The treatment of advanced cystadenocarcinoma of the ovary with gestronol and continuous oral cyclophosphamide.

Fifty-four patients with advanced cystadenocarcinoma of the ovary were treated with gestronol (Depostat), 200 mg/week, by intramuscular injection and continuous oral cyclophosphamide, the dose of the latter being varied to maintain the patient's white cell count at about 1.5 x 10(9)/l. A combined partial and complete remission rate of 76 per cent was obtained. The length of survival depended upon the type of remission and the type of remission was markedly affected by the amount of tumour removed at initial laparotomy.

Cyclic GMP in urine to monitor the response to ovarian cancer to therapy.

Guanosine 3':5'-phosphate (cycle GMP) in urine has been used to monitor the response of patients with ovarian cancer to treatment. Changes in the cyclic GMP level appear to correlate well with clinical status in that the disappearance of clinically detectable tumour is associated with a drop in the level whereas a tumour recurrence is associated with an elevation. Serially measured cyclic GMP is valuable for detecting a recurrence of tumour growth in patients in clinical remission and can predate any clinical signs by as much as 10 months. In patients who show no response to treatment, cyclic GMP levels in urine are elevated in the majority of specimens collected.

Use of progestogens as an adjuvant to surgery in the treatment of stage I adenocarcinoma of the uterine corpus.

Fifty-nine patients with proven stage I adenocarcinoma of the uterine corpus were treated by total abdominal hysterectomy and bilateral salpingo-oophorectomy, followed by gestronal hexanoate intramuscularly for 3 months and then medroxyprogesterone acetate orally for a prolonged period. In the 7-year period of study, there were no vaginal recurrences, but one patient suffered a recurrence in the inguinal lymph nodes and pelvis. Undesirable side effects did not occur. These results compare favourably with other reported studies in which surgery and radiotherapy were used.

Poly (epsilon-caprolactone) grafted with nano-structured chitosan enhances growth of human dermal fibroblasts.

Polyester films are modified with their bioactivity for tissue engineering by grafting a nano-structured bioactive material, nano-structured chitosan (nano-CS), on a model polymer, poly (epsilon-caprolactone) (PCL). The nano-CS was duplicated using a solvent-etched PCL mold and then grafted onto PCL using a selected solvent. The structure of the nano-CS/PCL surface was characterized using an atomic force microscope to observe the topography and determine the roughness. The centerline average roughness, Ra, of the surface of the nano-CS/PCL film is 106.0+/-4.0 nm whereas that of the surface of the CS-grafted PCL film (CS/PCL) is 3.6+/-0.4 nm. The latter is therefore very smooth. CS is known to swell following hydration, so the Ra values were determined again after immersion for 12 h in phosphate buffered saline. Although the centerline average roughness of the nano-CS/PCL was lower, it still markedly exceeded that of the CS/PCL film. Cells grown on nano-CS/PCL, CS/PCL, nano-structured PCL (nano-PCL), and PCL films were observed by fluorescent staining and analyzed by MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) viability assay following 3 and 7 days of culture, to evaluate the effects of the design on the growth of fibroblasts. The viability assay of the cells reveals that the growth rate of cells on both CS/PCL and nano-CS/PCL films significantly exceeds (P<0.001) those of PCL and nano-PCL films on both cultural days. Additionally, the growth rate and proliferation of fibroblasts on nano-CS/PCL films significantly exceed (P<0.001) those on CS/PCL films after both periods of culturing, suggesting that the bioactive surface following a nano-structured treatment promotes the growth rate of cells. However, nano-PCL films do not have the same effects as nano-CS/PCL films do. In conclusion, a novel biomaterial, nano-CS/PCL, is developed by grafting a nano-structured bioactive surface, CS, onto the PCL surface to promote the the growth rate of fibroblasts. This work elucidates a new concept for designing films or scaffolds for tissue engineering-the grafting of nano-structured bioactive biomaterials to the films or scaffolds to promote the growth of cells.

