[Study on the mass spectrometry of natural products. XI. The MIKES of fast atom bombardment mass spectra of sodium adduct ions of phenylpropanoid glycosides].

The fast atom bombardment (FAB) mass spectra and mass-selected ion kinetic energy spectra [MIKES] of sodium adduct ions [M + Na]+ of six phenylpropanoid glycosides have been studied. An intense sodium adduct appeared in their FAB mass spectra, which was more abundant than the corresponding protonated molecular ion [M + H]+ and may give more definite information about molecular weight of the sample. The MIKES of these [M + Na]+ ions provided a convenient way for sugar sequence analysis of such glycosides.

[A synthesis of heteroaromatic S- and N-beta-glycosides of N-acetylglucosamine under phase transfer conditions].

The use of crown ethers for a phase transfer-catalyzed synthesis of heteroaromatic glycosides of N-acetylglucosamine was studied. The solid-liquid system and the catalysis by 15-crown-5 were found to provide for both a 100% conversion of the alpha-D-glucosaminyl chloride peracetate and a high reaction rate. The interaction of alpha-D-glucosaminyl chloride peracetate and oxadiazole and triazole mercapto derivatives capable of thiol-thione tautomerism carried out at room temperature in acetonitrile in the presence of anhydrous potassium carbonate and crown ethers was shown to lead to both S- and N-glycosides. The structures of the compounds synthesized were confirmed by X-ray analysis and 13C and 1H NMR spectroscopy.

[Synthesis and protective activity of beta-glycosides of N-acetylmuramyl-L-alanyl-D-isoglutamine with alkylalicyclic and arylaliphatic aglycons].

The following glycosides of N-acetylmuramyl-L-alanyl-D-isoglutamine (MDP) were synthesized: beta-4-tert-butylcyclohexyl MDP, beta-2-(adamant-1-yl)ethyl MDP, beta-2,2-diphenylethyl MDP, and 3-2-(p-biphenyl)ethyl MDP. The starting peracetylated beta-N-acetylglucosaminides were prepared by the oxazoline method. They were converted into 4,6-O-isopropylidene-N-acetyl-D-muramic acids, which were coupled with L-Ala-D-Glu(NH2)OBn. The target glycopeptides were obtained after their deprotection. The stimulation of the antiinfection resistance of mice against Staphylococcus aureus by the MDP glycosides was studied. The enthalpies of hydrolysis of acyclic, monocyclic, and glycoside cyclic phosphate diesters.

The enthalpies of hydrolysis of acyclic, monocyclic, and glycoside cyclic phosphate diesters have been measured by flow microcalorimetry using a phosphohydrolase isolated from Enterobacter aerogenes as catalyst. The values so obtained (kilocalories per mol) (at 25 degrees) for sodium salts are: diethyl phosphate, minus 1.8 plus or minus 0.5; ethylene phosphate, minus 6.4 plus or minus 0.2; trimethylene phosphate, minus 3.0 plus or minus 0.2; tetramethylene phosphate, minus 2.2 plus or minus 0.1; methyl beta-D-ribofuranoside cyclic 3:5-phosphate, minus 11.1 plus or minus 0.2; methyl alpha-D-glucopyranoside cyclic 4:6-phosphate, minus 6.3 plus or minus 0.1; and cyclic adenosine 3:5-monophosphate (5-ester bond), minus 11.1 plus or minus 0.4 (10-minus 3 M Mg-2+). The enthalpy of hydrolysis of the 3-ester bond of cyclic adenosine 3:5-monophosphate (10-minus 3 M Mg-2+) has been revised to minus 11.1 plus or minus 0.2 kcal/mol from the value of minus 13.2 plus or minus 0.4 kcal/mol reported previously (greengard, p., rudolph, s.a., and sturtevant, j. m. (1969) j. biol. Chem. 244, 4798). All these values pertain to the hydrolysis of singly charged diesters to form singly charged monoesters. The data for the acyclic and monocyclic phosphodiesters are in qualitative agreement with their hydrolytic reactivities. The enthalpies measured for the hydrolysis of the glycoside cyclic phosphates cannot now be explained on the basis of their structures or reactivities; perhaps a contribution to the enthalpies by solvation or a previously unrecognized geometric strain effect may be responsible for the large exothermic enthalpies of these cyclic phosphate diesters. Changes in the heat capacity, increment Cp, for some of the hydrolytic reactions were also measured.

[Effect of multi-glycosides of tripterygii on latent autoimmune diabetes in adults in early stage].

OBJECTIVE: To observe the effect of multi-glycosides of tripterygii (MT) on latent autoimmune diabetes of adults (LADA) in early stage. METHODS: The diabetic patients were divided randomly into the control group treated with insulin alone and the treated group treated with insulin and MT. Levels of insulin, C-peptide, glutamic acid decarboxylase antibody (GADAb) and islet cell antibody (ICA) were detected and clinical features of the disease were observed. RESULTS: There was no difference between the control group and the treated group in body mass index (BMI), the occurrence of diabetic keto-acidosis and the function of liver and kidney (P >0.05). After 6 months' treatment, the positive rate of GADAb and ICA decreased, plasma levels of fasting and 2 hrs post-prandial C-peptide and also 2 hrs post-prandial true insulin in the treated group increased (P <0.01), while all the above indexes improved more significantly after 1 year's treatment (P <0.01). CONCLUSION: Compared treatment of insulin with multi-glycosides of tripterygii in early stage of LADA has better effects in relieving autoimmune injury and recovering function of pancreatic island than insulin alone.

[Synthesis and protective anti-infective action of anomeric lipophilic glycosides of N-acetylmuramyl-L-alanyl-D-isoglutamine].

