Some statistical properties of the miniature endplate potentials.

I will demonstrate that series of miniature endplate potentials (m.e.p.ps) showing a high proportion of so-called giant m.e.p.ps (g.m.e.p.ps) have different statistical structures from series where the proportion of g.m.e.p.ps is low. The nature of the different structures will be discussed on the basis of two statistical models, one for the distribution of the m.e.p.p. amplitudes and one for the series of point events occurring in time.

Inhibitory miniature synaptic potentials in rat motoneurons.

In the newborn rat spinal cord, spontaneous potentials were recorded, with KCl electrodes, from motoneurons in the presence of tetrodotoxin (10(-6) g ml-1) to abolish nerve impulses. These potentials occurred at low frequencies (less than 2 Hz), and their mean amplitude was a fraction of 1 mV. An increase of osmolarity with sucrose or an increase of extracellular K+, increased the frequency of miniature synaptic potentials. The amplitude of the spontaneous potentials was increased by intracellular injection of Cl-. Strychnine (2-25 microM) completely abolished the spontaneous potentials. It is suggested that these potentials are produced by the spontaneous release of packages of inhibitory transmitter at synapses on motoneurons.

Conformal miniaturization of domains with low domain-wall energy: monoclinic ferroelectric states near the morphotropic phase boundaries.

A theory is developed for intermediate monoclinic (FE(m)) phases near morphotropic phase boundaries in ferroelectrics of complex oxides. It is based on the conformal miniaturization of stress-accommodating tetragonal domains under the condition of low domain-wall energy density. The microdomain-averaged lattice parameters are determined and attributed to the parameters of an adaptive monoclinic phase. The theory is applied to the temperature, electric field, and compositional dependent FE(m) lattice parameters. The predictions of the theory are rigidly obeyed over the entire FE(m) stability range.

Insertion preference of maize and rice miniature inverted repeat transposable elements as revealed by the analysis of nested elements.

A 128-bp insertion into the maize waxy-B2 allele led to the discovery of Tourist, a family of miniature inverted repeat transposable elements (MITEs). As a special category of nonautonomous elements, MITEs are distinguished by their high copy number, small size, and close association with plant genes. In maize, some Tourist elements (named Tourist-Zm) are present as adjacent or nested insertions. To determine whether the formation of multimers is a common feature of MITEs, we performed a more thorough survey, including an estimation of the proportion of multimers, with 30.2 Mb of publicly available rice genome sequence. Among the 6600 MITEs identified, >10% were present as multimers. The proportion of multimers differs for different MITE families. For some MITE families, a high frequency of self-insertions was found. The fact that all 340 multimers are unique indicates that the multimers are not capable of further amplification.

The Maize Invertase-Deficient miniature-1 Seed Mutation Is Associated with Aberrant Pedicel and Endosperm Development.

Genetic evidence is presented to show that the developmental stability of maternal cells in the pedicel at the base of maize seeds is determined by the genotype of the developing endosperm. An early degeneration and withdrawal of maternal cells from the endosperm of homozygous miniature (mn mn) seed mutants were arrested if mn plants were pollinated by the wild-type Mn pollen. Similarly, the stability of the wild-type, Mn mn, maternal cells was also dependent on whether or not these cells were associated with the normal (Mn) or the mutant (mn) endosperm on the same ear. Biochemical and cellular analyses indicated that developing mn kernels have extremely low (<0.5% of the wild type) to undetectable levels of both soluble and wall-bound invertase activities. Extracts from endosperm with a single copy of the Mn gene showed a significant increase in both forms of invertases, and we suggest it is the causal basis of the wild-type seed phenotype. Collectively, these data provide evidence that invertase-mediated maintenance of a physiological gradient of photosynthate between pedicel and endosperm constitutes the rate-limiting step in structural stability of maternal cells as well as normal development of endosperm and seed.

Long-term biocompatibility of a miniature stimulator implanted in feline hind limb muscles.

Chronic foreign-body responses and muscular changes were examined following the implantation of active miniature stimulators into the hind limb muscles of cats for periods of up to three months. The radio-frequency (RF)-powered stimulators were injected into muscles through a 12-gauge hypodermic needle. The tissue responses around the active stimulators were compared histologically to those provoked by passive devices, broken glass, silicone tubing, polyester suture material coated with polybutylate, and two of the internal components of the stimulator (ferrite, integrated circuit chip). Active and passive stimulators produced similar, benign foreign-body reactions that resulted in an essentially identical fibrous capsule over time. The responses were similar to those produced by the internal components and the suture material, and were more modest than those produced by the broken glass. The capsule did not appear to interfere with the functionality of active devices because thresholds measured during the post-implantation survival period did not change significantly over time. Unexpectedly, the severity of the reaction differed significantly amongst the various target muscles. Medial gastrocnemius exhibited the most severe response, whereas tibialis anterior had the least reaction.

