Evaluation of eye movement variables of Chinese and American readers. Information abstraction by Chinese and American readers.

Eye movements were recorded while competent readers of English and Chinese read in their respective languages. There were no important differences in oculometric patterns of fixation pause durations for Chinese and American readers, suggesting that cognitive aspects of reading are more important than perceptual ones in determining eye movement patterns and fixation pauses of competent readers. An analysis of undershoot saccades associated with return of the eyes from the end of one line to the beginning of the next line suggests that these occur more frequently during reading than during the performance of similar eye movements that are more perceptually determined, however, the amount of undershoot does not discriminate between readers of alphabetic and logographic texts.

A comparative study of the use of primary Chinese hamster liver cultures and genetically engineered immortal V79 Chinese hamster cell lines expressing rat liver CYP1A1, 1A2 and 2B1 cDNAs in micronucleus assays.

Liver microsome preparations (S9 mix) have been extensively used for in vitro genotoxicity studies to provide the capacity for the activation of indirect genotoxins. However, the use of S9 preparations with mammalian cell cultures has raised considerable toxicity problems which limit their use to exposure times which are only a small fraction of the cell cycle. In addition, false negative results may be obtained if reactive metabolites are unable to penetrate the cell membrane or have short half-lives. The generation and detection of a promutagen within a single cell would therefore be advantageous. To this end, we have studied the bioactivation of a panel of promutagens (benzo[a]pyrene, cyclophosphamide, 2-aminoanthracene and sterigmatocystin) in low passage Chinese hamster fibroblasts of hepatic origin (LiC2 cells) and in a series of V79 Chinese hamster cell lines genetically engineered to express rat liver cytochrome P450 cDNAs. These include strains XEM2 (expresses CYP1A1), SD1 (CYP2B1) and strains XEMd-MZ and XEMd-NH which express CYP1A2. The end point selected for study was the induction of micronuclei. The protocol incorporated a cytochalasin B-induced cytokinesis block and the enumeration of micronuclei in the resulting binucleate cells which have undergone one nuclear division following the induction of chromosome damage. Micronuclei containing whole chromosomes and chromosome fragments were distinguished by the use of CREST antibody specific for kinetochore protein as a measure for the presence of centromeres. Micronuclei were induced by the test agents in low passage liver fibroblasts and in immortal V79 cultures only in the presence of Aroclor-induced S9 preparations. The data obtained from micronucleus assays of the genetically engineered V79 cell lines demonstrated the utility of each strain for the optimal detection and quantification of the activity of the individual test compounds. Kinetochore antibody demonstrated differences in the kinetics of induction of micronuclei containing chromosome fragments and whole chromosomes with chemicals such as benzo[a]pyrene. As part of this cytogenetic study, we also conducted karyotypic analyses and spindle fidelity assays of the V79 cell lines to investigate the presence of chromosomal instabilities which may arise as a consequence of the genetic engineering procedure. Such studies represent an important quality control step in the validation of the suitability of each cell line prior to their use in genotoxicity studies.

Chinese and Americans see opposite apparent motions in a Chinese character.

The perceived direction of apparent motion can be influenced by both "top-down" factors, such as expectation, and by "bottom-up" or stimulus-driven factors, such as grouping (Tse, P., Cavanagh, P. & Nakayama, K. (1998). The role of parsing in high-level motion processing. In T. Watanabe, High-level motion processing - computational, neurobiological and psychophysical perspectives. Cambridge, MA: MIT Press). Here we report the results of a single experiment that pitted top-down cues against bottom-up cues in an apparent motion sequence over the successive strokes of a Chinese character. Although each stroke was in fact presented all at once, subjects raised in China tended to see apparent motion over a single stroke in the direction it would have taken when drawn by hand, even though bottom-up cues drive a percept of apparent motion in the opposite direction for observers unfamiliar with the Chinese language. There is therefore a learned component to motion perception arising from top-down expectations capable of overriding bottom-up cues to motion.

Conceptualizing menopause and midlife: Chinese American and Chinese women in the US.

