Chloride-bicarbonate exchange through the human red cell ghost membrane monitored by the fluorescent probe 6-methoxy-N-(3-sulfopropyl)quinolinium.

The exchange of chloride and bicarbonate across the human red cell membrane has been characterized by quenching of an intracellular fluorophore, 6-methoxy-N-(3-sulfopropyl)quinolinium (SPQ). In 20 mM Hepes, pH 7.4, and varying concentrations of chloride and bicarbonate with total anion concentration of 150 mM, SPQ is quenched dynamically by chloride with an apparent Stern-Volmer quenching constant 0.071 +/- 0.016 mM-1 at 25 degrees C. Hepes alone quenched SPQ fluorescence with a quenching constant of 0.030 +/- 0.003 mM-1. Stopped-flow kinetic experiments give fluorescence time courses that, when corrected for Stern-Volmer quenching, are first order. Disulfonic stilbenes (inhibitors of anion exchange) decrease the rate of fluorescence equilibration. Transport of bicarbonate via hydration-dehydration of CO2 does not contribute to the observed kinetics. The chloride-bicarbonate exchange rate is independent of the anion concentration gradient, but increases at 25 degrees C from 1 to 4 s-1 as equilibrium chloride concentration increases from 20 to 130 mM (with concomitant decrease in bicarbonate concentration from 130 to 20 mM). The data indicate that the translocation rate of the chloride-loaded transport protein is greater than that of bicarbonate-loaded transport protein and that bicarbonate has a higher affinity for the transport protein than chloride. Our results validate the use of SPQ to measure quantitatively chloride-bicarbonate exchange in red cell ghosts. The methods we describe should be applicable to other systems exhibiting chloride-bicarbonate exchange.

Improved radiolabeled substrates for soluble epoxide hydrolase.

Two rapid assays for the soluble epoxide hydrolase (sEH) are described. First, a sensitive radiometric assay based on thin-layer chromatography of [(14)C]-cis-9,10-epoxystearic acid and its corresponding diol ((14)C]-9,10-dihydroxystearic acid) is described. The cis fatty acid oxide exhibits higher specific activity of hydration with sEH from mouse, rat, human, and potato compared to trans-stilbene oxide (TSO). The K(m) and V(max) obtained for [(14)C]-cis-9,10-epoxystearic acid with mouse sEH are 11.0 microM and 3460 nmol/min/mg protein, respectively. [(14)C]-cis-9,10- Epoxystearic acid might more closely mimic the structures of natural substrates for sEH. Second, [2-(3)H]-trans-1,3-diphenyl-propene oxide ([(3)H]-tDPPO) and [2-(3)H]-cis-1,3-diphenylpropene oxide ([(3)H]-cDPPO) were synthesized and rapid radiometric assays for epoxide hydrolases (EHs) were developed by differential partitioning of the epoxide into iso-octane and its corresponding diol into aqueous phase containing methanol. It was shown that sEHs from mouse, rat, human, and potato rapidly hydrolyze [(3)H]-tDPPO and in comparison to TSO have 20-,49-,28-, and 7-fold higher rates, respectively. Mouse sEH hydrates [(3)H]-tDPPO at 26,200 nmol/min/mg protein, and a K(m)p4 of 2.80 microM is observed.

Characterization of a Cl(-)-channel from rabbit transverse tubules in the planar lipid bilayer system.

A Cl(-)-channel of the transverse tubule(TT) was incorporated into artificial planar lipid bilayers and its properties were investigated. The channel was found to have the following properties. (1) The single-channel conductance is 40 pS in choline-Cl solution. (2) The rate of gating of the channel does not depend on the membrane voltage. (3) The channel is blocked by stilbene derivatives (DIDS and SITS), which are known as inhibitors of voltage-dependent Cl(-)-channels of the Torpedo electric organ, from both sides of the membrane. (4) An inhibitor of voltage-dependent Cl(-)-channels of skeletal muscle, 9-anthracene carboxylic acid(9-AC), inhibits Cl(-)-current from the cis-side to which the TT vesicles were added.

Microsomal glutathione S-transferase (GST) isozymes in Aspergillus ochraceus TS: induction by 3-methylcholanthrene.

