Inhibition of azoxymethane-induced rat colon carcinogenesis by potassium hydrogen D-glucarate.

While calcium D-glucarate was shown to inhibit chemical carcinogenesis in various animal models, the effect of potassium hydrogen D-glucarate has not been extensively investigated. In the present study, potassium hydrogen D-glucarate markedly inhibited azoxymethane (AOM)-induced colon carcinogenesis in male F344 rats. Potassium hydrogen D-glucarate (PHG) or potassium hydrogen carbonate (PHC) were administered to rats in a diet (140 mmol/kg). Continual post-initiation treatment with potassium hydrogen D-glucarate reduced both tumor incidence and multiplicity at sacrifice by ca. 60%, while PHC had no effect. amelioration of overexpression of the betaG gene in rat colon carcinomas was observed using RT-PCR and Northern blot analysis. We hypothesize that previously demonstrated conversion of PHG to D-glucaro-1,4-lactone, a potent inhibitor of beta-glucuronidase (betaG), may be responsible for this effect. The mechanism of PHG inhibition of colon carcinogenesis may also involve suppression of cell proliferation and possibly alterations in cholesterol synthesis or cholesterol metabolism to bile acids. In conclusion, PHG possesses excellent potential as a natural, apparently non-toxic inhibitor to prevent colon cancer.

Nitric oxide and NO-synthases in mammals in different functional states.

This review deals with the physical and chemical properties of nitric oxide as well as with the mechanisms and enzymes synthesizing this compound in animals including humans. The cytotoxic, vasodilatory, neuromediatory, and other properties of NO are analyzed. Polyfunctionality of NO in the norm and in pathologies of different genesis is shown. It is suggested that the analysis of the mechanisms of cyclic conversion of nitric oxide and the elucidation of the role of all NO metabolism products in living organisms would allow us to approach a more profound understanding of the NO problem in biology and medicine. It is reasonable to think the knowledge obtained in the course of the studies will permit an elaboration of the strategy and tactics of medical treatment of many diseases occurring on the background of disturbance in the mechanisms of formation and utilization of this compound.

Laparoscopic excision of retroperitoneal tumors: technique and review of the laparoscopic experience.

A technique for laparoscopic excision of benign retroperitoneal tumors, including a teratoma and two cystic lesions, is described. Laparoscopic resection of a 12-cm retroperitoneal teratoma was accomplished with the patient in the left lateral decubitus position. Medial mobilization of the ascending colon and the duodenum was required for access to the lesion. Resections of two cystic lesions (measuring 20 cm and 12 cm) were performed with the patients in the lithotomy position. The colon required medial mobilization in both cases to gain access to the cysts. Once the cysts were dissected from surrounding structures, they were punctured, and the aspirated fluid was sent for cytologic analysis. There were no complications or conversions. Mean operating time was 122 minutes (range, 80-190). Patients were discharged 1 day after surgery, requiring only nonsteroidal anti-inflammatory medications for analgesia. Retroperitoneal tumors can be resected laparoscopically with careful preoperative investigation and meticulous laparoscopic technique. A major advantage of laparoscopic resections is that the patient recovers rapidly with minimal morbidity.

[AM signals' digital demodulation and its application in physiological signals' remote transmission].

Taking advantage of the characteristics of digital systems, a new digital method is given in this article to overcome the difficulties in demodulating AM signals using digital methods. Physiological signals can't be transmitted through telephone networks because they are full of ultra-low frequency components. However, if they are modulated (such as AM), they can be transmitted to the receiving end through telephone networks. Then phaselocked loops can be employed to trace the frequency of the signals' carriers so that we can demodulate and transmit them in the end.

Detection of hypermethylated RIZ1 gene in primary tumor, mouth, and throat rinsing fluid, nasopharyngeal swab, and peripheral blood of nasopharyngeal carcinoma patient.

