[Differential analysis of mitral and tricuspid regurgitation by continuous Doppler].

The following parameters have been measured by continuous Doppler in 40 patients with both mitral failure (MF) and tricuspid failure (TF): effusion time (ET), peak time (PT), maximum effusion speed (S. max), maximum anterograde speed (S. ant), taking into account the cardiac rate and the ejection fraction. Only the S. max. and the S. ant. are significantly different and enable the determination of the effusion origin with a percentage error of about 4%. When a straight line is drawn to joint 3.7 m/s on the ordinate (S. ant) and 5 m/s on the abcissa (S. max), the TF lie below the line and the MF above it. The selection of the sub-groups with low ejection fraction (less or equal to 30%) or with arterial pulmonary hypertension (S. max MF higher or equal to 3 m/s) does not modify the precision of the discrimination. The graph suggested seems therefore to be a rapid and reliable means of characterizing TF in order to assess the arterial pulmonary hypertension of dilated hypokinetic cardiomyopathies.

[Submicroscopic study of adenohypophyseal cells with the hypophysis incubated in a hypertonic solution. II. Differential analysis of the intracellular distribution of arylsulfatase and thiamine pyrophosphatase within a single cell].

In the rat pituitary gland exposed to an hour incubation in the medium containing 1.65 M sucrose activities of aryl sulphatase (AS) and thiamine pyrophosphatase (TPP) were successively defined within a single specimen. After the detection and registration of both enzyme activities with the electron microscope ultrathin sections were processed with 50% H2SO4 resulting in the complete removal of the reaction products for TPP. In the mammotrophs and somatotrophs, cisternae of the Golgi apparatus and rough endoplasmic reticulum showed either AS, or TTP or both the enzymatic activities. In the latter case the two enzymes could be restricted to a single cistern.

An organizational model of choice: a theoretical analysis differentiating choice, personal control, and self-determination.

Individuals experience choice when they select one option from among meaningful alternatives that possess relatively equal attractiveness and some degree of indeterminacy. Choice has been found to influence important psychological and behavioral outcomes. After differentiating among choice, personal control, and self-determination, the author offers a model of choice, with self-determination as the key mechanism regulating how choice influences intrinsic motivation. The model suggests specific types of choice-relevant information that should affect whether choice results in an internal (self-determined) or external (controlled) locus of causality. The individual characteristics of locus of control, self-presentation, self-esteem, and Type A personality are suggested as possible moderators of the effects of choice. Finally, the implications of the choice model for organizations and further areas of research are discussed.

Effect of methylamine and plasmin on the conformation of human alpha 2-macroglobulin as revealed by differential scanning calorimetric analysis.

Differential scanning calorimetric analysis was used as a probe of the conformational alteration in human alpha 2-macroglobulin (AM) upon its complex formation with methylamine and with the protease, human plasmin. The slow electrophoretic form of AM displayed a single thermal transition, characterized by a temperature midpoint (Tm) of 65.8 +/- 0.3 degrees, a calorimetric enthalpy (delta Hc) of 2,550 +/- 150 kcal/mol and a van't Hoff enthalpy (delta Hvh) of 140 kcal/mol. In the presence of sufficient methylamine to irreversibly disrupt the four thiol ester bonds in AM, a single thermal transition was obtained, characterized by a Tm of 62.8 +/- 0.3 degrees, a delta Hc of 1,700 +/- 100 kcal/mol, and a delta Hvh of 169 kcal/mol. These data suggest that a major conformational alteration is produced in AM upon complex formation with methylamine. When plasmin interacts with AM, the resulting thermogram displays Tm values for AM of 68-69 degrees and 77 degrees, also suggestive of a large conformational alteration in AM. However, this latter alteration appears dissimilar to the change induced by methylamine.

LimROTS: a hybrid method integrating empirical Bayes and reproducibility-optimized statistics for robust differential expression analysis.

MOTIVATION: Differential expression analysis plays a vital role in omics research enabling precise identification of features that associate with different phenotypes. This process is critical for uncovering biological differences between conditions, such as disease versus healthy states. In proteomics, several statistical methods have been used, ranging from simple t-tests to more advanced methods like DEqMS, limma and ROTS. However, a flexible method for reproducibility-optimized statistics tailored for clinical omics data has been lacking. RESULTS: In this study, we developed LimROTS, a hybrid method that integrates a linear regression model and the empirical Bayes approach with reproducibility optimized statistics, to create a novel moderated ranking statistic, for robust and flexible analysis of proteomics data. We validated its performance using twenty-one proteomics gold standard spike-in datasets with different protein mixtures, MS instruments, and techniques for benchmarking. This hybrid approach improves accuracy and reproducibility of complex proteomics data, making LimROTS a powerful tool for high-dimensional omics data analysis.
