Binding of Bodian's silver and monoclonal antibodies to defined regions of human neurofilament subunits: Bodian's silver reacts with a highly charged unique domain of neurofilaments.

Cleavage at cysteine and chymotrypsin digestion were applied to two human neurofilament (NF) subunits, low- and high-molecular-weight NF (NF-L and NF-H), to locate the regions reacting with Bodian's silver stain and with several monoclonal antibodies, including NF-specific antibodies and one that recognizes all intermediate filaments (anti-IFA). Our findings indicate that whereas anti-IFA recognizes the highly conserved rod domain, all the NF-specific antibodies, as well as Bodian's silver, react with the carboxy-terminal tailpiece of NF subunits. The silver binding sites in NF-L are located in a carboxy-terminal 12-Kd chymotrypsin fragment, a highly charged, unique domain of NF.

Topical chemoprophylaxis with silver sulphadiazine and silver nitrate chlorhexidine creams: emergence of sulphonamide-resistant Gram-negative bacilli.

Controlled trials of 0.5% silver nitrate compresses (SN), 1% silver sulphadiazine cream (SSD), and a cream containing 0.5% silver nitrate and 0.2% chlorhexidine digluconate (SNC) showed that all were comparably effective in protecting burns from infection. SN compresses were much less active against miscellaneous Gram-negative bacilli than the other preparations, and the mean morning and evening temperatures and respiration rates in the patients treated with SN compresses were higher then those of patients treated with SSD. Pseudomonas aeruginosa and Proteus spp, though rare in all groups, were less often found in the patients treated with SN compresses. Sulphonamide-resistant Gram-negative bacilli became predominant during the trial of SSD cream on extensive burns and the prophylactic effectiveness of that preparation was thus reduced in the later stages of the trial.

[Application of silver acetate autometallography in histopathology: a new detection method for use in immunogold silver staining, lectin histochemistry and in situ hybridization].

Modern histochemical techniques allow the specific detection of tissue constituents in situ. Routinely formalin fixed, paraffin embedded tissues may present problems to the pathologist since destruction of substances can lead to false negative results. Immunogold-silver staining (IGSS) can be a way to overcome some of these problems. A highly efficient method allows silver enhancement to be carried out without the necessity of a dark box (silver acetate autometallography). To test the new modification, antisera to a variety of antigens were used on paraffin and semithin resin sections. The system was also tested for its applicability in multiple immunostaining techniques and for the in situ hybridisation of viral DNA and for demonstration of carbohydrates by lectin histochemistry. It showed a very high detection efficiency with low background staining.

[The dependence of the quantity of silver granules detectable in interphase nuclei after the silver nitrate staining of the chromosomal nucleolus-organizer regions on the conditions for the hypotonic treatment of the cells and the effect on this index of cell treatment by a pharmacological agent causing dissociation of the nucleolar components].

We investigated effects of hypotonic treatment of cultured HL-60 cells and human fibroblasts, and of DRB on the quantity of AgNORs revealed following AgNO3 staining. The average number of grains increases after KCl hypotonic treatment. The lower the concentration of KCl in the solution, the higher the number of silver grains revealed in the nucleoli due to their dissociation into substructures. The use of adenosine analogue (DRB) before the hypotonic treatment of cells facilitates the following count of silver deposits. A maximum increase in the mean number of silver grains was observed after a 1.5 h incubation.

Argentophilicity and solvent-induced structural diversity in double salts of silver acetylide with silver perfluoroalkyl carboxylates.

A series of novel double salts of silver(I) were isolated by dissolving Ag(2)C(2) in a concentrated aqueous solution of R(F)CO(2)Ag (R(F) = CF(3), C(2)F(5)) and AgBF(4). Different ancillary solvento ligands such as H(2)O, CH(3)CN, and C(2)H(5)CN were found to affect the crystallization process that led to the assembly of various silver(I) cages with embedded C(2)(2-) ions. 2Ag(2)C(2) x 12CF(3)CO(2)Ag x 5H(2)O (1) consists of two independent C(2)@Ag(7) cages, each having the shape of a basket with a square base. Ag(2)C(2) x 6CF(3)CO(2)Ag x 3CH(3)CN (2) contains a zigzag chain of edge-sharing triangulated dodecahedra, and 4Ag(2)C(2) x 23CF(3)CO(2)Ag x 7C(2)H(5)CN x 2.5H(2)O (3) features an unusual double-walled silver column constructed from the fusion of four different kinds of irregular polyhedra. Ag(2)C(2) x 10C(2)F(5)CO(2)Ag x 9.5H(2)O (4), Ag(2)C(2) x 9C(2)F(5)CO(2)Ag x 3CH(3)CN x H(2)O (5), and Ag(2)C(2) x 6C(2)F(5)CO(2)Ag x 2C(2)H(5)CN (6) all contain an edge-sharing double cage with each single cage in the shape of a square antiprism, a capped square antiprism, and a triangulated dodecahedron, respectively.

