Olefination and group transfer reactions of an electron deficient tantalum methylidene complex.

The reactivity of an electronically unsaturated tantalum methylidene complex [TolC(NSiMe(3))(2)](2)Ta(CH(2))CH(3) supported by [TolC(NSiMe(3))(2)] amidinate ligands is described. Electrophilic addition and olefination reactions of the Ta=CH(2) functionality are reported. Alkylidene participates in group-transfer reactions not observed in sterically similar, but electronically saturated, analogues. Reactions with substrates containing unsaturated C-X (X = C, N, O) bonds yield [Ta]=X compounds and vinylated organic products; carbon-sulfur cleavage reactions to produce tantalum thioformaldehyde and tantalum sulfido complexes.

The enantioselective synthesis of elecanacin through an intramolecular naphthoquinone-vinyl ether photochemical cycloaddition.

Elecanacin, an unusual cyclobuta-fused naphthalene-1,4-dione derivative isolated from the bulbs of Eleutherine Americana Merr. et Heyne (Iridaceae) has been obtained, together with its epimer isoelecanacin, by a 2 + 2 cycloaddition resulting from irradiation of 5-methoxy-2-(2-vinyloxypropyl)naphthalene-1,4-dione. The synthesis of enantiopure elecanacin starting with (R)-propylene oxide has established the absolute configuration of the natural product and has revealed that the sample isolated from the bulbs possessed an enantiomeric excess of only 14%.

1,3-Dipolar character of 2-vinyl quinazoline 3-oxides; first and second generation cycloaddition products.

The reaction between the heteroaromatic N-oxides 1a, 1b and 1c with dimethyl acetylenedicarboxylate or methyl propiolate furnishes 1,3-benzodiazepines, the products of ring transformations of primarily formed cycloadducts. The structures of 8a and 10a have been confirmed by X-ray crystallographic analysis. The aldonitrone 1c also reacts with N-methylmaleimide and with phenyl vinyl sulfone to furnish the first examples of primary cycloaddition products from quinazoline 3-oxides.

8-vinyl reduction and chlorophyll a biosynthesis in higher plants.

A technique involving solid-phase extractions and polyethylene h.p.l.c. suitable for the routine compositional analysis of the total protochlorophyllide pool of plants is described. The resynthesis kinetics of the individual components of the pool have been studied in briefly illuminated etiolated tissue of wheat (Triticum aestivum) and cucumber (Cucumis sativus) during subsequent redarkening. The data are interpreted in terms of a precursor-product relationship between the di- and mono-vinyl analogues of protochlorophyllide during their reaccumulation in darkness. The interconversion is assumed to be catalysed by an 8-vinyl reductase, which shows greater activity in wheat than in cucumber. Analyses of the redox state of the nicotinamide nucleotide of the pool during the process are compatible with NADPH as the cofactor of the putative reductase.

Peptidyl vinyl sulphones: a new class of potent and selective cysteine protease inhibitors: S2P2 specificity of human cathepsin O2 in comparison with cathepsins S and L.

Peptidyl vinyl sulphones are a novel class of extremely potent and specific cysteine protease inhibitors. They are highly active against the therapeutically important cathepsins O2, S and L. The highest kinact/K1 values exceed 10(7)M(-1) x s(-1) for cathepsin S and 10(5)M(-1) x s(-1) for cathepsins O2 and L. To study the primary specificity site of the novel human cathepsin O2 and the effectiveness of this novel class of inhibitors, a series of peptidyl vinyl sulphones with variations in the P2 residue was synthesized. Leucine in the P2 position was proven to be the most effective residue for cathepsin O2 and also for cathepsins S and L. Cathepsins O2 and S share a decreased accessibility towards P2 hydrophobic non-branched residues such as aminohexanoic acid (norleucine), methionine and oxidized methionine, but are distinguished by their different affinity towards phenylalanine in the P2 position. In contrast, cathepsin S accepts a broader range of hydrophobic residues in its S2 subsite than cathepsins O2 and L. The primary specificity-determining subsite pocket S2 in cathepsin O2 appears to be spatially more restricted than those of cathepsins S and L.

Increased collagen synthesis and cross-link formation in the skin of rats exposed to vinyl chloride monomer.

Rats treated orally with vinyl chloride (30 mg/kg) daily for 2 years developed a syndrome which included thickening of the skin. Analysis of the skin showed a 26% increase in collagen deposition per unit weight, and a corresponding 30% increase in the levels of glycosylated lysine and hydroxylysine. There was a significant increase in the amounts of the labile borohydride reducible cross-links, such as the intermolecular hydroxylysinonorleucine (+31%) and histidinomerodesmosine (+42%) as well as the intramolecular allysine aldol cross-links (+17%). These results provide evidence that fibrosis of the skin is one of the pathological consequences of exposure to vinyl chloride.

A laboratory study of coronal microleakage using four temporary restorative materials.

AIM: The aim of this study was to compare the sealing abilities of Fermin and Canseal with the more popular temporary coronal filling materials, Cavit and Caviton. METHODOLOGY: Standardized access cavities were prepared in 160 intact human permanent molar teeth. They were divided into five groups consisting of 32 samples. The teeth were restored using one of the temporary filling materials, namely: Fermin, Canseal at two powder to liquid ratios, Caviton and Cavit. Thermal cycling and/or load cycling were applied on the samples. Assessment of microleakage utilized methylene blue dye penetration. Grading of the microleakage pattern was from 1 to 3, with 3 providing the best seal. Results were analyzed using two-way anova and by Fisher's PLSD post hoc test (P < 0.05). RESULT: Microleakage along Fermin, Caviton and Cavit samples did not go beyond Leakage Grade 2. Dye penetration into these materials was noted. This was not observed in the two groups of Canseal tested. However, the two groups of Canseal exhibited total leakage notably after being subjected to thermal cycling. There was a statistically significant difference in the microleakage scores obtained between the materials and conditions tested (P < 0.0001). CONCLUSION: Fermin was found to exhibit the best seal amongst the four materials tested followed by Caviton, and Cavit. Thermal cycling influenced the seal of certain types of temporary filling materials more than load cycling.

