[Acid phosphatase and non-specific esterase activities in peripheral blood lymphocytes of coke plant workers].

In 32 workers of coking plants cytoenzymatic reaction was examined in lymphocytes peripheral blood to determine the presence of acid phosphatase and unspecific esterase expressed in total score values. Statistically significant decrease of unspecific esterase activity and lower mean values of acid phosphatase activity were found as compared to the results obtained in the control group. It seems that periodical determinations of cytoenzymatic activity of unspecific esterase in lymphocytes of peripheral blood may be useful in assessment of biological effects of occupational exposure in coking plants workers.

The influence of repeated oral administration of dichlorvos on circulating esterases in buffalo calves (Bubalus bubalis).

Alterations in circulating esterases induced by dichlorvos and their reversibility were investigated in male buffalo calves. Changes in blood glucose and total plasma proteins were also monitored. Animals received 4 or 8 mg dichlorvos/kg/day po for 28 consecutive d. Dichlorvos caused dose- and time- dependent significant (P less than 0.01) inactivation of blood esterases. Maximal inhibition of plasma cholinesterase (87-94%) and serum carboxylesterase (51-67%) was observed on the 28th day. An increase in blood glucose and total plasma proteins occurred during the dosing period; these parameters returned to control values in surviving animals within 7 d after the final dose. Activities of esterases returned to 45-70% of normal the 14th d after cessation of dichlorvos dosing.

[Non-specific esterase positive neutrophils in a case of refractory anemia with excess of blasts].

A 61-year-old woman was admitted with complaint of fever. The peripheral blood showed pancytopenia and bone marrow aspirate showed dysplasia in trilineage blood cells with increased blasts (18.2%). Bone marrow chromosome study revealed a karyotype of 46XX, -6, 3q-, +mar in 19 cells of 20 analyzed. She was diagnosed as refractory anemia with excess of blasts (RAEB). 95% of neutrophils in the bone marrow and 84% of that in the peripheral blood were stained with non-specific esterase using alpha-naphthyl acetate as substrate. On the other hand, the positivity of neutrophils for peroxidase. Sudan black B or chloroacetate esterase was markedly decreased. The phagocytotic activity of neutrophils was increased in comparison with normal control cells. Surface marker analysis of peripheral blood myeloid cells revealed increased expression of monocyte specific markers. These findings suggested that the patient's neutrophils, which were surely neutrophils in morphology, shared also monocyte-specific characters. After treatment with low dose Ara-C, pancytopenia was recovered and blasts in the bone marrow were reduced. Also was decreased non-specific esterase positive neutrophils, indicating that the neutrophils were derived from abnormal myeloid clone.

Antigen-specific, but not natural killer, activity of T cell receptor-gamma delta cytotoxic T lymphocyte clones involves secretion of N alpha-benzyloxycarbonyl-L-lysine thiobenzyl ester serine esterase and influx of Ca2+ ions.

Analysis of Ag specificity of TRC-gamma delta+ T cells in humans has been hampered by the fact that cloned lines of these cells expanded in IL-2 generally display high NK-like cytotoxic activity. A TCR-gamma delta+ CTL clone, isolated in IL-4, strongly lysed a specific stimulator cell, the EBV-transformed cell line JY, but failed to lyse K562 and other target cells sensitive for NK cell activity. Subsequent culture of this clone (CD124) in IL-2 induced high cytotoxic activity against the NK sensitive target cells. K562 cells were unable to induce the secretion of N alpha-benzyloxycarbonyl-L-lysine thiobenzyl ester [(BLT)-serine esterase] or influx of Ca2+ ions in clone CD124 cultured in either IL-4 or IL-2. In contrast, JY cells induced high BLT-serine esterase secretion and an increase of cytosolic Ca2+ levels. By using a combination of a 51Cr-release assay and a BLT-serine esterase secretion assay, the reactivity of clone CD124 against a limited number of target cells was analyzed. CD124 which expresses HLA-A2 and -B7, recognized an Ag shared by JY (HLA-A2; B7; C blank; DR4,6) and one haplotype expressed by the cell line SPS (HLA-A1; B14; Cw6; DR4). The only specificity shared by SPS and JY was HLA-DR4. However, clone CD124 failed to lyse 5 other HLA-DR4+ target cells. The cytotoxic activity of clone CD124 was inhibited by the class I MHC specific mAb W6/32 and the anti-beta 2m mAb A88, but not, or only marginally, by the anti HLA-DQ mAb SPV-L3 or the anti-HLA-DR mAb 135. These data strongly suggest that clone CD124 recognizes a class I MHC Ag different from HLA-A, -B, or -C.

