Fibrosarcoma of the vulva: case reports and discussion.

Three cases of fibrosarcoma of the vulva are reported. In two, surgical resection resulted in remission lasting over four years. In the third case, presenting with metastasis of the lung and liver, the patient died without surgery having been carried out. The low mitotic index found in the first two cases seems to lead to a favourable prognosis, whereas the inferior vulvar localization found in the last case seems to be more dangerous because of its diffusion into the paravaginal space.

[Primary fibrosarcoma of the lung].

A case of primary pulmonary fibrosarcoma in a women of 50 is described as a rare disease which is difficult to diagnose intravitally due to the absence of pathognomonic clinical and roentgenological data. The danger of prolonged time-course observation of the patient with a rounded shadow in the lung is emphasized. The importance of electron microscopic study to specify the morphological diagnosis is demonstrated.

Effect of cyclophosphamide on mice bearing methylcholanthrene-induced fibrosarcomas.

Mice bearing large methylcholanthrene-induced fibrosarcomas lost the ability to respond in vitro to mitogen stimulation and to specifically neutralize autologous tumor cells in vivo. This depressed immune capability was due to active suppression, since spleen cells from advanced tumor-bearing mice could suppress the mitogen response of normal spleen cells and could inhibit tumor rejection when adoptively transferred to mice previously immunized against the tumor. Treatment with cyclophosphamide (CY) was found to affect the immune capability of the host, in addition to have a direct effect on the tumor. The number of cells in the lymph nodes and spleen, as well as their response to concanavalin A and lipopolysaccharide (but not phytohemagglutinin), decreased initially but returned to normal by Day 14. Most importantly, when CY was administered one day after tumor inoculation, the treated animals developed the ability to neutralize tumor at the same time as untreated controls but retained this capability as the tumors became advanced. Treatment with a single dose of CY as late as 11 or 20 days after tumor inoculation maintained or restored the tumor-neutralizing capacity of spleen cells. CY appears to alter the antitumor response of the host by inhibiting both cytotoxic and suppressor cells, but the cytotoxic cells recover rapidly, whereas the suppressor cells do not.

[Fibrosarcoma of the pancreatic head].

A postmortem examination of a 60 year old female patient proved the presence of fibrosarcoma of the head of pancreas. The tumour first became manifest through an accidentally diagnosed pulmonary metastasis two years before death. 14 weeks before death, the patient had been operated on for a metastasis in the brain. Jaundice lasted 10 weeks.

[Fibrosarcoma of the right heart. Contribution to the problem of primary heart tumors].

Report on a fibrosarcoma of the right ventricle in a 70-year-old woman. The tumour was diagnosed by autopsy. Clinically were of importance the chronic insufficiency of the right heart, in the ECG the low voltage, a bifascicular block and a subepicardiac lesion in the area of the anterior wall. Roentgenogram of the thorax and the laboratory findings corresponded to a massive cardiac decompensation. The cytology from the effusion of the pericardium which often occurred gives a possibility of making a diagnosis intra vitam, if it is thought of a cardiac tumour.

Fibrosarcoma of the lung with extrapulmonary manifestations: case report.

A 50-year-old female presented with a five months history of recurrent attacks of dizziness, sweatiness, tremors and fainting with loss of consciousness. These were found to be due to hypoglycaemic episodes with blood sugars less than 1 mmol/l and were treated as such. A diagnosis of insulinoma was initially considered, but the patient turned out to have fibrosarcoma of the lung, a rare lung tumour. She also had finger and toe clubbing and features of hypertrophic pulmonary osteoarthropathy.

Response of the FSaII fibrosarcoma to antiangiogenic modulators plus cytotoxic agents.

