely used as a means of capturing the personal characteristics associated with various target groups (e.g., women vs. men, young adults vs. old adults). The purpose of this research was to develop a system for scoring the ACL items in terms of the five-factor model of personality. In Study 1, five groups of introductory psychology students served as judges, with each group of approximately 100 persons rating the 300 ACL items for one of the five factors. The ratings of each factor were highly reliable. When corrected for favorability, the intercorrelations among the five factors were quite low, as expected, except for the positive correlation of Openness and Extraversion. Good convergence was found between our ratings and the indicative and counterindicative items identified by John's (1989) graduate student judges. In Study 2, convergent validity was demonstrated between the five-factor scores obtained from self-descriptive ACLs and corresponding factor scores obtained from Costa and McCrae's NEO-PI-R and NEO-FFI instruments (Costa & McCrae, 1992). Data from earlier cross-cultural studies of gender and age stereotypes were rescored using the new ACL-FF system to illustrate its potential utility as a research tool.

Novel neural adhesion molecules in the Contactin/F3 subgroup of the immunoglobulin superfamily: isolation and characterization of cDNAs from rat brain.

We cloned two kinds of cDNAs encoding novel Contactin/F3-subgroup adhesion molecules, which we termed NB-2 and NB-3. Nucleotide sequence analyses have shown that NB-2 and NB-3 are comprised of 1099 and 1028 amino acid residues, respectively. There was 51% similarity in the amino acid sequence of NB-2 and NB-3. NB-2 shared 46, 43, 55 and 55% identities with Contactin/F3, Tag-1, Big-1 and Big-2, respectively, at the amino acid sequence level. Likewise, the amino acid sequence of NB-3 exhibited 42, 44, 58 and 60% identities with Contactin/F3, Tag-1, Big-1 and Big-2, respectively. Expression of NB-2 mRNA was restricted to cerebrum, cerebellum and was hardly detectable, if any, in spinal cord. On the other hand, high expression of NB-3 mRNA was observed in spinal cord, as well as in cerebrum and cerebellum. In the other tissues, no expression of NB-2 and NB-3 mRNAs was detected.

Environmental impact of coal industry and thermal power plants in India.

Coal is the only natural resource and fossil fuel available in abundance in India. Consequently, it is used widely as a thermal energy source and also as fuel for thermal power plants producing electricity. India has about 90,000 MW installed capacity for electricity generation, of which more than 70% is produced by coal-based thermal power plants. Hydro-electricity contributes about 25%, and the remaining is mostly from nuclear power plants (NPPs). The problems associated with the use of coal are low calorific value and very high ash content. The ash content is as high as 55-60%, with an average value of about 35-40%. Further, most of the coal is located in the eastern parts of the country and requires transportation over long distances, mostly by trains, which run on diesel. About 70% oil is imported and is a big drain on India's hard currency. In the foreseeable future, there is no other option likely to be available, as the nuclear power programme envisages installing 20,000 MWe by the year 2020, when it will still be around 5% of the installed capacity. Hence, attempts are being made to reduce the adverse environmental and ecological impact of coal-fired power plants. The installed electricity generating capacity has to increase very rapidly (at present around 8-10% per annum), as India has one of the lowest per capita electricity consumptions. Therefore, the problems for the future are formidable from ecological, radio-ecological and pollution viewpoints. A similar situation exists in many developing countries of the region, including the People's Republic of China, where coal is used extensively. The paper highlights some of these problems with the data generated in the author's laboratory and gives a brief description of the solutions being attempted. The extent of global warming in this century will be determined by how developing countries like India manage their energy generation plans. Some of the recommendations have been implemented for new plants, and the situation in the new plants is much better. A few coal washeries have also been established. It will be quite some time before the steps to improve the environmental releases are implemented in older plants and several coal mines due to resource constraints.

[Physical development of schoolchildren in the city of Moscow during the recent decade and a related assessment].

Data on the long-term and short-term monitoring over the growth and development of schoolchildren in the city of Moscow during the second half of the last century are presented (including around 10,000 persons). According to the purpose-oriented 40-year observations of the growing generation in Moscow, the accelerated physical development started in the 60-ies with its peak falling on the middle 70-ies; the elements of deceleration set on in the 80-ies and continued through the 90-ies. As for now, a deficit of the body weight, reduced width and span parameters and a decreased power abilities as well as shifts of the menarche time in girls towards an older age are typical of contemporary schoolchildren in Moscow. The detected trends are common regularities typical of children in all Russia's big cities.

