The asymmetry of life.

Stereoselective physical phenomena and their possible importance for the prevalence of D-sugars and L-aminoacids in living matter are reviewed. A classification is presented according to which a selective force provides a microscopic or macroscopic selection depending on its generality when taken over a macrosystem (a 'unitary biosphere' such as the Earth). The microscopic 'selections' are not genuine selections because the final sense of asymmetry is here determined by chance, in other words the initial choice is 'random', while it is 'directed' in the macroscopic selection. Two macroscopic selections appear possible: 1. selection due to an intrinsic energy difference between enantiomorph configurations, 2. selection accomplished by elliptically polarised radiation.

Radiolysis, racemization and the origin of molecular asymmetry in the biosphere.

An investigation has been undertaken to determine whether ionizing radiation might engender racemization of optically active amino acids, along with their usual radiolysis. As prototypes, crystalline D- and L-leucine, as well as aqueous solutions of their sodium salts were exposed to the radiation from a 3000 Ci 60Co gamma-ray source. Gamma-ray doses which caused about 68% radiolysis of solid leucine left a residue which was about 5% racemized, while racemization proved even greater at lower doses for the dissolved sodium salts. In aqueous solution both percent degradation and percent racemization of the sodium salts were proportional to gamma-ray dosage within the range employed (1--27 x 10(6) rads). Implications of these observations for the origin of molecular asymmetry by the beta-decay parity violation mechanism are discussed.

The absolute configuration of SU 23397: a novel neuroleptic agent.

The absolute configuration of a novel chiral neuroleptic agent SU 23397 (I) was determined by ORD comparison of (+)-5-methoxy dihydro coumarilic acid (VIII), a synthetic precursor of SU 23397 (I), with (+)-dihydro coumarilic acid, whose absolute configuration is known. This assignment was confirmed by oxidative degradation of (+)-5-methoxy dihydro coumarilic acid VIII to D-(+)-malic acid.

Continuous optical assay of sucrase and other glucosidases.

A continuous optical method for the assay of glucose-releasing hydrolases is reported. Particular emphasis is given to the assay of purified sucrase from rabbit small intestine. The procedure requires glucose dehydrogenase and mutarotase. In the presence of the latter enzyme, the initial lag is substantially shortened when glucose is released as alpha-anomer. Under the test conditions used, the method shows a good proportionality up to an activity of 0.2 units/3 ml and may also be applied for measuring the activity in crude homogenates.

Cyclic and linear vasopressin V1 and V1/V2 antagonists containing arginine in the 4-position.

Substitution of arginine for glutamine in the 4-position of a vasopressin V1 antagonist has been reported to turn it into an agonist. We resynthesized this 4-arginine analog and synthesized additional cyclic and linear vasopressin antagonists containing a 4-arginine. The presence of a 4-arginine in the resynthesized and new analogs had relatively minor effects on their antivasopressin V1 and V2 antagonistic potencies.