The formation of a blood supply (angiogenesis) is critical to the growth of solid tumors. The naturally occurring steroid tetrahydrocortisol, the synthetic cyclodextrin derivative beta-cyclodextrin tetradecasulfate, and the tetracycline derivative minocycline have antiangiogenic activity. Tetrahydrocortisol (125 mg/kg) and beta-cyclodextrin tetradecasulfate (1000 mg/kg) in a 1:1 molar ratio by continuous infusion over 14 days and minocycline (10 mg/kg) administered i.p. daily from day 4 to day 18 postimplantation of the FSaII fibrosarcoma did not alter the growth of the tumor. These antiangiogenic modulators were not cytotoxic toward FSaIIC tumor cells or bone marrow CFU-GM when tumor-bearing animals were treated and cytotoxicity determined by colony formation in culture. The antiangiogenic modulators markedly increased the cytotoxicity of cyclophosphamide toward FSaIIC tumor cells and to a much lesser degree toward bone marrow CFU-GM. The cytotoxicity of CDDP and radiation was enhanced only by administration of the three modulators in combination. In tumor growth delay studies, the three modulator combination increased the effectiveness of CDDP by 1.5-fold, of cyclophosphamide by 1.9-fold and of radiation by 1.4-fold. Although the antiangiogenic therapies alone did not substantially reduce the number of lung metastases compared with the untreated controls, addition of the antiangiogenic agents to treatment with the cytotoxic therapies reduced not only the number of lung metastases formed from the primary tumor but also reduced the number of large metastases. Thus, antiangiogenic therapies can potentiate the efficacy of standard anticancer therapies.

The important role of PKC in controlling polyploidy formation in cultured fibrosarcoma cell line.

Induction of polyploidization by colcemid in cultured fibrosarcoma cells (Meth-A cells) was examined. Activators of protein kinase C (PKC), phorbol 12-myristate 13-acetate (PMA) and ATP, inhibited colcemid-induced polyploidization, but not colcemid-induced cell proliferation cessation. These findings suggest that a down-regulation of PKC activity results in checkpoint "dysfunction" which induces polyploidization and that inhibition of polyploidization induction by PMA and ATP is not a result of the inhibition of colcemid-induced depolymerization of tubulin.

Comparison of fibrosarcomas that developed at vaccination sites and at nonvaccination sites in cats: 239 cases (1991-1992).

Questionnaires were sent to veterinarians who had submitted a fibrosarcoma from a cat to the surgical pathology services of the veterinary schools of the University of Pennsylvania and Tufts University between Jan 1, 1991 and June 30, 1992. Questionnaire items included signalment, FeLV and feline immunodeficiency virus status, site of sarcoma, vaccination site, vaccines used, treatment, biologic behavior of the tumor, and final outcome. Data were analyzed, using Student's t-test for continuous data, chi 2 test for categoric data, and log-rank test for survival estimates. Comparing results for cats with vaccination-site (VS) tumors and nonvaccination-site (NVS) tumors, we determined that VS tumors developed in younger cats and were larger than NVS tumors. Although VS sarcomas were biologically aggressive and redeveloped more often than NVS sarcomas, metastasis was not detected, and cats with VS tumors survived longer than cats with NVS tumors. Vaccination-site sarcomas developed in cats after injection of many types of vaccines, administered singularly or in combination. Of the cats in the VS group administered a single vaccine, 37% were given rabies, 33% were given feline viral rhinotracheitis/calicivirus/panleukopenia virus, and 30% were given FeLV vaccines. Cats with VS tumors were more likely to have received FeLV vaccine and less likely to have received rabies vaccine than those with NVS tumors. Although vaccines produced by certain manufacturers were used most often in cats with VS and NVS sarcomas, it was believed that this probably represented marketing practices and brand popularity.(ABSTRACT TRUNCATED AT 250 WORDS)

Use of the stress-inducible grp78/BiP promoter in targeting high level gene expression in fibrosarcoma in vivo.

