Chemical speciation and mobility of heavy metals in municipal solid waste incinerator fly ash.

Chemical speciation is a significant factor that governs the toxicity and mobility of heavy metals in municipal solid waste incinerator fly ash. Sequential extraction procedure is applied to fractionate heavy metals (Pb, Zn, Cd, Cu, and Cr) into five defined groups: exchangeable, carbonate, Fe-Mn oxide, organic, and residual fractions. The mobility of heavy metals is also investigated with the aid of toxicity characteristic leaching procedure. In the fly ash sample, Pb is primarily presented in the carbonate (51%) and exchangeable (20%) fractions; Cd and Zn mainly exist as the exchangeable(83% and 49% respectively); Cu is mostly contained in the last three fractions (totally 87%); and Cr is mainly contained in the residual fraction (62%). Pb, Zn and Cd showed the high mobility in the investigation, thus might be of risk to the natural environment when municipal solid waste incinerator fly ash is landfilled or reutilized.

Application of ultrasonic to speciation analysis of heavy metals in soil.

In order to reduce the operation time and improve the extraction efficiency, ultrasonic energy by means of ultrasonic bath was used to the modified Tessier sequential extraction for speciation analysis of heavy metals in soil. Extractable contents of Cu, Fe, Mn, Ni, Pb and Zn were measured by atomic absorption spectroscopy (AAS). The merit of the ultrasonic extraction (UE) applied to the modified Tessier method is not only that the operation time for the first 4 fractions was reduced from ca. 18 h to 8 h, comparing with conventional extraction (CE), but also the extraction efficiency was higher. The results for both of UE and CE were consistent. The extractable Cu, Ni and Zn in the sample No. 1 were mainly associated with the third fraction (Fe-Mn oxides fraction), and fourth fraction (organic matter fraction) in the sample No.2. The extractable Fe and Mn were all mainly associated with the third fraction, and Pb the fourth fraction in both of the samples. The effects of concentration of hydroxylamine chloride on the capability for the extraction of studied metals were also studied.

Ecological factors influencing tetraploid speciation in snow buttercups (Ranunculus Adoneus): niche differentiation and tetraploid establishment.

Chromosome doubling plays an important role in generating new species of flowering plants. However, reproductive incompatibilities between newly formed tetraploid plants and their diploid progenitors are expected to create a significant barrier to the persistence and establishment of neopolyploid populations. Ecological differentiation can reduce this barrier via prezygotic isolation arising from spatial separation. Alternatively, superior viability or fecundity of neotetraploid plants might compensate for the reproductive cost of incompatible pollen from diploid neighbors. The performance of plants of both cytotypes can be assessed in their respective habitats through reciprocal transplants, although such experiments have not been used previously in the study of tetraploid speciation. We used a series of seed and seedling transplant experiments to assess ecological differentiation and competitive ability during early establishment phases for tetraploid and diploid forms of the snow buttercup (Ranunculus adoneus). At two sites, seeds from diploids and tetraploids had similar germination probabilities. Tetraploid snow buttercup seedlings had a significant growth advantage in a controlled environment chamber experiment. However, in the field diploid and tetraploid buttercup seedlings did not differ consistently in survival or growth, nor did the two cytotypes show reciprocal advantages in performance, as expected if ecological differentiation has occurred. At the seed and seedling stages, neither niche differentiation nor tetraploid competitive superiority appears sufficient to explain neotetraploid success in the presence of their diploid progenitors.

Speciational history of Australian grass finches (Poephila) inferred from thirty gene trees.

Multilocus genealogical approaches are still uncommon in phylogeography and historical demography, fields which have been dominated by microsatellite markers and mitochondrial DNA, particularly for vertebrates. Using 30 newly developed anonymous nuclear loci, we estimated population divergence times and ancestral population sizes of three closely related species of Australian grass finches (Poephila) distributed across two barriers in northern Australia. We verified that substitution rates were generally constant both among lineages and among loci, and that intralocus recombination was uncommon in our dataset, thereby satisfying two assumptions of our multilocus analysis. The reconstructed gene trees exhibited all three possible tree topologies and displayed considerable variation in coalescent times, yet this information provided the raw data for maximum likelihood and Bayesian estimation of population divergence times and ancestral population sizes. Estimates of these parameters were in close agreement with each other regardless of statistical approach and our Bayesian estimates were robust to prior assumptions. Our results suggest that black-throated finches (Poephila cincta) diverged from long-tailed finches (P. acuticauda and P. hecki) across the Carpentarian Barrier in northeastern Australia around 0.6 million years ago (mya), and that P. acuticauda diverged from P. hecki across the Kimberley Plateau-Arnhem Land Barrier in northwestern Australia approximately 0.3 mya. Bayesian 95% credibility intervals around these estimates strongly support Pleistocene timing for both speciation events, despite the fact that many gene divergences across the Carpentarian region clearly predated the Pleistocene. Estimates of ancestral effective population sizes for the basal ancestor and long-tailed finch ancestor were large (about 521,000 and about 384,000, respectively). Although the errors around the population size parameter estimates are considerable, they are the first for birds taking into account multiple sources of variance.

