[The boundary design of mandibular model by means of the three-dimensional finite element method].

OBJECTIVE: To construct the mandibular model and simulate the boundary conditions by means of the three-dimensional finite element method. METHODS: A juvenile mandible, including TMJ, was scanned transversely by CT, then a three-dimensional finite element model of mandible was constructed. At the same time, the functions of stomatologic muscles and ligaments and the functional actions of temporomandibular joint (TMJ) were simulated by means of cable elements and compressive gap elements respectively. RESULTS: The cable elements were consistent with the functions of stomatologic muscles and ligaments, and the gap elements were similar to the boundary conditions of the TMJ and teeth. CONCLUSION: Cable and gap elements are reasonable boundary conditions of the finite element model of mandible, which will improve the similarity of the model.

[The effects of fluoride-containing trace element agents on the growth of mutants of S. mutans].

OBJECTIVE: To compare the influence of 6 trace element agents containing fluoride versus 6 trace element agents not containing fluoride on the growth of mutants of S. mutans and detect the interaction between fluoride and trace elements. METHODS: Six trace element agents containing fluoride [SnF2, ZnF2, SrF2, LaF2, (NH3)2MoF8, NaF] and 6 trace element agents not containing fluoride [SnCl2, SrCl2, LaCl2, ZnAc2, (NH3)2MoO4, NaCl] were selected. The continuous anaerobic cultivating technique and the absorbency of bacteria liquid and the count of bacteria were used to assess the effects of different agents on the growth of S. mutans. RESULTS: NaF, SnF2, SnCl2, ZnF2, ZnAc2, (NH3)2MoF8 and (NH3)2MoO4 were found to have strong inhibition effects on the growth of mutant of S. mutans(P < 0.01). SnF2 had more stronger inhibition effects than SnCl2; ZnF2 had more stronger inhibition effects than ZnAc2, no significant differences were seen between (NH3)2MoF8 and (NH3)2MoO4. SnF2 and ZnF2 had most potent inhibition effects(P < 0.01). No significant differences were observed between SrF2, SrCl2, LaCl2, LaF2, NaCl and the control groups(P > 0.05). CONCLUSION: The agents containing fluoride and trace elements, such as stannum and zinc had the effects of prohibiting the growth of mutant of S. mutans, As far as caries prevention is concerned, the agents containing both fluoride and trace elements are more effective than mono-fluoride, the potential mechanism may be the synergistic action between fluoride and trace elements.

[Elemental analysis of hilar and mediastinal lymph nodes in idiopathic pulmonary fibrosis].

Elemental analysis of hilar and mediastinal lymph node tissues was performed to determine whether some inhaled atmospheric elements are implicated in the pathogenesis of idiopathic pulmonary fibrosis (IPF). IPF was divided into two types; typical and atypical, according to our criteria. Lymph nodes were selected as the study material rather than lung tissue, because inhaled elements are concentrated in the hilar and mediastinal lymph nodes, and increase in lung weight due to fibrosis may influence the estimated values of element per unit weight of lung tissue. The hilar and mediastinal lymph nodes collected from 40 IPF cases and 43 non-IPF control cases were examined to determine the concentration of 13 elements in 100 mg dry tissue, using the X-ray fluorescent analysis method. For aluminium, 50 mg dry tissue was examined by an atomic absorption analysis method. Among the 14 elements, Ni in IPF lymph nodes showed a significantly higher level than in control nodes (mean +/- SD: 8.2 +/- 21.3 ppm versus 1.1 +/- 2.1 ppm, respectively. p < 0.01). No difference in each of the examined elements was noted between smokers and non-smokers or between typical and atypical IPF cases. Exogenous elements such as Ni may play a role as triggering substances in the pathogenesis of IPF.

[Analysis and discuss of trace elements in the teeth].