Duration selectivity organization in the inferior colliculus of the big brown bat, Eptesicus fuscus.

Duration selectivity of auditory neurons plays an important role in sound recognition. Previous studies show that GABA-mediated duration selectivity of neurons in the central nucleus of the inferior colliculus (IC) of many animal species behave as band-, short-, long- and all-pass filters to sound duration. The present study examines the organization of duration selectivity of IC neurons of the big brown bat, Eptesicus fuscus, in relation to graded spatial distribution of GABA(A) receptors, which are mostly distributed in the dorsomedial region of the IC but are sparsely distributed in the ventrolateral region. Duration selectivity of IC neuron is studied before and during iontophoretic application of GABA and its antagonist, bicuculline. Bicuculline application decreases and GABA application increases duration selectivity of IC neurons. Bicuculline application produces more pronounced broadening of the duration tuning curves of neurons at upper IC than at deeper IC but the opposite is observed during GABA application. The best duration of IC neurons progressively lengthens and duration selectivity decreases with recording depth both before and during drug application. As such, low best frequency neurons at upper IC have shorter best duration and sharper duration selectivity than high best frequency neurons in the deeper IC have. These data suggest that duration selectivity of IC neurons systematically varies with GABA(A) receptor distribution gradient within the IC.

A novel insertion mutation in spastin gene is the cause of spastic paraplegia in a Chinese family.

A total of eight loci for autosomal dominant hereditary spastic paraplegia (ADHSP) has been mapped to chromosome 14q, 2p, 15q, 8q, 10q, 12q, 19q, 2q, respectively, among which the SPG4 gene on chromosome 2p21-22 encoding spastin, an ATPase of the AAA family, accounts for 40-50% of all ADHSP families and is expressed in both adult and fetal tissues. In this work, we reveal a novel insertion mutation in exon 11 of the SPG4 gene found in a big Chinese family composed of 47 members, including 20 affected ones, using linkage analysis. The mutation was well demonstrated to be the cause of loss of production of the functional protein by pre-termination of translation in AAA cassette region. To our knowledge, this is the first report of spastin mutation in China.

Experience-dependent corticofugal adjustment of midbrain frequency map in bat auditory system.

Recent studies of corticofugal modulation of auditory information processing indicate that cortical neurons mediate both a highly focused positive feedback to subcortical neurons "matched" in tuning to a particular acoustic parameter and a widespread lateral inhibition to "unmatched" subcortical neurons. This cortical function for the adjustment and improvement of subcortical information processing is called egocentric selection. Egocentric selection enhances the neural representation of frequently occurring signals in the central auditory system. For our present studies performed with the big brown bat (Eptesicus fuscus), we hypothesized that egocentric selection adjusts the frequency map of the inferior colliculus (IC) according to auditory experience based on associative learning. To test this hypothesis, we delivered acoustic stimuli paired with electric leg stimulation to the bat, because such paired stimuli allowed the animal to learn that the acoustic stimulus was behaviorally important and to make behavioral and neural adjustments based on the acquired importance of the acoustic stimulus. We found that acoustic stimulation alone evokes a change in the frequency map of the IC; that this change in the IC becomes greater when the acoustic stimulation is made behaviorally relevant by pairing it with electrical stimulation; that the collicular change is mediated by the corticofugal system; and that the IC itself can sustain the change evoked by the corticofugal system for some time. Our data support the hypothesis.

Diuretic-natriuretic actions and pressor effects of big-endothelin (1-39) in phosphoramidon-treated rats.