OBJECTIVES: The purpose of this qualitative project was to describe and examine expectations and experiences of menopause and midlife in pre- and postmenopausal Chinese American and Chinese women in the United States. METHODS: Four focus groups were formed from a total of 44 women: two groups of premenopausal Chinese/Chinese American women (one conducted in English and one in Cantonese) and two groups of postmenopausal Chinese/Chinese American women (one conducted in English and one in Cantonese). Qualitative data, in the form of transcripts, were interpreted using text-based content analysis. RESULTS: The major thematic findings were: meanings of menopause are inextricably bound with meanings of midlife; the borders and timing of the menopausal transition are ambiguous; the menopausal transition represents a natural progression through the life cycle; the expectations of the premenopausal women did not match the experiences of the postmenopausal women; menopause is viewed as a marker for aging; and the menopausal transition must be prepared for and managed. CONCLUSIONS: Study findings indicate that the participants did not share the strictly medicalized view of menopause as a discrete, biological entity. Menopause was typically described as a natural transition that was virtually interchangeable with midlife. While most of the participants characterized menopause as signaling the end of fertility and virtually synonymous with old age, some women described it as a new opportunity and a second chance at life. Participants felt a sense of their own agency in addressing what they viewed as a complex life stage, the experience of which could be manipulated.

Agitation in Chinese elderly: validation of the Chinese version of the Cohen-Mansfield Agitation Inventory.

The reliability and validity of the Chinese version of the Cohen-Mansfield Agitation Inventory (CCMAI) were studied in 164 demented patients. The agitation pattern in Chinese elderly was also examined in this study. The CCMAI demonstrated high validity, test-retest reliability, and interrater reliability. Eighty-five percent of demented patients manifested one or more agitated behaviors at least once a week. Factor analysis yielded three subtypes of agitation: physically aggressive behaviors, physically nonaggressive behaviors, and verbally agitated behaviors. There was a linear upward trend in physical aggression as one progressed from one Global Deterioration Scale stage to the next. Community-living patients showed higher physically nonaggressive and verbally agitated behaviors, whereas institution residents exhibited a significantly higher level of physical aggression. The differences between residential and institutionalized patients need further research. Future study should focus not only on the behavioral disturbances; medical, psychological, and environmental factors should be taken into account to provide a more accurate profile.

The gain-of-function Chinese hamster ovary mutant LEC11B expresses one of two Chinese hamster FUT6 genes due to the loss of a negative regulatory factor.

The LEC11 Chinese hamster ovary (CHO) gain-of-function mutant expresses an alpha(1,3)fucosyltransferase (alpha(1,3)Fuc-T) activity that generates the LeX, sialyl-LeX, and VIM-2 glycan determinants and has been extensively used for studies of E-selectin ligand specificity. In order to identify regulatory mechanisms that control alpha(1,3)Fuc-T expression in mammals, mechanisms of FUT gene expression were investigated in LEC11 cells and two new, independent mutants, LEC11A and LEC11B. Northern and ribonuclease protection analyses, using probes that span the coding region of a cloned CHO FUT gene, detected transcripts in each LEC11 mutant but not in CHO cells or other gain-of-function CHO mutants that express a different alpha(1,3)Fuc-T activity. Coding region sequence analysis and alpha(1,3)Fuc-T acceptor specificity comparisons with recombinant human Fuc-TV and Fuc-TVI showed that the cloned FUT gene is orthologous to the human FUT6 gene. Southern analyses identified two closely related FUT6 genes in the Chinese hamster, whose evolutionary relationships are discussed. The blots showed that rearrangements had occurred in LEC11A and LEC11 genomic DNA, consistent with a cis mechanism of FUT6 gene activation in these mutants. By contrast, somatic cell hybrid analyses revealed that LEC11B cells express FUT6 gene transcripts due to the loss of a trans-acting, negative regulatory factor. Sequencing of reverse transcriptase-polymerase chain reaction products identified unique 5'- and 3'-untranslated region sequences in FUT6 gene transcripts from each LEC11 mutant. Northern and Southern analyses with gene-specific probes showed that LEC11A cells express only the cgFUT6A gene (where cg is Cricetulus griseus), whereas LEC11 and LEC11B cells express only the cgFUT6B gene. In LEC11A x LEC11B hybrid cells, the cgFUT6A gene was predominantly expressed, as predicted if a trans-acting negative regulatory factor functions to suppress cgFUT6B gene expression in CHO cells. This factor is predicted to be a cell type-specific regulator of FUT6 gene expression in mammals.

Attitudes toward life and death among Chinese adolescents: the Chinese version of the multi-attitude suicide tendency scale.