Current advances in human gene therapy open up new frontiers for molecular therapies of cancer. However, one major limitation in cancer gene therapy is the lack of a general tumor-specific promoter which allows stringent and high level expression of the therapeutic reagent in malignantly transformed but not normal tissues. Hallmark features of solid tumors such as glucose deprivation, chronic anoxia, and acidic pH induce the glucose-regulated proteins, in particular, GRP78/BiP, a M(r) 78,000 endoplasmic reticulum-localized protein with chaperone and calcium-binding properties. We report here that a truncated rat grp78 promoter with most of the distal basal elements removed can be utilized as a potent internal promoter in a retroviral vector to drive high level expression of a reporter gene in a murine fibrosarcoma model system. The stress-inducible grp78 promoter offers a novel approach for gene delivery systems targeting transcription in tumorigenic cells.

[Simultaneous hepatic cholangiocarcinoma and bone fibrosarcoma 45 years after ingestion of Thorotrast].

This case of a 63 year old man reports the simultaneous development of hepatic cholangiocarcinoma and fibrosarcoma of the sacrum 45 years after the systemic injection of Thorotrast. The characteristic radiologic aspect was an important criteria for diagnosis. Biopsies have confirmed the histology of both tumors. We describe the way to thorotrastosis diagnosis and characteristic malignant tumors, especially cholangiocarcinoma, induced by Thorotrast.

Mediation by an androgen receptor of the stimulatory effect of testosterone on the growth of a fibrosarcoma in the rat.

We studied the effect of testosterone on the growth of a 20- methylcholanthrene-induced transplanted fibrosarcoma and assayed androgen receptors in this tumor. The effect of an antiandrogen in male rats and the comparative tumor growth in females confirmed the androgen sensitivity. A synthetic androgen that strongly binds to the androgen receptor was used to characterize the binding activity in nuclear extracts of tumor. Scatchard analysis showed that the dissociation constant of the hormone receptor complexes (Kd) was 26 +/- 2 nmol/l and the number of binding sites was about 95 +/- 12 fmol/mg of protein. The hormone-receptor complexes sedimented in the region of 3.7 S.

Coagulation activation by MC28 fibrosarcoma cells facilitates lung tumor formation.

Tumor cells interact with the hemostatic system in various ways and may thus influence malignant growth and spread. MC28 fibrosarcoma cells possess a potent procoagulant activity (PCA) and form lung tumors following intravenous injection. The aim of this work was to study the relationship between PCA, intravascular coagulation and lung seeding in the MC28 model. MC28 cells were injected into control, warfarinized and heparinized hooded Lister rats. Coagulation changes were monitored by thromboelastography (TEG) and Sonoclot analysis (SA), lung fibrin formation by light and electron microscopy, tumor seeding by macroscopic counting and tumor cell and platelet deposition in the lungs by radiolabelling. PCA was measured by chromogenic assay. MC28 PCA was characterized as a tissue factor-factor VIIa complex that probably arose during cell culture or disaggregation of solid tumors. Injection of tumor cells caused marked coagulopathy and was rapidly (within 30 min) followed by fibrin deposition in the lungs and accumulation of radiolabelled platelets. Heparin and warfarin significantly reduced lung seeding (p < 0.001) and reduced retention of radiolabelled tumor cells in the pulmonary circulation (p < 0.01). Inhibition of cellular PCA by prior treatment with concanavalin A markedly reduced intravascular coagulation and lung seeding. We conclude that MC28 cells cause intravascular coagulation as a direct result of their procoagulant activity. The data suggest that tumor cells form complexes with platelets and fibrin which are retained in the lungs long enough for extravasation and seeding to occur.(ABSTRACT TRUNCATED AT 250 WORDS)

Altered topoisomerase I and II activities in suramin-resistant lung fibrosarcoma cells.

