Histological studies on the effect of ACTH and flavin adenine dinucleotide on the rat adrenal cortex atrophied by hypophysectomy.

The present study describes the morphological changes in the adrenals of hypophysectomized rats injected 0.03 mg of ACTH and/or 10 mg of FAD intraperitoneally for 5 successive days starting 24 hrs after the operation. The administration of only FAD to intact or hypophysectomized rats showed no morphological change in the adrenal cortex. Simultaneous administration of FAD and ACTH to the hypophysectomized rats is more effective for preventing the atrophy induced than the administration of only ACTH. In these animals, similar morphological observations to those in the control, such as the increased nuclear volume, an increase of clear cells, diminuted numbers of lipid droplets and frequent occurrence of various types of granules, were obtained. FAD seems to be effective, through coexisting with ACTH, for preventing atrophy of the adrenals induced by hypophysectomy.

[Influence of destruction of the subcommissural organ on the glomerular zone of the adrenal cortex].

Experiments were conducted on 30 rats; a study was made of the morphometric and quantitative enzymatico-chemical changes in the glomerular zone of the adrenal cortex in stereotaxic disturbance of the subcomissural organ; there was established a marked atrophy of the glomerular zone, a significant reduction of the diameter of the nuclei of the adrenocorticocytes and depression in these structures of the enzymatic activity of the energy metabolism. Histophysiological characteristics of the glomerular zone can be assessed as its hypofunctional condition and indicates an association between the functional activity of the subcommissural organ and the production of aldosterone.

Gestational changes in hamster adrenal cortex: morphometric and ultrastructural stereologic studies.

In the hamster, the weight of the adrenal glands increases during the course of gestation, with the highest value at day 5. In comparison to non-pregnant control animals, there were no changes in the volume of the zona glomerulosa (ZG) and zona fasciculata (ZF), while the volume of the zona reticularis (ZR) increased notably. The average volume of ZG-cells rose at day 5 of pregnancy and thereafter gradually decreased to that of control hamsters. A marked drop in the volume of ZF-cells was seen at days 5 and 10 of pregnancy, whereas at day 15 the cells were larger than in controls. At day 5 of pregnancy, a conspicuous increase in the cell volume was found in ZR, followed by lower values at day 10 and again higher than in control hamsters at day 15. The total number of parenchymal cells in hamster adrenal cortex increased at day 10 of gestation, then underwent a marked decrease, reaching the control value at the final day of pregnancy; this drop was mainly due to a reduction in the number of ZF-cells. The changes in the cell volume were paralleled by rather proportional changes in the volume of the mitochondrial compartment and in the quantity of smooth endoplasmic reticulum. The volume of the lipid-droplet compartment significantly rose in the course of gestation in both ZF- and ZR-cells. The cortisol output by adrenal homogenates gradually decreased during pregnancy.

Subcellular steroid distribution in the rat adrenal cortex.

The subcellular distribution of main steroids (pregnenolone, progesterone, corticosterone) and of cholesterol -- this last after correction according to exchanges occuring during differential centrifugation -- is presented in the rat adrenal cortex. The distribution patterns are not necessarily the same as established according to the localization of hydroxylating enzymes.

[Effect of cholesterol administration on the state of the adrenal cortex in severe stress].

The two weeks' effect of cholesterol on the rat adrenal cortex response under conditions of a severe stress was studied. In comparison with control, the degree of the gland activation and its alteration was found to decrease. It is suggested that the effect of cholesterol depends upon the action on the hypothalamus, and upon the adrenocortical tissue as well. The action of cholesterol on the adrenocortical tissue is bound with the cholesterol capacity to retard peroxydation reactions.

Effect of cortisol on cyclic AMP production stimulated by 1-24 ACTH in adrenal cortex membranes.

The effect of cortisol on cyclic AMP production by crude bovine adrenal cortex membrane preparations has been investigated. Results demonstrate that in the presence of cortisol, cyclic AMP production in response to 1-24 ACTH was enhanced up to a concentration of about 74 microM cortisol. At higher concentrations the effect was reversed. Cortisol had no effect on cyclic AMP production in the absence of 1-24 ACTH, and cyclic AMP production was completely inhibited when 5 mM EDTA was added to the incubation tubes with the cortisol.
