Effects of estradiol and tamoxifen on feeding, fattiness, and some endocrine criteria in hypothalamic obese hens.

In White Leghorn hens, basomedial hypothalamic (BMH) lesions result in two syndromes: a) obese, functionally castrated (OFC) hens, in which both the ventromedial hypothalamic nucleus (VMH) and the mammillary nuclei are damaged and plasma estrogen is very low; and b) obese laying (OL) hens, which have normal levels of plasma estrogen and are less obese than the former, and whose lesion is limited to the VMH. In the present study, the involvement of estrogen in regulation of fattiness and energy metabolism was assayed in OFC, OL, and control (CONT) hens. BMH lesions were made at 13 weeks of age. When the typical syndromes reached the static phase, 20 weeks later, CONT, OFC, and OL hens were divided into three subgroups and were injected for 10 weeks on each alternate day, with either 10 mg tamoxifen (TAM)/kg, 2 mg estradiol benzoate (E2)/kg, or the vehicle, corn oil (0.5 ml). E2 raised plasma total lipids and reduced plasma glucose, insulin, and hematocrit in all treated hens, and increased liver weight in OL and OFC, but not in CONT hens. In OFC hens only, E2 reduced food intake (FI) and fattiness. In OL and CONT hens, E2 increased plasma T3, but raised the resting metabolic rate (RMR) only in CONT ones. In OFC hens, E2 reduce plasma T3 and T4 without affecting RMR. E2 reduced comb weight and egg production in CONT and more severely in OL hens. In the latter, E2 diminished ovarian and oviduct weights, whereas in OFC hens it increased the size of the atrophied oviduct. TAM had no visible effect on OFC hens.(ABSTRACT TRUNCATED AT 250 WORDS)

Combined effects of interferon and steroid hormones on 2',5'-oligoadenylate synthetase activity in chronic lymphocytic leukemia cells.

2',5'-oligoadenylate synthetase (OAS) has been implicated in the effects of interferons (INF) and steroid hormones on cell growth and differentiation. We studied the combined effects in vitro of hormones and INF on OAS activity in cells from 10 patients with chronic lymphocytic leukemia. IFN enhanced OAS activity in all cells studied and also induced morphologic transformation. Diethylstilbestrol was also found to induce OAS activity, but not morphologic transformation. Progesterone, tamoxifen and dihydrotestosterone had no effect on enzyme activity nor on morphology. In five samples, all with estrogen receptor activity, DES and INF were synergistic in inducing OAS activity. TAM-INF synergism was observed in one sample. DES and TAM did not, however, significantly enhance IFN-induced morphologic transformations. Hydrocortisone reduced OAS activity, and antagonized INF-induced enzyme activity and morphologic transformations. We conclude that hormones can modulate OAS activity in CLL cells. These findings may be of importance in the design of INF-based therapies of CLL.

The origin and function of tumor-associated macrophages.

Tumor-associated macrophages (TAM) have a complex relationship with the neoplastic cells of the tumor. On the one hand, the two cell types produce reciprocal growth factors and may be considered to have a symbiotic relationship. On the other hand, TAM can be activated to inhibit tumor growth and destroy neoplastic cells. Here, Alberto Mantovani and colleagues describe this delicate balance and the prospects for its therapeutic manipulation.

Flexor tendon repair in zone 2C.

A randomized prospective clinical study was carried out in 33 patients (37 fingers) with lacerations of both FDS and FDP tendons in the area covered by the A2 pulley, that is, zone 2C in Tang's subdivision of no man's land. Both lacerated tendons were repaired in 19 fingers and repair of only FDP with regional excision of FDS were performed in 18 fingers. Follow-up of average 12 months revealed that there was no significant difference in the end results evaluated according to the TAM system. The average TAM was 204 degrees in the fingers with suture of FDP only and 187 degrees in those with suture of both tendons. The fingers with suture of both tendons showed a higher rate of re-operation due to adhesions or rupture of repair. This study suggests that it is better to repair only FDP with regional excision of FDS when both tendons are injured in zone 2C.

Accumulation of benzoporphyrin derivative in malignant and host cell populations of the murine RIF tumor.

The accumulation of benzoporphyrin derivative (BPD) in malignant and host cell populations of the mouse RIF tumor was studied by flow cytometry analysis of cells dissociated from the tumor tissue. The highest cellular BPD levels were found in a subpopulation of tumor associated macrophages (TAMs) characterized by a highly elevated expression of interleukin 2 receptor (F4/80+CD25++), but this selectivity was less pronounced than with photofrin. The BPD levels in tumor associated immune cells other than TAMs were lower than in malignant cells. No significant difference in cellular distribution of BPD was observed with the photosensitizer delivered in liposomes compared to the aqueous solution.