The interaction of glass-ionomer cements containing vinylphosphonic acid with water and aqueous lactic acid.

A glass-ionomer cement containing an acrylic acid/vinyl phosphonic acid copolymer, has been investigated for its interaction with water and with aqueous lactic acid and the results compared with those from conventional glass-ionomers based on polyacrylic acid. Cylindrical specimens (12 mm high x 6 mm diameter) were placed in 8 cm3 of aqueous lactic acid (20 mmol dm(-3); pH 2.7) for 1 week, at the end of which the pH was determined. Each specimen was then placed in a fresh 8 cm3 volume of lactic acid and the pH determined after a further week. This procedure was continued for a total of 13 weeks for each specimen. Experiments were also carried out on similar specimens exposed to 8 cm3 of water for 1 week only. Further experiments were carried out in which discs of cement were exposed to thin films (0.15 mm) of lactic acid at pH 4.5, with pH values determined at 30 s, 1, 2, 5 and 10 min. After 1 week, cements had changed the pH of the lactic acid to a mean value of 3.63 (SD 0.08) while in weeks 2-13 they changed it to a mean value of 3.31 (SD 0.11). After 1 week in aqueous lactic acid the cements had gained mass by a mean of 1.26% (SD 0.59%) compared with 1.22% (SD 0.14%) in water. After 13 weeks in lactic acid, cements had lost a mean of 2.83% (SD 0.74%) in mass. Thin films of aqueous lactic acid changed from pH 4.5-5.1 at 30 s and to a steady value of 5.6 (SD 0.3) between 2 and 10 min. These results were similar to those for conventional glass-ionomer cements based on carboxylic acid polymers. Hence, it was concluded that the presence of the vinylphosphonic acid units made no significant difference to the interaction of cements with aqueous solutions.

Serum fluoride concentrations, biochemical and histopathological changes associated with prolonged sevoflurane anaesthesia in horses.

The volatile anaesthetic sevoflurane is degraded to fluoride (F-) and a vinyl ether (Compound A), which have the potential to harm kidney and liver. Whether renal and hepatic injuries can occur in horses is unknown. Cardiopulmonary, biochemical and histopathological changes were studied in six healthy thoroughbred horses undergoing 18 h of low-flow sevoflurane anaesthesia. Serum F- concentrations were measured and clinical laboratory tests performed to assess hepatic and renal function before and during anaesthesia. Necropsy specimens of kidney and liver were harvested for microscopic examination and compared to pre-experimental needle biopsies. Cardiopulmonary parameters were maintained at clinically acceptable levels throughout anaesthesia. Immediately after initiation of sevoflurane inhalation, serum F- levels began to rise, reaching an ongoing 38-45 micromol 1(-1) plateau at 8 h of anaesthesia. Serum biochemical analysis revealed only mild increases in glucose and creatinine kinase and a decrease in total calcium. Beyond 10 h of anaesthesia mild, time-related changes in urine included increased volume, glucosuria and enzymuria. Histological examination revealed mild microscopic changes in the kidney involving mainly the distal tubule, but no remarkable alterations in liver tissue. These results indicate that horses can be maintained in a systemically healthy state during unusually prolonged sevoflurane anaesthesia with minimal risk of hepatocellular damage from this anaesthetic. Furthermore, changes in renal function and morphology observed after sevoflurane inhalation are judged minimal and appear to be clinically irrelevant; they may be the result of anaesthetic duration, physiological stressors, sevoflurane (or its degradation products) or other unkown factors associated with these animals and study conditions.

Ultrasonographic features of the endometrium and the ovaries in women on etonogestrel implant.

OBJECTIVE: To evaluate the ultrasound features of the endometrium and ovaries in women on etonogestrel implant, and to correlate these features with the bleeding pattern. METHODS: Observational study including 188 consecutive women presenting for follow-up transvaginal ultrasound examination after insertion of an etonogestrel implant contraceptive device. Thirty women had more than one follow-up examination. The bleeding pattern was considered abnormal if, in the last 3 months, there were more than five episodes of vaginal bleeding, or there was prolonged bleeding exceeding 14 consecutive days. RESULTS: At first follow-up examination, the mean age was 29.7 years and 47% of women had an abnormal bleeding pattern. Most bleeding episodes were of less intensity than menses. The mean endometrial thickness (ET) on ultrasound was 2.9 mm (standard deviation, 2.0). Ovarian follicle growth exceeding 5 mm was observed in 60% of the cases. Ovulation was demonstrated in one woman. Univariate analysis showed a positive association (P < 0.01) between ET, bleeding pattern, and bleeding intensity. Follicle growth was positively associated (P < 0.01) with ET, bleeding pattern, and interval between insertion and examination. Multivariate analysis showed that the ET was on average 1.25 mm greater in women with abnormal bleeding (P = 0.0001). The odds of finding follicle growth were 2.8 times higher (95% confidence interval, 1.2-6.2) in women presenting with a three-layer type of endometrial morphology. There was no association between the other patients' characteristics and the bleeding pattern. CONCLUSIONS: Abnormal uterine bleeding in women on etonogestrel implant was associated with follicle growth and a thicker, three-layer type of endometrium, suggesting incomplete ovarian inhibition and estrogen stimulation of the endometrium.
