Proton nuclear magnetic resonance study of the binary complex of cytochrome P450cam and putidaredoxin: interaction and electron transfer rate analysis.

A 1H nuclear magnetic resonance study of the complex of cytochrome P450cam-putidaredoxin has been performed. Isocyanide is bound to cytochrome P450cam in order to increase the stability of the protein both in the reduced and the oxidized state. Diprotein complex formation was detected through variation of the heme methyl proton resonances which have been assigned in the two redox states. The electron transfer rate at equilibrium was determinated by magnetization transfer experiments. The observed rate of oxidation of reduced cytochrome P450 by the oxidized putidaredoxin is 27 (+/- 7) per s.

Photoacoustic calorimetry of proteins.

We have described two examples of time-resolved photoacoustic calorimetry for the study of heme protein transient intermediates. Before photoacoustic calorimetry, determining thermodynamic information on short-lived intermediates was difficult. Along with being sensitive to enthalpic and volume changes, photoacoustic calorimetry can detect conformational changes in a time-resolved manner. In complex protein systems, the interpretation of the structural origins of a conformational change is sometimes difficult. Site-directed mutagenesis has been used successfully to identify the residues that play important roles in the ligand binding to both Mb and cytochrome P450cam. In both systems the hydration state of salt bridges gave rise to volume changes that were identified through mutagenesis of the residues involved. With its increasing popularity and the power of site-directed mutagenesis, time-resolved photoacoustic calorimetry is fast becoming a technique to probe conformational dynamics in proteins.

Effect of intracanal medicaments on the sealing ability of TERM.

An in vitro leakage study was performed to determine the effect of intracanal medicaments on the sealing ability of Temporary Endodontic Restorative Material (TERM). Fifty extracted mandibular molars with uniform access preparations were restored with TERM after placement of either a dry cotton pellet (control) or a pellet moistened with eugenol, formocresol, CMCP, or a paste of sodium perborate and superoxol. Multiple quantitative measurements were made on each intact specimen at five time intervals after restoration, and, in some cases, after thermocycling. Statistical analysis revealed no significant leakage in the control group, or in the groups which used eugenol, formocresol, or CMCP when they were compared with the control group. Analysis, however, did show significant leakage for the group containing the bleaching paste. The authors concluded that the sealing ability of TERM is not affected by eugenol, formocresol, or CMCP. However, it should not be used for the temporary restoration of teeth being treated by the walking bleach technique.