Combined effects of 13-cis-retinoic acid, tamoxifen and interferon on the growth of human breast cancer cells.

We studied the effect of 13-cis-retinoic acid (13-cRA) alone and in combination with interferons (IFNs) and tamoxifen (TAM) in two established human breast cancer cell lines: the estrogen-sensitive CG-5 and the estrogen-insensitive MDA-MB-453 cells. 13-cRA (10(-9)-10(-5) M) significantly reduced the growth of both cell lines in a dose-dependent fashion, after 3 and 6 days of treatment. When the retinoid (10(-9)-10(-5) M) was combined with natural beta-IFN (100-1000 IU/ml) for 6 days, we observed a growth inhibition more pronounced than that produced by each of the two single agents in both CG-5 and MDA-MB-453 cells. Only in the former model was the inhibitory effect synergistic at all the drug concentrations used. Association of 13-cRA (10(-9)-10(-5) M) and recombinant alpha2a-IFN (100-1000 IU/ml) or TAM (10(-7)-10(-6) M) did not determine an additive or synergistic effect on the growth of CG-5 cells.

Inflammatory infiltrates of experimental mammary cancers.

The purpose of this review was to summarize observations on the type and function of inflammatory infiltrates of mouse mammary tumors and to speculate on the underlying mechanisms and the significance of infiltrates to mammary tumor biology. Although the major conclusion is that much more work is needed, certain themes seem to be emerging. The number of infiltrating cells can be very high but is unrelated to biological behavior of the tumors. What seems to be important is the relative contributions of inflammatory cell subsets. In the case of T-cell subsets and NK cells, the infiltrates from tumors of long-term cell lines so far seem uninformative. The general characteristics are similar to those of infiltrates from rapidly proliferating, normal mammary tissues. These characteristics do not correlate with diverse biological behavior or malignant potential. A more informative model appears to be one in which the development of tumors from preneoplastic tissue can be observed. Here our attention is currently focused on NK cells. By contrast, the correlation between activated TAM and metastatic behavior suggests that our transplantable MMT lines may be biologically relevant in the study of infiltrating macrophages. We are especially interested in the role of TAM in the generation of tumor cell variability. Overall, our data indicate that the host infiltrate is another manifestation of both inter- and intra-tumor heterogeneity and, as such, is not simply a response to, but, rather, a part of the tumor ecosystem. Unraveling the cellular and molecular mechanisms that govern the inflammatory cell component of tumors should provide insight into the types of cellular interactions that result in tumor development and progression.

Biological and clinical aspects of prolactin receptors (PRL-R) in human breast cancer.

PRL has a definite activity in the induction and promotion of mouse and in the growth of rat mammary tumors. We and others have found that human PRL or growth hormone (GH) had a growth promoting effect on human mammary cancer cells. It has been shown that prolactin receptors (PRL-R) which are specific for all lactogenic hormones (hPRL, hGH, hPL) are present on mammary cancer cells in long-term tissue culture and also in tumor biopsies. We found that 43% of the tumors had free PRL-R (FPRL-R) and that 72% had total PRL-R (TPRL-R) which have been desaturated in vitro. A significant correlation (Spearman test) was found between PRL-R (especially TPRL-R) on the one hand, estradiol (P less than 0.001) and progesterone receptors (P less than 0.01) on the other. The demonstration of PRL-induced proteins (PIP) might be a better sign of PRL sensitivity than the existence or PRL-R; PIP have been found by Northern blot analysis in 47% of 70 breast cancers. Overall survival (OS) and relapse-free survival (RFS) analysis with a median duration of follow-up of 5.3 yr showed that TPRL-R had a significant prognostic value only in node positive patients (chi 2 = 5.61, P = 0.02). Neither FPRL-R or TPRL-R were a significant prognostic factor when studied by Cox analysis. This confirms our previous results. Since at least some human mammary cancers appear to be PRL-dependent we carried out a multicenter randomized trial comparing as the first hormonal treatment tamoxifen (TAM) (30 mg/day) + bromocriptine (B) (5 mg/day) vs TAM + placebo. 171 patients entered this trial. No difference was observed between the two groups in response rates, duration of response or survival. Recent studies are thus in favor of a role of lactogenic hormones during the course of breast cancer. However no improvement in therapy has been observed yet. The combination of drugs to achieve a total anti-lactogenic treatment, the use of anti-PRL-R antibodies are interesting areas of research; the recent cloning of PRL-R and GH receptors may open new clinical perspectives.