Induction of microsomal glutathione S-transferase isozymes in Aspergillus ochraceus TS is reported. Among the inducers of mammalian drug-metabolising enzymes 3-methylcholanthrene, benzo(a)pyrene and polychlorinated biphenyl enhanced the microsomal GST activity quite considerably. On the other hand phenobarbital, beta-naphthoflavone and trans-stilbene oxide were almost without any effect. To our knowledge this seems to be the first report on induction of microsomal GST in lower eukaryotes. A significant increase (7.9 fold) in GST activity was observed with the cells induced by 3-methylcholanthrene. Moreover, three new GST isozymes were found to be present in 3-methylcholanthrene induced microsomes. The isozymes are basic in nature. The GST isozymes were found to be catalytically distinct from each other.

DIDS binding 30-kDa protein regulates the calcium release channel in the sarcoplasmic reticulum.

The gating properties of the Ca2+ release channel in the heavy fraction of the sarcoplasmic reticulum (HSR) was monitored by measuring the choline permeation through the channel using a light scattering method. The choline permeation was increased by the treatment of the HSR vesicles with 4,4'-diisothiocyano-stilbene-2,2'-disulfonic acid (DIDS), and this effect was only observed at low extravesicular Ca2+ concentrations. This result indicates that DIDS locked the Ca2+ channel at the open state. An SDS-PAGE of the junctional face membrane (JFM) that was treated with 3H2-DIDS showed that 3H2-DIDS binding protein is not the Ca2+ release channel but the 30 kDa protein. Furthermore, it was found that this 30 kDa protein is also one of the calsequestrin interacting proteins. These results suggest that this 30 kDa protein regulates the Ca2+ release channel.

1 alpha,25(OH)2 vitamin D3-mediated stimulation of outward anionic currents in osteoblast-like ROS 17/2.8 cells.

1 alpha,25-dihydroxyvitamin-D3 [1 alpha,25(OH)2D3] is known to potentiate in osteoblast-like rat osteosarcoma cells (ROS 17/2.8) the influx of Ca2+ through voltage-activated L-type Ca2+ channels. In the present study, by means of the whole-cell patch-clamp technique, we found that physiological concentrations of 1 alpha,25(OH)2D3 (5-5000 x 10(-11) M) also stimulate, in a dose-dependent manner, outwardly rectifying anionic currents over the course of 1-5 minutes. These currents, recorded in the presence of 130 mM glutamate or approximately 140 mM C1- in the external solution, were blocked by 200 microM DIDS, a stilbene derivative selective for C1- channels. We conclude that, in addition to the already known effects of 1 alpha,25(OH)2D3 on Ca2+ channels, the hormone also enhances in ROS 17/2.8 cells an outward anion conductance. This is the first report concerning activation of anion channels by 1 alpha,25(OH)2D3.

Specific interaction of 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid with human cellular CD4.

We recently demonstrated that 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS) prevented the infection of T cells by human immunodeficiency virus type-1 (Cardin et al., J. Biol. Chem. 266, 13355, 1991). In the present study we have used a panel of monoclonal antibodies (mabs) against a variety of human leukocyte antigens to characterize the interaction of DIDS by flow cytometry with various T cell surface molecules. DIDS blocked the specific immunoreactivity of mabs OKT4A and Leu 3A with CD4 on human leukemic T cells (JM) and human mononuclear lymphocytes with an IC50 approximately 30 microM. The membrane distal (D1) and proximal (D3 and D4) domains of CD4 remained blocked for up to 5 hr of culture and returned to control levels of expression after 24 hr, reflecting the rate of membrane turnover of the CD4-DIDS complex. The binding frequencies (% positive) for anti-CD2, -CD3, -CD5, -CD6, -CD7, -CD8, -CD11a, -CD14, -CD18, -CD19, -CD45, -T cell receptor, and -HLA-DR were not significantly affected. However, there was a partial reduction in the antigen density of CD2, CD5, CD8, and CD11b. The selective interaction of DIDS with CD4 suggests that antagonism of the virus receptor may account in part for the antiviral properties of the stilbene disulfonate.

ATP-activated cationic current in rabbit sino-atrial node cells.