OBJECTIVE: Through the analysis and discussion of some trace elements in human's teeth, many related information were offered towards the researches and the cure in teeth diseases. METHODS: Using the ICP-AES method, some trace elements such as Cu, Zn, Cr, Mn, Fe, Al and Sr were determined respectively and the interfere caused by the principal element calcium to the above elements in this determination were studied. On the other hand, the changes of these trace elements were discussed between the normal teeth and the decayed teeth and the different ages and genders. RESULTS: It was found that the content of Cu was out of and that of Al was in direct and that of others elements were in inverse proportion to one's age; the ranges of content of all determined elements in the different age groups were as follows: Sr 70-226 micrograms/g, Zn 74-157 micrograms/g, Al 37-58 micrograms/g, Fe 3.24-11 micrograms/g, Cr 3.03-6.40 micrograms/g, Mn 2-3.39 micrograms/g and Cu 0.18-1.53 micrograms/g; the content of Sr in the female's teeth was obviously and that of Fe, Mn, Zn, Al were slightly higher than in male's, whereas the contents of Cu and Cr in the two genders were mainly consistent. CONCLUSION: Trace elements in teeth are a factor to indicate sorts of teeth diseases and provide the scientific references for some researches in teeth diseases.

Perturbation of axonal elemental composition and water content: implication for neurotoxic mechanisms.

The concentration and distribution of labile elements in nerve cells is tightly regulated by multiple membrane transport processes and by binding to lipids and proteins. The multifaceted nature of elemental regulation provides numerous sites at which toxicants or disease processes might act to disrupt this regulation. Such disruption can affect cytoskeletal integrity, macromolecular synthesis, energy production, osmoregulation and other cellular processes. The possible role of perturbed elemental homeostasis in the mechanism of nerve injury caused by certain chemicals (e.g., acrylamide, 2,5-hexanedione) and neuropathic diseases (e.g., diabetes) has not been determined. To investigate this possibility, we have used electron probe x-ray micro-analysis (EPMA) to measure the distribution of elements and water in cellular compartments of myelinated axons (axoplasm, mitochondria) and glial cells (cytoplasm, myelin) in normal rat central and peripheral nervous systems. Results indicate that each compartment exhibits a characteristic composition of elements and water which might reflect function of that anatomical region or organelle. Injury-induced changes in elemental content of PNS axons and Schwann cells have been identified using several neurotoxic models (i.e., acrylamide, axotomy, diabetic neuropathy). Each type of injury initiated early alterations in element and water composition of both axons and glial cells. Compositional changes were specific and developed sequentially instead of simultaneously. Results of these studies suggest that, rather than being an epiphenomenon, altered elemental regulation might represent a primary component of many neurotoxic mechanisms.

Chemical characteristics of aerosols at coastal station in Hong Kong. II. Environmental behavior of trace elements during the April 1995 to April 1996.

An one-year of sampling aerosol program was carried out during the period of April 1995 to April 1996 at coastal region, Cape D'Aguilar, in Hong Kong. The trace element composition of aerosol particles (TSP and PM10) were measured and analyzed by using the Instrumental Neutron Activation Analysis (INAA) and Inductive Couple Plasma-Mass Spectrometry (ICP-MS) techniques. The results showed that the polluted elements such as: V, Zn, As, Pb, Cd, Sb, I, Se, Bi, with high enrichment factor (E. F) values, are derived from anthropogenic sources, while Sc, Al, Fe, Mn, Sm, La, Sr are less than 10 in E. F, these elements are related the crust and soil. The concentration of the trace elements are present seasonal variation i.e. summer low and winter high, whereas the V or noncrustal V (V*) present summer high and winter low, this phenomenon might be explained by local or regional oil combustion sources. Use three type of elemental ratio, e.g. La/Sm, Mn*/V* and Bi/Al indicate that both TSP and PM10 of La/Sm ratios are mainly related to soil and dust, Mn*/V* are probably represent fossil combustion and Bi/Al are possibly associated with refuse incineration, ferromanganese alloys and aluminum production. The varimax rotation factor analysis for trace elements was performed. By means of the absolute principal component analysis (APCA) and multiple regression, the contribution of trace elements to possible sources and overall means of trace elements were estimated. The results showed that the present station is impacted by the anthropogenic species, although the quantities are different during the summertime and wintertime.

[Characteristics analysis of micro-element contents in western Erdos soil and in Tetraena mongolica plant].