Anomeric pairs of alpha- and beta-dodecyl, alpha- and beta-(1-pentylhexyl), and alpha- and beta-cyclododecyl glycosides of N-acetylmuramyl-L-alanyl-D-isoglutamine (MDP) were synthesized. The starting beta-D-glucosaminides were obtained by the oxazoline method, and the corresponding alpha-isomers, by the mercuric iodide-catalyzed glycosylation of alcohols with alpha-glucosaminyl chloride peracetate in nitromethane at -90 degrees C. No reliable differences between the stimulation of mouse resistance to the infection with Staphylococcus aureus (doses of 2, 20, and 200 microg/mouse) and Escherichia coli (doses of 0.05, 1, and 20 microg/mouse) with the MDP alpha- and beta-glycosides were found.

Cardiac glycoside poisoning involved in deaths from traditional medicines.

Autopsy cases (from all areas of South Africa except the Cape Province) are referred for chemical investigation to the Johannesburg Forensic Chemistry Laboratory of the State Health Department. Over a 1-year period in 41 autopsies where death was presumed to have been caused by a herbal medicine, the presence of cardiac glycosides was sought and was found in 44%. Most of the cases were from the Transvaal, followed by Natal. Clinical histories of the patients revealed that gastrointestinal irritation was the most common syndrome experienced after traditional medicine administration (54%). It is concluded that in patients presenting with gastro-intestinal symptoms, presumably due to poisoning by traditional medicines, cardiac glycoside poisoning should be suspected.

[Stimulation of nonspecific antibacterial resistance of mice to opportunistic gram-negative microorganisms with triterpene glycosides from Holothuroidea].

A total fraction of triterpenic glycosides from Cucumaria japonica named cucumarioside was used as a stimulator of nonspecific resistance to bacterial infections in mice. After intraperitoneal administration to mice subsequently infected with various strains of E. coli and Proteus mirabilis, cucumarioside provided survival of 40 to 90 per cent of the infected animals against 100 per cent in the control group. The protective effect directly depended on the dose of cucumarioside. It was optimal to administer the preparation 3 days before the infection. When the preparation was administered at such periods LD50 for Neisseria meningitidis BT-2 and Salmonella minnesota SF 1111 lowered 5 and 4.3 times, respectively. Therefore, the total fraction of triterpenic glycosides from Far Eastern holothuria had a marked ability to increase natural resistance of the animals to infections caused by various gram-negative organisms.

[The contraceptive activity of triterpene glycosides--the total sum of holotoxins A1 and B1 and holothurin A in an experiment].

The sum of triterpene glycosides of the Far Eastern holothurian Stichopus japonicus Selenka (holotoxins A1 and B1) possesses the contraceptive activity. Glycoside holoturin A of the tropical holothurian Holoturia atra lacks these properties. The action of the sum of holotoxins is based on the estrogenic activity which was demonstrated both in the uterotropic and vaginotropic tests. The drug under study suppresses ovulation and stimulates the contractile activity of the uterus.

The in vitro biosynthesis of dhurrin, the cyanogenic glycoside of Sorghum bicolor.

A microsomal fraction from seedlings of Sorghum bicolor (Linn) Moench has been shown to catalyze the conversion of L-tyrosine to p-hydroxymandelonitrile via p-hydroxyphenylacetaldoxime. This transformation is consistent with the general pathway for cyanogenic glycoside biosynthesis proposed on the basis of in vivo experiments. When the microsomal fraction was combined with a protein fraction from the soluble portion of the cell and uridine diphosphate glucose, it was possible to demonstrate the synthesis of the cyanogenic glycoside dhurrin from L-tyrosine.

Effect of praziquantel on the cardiotonic action of cardiac glycosides.

The effect of praziquantel on isolated left guinea-pig atria has been studied. Praziquantel caused a concentration-dependent increase in the force of contraction of the isolated left atria, an effect which may be due to an increase in Ca++ fluxes. Praziquantel antagonized the cardiotonic effect of cardiac glycosides on guinea-pig atria. This antagonism is neither due to an alteration of Ca++ concentration in the cell, nor to a direct action on Na+, K+-ATPase, nor to inhibition of the binding of cardiac glycosides to heart tissue. This interaction may involve an enhancement of K+ fluxes in cardiac cells under the influence of praziquantel.

Cytokeratin 13 contains O-glycosidically linked N-acetylglucosamine residues.

The glycosylation of human cytokeratins was investigated in cultured human keratinocytes and A431 cells by metabolic labeling with [3H]glucosamine. In the presence of tunicamycin, keratinocytes incorporated [3H]glucosamine into a vitamin A-regulated acidic 53-kDa component of the cytoskeleton which was identified as cytokeratin 13 by one- and two-dimensional immunoblotting with specific monoclonal antibodies. This cytoskeletal component was also labeled with [3H]glucosamine in A431 cells but not in KB cells, which do not express cytokeratin 13. Its labeling was resistant to tunicamycin, suggesting that [3H]glucosamine had not been incorporated into N-linked oligosaccharides. Acid hydrolysis followed by paper and ion-exchange chromatography showed that the radioactivity in electrophoretically purified cytokeratin 13 was still present as glucosamine. Radioactivity was completely removed by treatment with beta-N-acetylglucosaminidase, suggesting that it was present in terminal N-acetylglucosamine residues. The labeled carbohydrate was released by alkaline borohydride treatment and was bound by a phenylboronic acid column, indicating an O-glycosidic linkage. On Bio-Gel P-2 columns, the beta-eliminated carbohydrate co-eluted with authentic N-acetylglucosaminitol. The results indicate that cytokeratin 13 contains single residues of N-acetylglucosamine O-glycosidically linked to the polypeptide chain.