Silver(I) 1,3-butadiynediide and two related silver(I) double salts containing the C4(2-) dianion.

Ag2C4, the second silver carbide to be fully characterized, has been synthesized as a light-gray powder (contaminated with metallic silver) that is explosive at high temperature (130 degrees C) and sensitive to mechanical shock, rather like the well-known prototype Ag2C2. In the pair of hydrated double salts Ag2C4.6AgNO3.nH2O (n = 2, 3), the nearly linear, centrosymmetric 1,3-butadiyne-1,4-diyl dianion C42-exhibits an unprecedented mu8-coordination mode, each terminal being capped by four sigma-bonded silver(I) atoms with pi-interaction to one of them.

Occurrence of Bunodera sacculata Van Cleave and Mueller, 1932 in Perca flavescens from Silver Creek and Silver Lake, Michigan.

Bunodera sacculata (Trematoda: Allocreadiidae) infected 65 (54%) of 120 yellow perch collected in 1997 and 40 (50%) of 80 perch from Silver Creek in 1998. and 17 (30%) of 56 perch in 2000 from Silver Lake in lower Michigan. Gravid B. sacculata occurred in the pyloric ceca and anterior intestine. There were differences in monthly trends (July through August) in prevalences, mean intensities, and mean abundances of B. sacculata in Silver Creek between 1997 and 1998. The mean intensity +/- SD (8.2 +/- 9.1) and mean abundance (4.1 +/- 7.8) of B. sacculata were significantly higher in 1998 than in 1997 and 2000. An additional 20 perch (0+ in age) collected from the creek in July 1998 were all infected and had the highest mean intensity of 21.3 +/- 14.1.

A silver tattoo of the nasal mucosa after silver nitrate cautery.

We report a silver tattoo of the nasal mucosa that occurred after silver nitrate cautery for nasal bleeding. This type of tattoo is a very rare potential mimic of melanoma and appears not to have been described before. It has similar features to an amalgam tattoo of the oral mucosa on histology and energy dispersive X-ray analysis (EDAX).

Gold(I) and silver(I) mixed-metal trinuclear complexes: dimeric products from the reaction of gold(I) carbeniates or benzylimidazolates with silver(I) 3,5-diphenylpyrazolate.

Trinuclear mixed-metal gold-silver compounds are obtained by the reaction of gold(I) carbeniate [Au(mu-C(OEt)=NC6H4-p-CH3)]3, TR(carb), or gold(I) imidazolate [Au-mu-C,N-1-benzyl-2-imidazolate]3, TR(bzim), with silver(I) pyrazolate [Ag(mu-3,5-Ph2pz)]3. The crystalline products are mixed-ligand, mixed-metal dimeric products [Au(carb)Ag2(mu-3,5-Ph2pz)2], [Au2(carb)2Ag(mu-3,5-Ph2pz)].CH2Cl2, [Au(bzim)2Ag2(mu-3,5-Ph2pz)], and [Au2(bzim)2Ag(mu-3,5-Ph2pz)]. They have been characterized by elemental analysis and 1H NMR and mass spectrometry. The X-ray structure of [Au(carb)Ag2(mu-3,5-Ph2pz)2] shows it to be a dimer with two Ag...Au contacts between the trinuclear units of 3.083(2) and 3.310(2) A and with average intramolecular Ag...Ag and Au...Ag distances of approximately 3.3 and 3.2 A, respectively. The structure of [Au2(carb)2Ag(mu-3,5-Ph2pz)].CH2Cl2 is a dimer with one intermolecular Au...Au attraction of 3.3354(10) A and a short Ag...Au distance of approximately 3.42 A and intramolecular Ag...Au and Au...Au contacts of approximately 3.2 and approximately 3.3 A, respectively. Packing diagrams of both complexes show that the dimeric units are independent, similar to their parent molecules. The dimers of trinuclear [Au(carb)Ag2(mu-3,5-Ph2pz)2] and [Au2(carb)2Ag(mu-3,5-Ph2pz)].CH2Cl2 crystallize in the triclinic space group P (Z = 2), a = 9.688(3) A, b = 15.542(4) A, c = 23.689(6) A, alpha = 82.560(5) degrees , beta = 87.887(6) degrees , gamma = 78.060(5) degrees , and the orthorhombic space group Pca2(1) (Z = 4), a = 29.644(4) A, b = 7.4582(10) A, c = 30.473(4) A, respectively. The structure of [Au(bzim)Ag2(mu-3,5-Ph2pz)2] is a dimer with two metallophilic Ag...Au interactions of 3.14 A. The complex crystallizes in the monoclinic space group C2/c (Z = 4), a = 26.368(5) A, b = 15.672(3) A, c = 17.010(3) A, beta = 102.206(3) degrees .