Further studies on the antigonadotropic mechanism of action of norethisterone.

To examine the molecular mechanisms involved in the antigonadotropic effects of norethisterone (NET) and two of its A-ring reduced metabolites the 5 alpha-norethisterone (5 alpha-NET) and the 3 beta, 5 alpha-norethisterone (3 beta, 5 alpha-NET) at the neuroendocrine level, a series of experiments were undertaken in adult castrated rats. Animals were primed either with 0.2 mg of tamoxifen (Tam) for 4 consecutive days or 1.0 mg of cyproterone acetate (CPA) for 7 days followed by a single subcutaneous injection of 0.5 mg of NET, 5 alpha-NET or 3 beta, 5 alpha-NET. Four hours later, they were sacrificed and blood obtained for the measurement of immunoreactive serum LH and FSH. The results indicated that antiestrogen (Tam) pretreatment precluded the inhibitory effects of NET and the 3 beta, 5 alpha-NET but not those of the 5 alpha-NET derivative. Pretreatment with CPA did not modified the antigonadotropic action of the 3 beta, 5 alpha-NET metabolite but it markedly reduced the inhibitory action of the 5 alpha-NET, thus indicating that in the experimental model used, the antigonadotropic effects of NET, are in part the result of its metabolic conversion to its A-ring reduced metabolites. While the 5 alpha-NET displayed an androgenic effect, the 3 beta, 5 alpha-NET exhibited estrogen-like effect at the neuroendocrine level.

Accumulation of a non-binding form of estrogen receptor in MCF-7 cells under hydroxytamoxifen treatment.

It is well known that MCF-7 cells, when incubated with hydroxytamoxifen (OH-Tam) loose their capacity to bind [3H]estradiol. By using Western blotting and [3H]tamoxifen aziridine labeling of KCl extracts from these cells we found that this loss in binding capacity was not associated with a disappearance of the estrogen receptor (ER) protein, an event known to occur after incubation with estradiol. Attempts to label under exchange conditions these ER molecules, which, on the basis of enzyme immunoassays appear to accumulate under OH-Tam treatment, were unsuccessful. Cell fractionation suggested that their origin is nuclear. Assessment of a few triphenylethylenic antiestrogens, as far as their inhibitory potency towards the in vitro MCF-7 cell growth is concerned, indicated a correlation between accumulation of these non-binding ER molecules and the antiestrogen antiproliferative action. However, we were unable to demonstrate absence of such an ER accumulation in two tamoxifen-resistant variants. Impaired folding of the ER protein or impaired phosphorylation of its hormone-binding domain are attractive hypotheses to account for these non-binding ER molecules. Whether these ER molecules have any physiological role, such as competition with the "normal" receptor molecules for the estrogen responsive elements on the DNA is unknown and deserves further study.

Sex steroids induced up-regulation of 1,25-(OH)2 vitamin D3 receptors in T 47D breast cancer cells.

There is evidence indicating that 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] through binding to its specific receptor (VDR) exerts an antiproliferative effect on breast cancer cells. Considering the importance of receptor regulation in modulating the target cell responsiveness to hormones, the effect of dihydrotestosterone (DHT) and estradiol-17 beta (E2) on the regulation of VDR number was investigated in T 47D human breast cancer cells that also express androgen and estrogen (ER) receptors. T 47D cells were grown in RPMI medium containing 10% charcoal-treated fetal calf serum and the receptor content was determined in cells at confluence. Whole cell binding studies confirmed the presence of highly specific, saturable (4.01 +/- 1.82 fmol/10(6) cells), high affinity (Kd = 0.079 +/- 0.058 x 10(-9) M) 1,25(OH)2D3 receptors in control cells. Exposure to 10(-7) M DHT for 72 h resulted in a significant increase in VDR levels. Similar results were obtained with 10(-7) M E2. DHT- and E2-induced up-regulation was completely suppressed by 10(-6) M tamoxifen (TAM) addition but unaffected by 10(-6) M flutamide. TAM treatment alone produced a significant dose-dependent increase in VDR content, that was maximal at 10(-6) M. Our data strongly suggest, for the first time, an up-regulation of VDR by DHT and E2 via an ER-mediated mechanism.

