We describe a method for the assay of urinary 4-hydroxy-3-methoxyphenylacetic acid (homovanillic acid), based on its separation on a microparticulate reversed-phase column and direct electrochemical detection. Patients with neuroblastoma, pheochromocytoma, and Parkinson's disease have increased amounts of this compound in their urine.

The rates of disappearance of dopamine, 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) in the brain of rats treated with the tyrosine hydroxylase inhibitor alpha-methyl-p-tyrosine were measured. Levels of dopamine. DOPAC and HVA in tissue were quantified in the striatum, nucleus accumbens and frontal cortex of the rat by means of high pressure liquid chromatography with electrochemical detection. Rats were sacrificed 1, 2 or 3 hr after the administration of alpha-methyl-p-tyrosine (200 mg/kg, i.p.). Levels of dopamine, DOPAC and HVA in tissue were logarithmically transformed, plotted against the duration of inhibition of tyrosine hydroxylase and the rate constant of the decline of dopamine and of its metabolites calculated. The rate constant of decline of DOPAC in each of brain studied was found to be greater than that for dopamine. The rate constant for the decline of dopamine was found to be greater in areas of the brain presenting the largest DOPAC/dopamine tissue ratios (frontal cortex greater than nucleus accumbens greater than striatum). The present results suggest that a substantial amount of DOPAC in brain derives from a newly formed pool of dopamine.
A rapid and simple procedure is described for the concurrent determination of 4-hydroxy-3-methoxymandelic acid (HMMA) and homovanillic acid (HVA). The acids are separated by chromatography and estimated by a scanning technique. The urinary output of HMMA and HVA over 24 h from 123 normals (80 males, 43 females) gave mean values of 3.3 +/- 0.94 and 4.9 +/- 1.3 mg, respectively. There was no significant difference in the 24 hour urinary output of HMMA and HVA between the male and femal groups.
Plasma homovanillic acid concentration was assessed in 60 young schizophrenic patients, with and without first-degree relatives with schizophrenia, before treatment, and 3 days after starting haloperidol treatment. The baseline concentration of homovanillic acid in plasma was no different in the two groups before treatment; it was, however, significantly higher in the patients with relatives than in those without relatives diagnosed of schizophrenia after 3 days of haloperidol treatment.
