A histologic study was done on the spontaneously occurring hyperplastic and neoplastic lesions of the uterus, cervix, and vagina taken from a variety of inbred, hybrid, and backcross female mice. The most commonly observed neoplasm of the endometrium was stromal cell sarcoma, followed by adenocarcinoma, which occurred approximately half as frequently. Hyperplasia was the most commonly observed endometrial lesion occurring either as glandular, stromal, or both and was often cystic with the formation of polyps. Among the benign and malignant lesions other than endometrial, leiomyomas, hemangioendotheliomas and leiomyosarcomas were the most commonly encountered. Lesions in the mucosal cervix and vagina were comparatively rare and were observed in only a few animals. Females with hyperplastic and neoplastic lesions of the uterus, cervix, and vagina in this survey represented a small fraction of the large number of animals necropsied, emphasizing the relative infrequency of these spontaneous lesions in mice.
To study the development of the reproductive tract in heifers, the ovaries, uterus, cervix and vagina were examined by transrectal ultrasonography every 2 weeks, from 2 to 60 weeks after birth. First ovulation occurred at 63.7 +/- 1.1 weeks of age. Ovarian dimensions increased rapidly from 2 to 14 weeks of age, and increased again after 34 weeks of age (P<0.05). The size of the largest ovarian follicles increased from 8 to 14 weeks of age, from 38 to 42 weeks of age, and finally from 52 to 60 weeks of age (P<0.05). The number of follicles > or =3 mm in diameter tended to increase from 6 to 14 weeks of age (P<0.10) and increased significantly from 6 to 60 weeks of age (P<0.05). Mean numerical pixel values of the ovarian images decreased from 4 to 26 weeks of age, and then rose to 44 weeks of age (P<0.05). Diameter of the uterine body, cervix and vagina increased from 2 to 20-24 weeks of age, and again after 32 weeks of age (P<0.05). Mean numerical pixel values for the uterus and vagina decreased initially (uterus: 4-8 weeks and vagina: 6-22 weeks of age) and then increased (uterus: 14-42 weeks and vagina: 22-32 weeks of age; P<0.05). Pixel heterogeneity showed a consistent peak at 20-22 weeks of age for the uterus, cervix and vagina (P<0.05). In summary, in the heifer calf, the marked growth of the reproductive tract in the first few months of age, and prior to first ovulation, reflects phases of increased ovarian follicle (> or =3 mm in diameter) numbers and size. Ultrasonographic image analysis revealed patterns of numerical pixel values and heterogeneity that may be useful in determining important stages of growth and differentiation of the reproductive system.

This study examined responses of 35 neurons in the caudal intralaminar (IL) thalamic nuclei in 12 adult female virgin rats to mechanical stimulation of the skin (brush, pressure, pinch) and to 4 different visceral stimuli (noxious distension of the uterine horns and vaginal canal; gentle distension of the colon and probing the cervix). As in male rats and other species, many IL neurons (24/35) responded to frankly noxious somatic stimuli applied to several bodily regions. Some of these (16/24) also responded to one or more of the visceral stimuli (mainly the noxious ones), while 4/35 responded only to a visceral stimulus. Thus, unlike neurons in lateral thalamus studied under identical conditions, IL neurons appear to be signalling information primarily when intense somatic and visceral stimuli are frankly above the noxious threshold.

Administration of lindane (20 mg/kg . d) for 30 d to ovariectomized rats caused no significant change in the weight of the uterus, cervix, or vagina. Histological changes in these organs were comparable to those in ovariectomized control rats. Treatment with estradiol dipropionate alone and in combination with lindane induced a significant increase in the weights of these organs. Microscopically, these organs appeared normal. Lindane induced a significant increase in the glycogen content of the uterus, cervix, and vagina of ovariectomized rats compared to ovariectomized control rats, whereas a two- to fourfold increase in glycogen was observed in ovariectomized rats treated with estradiol dipropionate or estradiol dipropionate plus lindane. There was a significant increase in total erythrocytes and hemoglobin in ovariectomized rats treated with lindane, estradiol dipropionate, or lindane plus estradiol dipropionate. An increase in neutrophils with a corresponding decrease in lymphocytes was also observed in rats treated with lindane or lindane plus estradiol dipropionate.
Sensory nerve fibres containing calcitonin gene-related peptide (CGRP) innervate neurons of the paracervical ganglion (PCG) in the female rat pelvic plexus. We have combined retrograde tracing with immunocytochemistry to investigate whether CGRP-immunoreactive (-IR) fibres supply neurons targeting the genital tract. Of the total neurons projecting to either the uterine horns or the cervix, 38 and 41% received CGRP-IR innervation, respectively. All these neurons displayed choline acetyltransferase-IR, thus are cholinergic. They were found throughout the PCG and other pelvic plexus ganglia, namely accessory ganglia (AG) and hypogastric plexus (HP). Pelvic nerve section showed that afferent fibres in these nerves provided most of the CGRP-IR fibres supplying uterine- or cervical-related neurons in the PCG/AG, none in HP. It is suggested that such sensory-motor network may provide a local pathway for reflex control of genital tract activity, acting through cholinergic nerve projections.