This paper studied why the ex-situ conservation of Tetraena mongolica isn't successful. Through measuring and analyzing the micro-element contents in the soil where Tetraena mongolica is naturally distributed and where Tetraena mongolica is introduced in western Erdos. The results showed that among the areas where Tetraena mongolica is native to and the ex situ protection area, the average contents of 9 micro-elements, except B, are not only obviously higher than the average values in China's soil, but also higher than the values in desert areas. The contents of all micro-elements, except Cu, in the ex situ protection area are higher than desert and the natural distribution areas of Tetraena mongolica. Among the contents of the micro-elements in Tetraena mongolica plant, Fe, Cu, Zn, B, Se and Co are quite higher than that in other plant species on steppes, and Mo is higher than that in other plant species but is only 50% to the desert plants. The correlation coefficient for the contents of micro-elements between soil and plants (including adults and seedlings) are not significant and the contents in Tetraena mongolica plant are much higher than available contents of the counterparts in soil. By comparing the contents of micro-elements in Tetraena mongolica, grassland plants on steppe and desert and the contents in soil, it can be seen that every micro-element was enriched by Tetraena mongolica during its growth. The results do not support the suggestion that the contents of micro-elements in soil is the factor restricting the growth of Tetraena mongolica in the ex situ protection area.

Active finite element analysis of skeletal muscle-tendon complex during isometric, shortening and lengthening contraction.

An active finite element model was developed to predict the mechanical behaviors of skeletal muscle-tendon complex during isometric, shortening and lengthening contraction. The active finite element was created through incorporation of a user-defined material property into ABAQUS finite element code. The active finite element is controlled by a motor element that is activated by a mathematical function. The nonlinear passive behavior of the muscle was defined by the viscoelastic elements and can be easily altered to other properties by using other elements in the material library without the need of re-defining the constitutive relation of the muscle. The isometric force-length relationship, force-strain relations of the muscle-tendon complex during both shortening and lengthening contraction and muscle relaxation response were predicted using the proposed finite element model. The predicted results were found to be in good agreement with available experimental data. In addition, the stress distribution in the muscle-tendon complex during isometric, shortening and lengthening contractions was simulated. The location of the maximum stress may provide useful information for studying muscle damage and fatigue in the future.

[Melatonin inhibits the proliferation of pituitary prolactin-secreting tumor by suppressing the enhancer elements mutation of PRL gene in the rat].

In order to investigate the molecular mechanisms of the inhibition of the proliferation of 17-beta-estradiol (E(2))-induced pituitary prolactin-secreting tumor (prolactinoma) by melatonin (MLT) in the rat, we examined the inhibitory effects of MLT on the proliferation of E(2)-induced prolactinoma of the rat and the suppressing effects of MLT on the enhancer elements mutation of PRL gene in vivo and in vitro. The results showed that the weights of prolactinomas in MLT groups, in which 0.25 mg or 0.50 mg per day per rat of MLT was administered subcutaneously at 18:00, were decreased significantly. Out of the dosage of MLT, such as 0.05, 1.00 mg and 2.00 mg per day per rat, the antitumor action of MLT is less or disappointing. Polymerase chain reaction (PCR) and DNA sequencing showed five mutations in the enhancer elements of PRL gene in prolactinoma, such as -1885 point mutation (C --> G), -1857 - -1855 substitution (ACA --> G), -1792 - -1791 insertion G, -1383 - -1382 insertion (GGTGTGTG), -1265 - -1250 deletion (GTGTGTGTGTGTGTGT). Excluding of -1885 point mutation (C --> G), the mutation in the prolactinoma treated with 0.25 mg per day per rat MLT was decreased, such as -1792 - -1791 without insertion of G, -1856 - -1855 deletion AC, -1385 - -1384 deletion TG, -1250 - -1253 deletion GTGT. Firefly luciferase reporter gene systems showed that the luminosity of enhancer elements-luciferase reporter fusion gene in normal pituitary, prolactinoma treated without or with 0.25 mg per day per rat MLT were (13448.17+/-3012.74), (161831.67+/-60996.01), and (10212.17+/-634.71) OD units. Compared with the normal pituitary, the activity of PRL gene enhancer elements in prolactinoma was increased by 11 times (P<0.001). Compared with the prolactinoma, the activity of PRL gene enhancer elements in prolactinoma treated with MLT was decreased by 93.69% (P<0.001). Analysis of the space structure of PRL gene enhancer elements showed that the bending index in prolactinoma was higher than that in prolactinoma treated with MLT, which was higher than that in the normal pituitary. These results demonstrate that one of the important molecular mechanisms of MLT inhibiting the proliferation of prolactinoma is related to the reduction of enhancer elements mutation of PRL gene. These data also suggest that MLT-induced attenuation of enhancer elements mutation of PRL gene is involved in decreasing the bending index and attenuating the higher expression of PRL gene.