The electrophysiological effect of extracellular ATP was investigated in voltage-clamped single cells from the rabbit sino-atrial node. External 0.5 mM ATP consistently activated a time-independent, weakly inwardly rectifying current (2.26 +/- 0.14 pA/pF at -100 mV, n = 23). This external ATP-activated current increased in a dose-dependent manner over the concentration range 0.01 to 1 mM ATP. However, non-hydrolysable ATP analogs (a, beta-methylene ATP and beta,r-methylene ATP) failed to activate this current. Moreover, adenosine, adenosine monophosphate and adenosine diphosphate were all ineffective in eliciting the ATP-activated current over the concentration range 0.5 to 1 mM, suggesting that it is regulated via the P2-purinoreceptor. The permeability sequence for the ATP-activated current among different cations (determined from reversal potential measurements) was 1.09: 1: 1: 0.24: 0.22 for K+, Na+, Cs+, Tris and N-methyl-D-glucamine. The current component was not affected by substitution of external Cl- by aspartate or by the application of a stilbene-derivative Cl- channel blocker. Thus, the ATP-activated current in rabbit sino-atrial node cells was non-selective for monovalent cations, and could be carried by large molecular ions, such as N-methyl-D-glucamine or Tris. We suggest that it might make some contribution to the chronotropic effects seen on release of transmitters from autonomic nerve endings in the heart.

Inhibition of bicarbonate transport protects embryonic heart against reoxygenation-induced dysfunction.

It has not been well established whether the mechanisms participating in pH regulation in the anoxic-reoxygenated developing myocardium resemble those operating in the adult. We have specially examined the importance of Na+/H+ exchange (NHE) and HCO3-dependent transports in cardiac activity after changes in extracellular pH (pHo). Spontaneously contracting hearts isolated from 4-day-old chick embryos were submitted to single or repeated anoxia (1 min) followed by reoxygenation (10 min). The chronotropic, dromotropic and inotropic responses of the hearts were determined in standard HCO3- buffer at pHo 7.4 and at pHo 6.5 (hypercapnic acidosis). In distinct experiments, acidotic anoxia preceded reoxygenation at pHo 7.4. NHE was blocked with amiloride derivative HMA (1 micro mol/l) and HCO3-dependent transports were inactivated by replacement of HCO3 or blockade with stilbene derivative DIDS (100 micro mol/l). Anoxia caused transient tachycardia, depressed mechanical function and induced contracture. Reoxygenation temporarily provoked cardiac arrest, atrio-ventricular (AV) block, arrhythmias and depression of contractility. Addition of DIDS or substitution of HCO3 at pHo 7.4 had the same effects as acidosis per se, i.e. shortened contractile activity and increased incidence of arrhythmias during anoxia, prolonged cardioplegia and provoked arrhythmias at reoxygenation. Under anoxia at pHo 6.5/reoxygenation at pHo 7.4, cardioplegia, AV block and arrhythmias were all markedly prolonged. Interestingly, in the latter protocol, DIDS suppressed AV block and arrhythmias during reoxygenation, whereas HMA had no effect. Thus, intracellular pH regulation in the anoxic-reoxygenated embryonic heart appears to depend predominantly on HCO3 availability and transport. Furthermore, pharmacological inhibition of anion transport can protect against reoxygenation-induced dysfunction.

Inorganic mercury absorption in mature and immature rat jejunum: transcellular and intercellular pathways in vivo and in everted sacs.

The objective of the present work was to test the hypothesis that toxic and nonessential heavy metals in general, and inorganic Hg in particular, are taken up in rat jejunum by a mechanism similar to that previously proposed for Cd. The initiating step in Cd absorption involves electrostatic interaction of the cation with negative membrane charges. In contrast, Hg appears to react with apical membranes as an anion, although overall uptake is not inhibited by the stilbene inhibitor of anion transport DIDS. Further, the ratio of Cd/Hg uptake falls along the jejunum. However, several similarities between Cd and Hg uptakes were observed. Neither process depends on reactive sulfhydryl groups or on oxidative metabolism. Although tissue Hg, unlike Cd, could not be rigorously separated into membrane-bound and intracellular compartments, uptake of both metals includes their relatively temperature-insensitive and rapid influx into a pool readily accessible to suitable extracellular chelators. Another more temperature-sensitive and slower component leads to the filling of a relatively chelation-resistant compartment, identified in the case of Cd as intracellular. Non-specific membrane properties such as surface charge and perhaps membrane fluidity can thus account for mucosal Cd and Hg uptakes; no contribution of specific transport systems need be postulated. Mucosal permeability, calculated in reference to ethanol uptake as measure of absorbing surface area, falls to the same extent for Cd and Hg during postweaning development. Because this fall, however, parallels a diminution in polar diffusion pathways, the mechanism of the faster metal absorption in weanlings may have little bearing on the slower transcellular process in adults. Incidentally, polar diffusion pathways increase in mature jejunum upon preparation of everted sacs, a further limitation on the usefulness of this preparation for the quantitative study of normal intestinal solute transport.