[Speciation analysis of mercury by capillary electrophoresis on-line coupled with inductively coupled plasma mass spectrometry based on a micro mist nebulizer and a removable interface].

A method for mercury speciation analysis was developed by CE-ICP-MS with a Micro Mist nebulizer and a removable interface. Contamination of ICP-MS detector by the rising solution was avoided with the use of a removable interface during capillary rinsing. Baseline separation of methylmercury (MeHg) and inorganic mercury (Hg2+) was obtained by CE in a 45 cm x 75 microm i.d. fused-silica capillary at 22.5 kV, while a mixture of 30 mmol(-1) H3BO3 + 10% CH3OH (pH 8.7) acted as running electrolyte. The precisions (RSD, n=5) of migration time and peak area for the mercury species were in the ranges of 3.5%-3.8% and 2.3%-5.1%, respectively. The limits of detection (3sigma) of MeHg and Hg2+ were 47 and 48 microg x L(-1), respectively.

Congruent and synchronic patterns in biogeography and speciation among seabirds, pinnipeds, and cestodes.

Congruence in biogeographic patterns among diverse assemblages of taxa indicates uniformity in the historical determinants of biotic distributions. Comparisons of host and parasite phylogenies and the elucidation of distributional area relationships are requisite components of analyses in historical biogeography. Host-parasite associations with broad geographic ranges are often archaic and have been structured largely by coevolutionary processes. In contrast, the origins and radiation of the primary cestode faunas of some seabirds (Alcataenia spp./Alcidae) and pinnipeds (Anophryocephalus spp./Phocidae and Otariidae) are associated with colonization. These young colonizing faunas, in the Holarctic Region, were influenced by a common history during the late Pliocene and Pleistocene epochs. Periodic range contraction, with isolation in refugial centers, and subsequent expansion into postglacial habitats for hosts and parasites coincided with the cyclic pattern of stadials and interstadials. During the past 2-3 million years following colonization, these dramatic climatic fluctuations strongly influenced the continuity of ecological associations in marine habitats and appear to have been the determinants of congruent and synchronic patterns of speciation among these disparate taxa of marine homeotherms and eucestodes.

[Speciation analysis of arsenic in traditional Chinese medicine].

Some of traditional Chinese medicine (TCM) contain arsenide, such as realgar. The total amount of arsenic in the TCM exceeds the limits according to related regulations. But the roles of arsenic in TCM or its side-effects depend on its species existed in those therapies, not the total amount of arsenic. Therefore, in recent years, the analysis of arsenic in TCM focuses on the species of arsenic. The present paper summarized some methods and techniques in the speciation analysis of arsenic in TCM, in order that optimal methods can be chosen and the roles of arsenic could be evaluated properly.

[Speciation of iron using capillary electrophoresis inductively coupled plasma atomic emission spectrometry].

The separation of iron species, e. g. Fe2+ -Fe3+, Fe2+ -Fe(phen)3(2+), and Fe2+ -Fe3+, by mixing with complexing reagents of o-phenanthroline and EDTA, and Fe2+ -Fe3+ with o-phenanthroline was developed by capillary electrophoresis inductively coupled plasma atomic emission spectronetry (CE-ICP-AES). The effects of electrophoresis conditions such as voltage, buffer solution pH, and complex concentration on iron species are discussed. Satisfactory separation conditions for iron speciation have been acquired. Cations and anions, containing complexing agents, do not interfere in the separation of iron species. The method has advantage to CE-UV.

[Molecular markers of nuclear DNA in the study of evolution and speciation process in an example of "Lacerta agilis complex" (Sauria: Lacertidae)].

