The Di Guglielmo syndrome: a study of 17 cases. II. Periodic-acid schiff staining of the erythroblasts.

In 17 patients with Di Guglielmo's syndrome the PAS-stained erythroblasts in bone marrow smears have been assessed by a scoring system. Most of the PAS scores were either moderately high or very high--much higher than those normally seen in disorders with benign erythropoietic hyperplasia. On the other hand, very low scores were obtained in a few cases. Whereas a high PAS score always lends strong support to a diagnosis of Di Guglielmo's syndrome, a low score, or even a negative reaction, does not exclude this diagnosis. The highest scores were obtained in cases with many pathological erythroblasts. Staining after diastase digestion confirmed that the positive PAS reaction is due to glycogen. In this test, experience showed that a period of at least 2 hours is required for complete digestion of the glycogen.

Production and partial characterization of interleukin 2 induced by periodic acid oxidation of lymphocyte membranes.

Murine splenocytes, when stimulated to undergo blastogenesis by H5IO6 oxidation, produced a lymphokine with a spectrum of properties identical to that generally ascribed to IL-2 released by lectin-stimulated cells. These included the sustained propagation of IL-2-dependent CT6 cells, as well as identical thermal and enzymatic stabilities. Cell membrane carbonyls generated in situ by the oxidation of cell membranes served as triggers for subsequent IL-2 production by the activated cells. Reduction of membrane carbonyls by NaBH4, and their addition reaction with NaHSO3 and NH2OH abrogated cell activation and inhibited, but did not abolish, IL-2 production. None of the specific carbonyl reagents, e.g., NaBH4, NaHSO3, and NH2OH, have by themselves induced blastogenic transformation, although they did elicit IL-2 production. It is therefore concluded that cell membrane carbonyls serve as triggers for IL-2 production by H5IO6-transformed cells, although an increased rate of DNA synthesis per se is not an indispensable precondition for IL-2 synthesis.

Enhancement of histological detail using metanil yellow as counterstain in periodic acid Schiff's hematoxylin staining of glycol methacrylate tissue sections.

Histological detail in sections from tissues embedded in glycol methacrylate was improved by counterstaining PAS/iron-hematoxylin stained sections with a dilute solution of metanil yellow. The addition of the counterstain increases contrast in tissue sections and highlights PAS-positive entities. The staining protocol provides sharp definition of tissue morphology, differentiates cell types and other tissue components and does not produce background staining.

Periodic acid Schiff--p phenylenediamine staining of glycogen in chondrocytes. A new combination which improves both cellular detail and glycogen identification.

This study reports a method whereby glycogen is identified in the chondrocytes of the secondary center of ossification prior to mineralization. The use of new fuchsin rather than basic fuchsin on one micron Spurr sections of femoral head cartilage fixed with potassium ferrocyanide-reduced osmium produced excellent identification of glycogen and when followed by p phenylenediamine, intensified cellular detail.

Periodic acid Schiff-positive non-glycogenic globules in hepatocytes. Differential diagnostic aspects in screening for alpha-1-antitrypsin globules in an autopsy material.

Thirty-eight subjects with diastase-resistant PAS-positive cytoplasmic globules in hepatocytes were found among 238 autopsies. In 15 of the 38 subjects the globules were antigenically alpha-1-antitrypsin, in 23 subjects they were not. The latter globules were found in centrilobular regions, the alpha-1-antitrypsin globules mainly in periportal regions. The non-alpha-1-antitrypsin globules showed less differences in size (6-10 mu) and a smaller number per hepatocyte (1-7) than the alpha-1-antitrypsin globules (1-40 mu and 1-30 per cell). The non-alpha-1-antitrypsin globules were only demonstrated in livers with centrilobular sinusoidal dilatation having, in all cases but one, also centrilobular, confluent necrosis. This type of globules can be assumed to be of differential diagnostic importance mainly in an autopsy material. The nature of these globules is discussed.

Periodic acid Schiff (PAS) positivity in fetal erythroblasts.

The smears of 26 fetal blood samples taken for prenatal diagnosis were stained for the PAS reaction. Twelve out of 26 cases were subsequently found to be normal and 14 were found to be affected by different abnormalities; 83.4% of normal cases and 92.8% of abnormal cases showed moderate positivity to the PAS reaction. A higher percentage of PAS positive erythroblasts was found in the abnormal cases (p less than 0.01). Five cases with a percentage greater than 10% also showed a very high percentage of erythroblasts. No linear correlation was found between PAS positivity and erythroblastosis either in the normal or the abnormal cases, but pooling them resulted in a correlation between PAS positivity and erythroblastosis (r = 0.536; p less than 0.005). PAS positivity was most often diffuse and scored 1+. Since erythroblastosis is considered an indirect sign of fetal liver erythropoiesis, the hepatic environment and its peculiar hemopoiesis could play a role in producing a cohort of PAS positive erythroblasts.