Factors affecting the intracellular generation of free radicals from quinones.

Isolated hepatocytes do not liberate appreciable amounts of superoxide into the external medium. Simple quinones stimulate the release of superoxide up to 15 nmol/min/10(6) hepatocytes. Superoxide release stimulated by a variety of simple quinones and more complex antitumor quinones was maximal at a quinone one-electron reduction potential of -70 mV. This was qualitatively similar to the pattern of superoxide formation seen with NADH-cytochrome b5 reductase and NADH: ubiquinone oxidoreductase. Superoxide production by NADPH-cytochrome P-450 reductase was maximal at a quinone single-electron reduction potential at -200 mV. Phenobarbital pretreatment had no effect on superoxide formation by hepatocytes suggesting that NADPH-cytochrome P-450 reductase activity is not rate limiting for quinone stimulated superoxide formation. Sulfonated stilbenes, specific inhibitors of anion exchange, had no effect on the release of superoxide by hepatocytes suggesting that superoxide is not transported through anion channels in the plasma membrane. Pretreatment of hepatocytes with 10(-5) M diethyldithiocarbamate produced over a two fold increase in the release of superoxide.

Organ specificity of induction of activating and inactivating enzymes by cigarette smoke and cigarette smoke condensate.

Inhalation of cigarette smoke specifically induces the rat lung and kidney aryl hydrocarbon hydroxylase (AHH) in less than 4 h. The epoxide hydratase (EH) and the glutathione S-transferase are not significantly modified by a similar treatment in any of the rat tissues. Compared to the kidney AHH, the lung hydroxylase is 3--4 times more sensitive to small concentrations of cigarette smoke and seems to have a longer biological half-life. In both tissues, the induced AHH presents the same in vitro sensitivity to various inhibitors as a polycyclic hydrocarbon induced AHH. In primary fetal rat liver cell culture, the cigarette smoke condensate fractions (CSCF) induce both the AHH and EH activity. Nevertheless, the AHH activity responds faster and to lower concentrations of CSCF than the EH activity. The liver cell culture constitutes a unique tool for a comparative study of the AHH and EH induction mechanism. Low concentration (10 muM) of benz(a)anthracene induces only the AHH activity while trans-stilbene oxide enhances selectively the EH activity. Appropriate concentrations of CSCF or of phenobarbital (PB) determine a parallel induction of both enzymes. The results are discussed on the basis of (a) the existence of specific mechanisms of AHH regulation in the lung and in the kidney and (b) the existence of coordinated or independent biochemical control of the AHH and EH activity.

Role of extent and persistence of DNA modifications in chemical carcinogenesis by aromatic amines.

The initial step in tumor formation with aromatic amines is assumed to involve the modification of nucleic acids. Reactive metabolites bind covalently to purine and pyrimidine bases. The extent of formation of DNA adducts and their persistence is therefore expected to correlate with the biological effect. Using [3H]trans-4-amino-stilbene derivatives as model compounds, several parameters have been measured in susceptible and nonsusceptible rat tissues: total initial DNA binding, initial pattern of adducts, persistence of DNA adducts, accumulation of DNA binding after repeated doses, and persistence of DNA adducts after repeated doses. They did not correlate with tissue susceptibility. The tissue burden decreased in the order: liver greater than kidney greater than lung greater than glandular stomach greater than Zymbal gland. The latter tissue is the primary target for tumor formation. Comparison with other aromatic amines shows that aminostilbene derivatives are no exceptions. The role of specific nucleic acid modifications for mutagenic and carcinogenic effects is also difficult to evaluate in these cases. It is therefore emphasized that modulation of secondary steps in the multistage process may largely influence the final outcome. The role of cell proliferation and its stimulation as well as tumor promoting effects are discussed. Aminostilbene derivatives appear to produce primary lesions quite efficiently in rat liver but lack cytotoxic and promoting properties for this tissue.