Free radical theory of aging: effect of age, sex and dietary precursors on rat-brain docosahexanoic acid.

Increasing the peroxidizability of dietary fat has an adverse effect on the function of the central nervous system (CNS) in the rat. This effect may be influenced by the level of docosahexanoic acid, a highly unsaturated fatty acid, selectively concentrated in the phospholipids of brain membranes. This study was aimed at determining the influence of age, sex, and the nature of a dietary lipid supplement - linolenic acid (18:3w3), docosahexanoic acid (22:6w3), or the same amount of 22:6w3 in the form of menhaden oil triglycerides - on the rate of increase in the percentage of 22:6w3 in the whole-brain fatty acids of rats between the ages of 1 and 12 months. The dietary lipid supplements were reflected in linear increases in the brain 22:6w3 of female rats throughout the study. Between 6 and 12 months of age, the rate of incorporation of dietary 22:6w3 and its precursors into the brain 22:6w3 of male rats dropped. At 12 months it was about half that for females in the case of 22:6w3 and menhaden oil, and about zero for 18:3w3. It is suggested that dietary 22:6w3 and its precursors may modify CNS function by altering membrane function and peroxidizability through changes in the concentration of 22:6w3 in membrane phospholipids.

Biological activities of melanotropic peptide fatty acid conjugates.

Four fatty acids (FA, palmitic, myristic, decanoic, hexanoic) were individually conjugated to the N-terminus of the alpha-MSH fragment analog, H-Asp5-His6-D-Phe7-Arg8-Trp9-Lys10-NH2. This resulted in enhanced potency of the conjugates (compared to the unconjugated melanotropin analog) as determined in the lizard skin bioassay and in the mouse melanoma cell tyrosinase bioassay. The shorter conjugates of hexanoic and decanoic acid were at least equipotent to alpha-MSH in the lizard skin bioassay, whereas the longer myristoyl and palmitoyl analogs were 100 times less active. The myristoyl and palmitoyl conjugates exhibited a "creeping" potency in the lizard skin bioassay-that is, potency of the peptides increased with time in contact with the skins. These observations may be related to the more lipid nature of these FA-conjugates. In the tyrosinase assay, the conjugates were 10-100 times more active than alpha-MSH or the unconjugated analog. Each of the FA-melanotropic peptide conjugates exhibited prolonged (residual) melanotropic activity in both the lizard skin and melanoma cell bioassays. In other words, after removal of the melanotropin conjugates from contact with the skins or cells, responses were still manifested for hours or days thereafter. As little as 1 hr of contact with melanoma cells resulted in enhanced enzyme activity as measured 48 hr later. Since the conjugates, but not H-[Asp5, D-Phe7, Lys10]alpha-MSH5-10-NH2, exhibited prolonged activity, the conversion of reversible agonists to irreversible agonists was demonstrated.

Distribution of 2-ethylhexanoic acid in mice and rats after an intraperitoneal injection.

The distribution of 2-ethylhexanoic acid (2-EHA), a new wood preservative agent was studied in mice and rats. 2-14C-EHA in rat blood, brain, liver and kidney was quantitated by liquid scintillation analysis and by wholebody autoradiography in mice. A single intraperitoneal dose of 2-14C-EHA was injected in both species. Animals were sacrificed 30 min., 2 and 6 hr after the administration of 2-14C-EHA in autoradiography experiments. The highest uptake of 2-14C-EHA was observed in the liver, kidney and blood of mice. In contrast, low uptake of 2-14C-EHA was seen in the brain. 2-14C-EHA was well detectable in the olfactory bulb and in the salivary gland. In rats, at 2 hr after administration the highest concentration of 2-14C-EHA occurred in blood (0.3% of the total dose/g tissue). The radioactivity in the liver (0.2%) and kidney (0.1%) was also relatively high. The concentration of 2-14C-EHA was low in the brain (0.02%). By 6 hr. the radioactivity had decreased rapidly and was hardly measurable at 24 hr after the administration. The results suggest that 2-EHA is rapidly cleared from the tissues.