To better understand the molecular basis for the cytotoxic effects of suramin, we have developed suramin-resistant DC-3F/SU 1000 cells by continuous exposure of fibrosarcoma cells to increasing concentrations of suramin. The suramin resistance (approximately 10-fold) is not associated with changes in uptake or intracellular distribution of the drug. The sensitivity to actinomycin D, cytarabine, aphidicolin, hydroxyurea, vincristine, and 5-fluorouracil is unaltered. In contrast, DC-3F/SU 1000 cells are about 2-fold resistant to classical DNA topoisomerase II inhibitors such as doxorubicin, amsacrine, and etoposide, whereas the cells are 1.5-fold more sensitive to the topoisomerase I inhibitor camptothecin. The cross-resistance to topoisomerase II inhibitors occurred earlier than the collateral sensitivity to camptothecin. Amsacrine- and etoposide-induced DNA-protein complex formation is reduced about 2-fold in DC-3F/SU 1000 cells, compared with DC-3F cells, whereas camptothecin-induced DNA-protein complex formation is increased 1.5-fold. Western blot analysis of cellular lysates from the two cell lines shows no significant differences in the level of topoisomerase II, whereas the level of topoisomerase I is increased 2.5-fold in DC-3F/SU 1000 cells. The catalytic activities of topoisomerases I and II in nuclear extracts from DC-3F/SU 1000 cells are both about 2-fold higher than those in extracts from DC-3F cells, whereas amsacrine- and etoposide-induced DNA-protein complex formation is comparable between the two cell lines. Taken together, our results support the involvement of DNA topoisomerases in the cytotoxic activity of suramin. We further believe that the DC-3F/SU 1000 cells may be a useful model for the elucidation of factors that lead to low, clinically relevant, levels of resistance to topoisomerase II inhibitors.

[Apoptosis of mouse MS-2 fibrosarcoma cells induced by photodynamic therapy with LDL-administered zinc-phthalocyanine].

Photodynamic destruction of MS-2 fibrosarcoma cells in mice induced by LDL-administered Zinc-phthalocyanine and red light irradiation was studied by electron microscopy. The pronounced structural changes such as chromatin condensation, disappearance of nuclear pores, karyopyknosis, karyorrhexis, leakage of chromatin aggregates, autophagocytosis, bleb formation on the cell surface, cytoplasmic vacuolization and cell fragmentation suggest that the tumor cell death was induced by apoptosis. However, its exact mechanism and regulating pathways remain to be further investigated.

Effect of the chemoprotective agent WR-2721 on disposition and biotransformations of ormaplatin in the Fischer 344 rat bearing a fibrosarcoma.

The effects of the phosphorothioate agent, WR-2721, have been investigated with respect to the biotransformations of ormaplatin in the Fischer 344 rat bearing a transplanted fibrosarcoma. A number of different paradigms of dosing route and schedule for the administration of the two agents have been investigated. In the first group of experiments, WR-2721 (200 mg/kg, i.p.) was administered 30 min before ormaplatin (12.5 mg/kg, i.p.), and then peritoneal fluid, plasma, and tissues were harvested at 30 min after the ormaplatin administration. Our results suggest that a significant interaction between WR-2721 and ormaplatin is occurring in the peritoneal cavity. The interaction was evident in terms of both effects on distribution and disposition of total platinum and in alterations of the profiles of biotransformation products formed in the various tissues and fluids. Plasma protein binding of ormaplatin was decreased by 50% in the presence of WR-2721. Total platinum in the spleen was decreased by 66% and in the liver by 50%. There were no trends among the findings that would indicate any selectivity between tumor and nontumor tissue with respect to the effects of WR-2721 on the parameters measured. Subsequent investigations examined the effects of dosing the WR-2721 by the i.v. route while continuing with the i.p. administration of the ormaplatin. WR-2721 was administered either 30 or 5 min before the ormaplatin, and the plasma and tissues were harvested at 15, 30, or 60 min after ormaplatin administration. The reverse-phase HPLC peak, which behaved chromatographically as a Pt(dach)(WR-1065) standard, was less prominent after the i.v. administration of WR-2721 than it was after i.p. administration under any of the paradigms tested. There was again no evidence for selectivity between tumor and nontumor tissue in the findings from any of the paradigms. It is concluded that if WR-2721 is capable of selectively protecting nontumor tissue from the toxicities of platinum-based chemotherapy, it is doing so by some mechanism other than its selective uptake into normal tissue and subsequent nonspecific inactivation of any reactive cytosolic platinum species formed. Other possible mechanisms are briefly discussed.