In order to elucidate the molecular-genetic relations of some Lacerta s. str. lizard populations, subspecies and species in comparison with some other genera we used methods revealing four types of nuclear DNA markers. Among these were taxonprint, RAPD, Inter-MIR-PCR markers and also satellite DNA monomer sequences. The aim was to compare the phylogeny and systematics of this reptilian group based on morphological and molecular criteria. This problem has a general importance for understanding a speciation process. Our results show a good correlation between both approaches when genera and species levels were studied. Systematic status of five subspecies of L. agilis were supported but not in all cases, some subspecies have no meaningful genetic differences by three types of molecular markers, but all of them were differed by RAPD markers. The data confirm the subdivision of L. agilis populations into west and east clades proposed by other authors earlier on the basis of mitochondrial DNA and morphology. The population structure of one of the subspecies--L. agilis exigua, was studied on a number of populations distributed from Ural region up to Kabardino-Balkaria by IMP method. There were no significant differences among these 14 populations investigated. The data testify the rapid distribution of the species after the end of Pleistocene glaciation.

[Effect of submerging and wetting-redrying on Cd speciation and uptake by Sorghum hybrid Sudangrass in two paddy soils under spiked Cd].

Effect of continuous submerging and wetting-redrying on cadmium speciation and uptake by Sorghum hybrid Sudangrass in a Ferric-accumulic Stagnic Anthrosol and a Tipical Hapludult collected from the Taihu Lake region and the rolling downs of Yingtan, Jiangxi, China respectively, was studied by pot experiment with Cd spike in 2003. Compared to that under wetting-redrying treatment, the MgCl2-extracted pool of Cd in Huangnitu with spiked Cd of 5mg x kg(-1) and 10mg x kg(-1) was decreased from 21.8% and 28.5% to 8.0% and 16.9% while NH2OH x HCl-extracted pool increased from 17.7% and 17.3% to28.1% and 37.5% under continuous submerging. However, in the Typical Halpudult, NaAC-extracted pool of Cd decreased from 20.8% and 29.6% to 11.6% and 12.6%. Cd uptake by Sorghum hybrid Sudangrass was found in negative correlation with the MgCl2 extracted pool of Cd. Total Cd uptake was significantly reduced by the pretreatment of continuously submerging in Huangnitu.

Simple and economical method for speciation and resistotyping of clinically significant coagulase negative staphylococci.

An attempt was made to speciate 102 clinically significant isolates of coagulase negative staphylococci (CoNS) by a practical scheme adapted from various references. This scheme utilizes slide and tube coagulase test, urease test ornithine decarboxylase, novobiocin susceptibility and aerobic acid from mannose for assigning species group. Inclusion of one or two additional tests in a species group could identify the isolates to species level. Ninety eight (97%) isolates were conveniently identified as S. epidermidis (41%), S. saprophyticus (16.6%), S. haemolyticus (14.7%), S. hominis (14.7%), S. lugdunensis (4.9%), S. schleiferi (1.9%) and S. capitis (1.9%). Only four isolates were not identified to the species level, two of which were probably S. capitis subspecies ureolyticus / S. warneri / S. simulans . Antibiotic susceptibility testing showed maximum resistance to ampicillin (89%) followed by cefotaxime (59%) with no resistance to vancomycin. The increasing recognition of pathogenic potential of CoNS and emergence of drug resistance amongst them denotes the need to adopt simple laboratory procedures to identify and understand the diversity of staphylococci isolated from clinical material.

Molecular systematics of five Onchocerca species (Nematoda: Filarioidea) including the human parasite, O. volvulus, suggest sympatric speciation.

The genus Onchocerca (Nematoda: Filarioidea) consists of parasites of ungulate mammals with the exception of O. volvulus, which is a human parasite. The relationship between O. volvulus, O. ochengi and O. gibsoni remains unresolved. Based on morphology of the microfilariae and infective larvae, vector transmission and geographical distribution, O. ochengi and O. volvulus have been placed as sister species. Nevertheless, the cuticle morphology and chromosomal data (O. volvulus and O. gibsoni have n=4 while O. ochengi is n=5) suggest that O. gibsoni could be more closely related to O. volvulus than O. ochengi. Sequences from the 12S rRNA, 16S rRNA and ND5 mitochondrial genes have been used to reconstruct the phylogeny of five Onchocerca species including O. volvulus. Analyses with maximum likelihood and maximum parsimony showed that O. ochengi is the sister species of O. volvulus, in accordance with the classification based on morphology and geographical location. The separate specific status of the species O. gutturosa and O. lienalis was supported, although their phylogenetic relationship was not well resolved. The analyses indicated that the basal species was O. gibsoni, a South-East Asian and Australasian species, but this result was not statistically significant. The possible involvement of sympatric speciation in the evolution of this group of parasites is discussed.