Detection of organophosphate detoxifying esterases by dot-blot immunoassay in Culex mosquitoes.

A dot-blot immunoassay, using antiserum raised against esterase B1 responsible for organophosphate (OP) resistance in Culex quinquefasciatus, was used to study different laboratory strains and field collections of this species, as well as of Cx. pipiens, Cx. tarsalis, Anopheles albimanus and An. stephensi. The frequency of esterase B1 positive individuals revealed by this test was in agreement with the data obtained by other methods, indicating the potential of this method for the detection and monitoring of resistance due to increased detoxification by esterases.

Distribution of neurotoxic esterase in certain brain regions of laboratory animals.

Certain organophosphorous compounds caused the inhibition of 'neurotoxic esterase' present in central nervous system. The role of this enzyme is different from that of cholinesterase. The level of neurotoxic esterase in brain, corpus striatum and spinal cord of rats, mice, guineapigs and hens was measured. Maximum level of the enzyme was found in hens, followed by guineapigs, rats and mice in the order. The concentration of the enzyme was higher in corpus striatum greater than whole brain greater than spinal cord. The determination of the normal level of neurotoxic esterase may be useful in monitoring the exposure to organophosphorous compounds.

[Molecular interaction between synthetic spirobutenolides and C1 esterase normal inhibitor].

Previously we have found that spirobutenolide synthetic derivatives show an inhibiting effect on C1 esterase activity. In contrast to these compounds some derivatives in the same series reveal an esterase activity when they are added to a C1 esterase preparation in presence of normal serum. The present work shows that this effect reflect an interaction with natural inhibitor C1 INH.

Characterization of three serine esterases isolated from human IL-2 activated killer cells.

Human peripheral blood mononuclear cells, activated for 14 to 20 days with 1000 U/ml rIL-2, develop strong cytotoxicity for NK sensitive and resistant targets. This process is accompanied by the acquisition of cytoplasmic granules in approximately 60% of the cells and by the expression of esterase activity cleaving the synthetic substrate BLT. The esterase activity, localized in the cytoplasmic granules, was purified and characterized. Three proteins with 3H-DFP binding activity were isolated and had the following properties. Following the proposed nomenclature by Masson et al., the esterases were named human granzymes 1, 2, and 3. Human granzyme 1 on SDS-PAGE has an unreduced relative m.w. of 43,000 and can form disulfide-linked oligomers of relative higher m.w. All forms of granzyme 1 bind 3H-DFP. Upon reduction, granzyme 1 migrates with Mr 30,000 on SDS-PAGE. Additional proteolytic fragments of Mr 24,000 and Mr 28,000 are observed in some reduced preparations. Granzyme 1 cleaves the substrate BLT and appears homologous with murine granzyme A. Human granzyme 2 has an unreduced relative m.w. of 30,000; after reduction, it migrates at Mr 32,000. Even though granzyme 2 binds 3H-DFT, it does not cleave BLT. Human granzyme 2 has properties similar to those of murine granzymes B-H. Human granzyme 3 has unreduced and reduced relative m.w. of 25,000 and 28,000, respectively. It is active in cleaving the substrate BLT. A murine analog for human granzyme 3 has not been described previously. N-terminal sequencing of the purified human granzymes revealed that human granzyme 1 is the gene product of human Hanuka factor cDNA clone and that it represents the human homolog to murine granzyme A. Similarly, human granzyme 2 revealed absolute identity with cDNA-derived N-terminal sequence of a putative human lymphocyte protease cDNA clone.

Plasma kallikrein during experimentally induced allergic rhinitis: role in kinin formation and contribution to TAME-esterase activity in nasal secretions.