Multielement analysis of biological samples by inductively coupled plasma-mass spectrometry. II. Rapid survey method for profiling trace elements in body fluids.

A rapid survey of the elements in biological materials, covering most of the elements in the periodic table, is possible by using available software for semi-quantitative analysis (SEMI-QUANT) by inductively coupled plasma-mass spectrometry. The procedure takes 5 min after sample preparation and gives results with a precision (CV) of approximately 20%. At a 10-fold dilution, 13 elements can be consistently and reliably detected in serum and 15 elements in whole-blood samples. At present the most important limitation of this method is mass overlap by polyatomic species for some elements of interest (e.g., Cr, Mn, and V). However, for the set of elements that can be reliably determined at endogenous concentrations, including Li, B, Mg, Fe, Cu, Zn, Rb, and Sr, the rapid scanning capability may be useful. Although matrix effects limit the direct interpretation of the semi-quantitative output, reasonable estimates of concentration are attainable by using matrix-matched standards or by adding a multielement standard to an aliquot from one sample in the set. We also present an example of determination of 25 elements in saliva from a patient with extensive dental work: Components of many of his dental alloys were readily identified. The method may also prove useful for screening multiple toxic exposures to heavier elements, such as Pb, Tl, Cd, and Hg.

[Mobile elements in Drosophila natural populations in Azerbaijan].

The distribution of four genome DNA fragments containing different mobile elements in chromosomes of Drosophila melanogaster individuals from Azerbaijan coast and mountain populations was studied. The average copy number of each element was shown to be approx. equal in both populations. Sites of preferential localization, where the elements were present in no less than in half of individuals could be revealed in each population for all the elements. A part of these sites coincided with regions of intercalary heterochromatin, the number of such coincidences being larger in mountain populations. The copy number of the mobile elements under study in X-chromosomes of individuals from natural populations as well as from laboratory strains was less than in autosomes. X-chromosomes of different individuals differed in mobile elements' localization more than autosomes. It was assumed that peculiarities of mobile elements' distribution in X-chromosome could reflect the effect of decondensed structure of chromatin in male X-chromosome on the transposition of mobile elements.

Repression of polyoma virus DNA replication by 5'-flanking region of mouse DNA polymerase beta gene containing transcriptional silencer elements.

Dual cis-acting silencer elements are located upstream of the mouse DNA polymerase beta gene (Yamaguchi, M., Hayashi, Y., and Matsukage, A. (1989) J. Biochem. (Tokyo) 105, 79-83). In order to examine possible involvement of transcriptional silencer elements in the regulation of DNA replication, we have utilized a transient replication system of the plasmid DNA carrying replication origin of polyoma virus DNA in mouse MOP8 cells, which is constitutively producing polyoma virus large T-antigen. The polyoma virus origin of DNA replication is composed of three cis-acting genetic elements called alpha, beta, and core, in which alpha and beta elements correspond to enhancer domains. When the 5'-flanking regions of the DNA polymerase beta gene containing silencer elements were placed at the late gene border of alpha element, they effectively repressed the DNA replication. However, when placed at the early gene border of core element, it only marginally repressed the DNA replication. These results suggest that the silencer elements at cis position repress polyoma virus DNA replication by impeding the enhancer function that activates the DNA replication.

Essential and toxic element concentrations in fresh and formalin-fixed human autopsy tissues.