PURPOSE: The aims of this study are to examine the methylation status of the Fn2 RIZ1 gene in nasopharyngeal (NP) tumors and nasopharyngeal carcinoma (NPC) cell lines and to evaluate the clinical value of methylated RIZ1 gene in body fluid samples of NPC patients. EXPERIMENTAL DESIGN: Methylation status of RIZ1 was evaluated by MSP on CNE-2 and M1 cell lines, 30 tumor biopsies, and their matched body fluid samples, including mouth and throat (M & T) rinsing fluid, NP swabs, plasma, and buffy coat. Normal controls included 8 normal NP biopsies and body fluid samples from 29 healthy volunteers. Sequencing was performed on MSP products from one NP tumor and one M & T rinsing fluid. Transcription of the RIZ1 gene before and after 5-aza-2'-deoxycytidine treatment was examined on CNE-2. RESULTS: The methylated RIZ1 gene was detected in both CNE-2 and M1, 18 (60%) NP tumors, but not in any of the normal controls. Of 30 matched body fluid samples, methylated RIZ1 DNA was found in 11 (37%) NP swabs, 9 (30%) M & T rinsing fluid, 7 (23%) plasma, and 3 (10%) buffy coat samples. Sequencing analysis confirmed all cytosines to uracils conversion, excluding cytosines in CpG dinucleotides in methylated PCR products. Promoter methylation correlated with loss of RIZ1 mRNA expression, and 5-aza-2'-deoxycytidine treatment restored its expression in CNE-2. CONCLUSION: Our results suggest that promoter hypermethylation of the RIZ1 gene is commonly found in NPC. Its detection in body fluid samples of NPC patients but not in normal controls indicates that it is worth to further evaluate its clinical application in assisting screening of NPC and monitoring recurrence after treatment.

[Clinical and genetic characteristics of a Chinese family of primary pulmonary hypertension].

OBJECTIVE: To investigate the clinical and genetic characteristics of familial primary pulmonary hypertension (PPH) in Han nationality. METHODS: The clinical and laboratory features of patients of familial PPH in a family of Han nationality in Zhumadian, Henan Province, including the propositus, female, aged 37, her 29-years-old brother, and her 14-years-old daughter, were summarized. Samples of peripheral blood were collected from all family members and 100 healthy volunteers. Genomic DNA of the peripheral white blood cells was extracted from the samples. Primers for the exon 1 - 13 including the lateral intron of bone morphogenetic protein receptor-II (BMPR2) gene were designed. Then the genomic DNA was amplified by PCR. The PCR products were purified, sequenced, and compared with the sequence of normal BMPR2 gene. RESULTS: The 3 patients in this family, coming down with the illness at the ages of 35, 23, and 13 respectively, suffered from severe pulmonary hypertension and cor pulmonale with the clinical manifestations of cough, hemoptysis, heart enlargement, and cardiac function of class III. The propositus' mother came down with PPH in the age of 42 and died 1 year later. Sequence analysis showed codon 491 C-->T conversion in exon 11 in all three patients (heterozygote), which induces arginine to change to tryptophan (R491W). None BMPR2 mutation was identified in the 100 normal controls and other family members without PPH. CONCLUSION: As in the white people, the missense mutation of R491W in BMPR2 gene is also one crucial pathogenetic gene of familial PPH in Han nationality. There is no normal carrier of such genotype.

[Better listening, better information, better teaching. Lessons of our incompetence].

Short excerpts of conversations held during medical consultations permit analysis of a whole range of poorly adapted attitudes and thus demonstrate the possibility for improvement of affective contact, confidence and efficacy. It is recommended to: listen more attentively, avoid interruptions, excuses, simplifications, interpretations and premature information and to learn to repeat the flow of thoughts of the patient; inform more adequately without usage of medical terminology, abstain from confused explications, frequent change of subject, imprecision and attitudes forcing the patient into an inferior role; improve teaching of the patient by proposing distinct goals and by inviting him to find solutions to his problems by learning from errors, this without patronising and under consideration of the attitude of the patient towards his disease.

Protease-like sequence in hepatitis B virus core antigen is not involved in the cleavage processes of core protein in Escherichia coli.

A DNA fragment, coding for hepatitis core antigen (HBcAg), was amplified by polymerase chain reaction and inserted into a lambda PL promoter-derived expression vector. The recombinant plasmid was transformed into Escherichia coli and proteins produced after heat induction were analyzed. In addition to the 21 kDa HBcAg protein, several smaller related polypeptides, particularly one of 17 kDa in size, were also detected with rabbit anti-HBcAg antiserum. Whether the protease-like sequence of core protein involved in the self-cleavage process to form the 17 kDa polypeptide was investigated by a deletion experiment. Our results with a mutant in which 7 amino acids of the conserved protease-like region in the core protein have been deleted suggest that the cleavage does not depend on the presence of these protease-like sequence. In addition, the core protein synthesized from in vitro translation reaction was not cleaved. Core particles from E. coli lysate were purified by sucrose and cesium chloride density gradient centrifugations and subsequently treated with 0.2% of SDS and 0.2% of beta-mecaptoethanol. Immunoblotting analysis, however, did not reveal any conversion of the 21 kDa protein to smaller ones. In conclusion, our results suggest that the protease-like domain at the N-terminus of the core protein does not contain intrinsic autocleavage activity, nor could the HBcAg be converted to smaller antigens by detergent treatment.