Chalcone synthase-like genes active during corolla development are differentially expressed and encode enzymes with different catalytic properties in Gerbera hybrida (Asteraceae).

Recent studies on chalcone synthase (CHS) and the related stilbene synthase (STS) suggest that the structure of chs-like genes in plants has evolved into different forms, whose members have both different regulation and capacity to code for different but related enzymatic activities. We have studied the diversity of chs-like genes by analysing the structure, expression patterns and catalytic properties of the corresponding enzymes of three genes that are active during corolla development in Gerbera hybrida. The expression patterns demonstrate that chs-like genes are representatives of three distinct genetic programmes that are active during organ differentiation in gerbera. Gchs1 and gchs3 code for typical CHS enzymes, and their gene expression pattern temporally correlates with flavonol (gchs1, gchs3) and anthocyanin (gchs1) synthesis during corolla development. Gchs2 is different. The expression pattern does not correlate with the pigmentation pattern, the amino acid sequence deviates considerably from the consensus of typical CHSs, and the catalytic properties are different. The data indicate that it represents a new member in the large superfamily of chs and chs-related genes.

Phenylephrine and ATP enhance an amiloride insensitive bicarbonate-dependent alkalinizing mechanism in rat single cardiomyocytes.

To expel the excess protons generated during a cellular acidification and to fully recover basal intracellular pH (pHi), cardiac cells rely on the amiloride-sensitive Na/H antiport. We report that rat single ventricular cardiomyocytes, loaded with the fluorescent pH indicator Snarf-1 and treated with inhibitors of the Na/H antiport, amiloride or its analogues, partially restored their pHi through a bicarbonate-dependent mechanism following an acidosis (imposed by the ammonia-pulse technique). In the presence of ethylisopropylamiloride (10 microM) or amiloride (1 mM) and 25 mM bicarbonate in the extracellular solution, the average time that cells needed to recover half of their pHi, following the removal of 20 mM NH4Cl, was 3.4 min, while the rate of proton efflux was calculated to be 2.0 mM/min. The stilbene derivative, 4-4'-di-isothiocyanostilbene-2,2'-disulphonate (DIDS 200 microM), a known blocker of anion transporters, inhibited this recovery. Both phenylephrine (100 microM, 3 microM propranolol present), an alpha 1-adrenoceptor agonist, and ATP (10 microM), a purinergic agonist, significantly enhanced the rate of proton efflux that was due to this HCO3-dependent alkalinizing mechanism. Phenylephrine and ATP also shortened by three-fold the time that a myocyte needed to recover half of its initial pHi. This bicarbonate-dependent alkalinizing mechanism could provide an additional means by which cardiac cells recover their pHi from acidosis, especially under conditions in which the Na/H antiport is inhibited. Furthermore, catecholamines and ATP, which are released under various pathophysiological conditions often associated with intracellular acidosis, could play an important role in the modulation of pHi under these conditions.

Mutant genes of cytochrome P-450IID6, glutathione S-transferase class Mu, and arylamine N-acetyltransferase in lung cancer patients.