The concentrations of five essential elements and six potentially toxic elements were determined in seven organs collected at autopsy from 30 human subjects. Elemental analyses were carried out by graphite furnace atomic absorption spectroscopy, and inductively coupled plasma emission spectroscopy, and inductively coupled plasma mass spectroscopy, and concentrations in fresh and formalin-fixed tissues were compared. Formalin-fixation long-term storage has little effect on most element concentrations in tissue, except for Al and Mn, which changed with prolonged storage in formalin. The kidney and liver contained the greatest concentrations of toxic elements compared with other organs, whereas the essential elements were uniformly distributed among all organs. There was no more than a 10-fold difference in the tissue concentration of the elements studied among the organs, except for the concentration of Fe in liver, and Ca and Mg in bone. We also demonstrate that these elements are homogeneously distributed in tissues.

Multiple elements within the 5' distal enhancer of the mouse heme oxygenase-1 gene mediate induction by heavy metals.

A 268-base pair 5' distal fragment, SX2, which mediates basal level and inducer-dependent activation of the mouse heme oxygenase-1 gene, contains two activator protein-1 (AP-1) binding sites (Alam, J., and Zhining, D. (1992) J. Biol. Chem. 267, 21894-21900). Mutation of both AP-1 binding elements diminishes (by 50-70%), but does not abolish, the enhancer activity of SX2 in transient expression assays, suggesting that other sequences contribute to enhancer function. Directly upstream of the AP-1 binding sites are two copies of a sequence motif, TGAGGAAAT, which resemble elements found in cellular and viral genes that are known to interact with the CCAAT/enhancer-binding protein (C/EBP) family of transcription factors. These SX2 sequences bind specifically to liver-enriched, heat-stable nuclear proteins and confer C/EBP alpha-dependent transactivation of the heterologous chloramphenicol acetyltransferase (CAT) gene. Site-directed mutagenesis of these 9-base pair elements abolishes protein binding and transactivation, establishing these sequences as functional C/EBP binding sites. Stably transfected SX2/CAT fusion genes are induced between 37- and 44-fold in mouse hepatoma, Hepa, cells and between 52- and 111-fold in mouse fibroblast L929 cells in response to CdCl2 treatment. Subfragments of SX2 lacking the AP-1 binding elements do not mediate cadmium-dependent activation of the CAT gene, whereas subfragments containing the AP-1 binding elements, but lacking the C/EBP binding sites, exhibit only partial transcriptional activity. Site-directed mutagenesis of one or more of the C/EBP and AP-1 binding sites indicates that each of these elements is required for optimal activity of the SX2 enhancer fragment. The AP-1 binding elements, however, appear to be more important for induction as constructs containing multiple copies of either of the AP-1 binding elements, but not the C/EBP binding sequences, are readily activated by CdCl2. Treatment of Hepa cells with cadmium or heme does not alter the nuclear concentration of AP-1 or C/EBP binding activity.

Genetic and molecular investigations on the endogenous mobile elements of non-drosophilid fruitflies.

A syndrome of abnormal genetic effects, resembling Drosophila hybrid dysgenesis, occurs in Ceratitis capitata when strains of different origin are mated. The pattern of abnormal traits observed appears to be the phenotypic expression of a complex interacting dysgenic system of inducer and suppressor effects; probably more than one system is activated in the crosses. This suggests that different systems of mobile elements occur in different strains and populations of C. capitata. Using a PCR primer specific to the ITR sequence of a deleted element, full length mariner elements were isolated from C. capitata, Ceratitis rosa, and Trirhithrum coffeae. Very high similarities were found in inter- and intraspecific comparisons of the elements. The majority of these elements contained deletions and frame-shifts. However, one clone Ccmar1.18, from C. capitata, was found to possess an uninterrupted ORF coding for 338 amino acids with approximately 60% similarity to the Mos1 element of Drosophila mauritiana. Database searches and phylogenetic analyses showed that the mariner elements isolated in the present study are representatives of Robertson's mellifera mariner subfamily. The copy numbers of the elements within each species are very different, ranging from about 10 in T. coffeae to 5000 in C. rosa.

Pre-mRNA processing enhancer (PPE) element increases the expression of an intronless thymidylate synthase gene but does not affect intron-dependent S phase regulation.