The psychometric properties of the Chinese version of the Multi-Attitude Suicide Tendency Scale were examined in a sample of 415 Chinese adolescents. Confirmatory factor analyses of the present data disconfirmed the original 4-factor structure. Exploratory factor analysis was then conducted to explore the underlying structure of the data set. A revised 4-factor structure, with a number of differences in item factor loadings, was obtained for the present sample. The scale also demonstrated good internal consistency and convergent validity with the Depression Self-Rating Scale and Suicidal Behaviors Questionnaire. Distinct profiles of the scale were shown on non-suicidals, suicide ideators, and suicide attempters. Discrepant findings from previous work were discussed.

Assessing Chinese older adults' suicidal ideation: Chinese version of the Geriatric Suicide Ideation Scale.

The psychometric properties of the Chinese version of the Geriatric Suicide Ideation Scale (GSIS-C) were investigated in a sample of 154 Hong Kong Chinese older adults. The internal consistency, test-retest reliability, face validity, convergent and divergent validities of the GSIS-C were assessed and all of them were well above acceptable levels. The GSIS-C demonstrated excellent internal consistency including the four subscales and the test-retest reliability of the GSIS-C was also good. In terms of convergent validity, the GSIS-C correlated significantly and positively with depression (assessed by CES-D), loneliness (assessed by Revised UCLA Loneliness Scale), and hopelessness (assessed by Beck 'Hop'elessness Scale). The divergent validity of the GSIS-C was demonstrated by the negative but significant, association between the GSIS-C and two variables including self-rated health status and life satisfaction (assessed by Life Satisfaction Inventory--Version A).

A conserved region in human and Chinese hamster X chromosomes can induce cellular senescence of nickel-transformed Chinese hamster cell lines.

Cellular senescence is the genetically programmed cessation of cellular proliferation. We have recently mapped a putative senescence gene(s) on the X chromosome of Chinese hamster embryo (CHE) cells. In the present study, we have utilized microcell-mediated chromosome transfer (microcell fusion) to test whether: (i) the human X chromosome exhibits similar genetic potential to induce senescence and (ii) the deletion or inactivation of the X-linked senescence gene(s) in CHE cells is associated with nickel-induced immortalization. A normal CHE or human X chromosome was first introduced into mouse-cell hybrids, then transferred by microcell fusion into a nickel-transformed, immortal male CHE cell line (Ni-2/TGR) with an X deletion (Xq1). Microcell fusion of the normal CHE X chromosome into tumorigenic Ni-2/TGR cells yielded senescence of all X recipient clones. The normal human X chromosome induced dominant senescence of tumorigenic Ni-2/TGR cells in only 17% of the resulting microcell hybrids (14/81). Karyotypic analyses of 13 non-senescing human X chromosome-derived microcell hybrid clones revealed that none of these clones retained the complete X. A normal CHE X chromosome induced senescence of 75% of hybrids obtained with another immortal and tumorigenic nickel-transformed male CHE cell line (Ni-6/TGR), which exhibited no visible deletion of the X chromosome, while the normal human X chromosome, only induced senescence in 19% of these hybrids. Transfer of the normal CHE or human X chromosome into spontaneously transformed and tumorigenic cell lines, CHO/TGR or V79/TGR, had little or no effect on their growth. These data suggest that both human and CHE cells possess similar X-linked genetic activities that regulate the process of cellular senescence, and that in Chinese hamster cells nickel-induced immortalization but not that of CHO or V79 cells is associated with inactivation of an X-linked senescence gene.

Immunogenetic aspects of nasopharyngeal carcinoma. IV. Increased risk in Chinese of nasopharyngeal carcinoma associated with a Chinese-related HLA profile (A2, Singapore 2).

The results of this study of 110 Singapore Chinese with nasopharyngeal carcinoma (NPC) and 91 controls confirmed the association between the occurrence of HLA antigen Singapore 2 (Sin2) and NPC in the Chinese population, and indicated that their increased risk for NPC was confined to the joint occurrence of Sin 2 and A2 antigens. These findings suggested that the genotype of importance in susceptibility to NPC is the A2-Sin 2 haplotype.