[Chip-based capillary electrophoresis on-line coupled with atomic fluorescence spectrometry and its application to rapid speciation analysis].

Chip-based capillary electrophoresis (chip-CE) is under rapid progress due to its high efficiency, fast separation, little sample consumption and integration. Due to the short optical path (30-50 microm), the conventional UV absorbance cannot satisfy the chip detection needs. Atomic fluorescence spectrometer (AFS) is a selective and sensitive detector for hydride-forming elements. With the strong points of low cost and easy operation, AFS is attractive as an ideal detector of chip-CE for mercury, arsenic and selenium, etc. A few important practical aspects of coupling chip-CE to AFS are critically discussed. These include the design of the chip, chip-AFS interface, gas-liquid separator, AFS atomizer, and elimination of the effect of backpressure on separation. The application of the developed chip-CE-AFS to the mercury speciation analysis is highlighted.

[Application of ICP-AES to the chemical speciation of heavy metals in flyash].

Chemical speciation of seven heavy metals of flyashes in incinerator was quantitatively tested using ICP-AES. Results showed that ICP-AES procedure could carry out quick, exact and high precision experiments. RSD ratio for most detected metals was lower than 3% while few metals present a comparatively high RSD when whose content was near the detection limits. The recovery ratio was 85.7%-100.63% flyashes were found to have high content of Zn, Pb. Cd, Cu, Mn, Pb and Zn existed mostly as carbonates and were leachable, while Cr and Ni were combined to metal oxides substrates and present immobilization characteristics.

[Repetitive DNA, chromosome polymorphism and speciation of gerbils].

Restriction analysis of DNA and differential staining of chromosomes in three species of the genus Meriones: M. tristrami. M. meridianus. M. inguiculatus was carried out. High extent of homology of the genomes under study was found at the molecular and chromosomal levels. Speciation of M. tristrami followed by numerous fissions of metacentric chromosomes is assumed to be connected with appearance of the short BspRI sequence. Possible mechanisms of the mammal genome evolution are discussed.

A possible model for speciation by cytoplasmic isolation in the Culex pipiens complex.

Evidence so far available indicates that in the Culex pipiens complex there exists a genetic system with unique properties not comparable with those of the chromosomal gene system and probably involved in a mode of speciation peculiar to this complex. In a long series of backcrosses it has been shown that probably every Culex pipiens population contains a factor that is inherited through the cytoplasm, the only phenotypic expression of which is the crossability or non-crossability (incompatibility) of a given strain with other strains. That there is a whole series of such cytoplasmic factors is indicated by the twenty or so crossing types of Culex strains found throughout the world. In closed populations it seems likely that, as a result of genic diversification, a modification occurs that will eventually lead to differences in traits determined by genes.

Speciation in Yersinia.

Deoxyribonucleic acid (DNA) relatedness among members of the genus Yersinia and the biochemical reactions of these organisms were compared. These data were used to reassess speciation within the genus Yersinia, and three new species were proposed. These are Yersinia ruckeri sp. n., formerly called the redmouth bacterium; Yersinia intermedia, sp. n., formerly a rhamnose-positive, raffinose-positive, melibiose-positive biogroup within Yersinia enterocolitica; and Yersinia frederiksenii sp. n., formerly a rhamnose-positive, raffinose-negative, melibiose-negative biogroup within Y. enterocolitica.

Chromosomal speciation and folliculogenesis in the semisterile interspecific hybrids of the varying lemmings, genus Dicrostonyx.

Karyotypes of Dicrostonyx torquatus torquatus and D. vinogradovi that are very similar phenotypically differ at least by 13-14 chromosomal rearrangements including centric and tandem fusions, fissions and paracentric inversion. In their male hybrid F1 spermatogenesis is arrested on early pre-meiotic stages, the female hybrids are semisterile. Histological investigation of ovaries has demonstrated reduced number of follicles, higher percentage of atretic follicles and an acceleration of folliculogenesis in hybrids as compared with the parental forms. As a result of the acceleration, the number of mature follicles and ovulations is not significantly diminished in the hybrid females, and their semisterility is caused, to a great extent, by high prenatal loss which most likely arises from the chromosomal imbalance in F2 hybrids. Thus, the mechanism suggested by the concept of chromosomal speciation is active, but not alone in forming reproductive isolation between twin species of the varying lemming.