The pre-mRNA processing enhancer (PPE) element is an RNA sequence element derived from the intronless HSV-TK gene. Insertion of the element into the highly intron-dependent human beta-globin gene leads to efficient expression in the absence of splicing. We have analyzed the effect of the PPE element on the expression of mouse thymidylate synthase (TS) minigenes. We have previously shown that the expression of intronless TS minigenes is moderately (up to 20-fold) stimulated by the inclusion of introns. Furthermore, S phase-specific expression of TS minigenes in growth-stimulated cells depends on the presence of a spliceable intron as well as the TS promoter. The goal of our study was to determine if the PPE element would overcome the dependence on introns for efficient expression and for S phase-specific expression of transfected TS minigenes. We found that insertion of the PPE element into an intronless TS minigene partially overcame intron dependence. However, the increase in expression was much less than that observed for the intronless beta-globin gene. We also found that intronless TS or HSV-TK genes that contained the PPE element and that were driven by the TS promoter were expressed at a constant level in serum-stimulated cells. However, when an intron was included in these genes, they were expressed in an S phase-specific manner. Thus the PPE element was not able to overcome the dependence on introns for S phase-specific expression of TS minigenes.

Technical and clinical aspects of spectrometric analysis of trace elements in clinical samples.

The capabilities of ICP-MS far exceed the slow, single-element analysis of GFAAS for determination of multiple trace elements. Additionally, its sensitivity is superior to that of DCP, ICP, and FAAS. The analytic procedure for ICP-MS is relatively straightforward and bypasses the need for digestion in many cases. It enables the physician to identify the target trace element(s) in intoxication cases, nutritional deficiency, or disease, thus eliminating the treatment delays experienced with sequential testing methods. This technology has its limitations as well. The ICP-MS cannot be used in the positive ion mode to analyze with sufficient sensitivity highly electronegative elements such as fluorine, because F+ is unstable and forms only by very high ionization energy. The ICP mass spectrometers used in most commercial laboratories utilize the quadrupole mass selector, which is limited by low resolution and, thus, by the various interferences previously discussed. For example, when an argon plasma is used, selenium (m/e 80) and chromium (m/e 52) in serum, plasma, and blood specimens are subject to polyatomic and molecular ion interferences. Low-resolution ICP mass spectrometers can therefore be used to analyze many trace elements, but they are not universal analyzers. High-resolution ICP-MS can resolve these interferences, but with greater expense. With the advent of more research and development of new techniques, some of these difficulties may be overcome, making this technique even more versatile. Contamination during sample collection and analysis causes falsely elevated results. Attention and care must be given to avoid contamination. Proper collection devices containing negligible amounts of trace elements should be used. Labware, preferably plastic and not glass, must be decontaminated prior to use by acid-washing and rinsed with [table: see text] de-ionized water. A complete description of sample collection and contamination has been written by Aitio and Jarvisalo as well as by Chan and Gerson. Lutz et al observed the ranges in blood shown in Table 4. We have adopted the ranges listed in Table 5 in urines of healthy, ambulatory, and community-dwelling individuals through a limited in-house study and review of literature. In conclusion, differentiation of trace element abnormalities (primary intoxication or disease versus secondary underlying disease) can be made only by utilizing results from trace element analyses in clinical specimens, medical history, and careful observation of symptoms. Repeat analysis on a second specimen collection is recommended when contamination is suspected.

Inactivation of MED-1 elements in the TATA-less, initiator-less mouse thymidylate synthase promoter has no effect on promoter strength or the complex pattern of transcriptional start sites.

The mouse thymidylate synthase (TS) promoter is a member of a family of promoters that lack a TATA box as well as an initiator element and that initiate transcription at many sites over a broad initiation window. An element (MED-1) downstream of the initiation window of almost all promoters of this family has been proposed to be important for promoter activity, as well as for multiple start site utilization. Two consensus MED-1 elements are located downstream of the initiation window of the TS promoter. To determine the role of the MED-1 elements in the TS promoter, one or both elements were inactivated by site-directed mutagenesis and the effects on promoter function were determined. We found that inactivation of the MED-1 elements had no measurable effect on promoter strength, the boundaries of the initiation window, or the pattern of transcriptional start sites. Furthermore, inactivation of the elements did not affect the ability of the TS promoter to direct S phase-specific expression of the gene in growth-stimulated cells. We conclude that the MED-1 element does not play a significant role in TS promoter function and therefore is not an essential component of all TATA-less promoters with complex transcriptional initiation patterns.