Molecular cloning of Chinese hamster dihydrofolate reductase-specific cDNA and the identification of multiple dihydrofolate reductase mRNAs in antifolate-resistant Chinese hamster lung fibroblasts.

ds cDNA from antifolate-resistant Chinese hamster lung fibroblast subline DC-3F/MQ19 was ligated to Eco RI and Sal I oligonucleotide linkers and cloned into Eco RI and Sal I digested pBR322. Transformed colonies containing dihydrofolate reductase (DHFR)-specific recombinant plasmid were identified by Grunstein Hogness assay using a Chinese hamster DHFR-specific cDNA probe. A recombinant plasmid, pDHFR6, containing a 650 bp HFR insert was isolated and analyzed. This plasmid was used as a molecular probe in a Northern blot analysis of both cytoplasmic and polysomal DHFR, poly A+ mRNAs of the DC-3F/MQ19 subline, which over-produces a 20,000d DHFR 150-fold, and DC-3F/A3 subline, which over-produces a 21,000d DHFR 170-fold. This analysis revealed the presence of three DHFR mRNA species of 1350, 2200, and 3300 nucleotides in both independently-derived cell lines. The relative abundance of each species however varied strikingly between the two cell lines.

Relations of maternal style and child self-concept to autobiographical memories in chinese, chinese immigrant, and European american 3-year-olds.

The relations of maternal reminiscing style and child self-concept to children's shared and independent autobiographical memories were examined in a sample of 189 three-year-olds and their mothers from Chinese families in China, first-generation Chinese immigrant families in the United States, and European American families. Mothers shared memories with their children and completed questionnaires; children recounted autobiographical events and described themselves with a researcher. Independent of culture, gender, child age, and language skills, maternal elaborations and evaluations were associated with children's shared memory reports, and maternal evaluations and child agentic self-focus were associated with children's independent memory reports. Maternal style and child self-concept further mediated cultural influences on children's memory. The findings provide insight into the social-cultural construction of autobiographical memory.

Alcoholism among Taiwan aborigines defined by the Chinese Diagnostic Interview Schedule: a comparison with alcoholism among Chinese.

The prevalence of alcoholism was reported to be 0.1% in an aboriginal study on Taiwan using the census survey method in the 1950s. This study adopted a modified Chinese Diagnostic Interview Schedule to determine the prevalence of DSM-III-defined alcohol abuse (AA) and alcohol dependence (AD) in the Atayal, Paiwan and Yami ethnic groups of Taiwan aborigines. Stratified random sampling was used. The sample sizes of Atayal, Paiwan and Yami were 793, 656 and 106 respectively. The prevalence rates of DSM-III-defined AA and AD were 11.6%, 11.4% and 14.2%; and 9.0%, 8.1% and 6.4% respectively. No significant difference was found between the 3 ethnic groups. These prevalence figures are significantly higher than those for Chinese. In this comparative analysis, 2 distinct etiological hypotheses are proposed for the AA and the AD.

Correlations between scores on Chinese versions of long and short forms of the Geriatric Depression Scale among elderly Chinese.

A total of 187 elderly Chinese from the community were interviewed with the Chinese versions of the Geriatric Depression Scale-the long form and the short form. The scores on a proposed new version of the short form were also calculated. Cronbach alpha for the Geriatric Depression Scale-long form was .86. The correlations were significant between scores on the long and short forms (r = .94, p < .05) and between these on the long and new short forms (r = .91, p < .05). Cronbach coefficient alpha for the short form was .77 and for the new short form .81. Results of our study indicated that responding by these elderly persons on both short forms is acceptably internally consistent in comparison with those on the original Geriatric Depression Scale.

Whoever is influential is creative: how Chinese undergraduates choose creative people in Chinese societies.

This study surveyed 994 undergraduates in Guangzhou, Hong Kong, Nanchang, Nanjing and Xian about their nomination and evaluation of the most creative people in Chinese societies as well as their valuation of collectivistic and individualistic goals of creativity. Politicians and scientists/inventors were mostly nominated and were generally rated higher on social contribution than on creativity. Artists/musicians and writers/poets were rarely nominated and were mostly rated higher on creativity than on social contribution. Collectivistic goals of creativity were significantly more valued than individualistic goals of creativity. There appears to be a social validation of creativity such that meritorious salience of creativity and social influence of the creator were greatly emphasized. Merit-based attribution of creators may have a detrimental effect on perception and promotion of general creativity in Chinese societies.

[Inducing chromosome translocation and deletions by Chinese Spring-Agilops 2C disomic addition x Chinese Spring-Elytriga 5E disomic addition].

