[Mitochondria from rat brain purified by partition in biphases].

Rat brain mitochondria and synaptosomes can be separated by partition in polyethileneglycol-Dextran biphases. Mitochondria obtained by this method show very low contamination with synaptosomes and good respiratory activity. Their respiratory activity may be stimulated by exogenous potassium when they are obtained in potassium depleted media. These mitochondria show the highest purity described till now, in terms of the lactate dehydrogenase and acetylcholinesterase activities found in their preparations.

Functions of human monocyte and lymphocyte subsets obtained by countercurrent centrifugal elutriation: differing functional capacities of human monocyte subsets.

Two monocyte subsets from the peripheral blood of healthy humans were isolated by countercurrent centrifugal elutriation (CCE), and their functional heterogeneity was assessed. Cell volume of the smaller monocyte population was (332 +/- 32 mu 3), compared with the larger monocytes (380 +/- 30 mu 3). Superoxide release by large monocytes exposed to opsonized zymosan was 1.5-fold more active than those of small monocytes. Furthermore, the production of colony-stimulating factor (CSF) and myeloperoxidase activity by the larger monocytes was 2-fold and 1.4-fold greater than that of small monocytes, respectively. Thus, CCE provides a useful means to isolate 2 subsets of monocytes with similar functional properties but quantitatively different activities. This suggests that the 2 subsets may be closely related in their developmental spectrum.

[Immune status after tetanus vaccination. Possibilities and indications for a quantitative rapid determination of tetanus antibodies in human serum].

Active and passive immunization against tetanus is one of the most used vaccination procedures in our population. Detection of size and duration of protection till now failed due to expensive methods. Counter-Immuno-Electrophoresis (CIE) is presented as a simple and non-expensive method with adequate sensitivity, and allows to obtain data concerning immunity of tetanus-immunized persons in almost any case. Rarely positive results as to antitoxin level are obtainable only by more sensitive methods. Further detection methods, and examinations of tetanus-immunity of "problem patients" showing immunofailure and of collectives immunized on various schedules will be reported later.

[Partition of cell suspensions in 2-phase systems].

The physical principles of cell suspension partition in aqueous two-polymer phase systems are considered. The partition procedures and phase composition are analyzed. The results of application of analytic and preparative two-phase partition are presented.

[Sensitivity and specifitivity of the counterimmunoelectrophoresis for determination of precipitating antibodies (author's transl)].

Counterimmunoelectrophoresis (C.I.E.) is suitable for routine-work in clinical medicine to give quantitative evidence of precipitating antibodies. Sensitivity of this method is high (lowest level being 200 to 400 ng albumin/ml serum) and range of evidence being very broad (14-15 titer steps). The direction of migrations of proteins with anodal migration (Ig) is changed by carbamylation. The experience gained in more than 3,000 investigations, concerning methods and evaluation of the results, is reported in detail. Counterimmunoelectrophoresis is superior to immunoelectrophoresis and double diffusion (Ouchterlony) due to its higher sensitivity and to RAST or RIST by reason of its lower costs.

A rapid method for repurifying partly degraded radiolabeled steroids.

This rapid method for repurifying radiolabeled steroids from degraded products is based upon differential partitioning of intact and degraded molecules between aqueous-phase solution and anhydrous diethyl ether. Results compare favorably with those by liquid-chromatographic methods.

Physico-chemical properties of Staphylococcus epidermidis and Staphylococcus saprophyticus as studied by aqueous polymer two-phase systems.

The physico-chemical surface characteristics, i.e. charge, charge density and hydrophobic interaction liability, of two strains of Staphylococcus saprophyticus were studied by the partition in aqueous polymer two-phase systems and compared to that of three strains of Staphylococcus epidermidis. The two strains of S. saprophyticus, one with and one without sheep erythrocyte agglutinating ability, were found to be negatively charged at pH 7.2 and 5.5, but exhibited a charge reduction at the lower pH. The three strains of S. epidermidis were found to carry a lower charge and lost relatively more charge at pH reduction than did S. saprophyticus. All strains of both bacterial species had a poor hydrophobic interaction liability. Incubation of one strain of S. saprophyticus with specific immune serum did not influence on hydrophobicity, but reduced charge drastically. Charge reduction was found to be associated with an immune serum fraction containing IgG and IgA.