We have shown recently that kinins are generated during experimentally induced allergic rhinitis in man, and have demonstrated that substrates for kinin-forming enzymes are provided during the allergic response by a transudation of kininogens from plasma into nasal secretions. In light of this increased vascular permeability during the allergic reaction, we have extended our studies on the mechanisms of kinin formation to examine the potential involvement of plasma kallikrein. Allergic individuals (n = 7) and nonallergic controls (n = 7) were challenged intranasally with an allergen, and nasal lavages, obtained before and after challenge, were assayed for immunoreactive human plasma kallikrein/prekallikrein (iHPK). Post-challenge iHPK values were significantly elevated (p less than 0.01) in the allergic group (353 +/- 394 ng/ml; x +/- SD) as compared to the nonallergics (19 +/- 22 ng/ml), and correlated with increases in kinins, histamine, and N-alpha-tosyl-L-arginine methyl esterase (TAME-esterase) activity and with the onset of clinical symptoms. Gel filtration studies revealed that plasma prekallikrein is activated during the allergic response and contributes to kinin formation prior to interaction with plasma protease inhibitors. We also show that the majority of the TAME-esterase activity detected in nasal secretions during the allergic response is due to activities consistent with a plasma kallikrein/alpha 2-macroglobulin complex and with mast cell tryptase.

Differences in nonspecific esterase from normal and leukemic monocytes.

Nonspecific esterase (NSE) is used to identify normal and leukemic mononuclear phagocytes cytochemically. It can be demonstrated by the hydrolysis of alpha-naphthyl acetate or butyrate. NSE from leukapheresed leukemic monocytes were extracted with several types of detergent and grouped into two isoelectric point (pl) categories based on the ease of solubility: pl 5.7-6.3 (8 bands) greater than or equal to pl 6.6-7.6 (6 bands). Normal monocytes yielded only the pl 5.7-6.3 isozymes. Isozymes from both leukemic and normal monocytes were inhibited similarly by sodium fluoride, pH less than 4.7, and a serine active site inhibitor. All isozymes were bound by Sepharose-concanavalin A (Con-A) and displayed similar substrate preferences and pH v activity slopes. Under the conditions of detergent solubilization, the smallest mol wt species retaining enzymatic activity was 50 +/- 5 kd. Despite these similarities, the isozymes of pl 5.7 through 6.3 and pl greater than 6.3 exhibited different degrees or types of inhibition by phenylmethylsulfonyl fluoride (PMSF), resistance to heat and antigenic character. Thus, the esterase isozymes represent two families of glycoproteins, both of them probable cell surface enzymes, resembling classical liver carboxyl or B-esterases (EC 3.1.1 1).

Characterization of affinity-purified juvenile hormone esterase from Trichoplusia ni.

Juvenile hormone (JH) esterase was purified greater than 1000-fold in one step from hemolymph and whole larval homogenates from the last larval instar of Trichoplusia ni to give a single diffuse band that migrates at Mr = 64,000 on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The purification was based on an affinity chromatography procedure that employs trifluoromethyl ketone ligands. Isoelectric focusing of the purified preparations resulted in multiple bands that coincided to all significant hydrolysis of juvenile hormone detected in this manner. Kinetic experiments using optically pure enantiomers of JH II as substrates showed the two main electromorphs of JH esterase from the hemolymph to have apparently identical kinetic parameters as well as a similar capability to distinguish between substrates that differ in the orientation of the epoxide moiety of JH. However, the enzyme could hydrolyze esters lacking the JH structure. The proteins were shown to be monomers and to have asparagine-linked oligosaccharides, most likely of hybrid structure. Immunochemical and other evidence showed that the affinity-purified proteins were responsible for all significant JH esterase activity during periods of rapid esterolysis in vivo.

Localization and characterization of meperidine esterase of rats.

Meperidine esterase of the rat was studied. The enzyme activity was found in the liver microsomes but not in brain tissue or blood. The enzyme had an optimal activity between pH 8.0 to 9.5 and a temperature of 53 degrees-55 degrees C. The heat of activation was found to be 4.6 X 10(6) cal/mol and the KM 1.2 X 10(-4) M. The esterase activity of older male rats (200-350 g) was significantly higher than that of the younger group (150-175 g). There was no significant difference in specific activity of hepatic meperidine esterase between male and female rats, and between rats fasted for 20 hr and phenobarbital- and benzopyrene-treated rats and their respective control groups. However, the liver weight and the microsomal protein yield per g of liver of fasted rats were significantly smaller, and those of phenobarbital- and benzopyrene-treated rats were significantly larger, than those of the control group. The overall enzyme activity per liver of phenobarbital- and of benzopyrene-treated rats was therefore significantly increased. The specific enzyme activities in microsomes prepared by ultracentrifugation and by calcium aggregation were not significantly different, although the ultracentrifugation method resulted in a higher yield of hepatic microsomal protein. The enzyme activities in microsomes prepared by ultracentrifugation/or calcium aggregation and frozen at -20 degrees C up to 22 days were not significantly different from those of freshly prepared microsomes. The enzymatic activity was inhibited by cocaine hydrochloride, isocarboxazid, neostigmine methyl sulfate, alpha-naphthyl acetate, p-nitrophenyl acetate, paraoxon, propoxyphene hydrochloride, phenelzine sulfate, physostigmine sulfate, SKF 525-A, tranylcypromine sulfate, fluoride ions, methanol, and ethanol.

