Gallyas-Schiff stain for senile plaques.

Gallyas technique was modified by a direct application of Schiff's reagent after physical development, resulting in distinctive staining of amyloid deposits in argyrophilic structures. With this modified method, senile plaques in Alzheimer's disease are clearer. This method is easy to perform and suitable for routine neuropathological examination.

Silver ion triggers Ca2+ release from intracellular store sites in saponin-treated HL-60 cells.

We examined the effect of silver ion on Ca2+ mobilization from intracellular stores in permeabilized HL-60 cells using a filtration method and 45Ca2+. In HL-60 cells preloaded with Ca2+ in the presence of ATP, micromolar concentrations of AgNO3 elicited marked Ca2+ release within 1 min. The AgNO3-induced Ca2+ release was not affected by the free Ca2+ concentration in the medium. Equivalent concentrations of AgNO3 inhibited energy-dependent Ca2+ uptake as well as oxalate-supported Ca2+ uptake. In passive Ca2+ release experiments when ATP was completely depleted in the solution, AgNO3 also triggered Ca2+ release. Sulfhydryl protecting agents such as 2-mercaptoethanol, dithiothreitol, and glutathione (reduced form) blocked the AgNO3-induced Ca2+ release. From these results, we conclude that the apparent Ca2+ release induced by AgNO3 is mainly due to inhibition of the Ca2+ pump with increased permeability for Ca2+ and partly due to a direct effect on the Ca2+ release channel, probably by modification of sulfhydryl groups on these proteins.

Mercury, silver, and gold inhibition of selenium-accelerated cysteine oxidation.

In vivo, cysteine in proteins or glutathione is the major amino acid involved in sulfhydryl oxidation-reduction reactions. An in vitro model of cysteine oxidation accelerated by selenium compounds was used to study the interaction of selenocystine and sodium selenite with metal ions. The interaction of metal ions with selenium compounds inhibited cysteine oxidation. The ionic forms of three toxic soft-acid metals, mercury, silver, and gold, were the most effective inhibitors. The antiarthritic gold drugs, aurothiomalate and aurothioglucose, were of particular interest as they inhibit the activity of selenium-glutathione peroxidase. The effect of gold ligands on gold(I) inhibition of selenocystine-accelerated cysteine oxidation was tested. Sodium cyanide partially reversed inhibition and potassium iodide had no effect. Inhibition of selenium-accelerated oxidation-reduction reactions by soft-acid metal ions may be of biological relevance during toxicities or during antiarthritic gold therapy.

Multicomponent synthesis of 2-imidazolines.

[reaction: see text] A multicomponent reaction (MCR) between amines, aldehydes, and isocyanides bearing an acidic alpha-proton gives easy access to a diverse range of highly substituted 2-imidazolines. The limitations of the methodology seem to be determined by the reactivity of the isocyanide and by the steric bulk on the in situ generated imine rather than by the presence of additional functional groups on the imine. Less reactive isocyanides, for example p-nitrobenzyl isocyanide 25a, react successfully with amines and aldehydes, using a catalytic amount of silver(I) acetate. Some of the resulting p-nitrophenyl-substituted 2-imidazolines undergo air oxidation to the corresponding imidazoles. Differences in reactivity of the employed isocyanides are explained with use of DFT calculations. Difficult reactions with ketones instead of aldehydes as the oxo-compound in this MCR are promoted by silver(I) acetate as well.

An approach for fabricating self-assembled monolayer of Ag nanoparticles on gold as the SERS-active substrate.

In this paper, an approach for fabricating an active surface-enhanced Raman scattering (SERS) substrate is adopted. This approach is based on the assembling of silver nanoparticles film on gold substrate. Rhodamine 6G (R6G) and p-aminothiophenol (p-ATP) were used as probe molecules for SERS experiments, showing that this new active substrate has sensitivity to SERS response. Tapping-mode atomic force microscopy (AFM) was also used to investigate the surface morphology following the fabricating process of the active SERS substrate, which showed that large quantities of silver nanoparticles were uniformly coated on the substrate.

Evaluation of systems for quantitation of autoradiograms.

Automated systems for the evaluation of autoradiograms were compared wit the traditional method of manual counting. The automated systems included television image analysis systems, reflectance/densitometric systems, and semiautomatic digitizing pad systems. Highly automated analysis systems were considered applicable for quantitation of autoradiograms when the subcellular location of the radioactivity is not an important consideration, when background is low, and when operator interaction with the system can be kept at a minimum. In many cases the relationship between silver grains and cell compartments is important for interpretation of the results. Under these conditions, the most rapid, reproducible and reliable method would combine measurement of cellular dimensions with a digitizing pad and manual counting of silver grains by the observer. Such a systems should be connected to a computer for formatting and statistical evaluation of the data. Initial testing of a quantitative autoradiographic technique indicated that the autoradiographic slide is a reliable estimator of the radioactivity present in the tissue of experimental animals injected with tritium labeled compounds. Accuracy in counting was lost when silver became coincident. Thus, a system for the quantitation of silver grains in relation to cellular morphology requires careful selection of exposure time, computer assisted measurement of cellular dimensions, operator evaluation of silver grains and computerized statistical analysis of the data.