Distribution of disulfonated and tetrasulfonated aluminum phthalocyanine between malignant and host cell populations of a murine fibrosarcoma.

Levels of disulfonated and tetrasulfonated aluminum phthalocyanines (AlPcS2,4) were measured in cells derived from FsaR tumors (murine fibrosarcoma) using a fluorescence-activated cell sorter (FACS). The tumors were excised from animals injected with the sensitizer 24 h earlier and enzymatically dissociated. Before flow cytometry, the cells were stained with fluorescein isothiocyanate-conjugated anti-mouse monoclonal antibodies to specific immune cell membrane markers (Mac1, Fc receptor (FcR) or CD45). Staining to FcR and CD45 was combined with a DNA stain Hoechst 33342. This enabled concomitant discrimination to be made by the FACS between different populations of tumor-infiltrating host cells and malignant cells. The results showed on average 1.49 times higher AlPcS2 levels and 1.16 times higher AlPcS4 levels in Mac1-positive (Mac1+) compared with Mac1-negative (Mac1-) tumor cell populations. The same type of experiments performed with SCCVII tumor (squamous cell carcinoma) gave average Mac1+/Mac1- ratios of 1.75 and 1.45 for AlPcS2 and AlPcS4 respectively. The data using other antibodies and DNA staining are consistent with the conclusion that, based on average per cell content, elevated levels of AlPcS2, and to a lesser extent AlPcS4, are retained in tumor-associated macrophages (TAM). The levels of these photosensitizers in other leukocytes and in non-immune host cells were not substantially different from those in malignant tumor cells. It is also shown that elevated levels of AlPcS2 and AlPcS4 are not localized in all TAM, but rather in a fraction of this cell population characterized by extremely high photosensitizer content.

Inhibition of human plasma cholinesterase by malachite green and related triarylmethane dyes: mechanistic implications.

The inhibitory effects of the cationic triarylmethane (TAM+) dyes, pararosaniline (PR+), malachite green (MG+), and methyl green (MeG+) on human plasma cholinesterase (BChE) were studied at 25 degrees C in 100 mM Mops, pH 8.0, with butyrylthiocholine as substrate. PR+ and MG+ caused linear mixed inhibition of enzyme activity. The respective inhibitory parameters were K(i) = 1.9 +/- 0.23 microM, alpha = 13 +/- 48, beta = 0 and K(i) = 0.28 +/- 0.037 microM, alpha = 23 +/- 7.4, beta = 0. MeG+ acted as a competitive inhibitor with K(i) = 0.12 +/- 0.017 microM (alpha, infinity, beta, not applicable). The K(i) values were within the same range reported for a number of ChE inhibitors including propidium ion, donepezil, and the phenothiazines, suggesting that TAM+s are active site ligands. On the other hand, the alpha values failed to correlate with values previously reported for a number of ChE inhibitors. It appears that mixed inhibition is the combined result of more than one type of binding and S-I interference. The impact of ligands at the choline-specific and peripheral anionic sites (or, possibly, accessory structural domains) on BChE activity needs to be studied in further detail.

Breast cancer in the elderly: the role of adjuvant radiation therapy.

PURPOSE: To evaluate available data on breast conserving surgery (BCS) with or without radiation therapy (RT), and alternative options (e.g. Tamoxifen alone) (Tam), focusing specifically on the older patients. METHODS AND MATERIALS: The MEDLINE was searched for the terms elderly, BCS, adjuvant/postoperative RT, Tamoxifen, and randomized trials from 1992 to 2002. RESULTS: Authors reviewed papers of general interest on the elderly and breast cancer (BC), meta-analysis, randomized trials on BCS+/-RT and Tam+/-surgery without RT, retrospective non-randomized trials, and reported on prognostic factors for local recurrence in BCS alone, including biomarkers, attempts made to define a low-risk group, and methods of modification of the current, protracted standard adjuvant RT course. CONCLUSIONS: Postoperative RT after BCS has a firm rationale; in current clinical practice, if the BCS+RT is medically appropriate and the patient shares the choice, after a full information of the available options and their implications, it is clinically sound to propose this approach. However, there is controversy whether a subgroup of elderly patients could be safely spared: in this setting a randomized trial is clearly warranted.

Tumour-associated macrophages as a prototypic type II polarised phagocyte population: role in tumour progression.