Effects of regional stimulation using a miniature stimulator implanted in feline posterior biceps femoris.

The effects of placement of a miniature implantable stimulator on motor unit recruitment were examined in the posterior head of cat biceps femoris. The implantable stimulator (13-mm long x 2-mm diameter) was injected either proximally near the main nerve branch, or distally near the muscle insertion, through a 12-gauge hypodermic needle. Glycogen-depletion methods were used to map the distribution of fibers activated by electrical stimulation. Muscle fibers were found to be depleted at most or all proximodistal levels of the muscle, but the density of depleted fibers varied transversely according to the stimulus strength and proximity of the device to the nerve-entry site. Thus, muscle cross sections often had a "patchy" appearance produced because different proportions of depleted fibers intermingled with undepleted fibers in different parts of the cross section. In other preparations, the force of muscle contraction was measured when stimuli of varying strengths were delivered by the stimulator positioned at the same proximal or distal sites within the muscle. Devices placed close to the nerve-entry site produced the greatest forces. Those placed more distally produced less force. As stimulus current and/or pulse width increased, muscle force increased, often in steps, until a maximum was reached, which was usually limited by the compliance voltage of the device to less than the force produced by whole nerve stimulation.

Transformer miniaturization for transcutaneous current/voltage pulse applications.

A general procedure for the design of a miniaturized step up transformer to be used in the context of surface electrode based current/voltage pulse generation is presented. It has been shown that the optimum secondary current pulse width is 4.5 tau, where tau is the time constant associated with the pulse forming network associated with the transformer/electrode interaction. This criteria has been shown to produce the highest peak to average current ratio for the secondary current pulse. The design procedure allows for the calculation of the optimum turns ratio, primary turns, and secondary turns for a given electrode load/tissue and magnetic core parameters. Two design examples for transformer optimization are presented.

A miniaturized applanation tonometer.

A miniaturized hand-held applanation tonometer is introduced, in which a special cone prism is employed to be an applanation probe to flatten the eye vertically. The self-weight of the probe is just the applanation force, and the applanation area of the ocular cornea is monitored by the optoelectronic signal. The preliminary test demonstrates its good clinical acceptance and its accuracy meeting clinical needs.

Design of a miniaturized ultrasonic bladder volume monitor and subsequent preliminary evaluation on 41 enuretic patients.

Nocturnal incontinence (enuresis) affects 20% of children over four years old, and this figure typically decreases by 15% each year. At the age of 18, 1% of those people remain enuretic. Nocturnal enuresis can be treated by means of a conditioning device that awakens the patient once the urine level has reached a preestablished threshold of the capacity of his or her bladder. We have designed and implemented a portable miniaturized ultrasonic monitor, which permits estimation of the urine volume with an accuracy of 75%. Prototypes have been completed and validated on 41 patients (children) at Ste. Justine Hospital (Montréal). On the first group of 33 patients, we used a hand-held transducer to determine the volume detection range, which is accurate for volumes between 40 and 400 mL. With the second group of 8 patients, the device was mounted on an elastic belt around the abdomen. Measurements were taken in order to validate the accuracy of urine threshold detection and the activation of the corresponding alarm.

Measurement of stand-sit and sit-stand transitions using a miniature gyroscope and its application in fall risk evaluation in the elderly.

A new method of evaluating the characteristics of postural transition (PT) and their correlation with falling risk in elderly people is described. The time of sit-to-stand and stand-to-sit transitions and their duration were measured using a miniature gyroscope attached to the chest and a portable recorder placed on the waist. Based on a simple model and the discrete wavelet transform, three parameters related to the PT were measured, namely, the average and standard deviation of transition duration and the occurrence of abnormal successive transitions (number of attempts to have a successful transition). The comparison between two groups of elderly subjects (with high and low fall-risk) showed that the computed parameters were significantly correlated with the falling risk as determined by the record of falls during the previous year, balance and gait disorders (Tinetti score), visual disorders, and cognitive and depressive disorders (p < 0.01). In this study, the wavelet transform has provided a powerful technique for enhancing the pattern of PT, which was mainly concentrated into the frequency range of 0.04-0.68 Hz. The system is especially adapted for long-term ambulatory monitoring of elderly people.

A coaxial antenna with miniaturized choke for minimally invasive interstitial heating.

We present a new coaxial antenna for microwave interstitial coagulative therapy, working at 2450 MHz and endowed with a miniaturized sleeve choke in order to reduce back heating effects and make the system response less dependent on the antenna insertion depth into the tissue; the way the choke is implemented makes the overall transversal size minimum and allows small adjustments of the choke section length even during operation. We describe the main technical features of the antenna and show experimental results clearly proving the choke effectiveness. Numerical simulations well agree with experimental data, confirming the suitability of the proposed device for minimally invasive medical applications.