Relationship between inducible H-2 expression and the immunogenicity of murine skin neoplasms. I. Evidence that the immunogenicity of ultraviolet radiation-chemically induced tumors is associated with their susceptibility to gamma-interferon-mediated enhancement of H-2Kk expression.

Our objective was to determine the relationship between major histocompatibility complex class I molecule expression and the tumorigenic properties of cutaneous neoplasms induced by ultraviolet radiation or chemical carcinogens. All tumors tested were found to express low constitutive levels of MHC class I molecules in vitro as determined by indirect immunofluorescence and flow cytometry. Those tumors capable of growth in UVR-exposed but not in normal recipients (regressors) were found to express enhanced levels of H-2Kk following incubation in the presence of gamma-IFN. In contrast, only one of the tumors that were capable of growth in normal recipients (progressors) exhibited more than moderate enhancement of H-2Kk expression in response to gamma-IFN. Analysis of tumor variants obtained by conversion of a UVR-induced regressor tumor to the progressor phenotype by passage through sublethally gamma-irradiated hosts, or the generation of regressor tumors by mutagen exposure of a benz [A] pyrene (BAP) induced progressor tumor, further supported the direct relationship between tumor immunogenicity in vivo and the capacity to elevate H-2Kk expression in response to gamma-IFN. No correlation existed between H-2Dk expression by the tumors and their transplantation phenotype. Furthermore, we failed to observe MHC class II expression by any of the tumors tested. Finally, the growth rate of a regressor tumor implanted into UVR-exposed hosts was significantly reduced if the tumor was pretreated with gamma-IFN in vitro prior to inoculation. This result suggests that UVR-exposed animals may be deficient in their ability to enhance the expression of MHC class I molecules on developing tumors. This alteration may, in part, account for the state of tumor susceptibility caused by UVR exposure.

New sensitive assay for phosphatidylserine decarboxylase based on the detection of CO2 from nonradiolabeled phosphatidylserine.

A new rapid assay for phosphatidylserine decarboxylase, which is sensitive in the nanomolar range, is described. Synthesis of radiolabeled phosphatidylserine for use as a substrate is not required, since the assay, unlike previous ones, is based on the detection of CO(2) liberated from unlabeled phosphatidylserine. The assay employs a gas chromatographic procedure for the analysis of methane formed by catalytic conversion of the CO(2) produced as a product of the enzymatic reaction.

Arachidonic acid epoxides. Demonstration through [18O]oxygen studies of an intramolecular transfer of the terminal hydroxyl group of (12S)-hydroperoxyeicosa-5,8,10,14-tetraenoic acid to form hydroxyepoxides.

Purified 12-hydroperoxyeicosa - 5, 8, 10, 14 - tetrae noic acid (12-HPETE) containing deuterium atoms at 5, 6, 8, 9, 11, 12, 14, and 15 was prepared by incubating octadeuterated arachidonic acid with a platelet preparation in air or [18O]oxygen gas. A mixture of 12-HPETE containing 16O16O:18O18O (5:4) was subsequently prepared and incubated with hematin in phosphate buffer (pH 7.4); in another experiment the same mixture of 12-HPETE was incubated with a rat lung preparation (0-30% ammonium sulfate) which lacked epoxide hydratase activity. Gas chromatography-mass spectrometry negative ion chemical ionization analysis of the extracted products after conversion into pentafluorobenzyl-trimethylsilyl derivatives indicated that the products from both incubations, i.e. 12-hydroxyeicosa - 5,8,10,14 - tetraenoic acid (12 - HETE), 8 - hydroxy - 11,12-epoxyeicosa - 5,9,14 - trienoic acid (8H-11,12-EPETE), 10H - 11,12-epoxyeicosa - 5,8,14 - trienoic acid (10H-11,12-EPETE), and 8,11,12-trihydroxyeicosa-5,9,14-trienoic acid, all retained either two [16O]oxygen or two [18O]oxygen atoms of starting 12-HPETE and that no mixed species existed which contained one [16O]oxygen and one [18O]oxygen atom. These results demonstrate for the first time in the arachidonic acid series an intramolecular transfer of the terminal hydroxyl group of the hydroperoxide of 12-HPETE to the C-8 or C-10 alkyl positions to form the hydroxyepoxides 8H-11,12-EPETE and 10H-11,12-EPETE. This reaction carried out by both hematin in the absence of protein and the rat lung preparation is suggestive of a metal-hydroperoxide-olefin "cage" complex facilitating a concerted reaction in which the terminal hydroxyl group of the hydroperoxide is trapped by alkyl free radical centers.