Crossing between two disomic addition lines, Chinese Spring-E. elongata and Chinese Sping with two gametocidal chromosomes 2C (from Ae. cylindric), was carried out to investigate the function of gametocidal chromosome. After scrutinizing the meiosis of pollen mother cells (PMCs) in F1 hybrids, several results were concluded: (1) In seven of the crossing combinations, the number of univalents exceeded the expected and some trivalents and tetravalents appeared also in MI; lagging, breakage and bridge of chromosomes were observed in anaphase and telophase; considerable micronuclei formed in telophase and tetrads. These were mainly caused by the gametocidal chromosome 2C. (2) Chromosomes 6E and 7E were more susceptible to the effect of the gametocidal chromosome 2C. (3) The gametocidal chromosome 2C functioned in prophase viz. the period of forming synaptonemal complex. Four F1 lines, 5-14, 5-37, 5-67 and 5-71, were identified to be T5ES 4AST5EL 2BS, T5EL 3AS, T5ES 5BS translocation respectively by using C-banding and genome in situ hybrydization(GISH) analysis. Deletion was detected in line 5-17 (short arm of chromosome 2A), 5-27(6B), 5-18(4B and 5B), 5-72(4A) and 5-4(4B) by C-banding analysis. The statistic data showed that gametocidal chromosome could induce translocation with a high frequency of 5% and reacted on group B more efficiently than on groups A and D since translocation involving chromosome 4A, 2B, 3A, 5B and deletion involving chromosome 6B, 5B, 4B, 4A 2A according to Endo's work.

Measuring quality of life in Chinese cancer patients: a new version of the Functional Living Index for Cancer (Chinese).

INTRODUCTION: Since its translation into Chinese, the Functional Living Index for Cancer (FLIC) has not been widely received due to some of its difficulties. We modified its visual analogue scale (VAS) to an ordered categorical scale and changed some of the wording in the instrument. This study examined the measurement properties of the modified FLIC. MATERIALS AND METHODS: The modified version of FLIC and the Functional Assessment of Cancer Therapy (FACT-G Chinese version 4) were filled in by 140 patients recruited from the National Cancer Centre Singapore. The patients' FLIC scores were compared with their clinical characteristics to establish known-group validity. Convergent and divergent validity of FLIC were examined by correlation analysis with FACT-G and its sub-scales. Cronbach's alpha and relative efficiency were also examined. RESULTS: FLIC and most of its sub-scales could indicate a clear and statistically significant difference of quality of life (QOL) according to patients' performance status and treatment status. FLIC strongly correlated with FACT-G. The Physical, Psychological, and Symptoms sub-scales of FLIC converged to and diverged from FACT-G sub-scales as conceptually expected. Cronbach's alpha indicated a satisfactory level of reliability. FLIC appeared to be more efficient than FACT-G, meaning that a smaller sample size will be required for FLIC than for FACT-G to achieve the same research purpose. CONCLUSIONS: The modified version of FLIC was found to have achieved satisfactory measurement properties. This is a user-friendly alternative to the original FLIC.

[Chinese men's attitudes to life events and sexuality: prevalence of erectile dysfunction and related health concerns among Chinese men in Asian MALES study].

OBJECTIVE: The aims of Asian Men's Attitudes to Life Events and Sexuality (Asian MALES)-China study were to identify the prevalence of erectile dysfunction (ED) and related health concerns in the general male population in China, and to examine the attitudes and behaviors of men in relation to these health issues. METHODS: A total of 2055 men aged 20-75 years were included in Phase-I MALES study using a standardized questionnaire, and another 225 recruited from the sub-sample of Phase-I MALES participants with reported ED and some ED patients from other sources were involved in Phase-II Asian MALES- China study, with the prevalence of ED and men's associated attitudes as the main measure. RESULTS: The overall rate of ED in the Asian MALES-China sample was 5.9%. The rate of self-reported ED rose with age increase. A higher rate of ED was reported in men with co-morbid medical conditions and risk factors, including cardiovascular disease, hypertension, dyslipidemia, and depression, and so were the rates of these co-morbid conditions in ED patients. Data from Phase-II Asian MALES-China indicated that among men with reported ED, only 17% would seek treatment. CONCLUSION: The rate of ED is high in Chinese men and it is associated with co-morbid conditions. For Chinese men, it is necessary to know more about ED and to seek safe and effective treatment of its symptoms.