In vitro measurement of myocardial impedivity anisotropy with a miniature rectangular tube.

Due to rapid change of fiber orientation, it is difficult to measure myocardial impedivity separately in a longitudinal or transverse fiber direction without mutual influence in the two directions. Previously published values of the longitudinal and the transverse myocardial impedivity were derived indirectly from measurements that mixed the impedivity in all directions. Those values are questionable because the derivations were based on a simplified uniform myocardial fiber model. In this paper, a miniature rectangular tube was devised to facilitate direct measurement of myocardial impedivity in a uniform fiber direction. The average transverse-to-longitudinal ratio of the measured in vitro swine myocardial impedivity was about 1.66 from 1 Hz to 1 kHz and dropped to 1.25 at 1 MHz. The result is important for accurate modeling of the electrical property of myocardium in biomedical research of radio-frequency cardiac catheter ablation.

Design of miniaturized RF SAW duplexer package.

This paper provides a comprehensive methodology for accurate analysis and design of miniaturized radio frequency (RF) surface acoustic wave (SAW) duplexer package. Full-wave analysis based on the three dimensional (3-D) finite element method (FEM) is applied to get the package model. The die model is obtained by combining the parasitics and acoustics models. The modeling of bonding wire is also discussed. The models of package, die, and bonding wires are assembled together to get the total response. Based on this methodology, several novel ideas are proposed to significantly improve the isolation. Simulation and measurement results are compared, and excellent agreement is found. The technique developed in this paper reduces the design cycle time greatly and can be applied to various RF SAW device packages.

Calibration of a focusing transducer and miniature hydrophone as well as acoustic power measurement based on free-field reciprocity in a spherically focused wave field.

The free-field transmitting voltage response at the pressure focus of a spherically focusing transducer was defined and calibrated based on the reciprocity theorem of a free-field spherically focused acoustic wave. The acoustic power, the radiation conductance, and the pressure at the pressure focus were derived and measured accordingly from the transmitting current response on the imaginary mirror symmetric spherical surface of the radiating surface. A miniature hydrophone was calibrated by the self-reciprocity of the spherically focusing source. Comparison results show that the measured acoustic power deviation between the reciprocity method and the radiation force balance method are within +/- 5% for two air-backed focusing transducers at 1.53 MHz and 5.27 MHz, respectively, and the maximum deviation of a hydrophone calibration between the new method and the free-field plane wave reciprocity method is within 1.4 dB in the frequency range from 1 MHz to 2 MHz in experiments.

Hand-held miniaturized cardiac ultrasound instruments for rapid and effective bedside diagnosis and patient screening.

RATIONALE, AIMS AND OBJECTIVES: Technological progress in recent years has made it possible that ultrasound industry can now offer affordable, portable and battery-operated ultrasound systems the size of a laptop computer. The purpose of this study was to compare these hand-carried ultrasound instruments with standard echocardiography in order to investigate the facility of a rapid bedside diagnosis in patients with suspected or known cardiovascular disease. METHODS: Fifty consecutive patients were studied with miniaturized ultrasound equipment (SonoHeart) and a conventional scanner (Acuson Sequoia) in a blinded manner. All studies were performed by three board-certified cardiologists skilled and experienced in echocardiographic practice. Investigators were not aware of any previous medical reports. RESULTS: With the new system, adequate images could be obtained in all patients. Left ventricular and left atrial diameters measured with the hand-held system correlated well with those obtained with conventional scanning: r = 0.87, mean difference 3.12 +/- 2.7 mm and r = 0.84, mean difference 2.8 + 2.4 mm, respectively. The presence of left ventricular dysfunction, regional wall motion abnormalities, relevant valvular regurgitation (moderate or more) or valve stenosis was correctly diagnosed in all patients. However, there was a tendency towards underestimating the extent of wall motion abnormalities particularly in patients difficult to image. Discrepancies also frequently occurred in patients with trivial or mild regurgitation, where false-positive and false-negative findings were described. CONCLUSION: Currently available hand-held echocardiography systems can facilitate rapid bedside diagnosis and patient screening. However, this recent development in echocardiography also raises a number of questions and its actual impact on general clinical practice still remains to be evaluated.

Further miniaturization and automation of in vitro lymphocyte cultures.

A microtechnique (10 mul culture volume) for in vitro lymphocyte cultures is described, which, compared with current miniaturized techniques, permits a four to 20-fold reduction of both medium volume and cell numbers. Furthermore, two alternative procedures for harvesting and washing radioisotope labelled cells are described. The first, making use of a conventional harvesting device, collects cells on glass fibre filters in a two to three-fold shorter time than that needed for the current semi-automatic collectors. The second takes advantage of the possibility of washing and fixing the cells in their culture wells and allows processing of more cultures with fewer manual operations. Various technical aspects of the micro-culture system are described and discussed with special reference to automation problems.