The effects of phosphoramidon, a metalloproteinase inhibitor, on the pressor and renal actions of big-endothelin (BET), the precursor of porcine Endothelin-1 (ET), was studied in rats. In control rats, BET (0.3, 1.0, and 3.0 nmol/kg) elicited a marked increase in mean arterial blood pressure (from 110 +/- 7 to 105 +/- 7, 120 +/- 8, 147 +/- 6 mm Hg, respectively), and a prominent, dose-dependent, diuretic and natriuretic response (fractional sodium excretion (FENa) increased from 0.4 +/- 0.2 to 0.8 +/- 0.2, 3.1 +/- 0.1, and 8.5 +/- 1.7%, respectively). Pretreatment with phosphoramidon (10 mg/kg + 0.25 mg/kg/min) completely abolished the increase in blood pressure induced by BET, but the diuretic-natriuretic effects were only partially inhibited (FENa increased from 2.0 +/- 0.9 to 3.7 +/- 1.5, 3.9 +/- 1.3, and 4.3 +/- 1.2%, respectively, P < 0.05). Rats treated with phosphoramidon only had no natriuresis over time (FENa changed from 1.9 +/- 0.5 to 2.3 +/- 0.3, 1.6 +/- 0.4, 1.7 +/- 0.6 respectively, P--NS). The data suggest that, unlike the vascular type of the enzyme, the renal endothelin converting enzyme is relatively insensitive to phosphoramidon. Further, diuresis and natriuresis can be induced by BET in the absence of any pressor effect.

[Dynamic study of hepatocellular pathological change and uptake rate of 125I-insulin during experimental hepatocarcinogenesis].

This study was designed to observe the hepatocellular pathological change and uptake rate of 125I-insulin during experimental hepatocarcinogenesis in rats and clarify the possible mechanism of increasing uptake of 125I-insulin. 80 SD rats were divided into 2 groups, control and experimental groups. All of the rats were given common feed, and the rats of experimental group were given extra diethylnitrosamine (DENA) 70 mg/kg each week. At 6th, 11th, 15th and 20th week after start of the experiment, 10 control and 10 experimental rats were killed 1 hour post administration of 125I-insulin by tail vein. Blood, liver, lung, spleen, kidney, muscle and bone were collected; the radioactivity was measured and calculated %ID/g. The data of 2 groups were compared and examined with t-test. All of livers were pathologically examined. The results showed all livers of control group were normal. At 6th week, the surfaces of experimental rats' livers were coarse. Hepatocellular hyperplasia was observed, 125I-insulin-uptake rate was 1.86 time as much as that of control group. At 11th week, the liver's colour became lighter than that of control group. Hyperplasia and hepatocirrhosis were observed, 125I-insulin-uptake rate was 1.50 time as much as that of control group. At 15th week, hyperplastic nodules were observed in all experimental rats' livers. Hepatomacellulae were observed in 6 rat livers. 125I-insulin-uptake rate was 1.56 time as much as that of control group. At 20th week, the livers became enlarged out of shape. There were a lot of big or small greyish white nodules in all livers. Necrosis, liquefaction and hemorrhage were observed. Hepatomacellulae were observed in all of experimental rats livers. 125I-insulin-uptake rate was 1.46 time as much as that of control group. The differences of 125I-insulin-uptake rate between experimental and control groups were significant. These results demonstrated that the liver ability of uptaking 125I-insulin increased which mainly took place during the period of hepatocellular hyperplasia, and the hepatomacellulae kept this characteristic.

Mutation assays in male germ cells from transgenic mice: overview of study and conclusions.

Three confirmed mouse germ cell mutagens, ethyl nitrosourea (ENU), isopropyl methanesulphonate (iPMS) and methyl methanesulphonate (MMS), have been evaluated for their activity as mutagens to the germ cell DNA of two strains of transgenic mice (lac I, Big Blue and LacZ, Muta Mouse). Both testicular DNA and epididymal sperm DNA were evaluated. A range of sampling times was studied, from 3 days post-dosing to 100 days post-dosing. ENU and iPMS were mutagenic to both testicular DNA and epididymal sperm DNA. Mutant frequencies were higher for both chemicals in DNA recovered from testicular tissue than in epididymal sperm DNA. Likewise, mutant frequencies were higher for both DNA samples at the later sampling times. MMS was not mutagenic under any condition of test. A good level of qualitative agreement in test results was seen for the two assays and for the same assays conducted in different laboratories. The level of quantitative agreement was not as high, but was, nonetheless, generally good. Recommendations for the future conduct of transgenic rodent germ cell mutation assays are made. The test data are discussed within the context of the larger question of how such assays should be integrated into the chemical hazard assessment process.