Silver deposition in the cervix after application of silver nitrate as a cauterising agent.

Large amounts of metallic silver pigment were found in 10 cervical biopsy specimens taken for the histological grading of cervical intraepithelial neoplasia (CIN). The assessment of CIN was made much harder because the pigment obscured the morphological detail of the epithelial cells, and in some it was very difficult to determine whether koilocytosis or CIN I was present. The silver can easily be removed by a simple chemical method.

Microwaves applied to silver impregnations with ammoniacal silver carbonate.

Techniques for impregnation with ammoniacal silver carbonate provide valuable information on all types of tissue; however, the time investment required to impregnate a few sections has limited their application. We have shortened the impregnation times by using microwaves in techniques for reticular fibers, astrocytes, nerve fibers and chromaffin cells. The results were satisfactory with markedly reduced impregnation time and elimination of nonspecific silver deposits.

Mass spectrometric identification of proteins from silver-stained polyacrylamide gel: a method for the removal of silver ions to enhance sensitivity.

Mass spectrometry is a powerful technique for the identification of proteins at nanogram quantities. However, some degree of sample preparation prior to mass spectrometry is required, and silver-stained protein gel samples are most problematic. Here we report our strategy to obtain peptide mass profiles from silver-stained protein gel samples from one- or two-dimensional gels by destaining prior to enzymatic digestion. This study demonstrates that by using the destaining method, the sensitivity and quality of mass spectra is increased for matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) mass spectrometric analysis, permitting more proteins to be identified by peptide mass database analysis.

Optical properties of gas-phase tryptophan-silver cations: charge transfer from the indole ring to the silver atom.

We present a joint experimental and theoretical investigation of the electronic excitation spectra of the tryptophan-silver complex. The photodissociation spectrum of gas-phase [Trp-Ag]+ was measured from 215 to 330 nm using a quadrupole ion trap coupled to an optical parametric-oscillator laser. The calculated time-dependent density functional theory (TD-DFT) absorption spectra for different prototypes of structures are presented. Low-energy transitions that are experimentally observed are only calculated for the charge-solvation (CS) structures. These transitions are a signature of the metal-pi interaction in [Trp-Ag]+. The recorded spectrum is compared to a Boltzmann average of the absorption spectrum obtained from direct molecular dynamics (MD) simulations involving simultaneous transitions to excited states based on semiempirical configuration interaction (CI) calculations. The results demonstrate that charge transfer can be photoinduced from the indole ring to the silver atom.

Precipitation of silver-thiosulfate complex and immobilization of silver by Cupriavidus metallidurans CH34.

Cupriavidus metallidurans CH34 is a facultative chemolithotrophic bacterium that possesses two megaplasmids (pMOL28 and pMOL30) that confer resistance to eleven metals. The ability of Cupriavidus metallidurans CH34 to resist silver is described here. Electronic microscopy, energy-dispersive X-ray (EDX) and X-ray diffractometry (DRX) observations revealed that C. metallidurans CH34 strongly associated silver with the outer membrane, under chloride chemical form. Using derivate strains of C. metallidurans CH34, which carried only one or no megaplasmid, we show that this resistance seems to be carried by pMOL30.

Silver staining of proteins in polyacrylamide gels: increased sensitivity through a combined Coomassie blue-silver stain procedure.

A combined Coomassie blue-silver stain method has been developed in sodium dodecyl sulfate-polyacrylamide gels for the detection of proteins using the model compounds bovine serum albumin, lysozyme, and recombinant DNA-derived human insulin. Sensitivity was enhanced 2.2 to 8.6 times by the new method relative to that of silver staining alone. The new method may also be useful in enhancing detection sensitivities of other proteins.