Effect of sampling frequency on EMG power spectral characteristics.

During digitization of surface EMG, the signal is converted by an Analog-to-Digital (A/D) convertor at a specific sampling rate. This sampling rate should be selected cautiously because a high rate may include noise signals and a low rate may lead to errors. The present study was undertaken to show the effect of sampling rate on EMG power spectrum. The study was carried out on nine subjects and EMG record was done from deltoid muscle at sampling rates of 4, 6, 8 and 10 KHz during maximal voluntary contraction. Biopotential coupler with a response range of 5 Hz to 2 KHz was used during recording of the EMG signal which was processed later by Fast Fourier Transform software. The study showed a linear relationship between the sampling rate and the mean power frequency (MPF). But there was no distinct effect of sampling rate on mean and root mean square (RMS) of the digitized signal.

Noninvasive determination of bone stiffness for distraction osteogenesis by quantitative computed tomography scans.

Apparent density of canine tibial specimens was correlated to measured quantitative computed tomography (QCT) numbers, and a conversion equation was derived. Finite element analysis models of canine tibiae were constructed using spatial coordinates of converted QCT data. Two QCT-derived finite elemental analysis models were constructed for a pair of matched tibiae: one lengthened 15% by distraction osteogenesis and one contralateral control. Two equations relating the modulus of elasticity to apparent density were used. These models were then verified mechanically by measuring strains of the two specimens under applied load. Equation I correlated best with measured strains in the less-dense regenerate bone of the distracted tibia (R2 = 0.914, p = 0.0028, n = 6); Equation II performed best in the denser control tibia (R2 = 0.820, p = 0.0129, n = 6). These results demonstrate that the stiffness of tubular bones, including cortical and heterogenous cancellous structure, can be accurately predicted by a noninvasive QCT scan.

[Nutrition of juvenile prawn Macrobrachium carcinus (Crustacea: Decapoda) with diets of vegetable and marine residues].

Juvenile prawn Macrobrachium carcinus were fed two different diets: restaurant by-products (diet I) and fish and vegetable market by-products (diet II). These diets were evaluated by proximal analysis, assimilation efficiency and the factor conversion rate (FCR). Diet I registered a higher efficiency, but there was no difference in the growth rate. The growth mean (G. L.) for three months was 0.254 +/- 0.13 cm (diet I) and 0.191 +/- 0.1 cm (diet II). The conversion rate was good for both, suggesting that 6 to 7 kg of food are needed to obtain 1 kg of prawn. Survival was 76% and 100% for diets I and II, respectively.

Repeat-induced point mutation and the population structure of transposable elements in Microbotryum violaceum.

Repeat-induced point mutation (RIP) is a genome defense in fungi that hypermutates repetitive DNA and is suggested to limit the accumulation of transposable elements. The genome of Microbotryum violaceum has a high density of transposable elements compared to other fungi, but there is also evidence of RIP activity. This is the first report of RIP in a basidiomycete and was obtained by sequencing multiple copies of the integrase gene of a copia-type transposable element and the helicase gene of a Helitron-type element. In M. violaceum, the targets for RIP mutations are the cytosine residues of TCG trinucleotide combinations. Although RIP is a linkage-dependent process that tends to increase the variation among repetitive sequences, a chromosome-specific substructuring was observed in the transposable element population. The observed chromosome-specific patterns are not consistent with RIP, but rather suggest an effect of gene conversion, which is also a linkage-dependent process but results in a homogenization of repeated sequences. Particular sequences were found more widely distributed within the genome than expected by chance and may reflect the recently active variants. Therefore, sequence variation of transposable elements in M. violaceum appears to be driven by selection for transposition ability in combination with the context-specific forces of the RIP and gene conversion.
