[Penetration of 35S-lipoate into cells of animals subjected to overloading with the lipoate].

Penetration of 35S-lipoate into nucleated and nucleus-free erythrocytes, isolated cells (enterocytes) and accumulating tissue preparations from intestinal mucosa was studied in presence or in absence of other vitamins. Effect on this process of overloading the penetration of 35S-lipoate into rat and pigeon erythrocytes was found to be increased; under these conditions the enterocytes bind less 35S-lipoate than in control animals. Water-soluble vitamins (nicotinate, riboflavine, thiamin and, especially, panthotenate) decreased the rate of 35S-lipoate penetration into the cells. The mixture of the vitamins caused less distinct inhibitory effect, than the panthotenate only.

Effect of hyperbaric helium on vitamin uptake and utilization by micro-organisms.

Growth responses under hyperbaric helium of various procaryotic micro-organisms to graded levels of six water-soluble vitamins were measured. Vitamins included: thiamin, riboflavin, niacin, pantothenic acid, biotin, and folic acid (folacin). Growth response of each organism under hyperbaric helium was compared with that under 80% He-20% O2 and other He-O2 gas mixtures at atmospheric pressure, and with response in air at atmospheric pressure. For five of the six vitamins, no differences in response were observed. Growth response of Streptococcus faecalis to growth-limiting concentrations of folic acid was depressed during cultivation under He-O2 at 69 ATA. It is concluded that growth response of the organism is repressed by hyperbaric helium as a result of interference with uptake or utilization of the vitamin.

The effect of tumour growth on liver pantothenate, CoA, and fatty acid synthetase activity in the mouse.

Enzymatic, and microbiological assays were used to determine the hepatic contents of coenzyme A, acetyl CoA, fatty acid synthetase activity, and pantothenate in livers of tumour-bearing mice. Significant decreases in CoA and acetyl CoA were found in mice bearing TLX-5 lymphoma, sarcoma 180 or a fibrosarcoma. These changes were accompanied by significant decreases in pantothenate and increases in 4-phosphopantothenate suggesting an increase in pantothenate kinase activity due to reduction of CoA inhibition of the enzyme. In contrast, large increases were found in pantothenate and 4-phosphopantothenate in mice bearing TLX-5 lymphoma, i.p. or s.c. These changes could be due to a large reduction in the rate of conversion of an intermediate in the pathway of CoA, or increased production of pantothenate or 4-phosphopantothenate from the degradation of CoA or the phosphopantetheine residue in fatty acid synthetase. Activities of fatty acid synthetase in liver of mice bearing this tumour showed marked decreases, but were insufficient to account for the increase in pantothenate, and may reflect a reduction in cytosolic CoA needed for the conversion of the apo to the holoenzyme.

[Development of nutrient medium for the cultivation of Lactobacillus plantarum 8R-A3 ].

Optimal nutrient medium providing accumulation of cell biomass of Lactobacillus plantarum 8R-A3 has been developed. According to this index the proposed medium meets the nutrition requirements of bacteria under study and is comparable with the basal (MRS) medium. Dependence of growth properties of Lactobacillus plantarum 8R-A3 on concentration of the medium basal components has been studied. Maize extract (6 vol.%) and enzymolysate of fodder yeast (5 vol.%) are the most favourable sources of nitrogen nutrition. Addition of pantothenic, nicotinic acids or riboflavin to the nutrient medium increased the yield of Lactobacillus plantarum biomass.