Epidemiological studies suggested a protective effect of certain phenotypes of polymorphic foreign-compound-metabolizing enzymes in some types of cancer. Poor metabolizers (PM) of debrisoquine 4-hydroxylase (cytochrome P-450IID6, CYP2D6) were found to be underrepresented among patients with lung cancer. Recent advances in molecular genetic characterization of CYP2D6, glutathione S-transferase (GST) class Mu, and arylamine N-acetyltransferase enabled genotypical determination of mutant alleles in lung cancer patients. Restriction fragment length polymorphism (RFLP) with a cDNA gene probe of CYP2D6 was analyzed in 79 lung cancer patients who were phenotyped with debrisoquine. Mutant alleles were detected by allele-specific polymerase chain reaction (PCR). In the same individuals, genotype of GST class Mu was analyzed by PCR and correlated with ex vivo activity of glutathione conjugation towards trans-stilbene oxide. RFLP patterns allowed discrimination between the slow and fast genotype of N-acetyltransferase as well as the heterozygotes. Three phenotypical PMs of debrisoquine (3.8%) were confirmed by PCR and RFLP. No PM could be unambiguously recognized only by RFLP patterns. The PMs were characterized by PCR and RFLP as carriers of the 29B/29B (n = 1), 29A/29B (n = 1), and 29A/44 (n = 1) mutant alleles. Higher debrisoquine hydroxylase activities were found in the homozygous EMs, who possess two active genes, as compared to heterozygous EMs, who have only one active gene. The patients with phenotypically impaired GST Mu activity were confirmed as such by PCR. A complete correspondence between phenotyping of N-acetyltransferase (with caffeine) and genotyping was found. The new genetic techniques proved to be powerful tools for molecular-epidemiological studies aimed at establishing host factors of cancer susceptibility.

The mycotoxin ochratoxin A deranges pH homeostasis in Madin-Darby canine kidney cells.

Ochratoxin A (OTA) is a nephrotoxin which blocks plasma membrane anion conductance in Madin-Darby canine kidney (MDCK) cells. Added to the culture medium, OTA transforms MDCK cells in a manner similar to exposure to alkaline stress. By means of video-imaging and microelectrode techniques, we investigated whether OTA (1 mumol/liter) affects intracellular pH (pHi), Cl- (Cl-i) or cell volume of MDCK cells acutely exposed to normal (pHnorm = 7.4) and alkaline (pHalk = 7.7) conditions. At pHnorm, OTA increased Cl-i by 2.6 +/- 0.4 mmol/liter (n = 14, P < 0.05) but had no effect on pHi. At pHalk, application of OTA increased Cl-i by 8.6 +/- 2.6 mmol/liter (n = 10, P < 0.05) and raised pHi by 0.11 +/- 0.03 (n = 8, P < 0.05). The Cl-/HCO3- exchange inhibitor DNDS (4,4'-dinitro-stilbene-2,2'-disulfonate; 10 mumol/liter) eliminated the OTA-induced changes of pHi and Cl-i. OTA did not affect cell volume under both pHnorm and pHalk conditions. We conclude that the OTA-induced blockade of plasma membrane anion conductance increases Cl-i without changing cell volume. The driving force of plasma membrane Cl-/HCO3- exchange dissipates, leading to a rise of pHi when cells are exposed to an acute alkaline load. Thus, OTA interferes with pHi and Cl-i homeostasis leading to morphological and functional alterations in MDCK cells.

Band 3 antagonists, p-azidobenzylphlorizin and DIDS, mediate erythrocyte shape and flexibility changes as characterized by digital image morphometry and microfiltration.

Two nonpenetrating membrane probes, p-azidobenzylphlorizin (p-AzBPhz) and 4,4'-diisothiocyano-2,2'-stilbene disulfonate (DIDS), have been shown in earlier studies to induce dose-dependent changes in red blood cell (RBC) shape and volume at the same low concentrations that inhibit anion transport. In the present work, these ligand-induced morphology and rheology changes were studied using video digital image morphometry (VDIM) and microfiltration techniques. The results of these experiments corroborate our earlier investigation. RBCs were filmed using a Nomarski optics microscope with video camera attachment and cell size and shape changes were computer analyzed using VDIM. Low microM p-AzBPhz or DIDS levels caused collapse of the cell's biconcave structure and cell flattening occurred within 1-2 sec after drug exposure. Higher doses of either agent converted cells to a new steady-state in which a concurrent limited increase in erythrocyte volume and blunt membrane protrusions were produced. These changes were reversed in less than 2 sec by washing the drug from the membrane. Both ligands increased the deformability of RBCs in a dose-dependent manner as determined by filtration through Nuclepore polycarbonate filters (3 microns pore diameter). The improvement in deformability of drug-treated sickle cells was much more dramatic than for normal cells at low p-AzBPhz concentrations. These results support our earlier conclusions that the ligands, through a common interaction with band 3, induce volume-associated cytoskeletal alterations which lead to changes in morphology and flexibility.
