Dimethoate 40% organosphosphorous pesticide toxicity in Prochilodus lineatus (Prochilodontidae, Characiformes) eggs and larvae.

Toxicity tests using early life stages of fish are of great importance in assessing risks to growth, reproduction and survival in polluted environments and are important tools for good environmental monitoring. However, a small number of standard bioassays of this type have been developed in Brazil. Curimbatá (Prochilodus lineatus) is an abundant South American characid fish of great commercial interest. It was chosen for testing different concentrations of 40% dimethoate, an organophosphate insecticide and acetylcholinesterase enzyme (AChE) inhibitor used widely in Brazil. The 48-h LC50 for eggs is higher than 16.0 microg.L-1, whereas for recently hatched larvae it was found to be significantly lower (11.81 microg.L-1, ranging between 10.23 microg.L-1 - 13.65 microg.L-1) and also significantly lower than that for eggs by a Student t-test for independent samples (p = 0.03). The 96-h LC50 for 3-day old larvae was 10.44 microg.L-1 (8.03 microg.L-1 - 13.57 microg.L-1), similar to that of recently-hatched larvae (p = 0.76). Larval mobility was also found to be reduced by this insecticide.

Antidotal effect of glucoside extracted from Astragalus membranaceus on dimethoate intoxication in guinea pigs.

Therapeutic effect of the glucoside extracted from the root of Astragalus Membranaceus (AM) was studied in guinea pigs intoxicated with 600 mg/kg doses of Dimethoate (D). Four groups of guinea pigs, each consisting of two males and two females, were treated with (a) D, (b) D + Atropine, and (c) D + AM, (d) D + Atropine + AM. The survival time of the animals increased from an average of 70 minutes without AM to an average of 235 minutes with AM. Severe changes in ECG were observed prior to respiratory distress in groups without AM, and Atropine did not modify such changes. Differently, severe ECG disorders appeared only after respiratory distress in groups treated with AM. Prolongation of the Q-T interval and changing of the T wave configuration were significantly mitigated in the AM treated animals, while, arrhythmias were minimized and postponed. Moreover, muscular fasciculation and fibrillation, seizures and secretion in the respiratory tract were also significantly reduced by AM treatment. Results have shown that AM could be a promising drug to be used after cholinergic crisis in the treatment of cardiac complications with severe organophosphate intoxication.

Exposure to pesticides in agriculture: a survey of spraymen using dimethoate in the Sudan.

There is a need for objective data on the exposure of spraymen to pesticides in agriculture in order to assess minimum requirements for practicable protection in tropical areas. To provide data, a survey was carried out in the Sudan on spraymen using dimethoate. No cholinesterase depressions were found and the calculated dose received by each man per day was within safe limits in the circumstances of this survey. Respiratory exposure was only a minor part of total exposure. The methodology of such surveys is discussed and the need for the use of a standard protocol is emphasized.

[Effect of dimethoate on the expression of c-fos gene in skeletal muscle].

To elucidate the effect of organophosphate pesticides on the expression of immediate early gene in skeletal muscle, the concentrations of c-fos mRNA and protein were measured by RT-PCR and western dot blot techniques. The result showed that c-fos mRNA and protein were significantly increased in skeletal muscle of rat after dosing with dimethoate. These results indicated that c-fos might act as transcription factor and regulate other gene expression during the early period of organophosphate intoxication.

Teratogenicity testing of dimethoate containing insecticide formulation (BI 58 EC) in chicken embryos.

BI 58 EC insecticide formulation was tested for teratogenicity in chicken embryos after the treatment of embryonated eggs. The pesticide was diluted in water to 0.8% concentration level, and the emulsion was injected into air space in a volume of 0.1 ml/egg or hen eggs were treated by the immersion technique. The morphological examinations were done on the days 13, 15 and 19 of incubation of chicken embryos. BI 58 EC produced an increased embryo mortality after the treatment which was the most expressive on day 15 of incubation. The trend of embryo weight showed similarity in the control and treated groups after both treatments. The developmental anomalies were sporadic and dose-effect dependency was not seen. Light microscopic findings exhibited a degenerative change in the liver tissue of both treated groups. In summary, the 38% dimethoate containing pesticide formulation (BI 58 EC) was toxic to the developing embryo at 0.8% concentration in our study.

Effect of Dimethoate (Rogor 30% EC) on the brain neurosecretory cells of third instar grubs of Oryctes rhinoceros L. (Coleoptera : Scarabaeidae).

The brain neurosecretory cells of III instar grubs of Oryctes rhinoceros were exposed to insecticide Dimethoate (Rogor 30% EC) in the laboratory condition. The sublethal doses (0.125, 0.25 and 0.5%) of Rogor at time intervals of 8, 16 and 24 h have produced marked changes in the structure and the secretory activities of medial and lateral neurosecretory cells. Rogor stimulates the synthetic activity of these cells at the initial stages of its action and results in the accumulation of neurosecretory materials (NSM) in the cytoplasm. The decreased neurosecretion at later stages of the action was due to its transportation through the axons before the death of treated grubs. Similarly, vacuolization, shrinking and degeneration of cells were also observed in treated grubs.

Changes in the central nervous activity of rats treated with dimethoate in combination with other neurotoxicants in different phases of ontogenesis.