Macrophages are versatile, plastic cells which respond to micro-environmental signals with distinct functional programmes. In the tumour microenvironment, tumour-associated macrophages (TAM) polarise towards a type II phenotype, oriented to the promotion of tissue remodelling and repair. As polarised type II macrophages, TAM are a key component of the inflammatory circuits that promote tumour progression.

Homopterocarpanes as bridged triarylethylene analogues: synthesis and antagonistic effects in human MCF-7 breast cancer cells.

A series of new compounds structurally derived from 6a,12a-dihydro-6H,7H-[1]-benzopyran-[4,3-b]-benzopyran (homopterocarpane) was efficiently synthesized by reduction of the corresponding pyrilium salts obtained by treatment of selected flavanones and aldehydes with anhydrous HClO4. Cytotoxic effects on the human breast cancer cell line MCF-7 and antiestrogenic activity (only for compounds which resulted more active than tamoxifen (TAM)) on MCF-7 cells stimulated by 17beta-estradiol were evaluated. In vivo antiestrogenic activity and the relative binding affinity were also assessed. Some of the new compounds (4c, 4h, 4i and 4l) showed a biological activity in the micromolar range, and were more potent than TAM taken as the reference.

HIV-1 drug resistance in Thailand: before and after National Access to Antiretroviral Program.

BACKGROUND: The Ministry of Public Health (Thailand), MoPH, has had a program called National Access to Antiretroviral Program for People who have AIDS (PHA) or "NAPHA", to offer free antiretroviral drugs (ARV), which are locally produced in Thailand, to any HIV-1 infected patients with CD4<200 since 2002. This program may increase usage of ARV therapy and the emergence of HIV-1 drug resistance. OBJECTIVES: To monitor HIV-1 ARV drug resistant codon mutation in Thailand before and after the "NAPHA" program. MATERIALS AND METHODS: EDTA blood samples were collected from 542 HIV-1 infected subjects, who received ARV therapy in 1999 and 2001-2003, and perinatal chemoprophylaxis in 1998 and 2000. HIV-1 pol nucleotide sequences were analyzed. RESULTS: The percentage of drug resistant detection from the ARV therapy group in 1999 and 2001-2003 were 12.14 (34/280), 10.23 (9/88), 86.96 (20/23) and 57.55 (61/106), respectively. Of 332 NRTI drug resistant codon mutation, 226 (68.07%) were thymidine analogue mutations (TAMs). The percentage of TAMs detection in 1999 and 2001-2003 were 7.14 (20/280), 9.09 (8/88), 56.52 (13/23) and 43.34 (46/106), respectively. Of 105 NNRTI drug resistant codon mutation, 95 (90.48%) were related to nevirapine drug resistance. CONCLUSION: Thailand may need more appropriate monitoring of drug resistance in the free ARV therapy program to protect the future usage of drugs by minimizing the emergence of drug resistance.

Synergistic effect of estrogen and VEGF on the proliferation of hemangioma vascular endothelial cells.

PURPOSE: The aim of this study was to observe the influences of estradiol (E2), vascular endothelial growth factor (VEGF), and 4OH-tamoxifen (TAM) on the proliferation of hemangioma vascular endothelial cells (HVECs). METHODS: Two strawberry hemangiomas with positive estrogen receptor staining from 2 infants (cases 1 & 2) were used for HVECs culture. The HVECs of passage 3 were cultured in estrogen-free improved minimum essential medium and divided into 5 groups based on different treatments: group 1, no treatment; group 2, treated with E2; group 3, treated with VEGF; group 4, treated with both E2 and VEGF; group 5, treated with E2, VEGF, and TAM. Cell count (CC) and DNA proliferation index (PI) were determined on culture days 0, 3, 6, and 9. RESULTS: On day 9 in case 1, CC and PI were the following: in group 1, 1.15 +/- 0.18 x 10(5) mL and 19.96% +/- 1.45%, respectively, presenting no statistically significant changes; in group 2, 1.38 and 1.61 times those of group 1, respectively (P <.01); in group 3, 2.10 and 1.61 times those of group 2, respectively (P <.01); in group 4, 1.62 and 1.40 times as high as with group 3, respectively (P <.01); in group 5, down to the levels of group 1. The results in case 2 were similar to those in case 1. CONCLUSIONS: In vitro, the promoting effect of VEGF on HVECs proliferation is stronger than that of estrogen. Estrogen and VEGF enhance this proliferation in a synergistic fashion, which can be inhibited by tamoxifen.
