[Cultivation of Polygonum multiforum].

The experiments of some cultivated medthods of Polygonum multiforum show that the direct sowing of seeds is better than the cuttage, the index of reproduction and yield are higher, and the cultivated method can decrease expense. To cultivation period of 5 years, high yield and excellent qualty is available.

[Improving the method of processing Polygonum cuspidatum Sieb. et Zucc. and Spatholobus suberectus Dunn].

This paper is concerned with a comparison of Polygonum cuspidatum with Spatholobus suberectus in cutting, drying time, products, chief compositions and aqueous extracts. The results showed that fresh cut was better than the customary cut, i. e. re-cut pieces, for the former could not only raise the quality specifications and contents of ingredients of the drinking tablets, but also avoid repeated labour, save energy for drying and eliminate waste of drug materials.

[Contents comparison of resveratrol and polydatin in the wild Polygonum cuspidatum plant and its tissue cultures].

OBJECTIVE: To compare the contents of resveratrol and polydatin in some materials of Polygonum cuspidatum from various sources, so to screen and obtain the suitable cultures for the following metabolism regulation study. METHOD: RP-HPLC method was applied to simultaneously assay resveratrol and polydatin in different samples. RESULT: By the modified methods of extraction and determination, large amount of materials were screened. The results indicated that the contents of resveratrol and polydatin in root and rhizome were evidently higher than those in the leave and stems. The content of polydatin in the seedlings cultured indoor for three months was 1.27% and showed a 1.25-time increse than that in the wild plants, while the content of resveratrol (0.401%) approached that in the wild plants. Both of resveratrol and polydatin could be examined from different tissue cultures of P. cuspidatum, such as the sterile seedlings, callus, suspended cells and hairy roots, and the levels of them were closely related to the growth speed, physiological status and developmental phase. Hairy roots had the highest potentiality in several tested cultures and the increase rate of dry weight was 8.29 when cultured in vitro for 30 days, and showed a 8.4-fold and a 192.8-fold increase compared with those of natural roots and suspended cells, respectively. The content of polydatin in the hairy roots was up to 0.037% and that of resveratrol was 0.007%. CONCLUSION: The established analysis method is rapid, simple and accurate, especially adapted to the simultaneous determination of resveratrol and polydatin in massive biological samples. Hairy-root cultures have the superiority among the tested materials of P. cuspidatum and are suitable for the large-scale biomass and consistent production of efficient constituents.

[Orienting toxicologic studies in Wistar rats for the evaluation of knotgrass--(Polygonum convolvulus)--seed].

Black bindweed seed (Polygonum convolvulus) from the 1987 harvest was tested for possible toxicity in a 38-day range-finding test with Wistar rats which received feed rations on the basis of VDT-1 standard feedstuff and components of 5 or 20%. Histological liver findings as well as parameters recorded from laboratory diagnosis (plasma activities of ALAT, ASAT, and gamma GT as well as total protein and albumin levels in plasma) were indicative of incipient impairment of liver metabolism. More detailed and comprehensive chemico-analytical investigations are recommended, with due consideration of the unity between high-value primary components, on the one hand, and possibly toxic secondary constituent substances, on the other.

[Effect of Polygonum cuspidatum No 4 on the enhancement of cardiac function during burn shock].

A burn shock model was produced in rats by scalding the lumbus and lower extremities with 80 degrees C water for 30 seconds. The variable of cardiac functions were measured with a Cardiac-II-R cardio-output computer. It was shown that heart functions decreased as early as 10 min following scalding. Cardiac output (CO), cardiac index (CI), and stroke volume index (SVI) were reduced by 40% of normal, and stroke work (SW) by 47%. With the decrease of CO, total peripheral resistance (TPR) increased. Two hour later TPR was increased 2.6-fold more than pre-burn level, which made BP maintain nearly normal. TPR started to reduce at 3 hr post-burn, resulting in decrease of BP. The decreased heart function returned to near normal by administration of crystal of Polygonum cuspidatum No 4 (p.c), e.g, CO and CI were increased to 91.0% of pre-burn value, SW to 100.1%, and SVI to 102.0%. The increased TPR was reduced to near normal. Half of p.c.-treated animals survived within 24 hr, but all of simple burn animals and n.s.-treated animals died within 24 hr. Under the condition of no fluid infusion, administration of p.c. could restore the decreased heart function, which showed that besides loss of plasma, direct myocardial depression was one of the reason for the decrease of cardiac output during burn shock. The survival rate of animals with burn shock was enhanced by injection of p.c., which indicated that increasing heart function was a very important measure for the treatment of burn shock.

Determination of avicularin in Polygonum aviculare L. by square wave polarography.

A method for the determination of avicularin in Polygonum aviculare L. is described. Square wave polarography (SWP) with a static mercury drop electrode was used to observe the reduction of avicularin. The supporting electrolyte was an acetate buffer solution of pH 4. Avicularin gives a well-defined peak at a potential of -1.30 V (vs. Ag/AgCl). An excellent linear relationship was obtained between the concentration (1.20-3.59 x 10(-6) mol/L) and peak current. The method can be used to analyze small amounts of sample.

[Effect of crystal No. 4 of Polygonum cuspidatum on the restoration of pulse pressure and microcirculatory perfusion during shock].

A model of irreversible hemorrhagic shock was reproduced in rats. Synchronous video tape recording with 2 cameras was used to observe the relationship between the change of blood pressure and that of microcirculation of rat cremaster muscle during shock. It was shown that after antishock treatment the degree of capillary perfusion is closely related with the survival rate and the restoration of pulse pressure is positively correlative with the percentage of open capillaries. The injection of crystal No 4 of Polygonum cuspidatum could restore the pulse pressure to a state above normal level. With the changing of pulse pressure, a pulsatile blood movement occurred in the opening of capillary which impacted and dislodged the plugging or stationary blood cells, leading to reperfusion of capillaries and increase of survival rate.

Hepatitis related to the Chinese medicine Shou-wu-pian manufactured from Polygonum multiflorum.

Hepatitis developed in a 31-y-old pregnant Chinese woman after consumption of Shou-Wu-Pian, a proprietary Chinese medicine prepared from Polygonum multiflorum. Tests for viral hepatitis were negative and there was no evidence of other systemic disease. The herbal preparation is commonly available in the Orient and China towns in western countries. Cases of herb-induced hepatitis reported in China are reviewed.