Organophosphates are usually found in the environment with other pesticides and with pollutants of industrial origin can cause combined exposure involving unknown interactions between the agents. In this study, female Wistar rats were given 1/25 LD50 of dimethoate by gavage, combined with the same LD50 fractions of propoxur and cypermethrin or with arsenic (6.66 mg kg(-1)). The doses were given from day 5 to 15 of pregnancy, or that plus for the 4 weeks of lactation, or that plus 8 weeks for the male offspring after weaning. Control rats received distilled water. Electrophysiological recording was done when the male offspring reached 12 weeks of age. Spontaneous activity and evoked potentials from the somatosensory, visual and auditory cortex; and conduction velocity and absolute and relative refractory periods of the tail nerve were measured. The general trend was a shift of the spontaneous cortical activity to higher frequencies and increase in the evoked potential latency. The results showed that combined exposure to several environmental toxicants could be more harmful than the effects of each substance alone, indicating the importance of combination toxicology in modelling human effects. Furthermore, these results emphasize the importance of avoiding toxic exposures in pregnant and nursing women.

Changes in serum lipid profile and esterases of rats after sublethal daily doses of dimethoate.

Male Wistar rats were injected by dimethoate (10 mg/0.5 ml) daily for 8 successive days. Controls received the same amount of saline., A group of 5 rats were anesthetized at 0, 2, 4, 6 and 8 days of injection. Blood was withdrawn from heart. serum lipid components and 4 species of serum esterases were assayed for each group. A general decrease in the activities of serum esterases was observed. A marked decrease also was observed in lipids profile during the 8 days course of experiment.

[Behavior of serum cholinesterase and acetylcholinesterase activity in acute dimethoate poisoning].

With a patient who in suicidal intention had orally taken a larger quantity of Bi 58 EC (dimethoate) especially the behaviour of the serum cholinesterase activity and the whole blood acetylcholinesterase activity was observed over a period of 38 days and it was compared with the clinical appearance. For the serum cholinesterase activity and the acetylcholinesterase activity a three-phase course that with statistically proven adaption could be described only by a three-membered e-function could be shown. Till the 3rd day an extreme decrease of activity of 80-85 per cent occurred. Both cholinesterases provide the same information with regard to the consequences of intoxication. They are only quantitatively different from each other.

Evaluation of dimethoate (Cygon 4E) for teratogenic activity in the cat.

A commercial formulation (Cygon 4E), containing 47.3 percent dimethoate and 52.7 percent of unknown ingredients, was administered orally in single daily doses of time-mated cats between the 14th and 22nd day of pregnancy. The cats were necropsied on the 43rd day of pregnancy. Fetal values for survival, body weight, and skeletal and visceral anomalies were obtained following conventional methods. The highest dose, 12 mg/kg, was associated with polydactyl in eight of the 39 fetuses, whereas the 6 and 3 mg/kg doses produced no evidence of teratogenicity or embryotoxicity.

Suspected dimethoate toxicity in cattle.

Dimethoate, an organophosphorus insecticide, was the suspected cause of toxicosis in a group of young cattle grazing on pasture that had been sprayed 6 weeks before the onset of clinical signs. Affected animals had primarily nicotinic signs, such as muscle twitching, stiffness, weakness and paralysis, though muscarinic signs, such as diarrhea, salivation and pollakiuria, were also observed. Whole blood acetylcholinesterase activity was depressed in 3 animals. The atypical clinical syndrome and poor response to treatment with atropine and other anticholinergics may have been due to coexistent hypomagnesemia.

[Study of three biocides (dimethoate, parathion ethyl and zineb) on female rat neuro endocrinological balance (author's transl)].

The authors studied the neuro endocrinological effects of three pesticides. Two of them were organophosphorus compounds (ethyl parathion and dimethoate) and one was a carbamate (zineb). Mature female rats have been orally administered these compounds for 16 days at respectively 1/25, 1/10 and 1/10 of the LD50. The parameters recorded are: the gonadotropins, the length of estrous cycle and the weight of the anterior pituitary gland, the ovaries and the uterus. In contradistinction with the organochlorine compounds the organophosphorus did not induce any disturbances in the neuro endocrinological system, whereas the carbamate compound decreased the pituitary gonadotropins.

[Acute poisoning by Vantal (combination of dimethoate and DDT) in a group of longshoremen].

In this work the authors describe a case of acute intoxication by "VANTAL" (DDT and Dimethoate) occurred in a group of six longshoremen. The authors particularly consider the reports about the gastroenteric apparatus and the central and peripheric nervous system. It has in fact been possible to document all kinds of pathologic condition from gastritis to duodenal ulcer in all subjects; whereas only three subjects out of six showed alterations in E.E.G. and E.M.G.

EEG changes caused by dimethoate treatment in three generations of rats.

Wistar rats were continuously treated with 7.0, 10.5, 14.0, 28.0 mg/kg (1/100, 1/75, 1/50 and 1/25 LD50) of dimethoate per os including the pregnancy of dams and the lactation period of pups as well as in adulthood for three subsequent generations. The EEG of male rats of each generation was recorded at the age of 12-13 weeks. The mean frequency, mean amplitude, EEG index and the power spectrum were analysed. The data showed that the overall EEG activity of the treated rats was greater than of the controls. The mean frequency was higher, the mean amplitude and the EEG index were lower, the activity of the lower frequency wave bands decreased, that of the higher frequency wave bands increased. The changes of the mentioned EEG parameters were most pronounced in the third generation.

[Influence of chronic intoxication with phospho-organic insecticide dimethoate on the hemopoietic system in rats].

Toxic effect of insecticide dimethoate applied to 400 rats for two years in concentrations of 2 to 200 mg/kg fodder was investigated. During the last six months of observation the toxic influence of the larger doses of this preparation was manifested by a higher mortality of the animals. Neither disadvantageous influence upon the maturation of each individual haemopoietic series, nor action inducing pathologic proliferation in the haemopoietic system of rats were found.
