Immunogenetic studies of H-2 antigens of MCG 3 tumour cells.

MCG 3 is a B 10(H-2b) sarcoma induced as a solid tumour with 0.2 mg of methylcholantrene in our laboratory. The ascites form obtained was used to study the expression of H-2 antigeneic specificities in a postlabelling radioassay. H-2D.2, H-2K.33 (private specificities of H-2b) and H-2.28, H-2.35 (public specificities) were normally expressed for MCG 3 while H-2.5, H-2.29, H-2.36 (public specificities) were abnormally absent. These results were confirmed by quantitative absorption analysis using MCG 3 and positive-negative normal lymphoid cells for a particular specificity.

[Actual problems of the H blood group immunogenetics (author's transl)].

The quantity of H blood group antigen produced by the H gene product, 2-alpha-fucosyltransferase, has a key importance in the A, B and Leb antigen formation. The H blood group deficiency results always in a more or less expressed insufficiency fo the A, B and Leb antigen production. More thorough investigations of the "Bombay" and "Para-Bombay" blood groups have shown, that an absolute lack of the H antigen on the red cells of this type probably does not exist at all. The production of H antigen is only in one or both chemical forms more of less suppressed. The formation of the H antigen is under control not only of the structural gene and of its mutants, but also of the action of at least two types of regulator genes, one of them controlling the formation of the glycoprotein, the other one the glycolipid form of the H antigen. Considerations about the probable genetic background of the recessively inherited Oh or OHx, Ah, Bh and OHz, as well as of the dominant Hm phenotypes, are presented. The heterogeneity and individuality of the ABH blood groups of the same specificity is pointed out.

An immunogenetic analysis of the T-cell recognition of the major house dust mite allergen Der p 2: identification of high- and low-responder HLA-DQ alleles and localization of T-cell epitopes.

Cellular reactivity to Der p 2, a major allergen of the house dust mite (HDM) Dermatophagoides pteronyssinus, was studied in a group of 41 symptomatic HDM sensitive patients, using fresh peripheral blood mononuclear cells (PBMC) and assays of proliferation. Sixty per cent of the patients responded to Der p2, with reactivities being greater in patients with asthma as one of their clinical manifestations and also in those who had skin-test reactivity to a number of allergens. HLA-DR and -DQ serotyping was undertaken in 39 of the patients and the magnitude of T-cell proliferative responses to Der p 2 were found to be positively associated with DQ7 and negatively associated with DQ2. T-cell determinants within the Der p 2 molecule were identified by assays using a series of overlapping peptides (15- to 19-mers) spanning the entire protein. Fifty-nine per cent of the 41 HDM-sensitive patients responded to one or more of the peptides. All of the peptides were antigenic for at least one of the individuals, indicating the heterogeneity of the human repertoire reactive with Der p 2. There was a substantial variability in the number and location of epitopes recognized by T cells from the different allergic patients, the mean number per patient being 2.3 +/- 1.3 (SD). The most frequently recognized peptide was that spanning residues 111-129, being stimulatory in 66.7%, the other peptides were each recognized by between 8 to 25% of individuals. There was no correlation between the epitope recognized and the presence of particular HLA-DQ antigens.

[Ecological immunodeficiency: immunogenetic aspects of its etiology and correction].

The examinations of auto-transport Kazakh drivers have indicated that there is a significant reduction in some immunological parameters HLA-B5 and HLA-DR5-proliferative responses of lymphocytes to mitogens, production of interleukin-1 and interleukin-2, activity of NK and LAK-cells. It is suggested that these impairments occur with their long-term exposure to automobile transport effluents (ATE), since the same changes in immunological parameters were found previously in the experiments with animals exposed to ATE for a long time. Some of the detected immunoresponsive disorders are associated with the availability of definite HLA antigens, such as HLA-B5 and HLA-P5. The new immunomodulating agents thymohexin (TH) and phyto-extraction drugs C4 and C6 used in vitro substantially restored the lower functional activity of immunocompetent cells and production of cytokines (thymohexin was particularly effective). The most marked recovery was observed in the drivers with the phenotype HLA-B6+ and HLA-P5+, i.e. in persons with maximally ATE-reduced immunological parameters.

Erythrocyte antigens as immunogenetic markers of respiratory atopic diseases in Georgians.

The aim of this study was to investigate the role of erythrocyte antigens of several systems (ABO, Rh, MNSs, P, Duffy, Kell) in the predisposition to and development of respiratory atopic disease (RAD) in the Georgian population. Two hundred and ninety-three patients with RAD, 83 patients with intrinsic bronchial asthma (IBA) and 215 healthy subjects were included. Associations between blood groups and RAD (atopic bronchial asthma [ABA] and pollinosis), the severity of the disease and the type of sensitization were established, involving the ABO, MNSs and Duffy systems. The groups at higher risk for the development of RAD were carriers of phenotypes O(I), MN, O(I)MN, MNFya(-), O(I)Fya(-) and O(I)MNFya(-). Resistance to RAD was associated with phenotypes A(II), AB(IV), N, Fya(+) and B(III)N. Markers for ABA and pollinosis were also revealed. Allergy to house dust allergen was associated with phenotype MN, whereas pollen sensitization was associated with the M group of MNSs. The risk for the development of severe ABA was higher in patients with B(III), M and B(III)M phenotypes. For the patients with groups O(I) and MN, a mild to moderate degree of severity of ABA was more characteristic. In healthy Georgians, erythrocyte polymorphism was not associated with immune status. In cases of house dust allergy, erythrocyte antigen P1 was associated with high levels of IgE. In patients with RAD higher B-lymphocyte counts were associated with O(I) and lower counts with A(II).