The in vivo binding of antiestrogen in nuclei within the uterine stromal, epithelial and myometrial tissue compartments was compared to that of estrogen 2-48 h after injection. Tissue binding and retention of radioactivity was also studied. Immature rats were injected s.c. with 0.36 microgram [3H]hydroxytamoxifen [( 3H]TAM(OH] or 0.24 microgram [3H]estradiol [( 3H]E2) in oil. At 2, 4, 8, 12, 24 and 48 h after injection, uteri were processed for thaw-mount autoradiography and, along with other tissues, for liquid scintillation counting. After [3H]TAM(OH) injection total radioactivity in uterus, cervix, vagina and liver reached peak levels by 8 h then decreased slowly so that by 48 h radioactivity still remained in the tissues. At all time intervals, the levels of radioactivity in heart and muscle remained low. After [3H]E2 injection radioactivity in uterus, cervix and vagina reached peak levels between 2 and 4 h then decreased rapidly so that by 12 h radioactivity was low and essentially the same as in liver, heart and muscle. These results were paralleled by the time course of nuclear binding of [3H]TAM(OH) and [3H]E2 in the uterine cell types: peak levels occurred at 8 h and 4 h, respectively. Nuclear binding was still present 48 h after [3H]TAM(OH) injection but was absent by 24 h after [3H]E2 injection. Different uterine cell types bound different amounts of both the drug and the hormone. After [3H]TAM(OH) injection the decreasing order of labeling intensity in the tissue compartments was stroma, myometrium, epithelium; in contrast, that after [3H]E2 injection was stroma, epithelium, myometrium. The results indicate that the dissimilar nuclear binding kinetics of estrogen and antiestrogen are influenced by the pharmacokinetics but the uterine cell types may also influence binding kinetics.

Fifty hysterectomy specimens were studied by a multiple biopsy technique to determine the normal bacteriological flora of the uterine cavity and cervical canal. All the uterine cavities and 23 of the cervical canals were sterile. The lower half of the other 27 cervical canals contained organisms similar to those found in the vaginal vault.
The development of uterine oxytocin receptors is an important regulatory step in the initiation of labour. Paracrine production of oxytocin by uterine and placental tissues may also be involved in some species. Placentome, intercotyledonary endometrium, myometrium and fetal membranes were collected from 3-5 ewes each, at regular intervals throughout pregnancy and from eight ewes during labour. Localization of mRNA encoding oxytocin and its receptor was by in situ hybridization; oxytocin peptide concentrations were measured by radioimmunoassay and oxytocin receptor concentrations were measured by autoradiography and radioreceptor assay. In the intercotyledonary endometrium, mRNA encoding the oxytocin receptor was located in the luminal epithelium only. Both the epithelial and myometrial receptors were detected at low concentrations from the fourth week of gestation onwards, with a major increase associated with the onset of labour. In the placentomes, oxytocin receptors were localized to a stromal capsule surrounding the placental villi. Expression in this region was maximal in mid-gestation, declining in the second half of pregnancy and remaining low during labour. Cervical oxytocin receptors were detected at low concentrations in the epithelium and the muscular/connective tissue layers from day 22 of pregnancy onwards. There was no evidence for the local uterine production of oxytocin in the ewe; mRNA encoding oxytocin was undetectable and oxytocin concentrations were always < 23 pg g-1 wet mass of tissue. These results suggest that regulation of the timing of oxytocin receptor development varies between the different tissue types, despite a similar steroidal background. The receptors in the luminal epithelium are probably associated with the ability of exogenous oxytocin to induce the release of PGF2 alpha throughout most of pregnancy. The increase in receptors in both the intercotyledonary endometrium and myometrium at term suggest an involvement in labour, whereas their role in caruncular stroma in mid-pregnancy is unknown.