On the role of the C1-esterase inhibitor in cold urticaria.

Plasma samples from 20 patients with acquired cold urticaria were studied. The C1-esterase inhibitor activity was found to be low, but the total C1-esterase inhibitor concentration was normal. Prekallikrein, plasmin-alpha 2-antiplasmin complex, and kallikrein-like activity were also found to be within normal limits. Cold-promoted activation of coagulation factor VII occurred in 45% of the patient plasmas and resulted in a considerable drop in C1-esterase inhibitor activity.

The deacetylation of N-hydroxy-2-acetylaminofluorene by rat liver microsomes and carboxyl esterase.

We have demonstrated that incubation of rat liver microsomes with N-hydroxy-2-acetylaminofluorene (N-OH-AAF) leads to formation of a 2-nitrosofluorene-membrane lipid adduct. This adduct exists as a nitroxyl free radical, termed N-O-LAF, in its oxidized state. When microsomes were incubated with the sulfhydryl binding agent, rho-hydroxymercuribenzoate, a larger amount of N-OL-LAF formed. We interpret this as a slowdown in the rate of endogenous chemical reduction of carcinogen-membrane lipid adduct. In this paper we present evidence that N-OH-AAF is deacetylated by a microsomal enzyme to form N-hydroxy-2-aminofluorene and this is then oxidized to 2-nitrosofluorene which adds covalently to membrane lipid double bonds to form N-O-LAF. Various antioxidants, peroxidase inhibitors, and P450 substrates and inhibitors were ineffective in altering the amount of N-O-LAF formed from N-OH-AAF; but two esterase inhibitors, dietyl-rho-nitrophenylphosphate and alpha-toluene-sulfonyl fluoride, prevented N-O-LAF formation. Of the following purified enzymes tested: porcine liver carboxyl esterase, pepsin, chymotrypsin, cathepsin D, ficin, papain, leucine aminopeptidase, Naja naja phospholipase, acetylcholinesterase (type I), trypsin (type I and V) and epoxide hydrase; only carboxyl esterase was effective in deacetylating N-OH-AAF.

Atropine esterase status of laboratory mice.

Fifty eight inbred strains of laboratory mice and two species of wild mice [Mus castaneus (inbred) and Peromyscus maniculatus bairdii (closed colony)] were tested for the presence of atropine esterase by the microhematocrit tube test of Tucker and Beattie. The results show clearly that laboratory mice do not have significant levels of atropine esterase as determined either by the microhematocrit tube test or by the agar plate test, and therefore that under these conditions atropine esterase is not a polymorphic variant as has been demonstrated clearly in the rabbit.

Serum and urinary arginine-esterase activity in paediatric kidney diseases.

Serum and urinary kallikrein activities were determined from the arginine-esterase activities in various groups of kidney diseases and were compared with urinary beta-glucuronidase excretion, urinary output, urinary protein content and creatinine clearance. Serum arginine-esterase activity was significantly augmented in the active stage of diffuse renal diseases but was not related to the severity of parenchymal damage. The values improved during remission; the enzyme activity in chronic uraemic patient was as low as in the control sera. There was a positive correlation between urinary output and serum arginine-esterase activity, and consequently serum kallikrein might have an enhancing effect on diuresis.

Arginine esterase activity of the plasma in different types of bronchial asthma.

Spontaneous arginine esterase activity and esterolytic activity of plasma after a contact with kaolin, using a synthetic substrate BAEE in asthmatic patients were studied. Also activity of esterase inhibitors, the plasma kininogen level and fibrinolytic activity were determined. It was shown, that during an asthma attack a significant rise in the plasma esterolytic activity occurred. At the same time esterase activity displayed a fall after contact with kaolin as compared with controls. These changes were accompanied by a rise in the fibrinolytic activity of the plasma and a decrease in the kininogen level. The behavior of parameters studies was analyzed in relation to the immunological type of asthma and the clinical state of the patients.
