[Growth and metastasis of Lewis' lung carcinoma in mice under the pharmacological action on metabolism and the effects of kinins].

In experiments on C57 Bl mice the effects of growth and metastatic proliferation of Lewis lung carcinoma inoculated intramuscularly on metabolism and kinin activity have been studied during the application of pharmacologic agents (parmidin, cellulose sulphate, unithiol). Causing a weak inhibition of the primary tumour growth, parmidin if administered repeatedly in given doses, inhibits partially metastatic process, which is manifested in an essential decrease of the quantity, size and weight of lung metastases. Parmidin also inhibits an adverse effect of cellulose sulphate and unithiol on metastatic process. It is believed that the action of parmidin is of an indirect character and is mainly associated with the elimination of the kinin component of microcirculatory disorders in the lungs.

[Effect of unithiol and thymidine on the toxic and therapeutic action of adriamycin].

The influence of unithiol and thymidine treatment on the acute toxicity and therapeutic effect of adriamycin in hemoblastosis La was studied in CBA and C57B1 mice. Unithiol and thymidine were shown to markedly decrease the toxicity of different doses of adriamycin which resulted in a lower mortality and longer survival of the animals. When unithiol and thymidine were administered in combination, their antitoxic effect was added. Since unithiol and thymidine administration does not diminish the therapeutic effect of various doses of adriamycin, unithiol may be recommended for clinical use to decrease adriamycin toxicity in cancer patients.

The therapeutic use of 2,3-dimercaptopropane-1-sulfonate in two cases of inorganic mercury poisoning.

Two patients had heavy metal intoxication resulting from industrial exposure to mercury vapors. One patient remained asymptomatic despite high urinary mercury levels. The second presented with a documented toxic reaction, including abnormal electromyograms and hematuria. Treatment was begun with 2,3-dimercaptopropane-1-sulfonate, and investigational drug used under Food and Drug Administration guidelines (IND 19,276). Use in both patients resulted in a dramatic change in excretion half-life of the mercury, from 33.1 days before treatment to 11.2 days during therapy. No significant toxic effects related to drug therapy were observed. Clearing of hematuria and reversal of electromyographic abnormalities were prompt in the symptomatic patient. Therapy with 2,3-dimercaptopropane-1-sulfonate was effective in these two patients.

An analysis of arsenic exposures referred to the Blodgett Regional Poison Center.

Fifty-seven cases of acute arsenic exposure were referred to Blodgett Regional Poison Center from January 1985 through December 1985. We will summarize epidemiologic aspects, laboratory values and the types of arsenicals involved. Cases predominantly involved children ingesting sodium arsenate in the form of ant poison. A great majority were accidental exposures related to the home use of ant killers in contrast to previous reports of homicidal or industrial poisoning. Initial urine arsenic levels, symptoms and therapy will be included. Most cases were reported within hours with patients being asymptomatic or with relatively few complaints. The current Blodgett Regional Poison Center Arsenic Protocol and instructions for the preparation of D-penicillamine suspension adapted from the Blodgett Pharmacy Department will be included. Conclusions, outcomes and recommendations are discussed.

Growth of Mycobacterium leprae in a redox system: II. Further improvements in the system and growth efficiency.

Improvement of the Redox System for growth of M. leprae as brought about by modification in the concentration and mode of preparation of individual media constituents, and by addition of newer substances, is being reported. A structural modification in the construction of the Thunberg's tubes and flasks that are used as culture vessels, has been introduced for ease of handling. Vitamin E (alpha-tocopherol) has been found to be useful. Concentrations of Liposomes and Gelatin in the medium could be reduced by at least five folds, considerably easing thereby smearing and harvesting of cultures. Dimercaptopropanol British Anti-lewisite or BAL) has been used, but its usefulness or otherwise is yet to be determined conclusively. The basis of intracellular parasitism of M. leprae has been discussed.

[Comparative effectiveness of the basic drugs used in rheumatoid arthritis (long-term randomized study)].

Study of the comparative efficacy of the basic slow-acting drugs for the treatment of rheumatoid arthritis permits considering randomization as necessary for obtaining objective data. It has been established that gold salts (chrisanol and myochrysin) are the most efficacious drugs for the treatment of RA. D-Penicillamine and delagil, in particular, are less efficacious. Administration of levamisole is rather dangerous because of the frequency and intensity of side effects.

[DMSP treatment in acute sublimate (mercury chloride) poisoning].

A 19-year-old girl was treated for a heavy mercury chloride intoxication (3 g HgCl2) with 2,3-dimercaptopropane-1-sulfonate (DMSP) and dialyses. The anuric stage lasted for 10 days. With DMSP the mercury clearance by hemodialysis was 3,5-5,0 ml/min, with BAL 0,6 ml/min. DMSP is an effective watersoluble mercury-chelator and is subjectively better tolerated and less toxic than BAL. The anuric stage was probably not shortened.

[Action of unithiol on the toxic effect of antitumor preparations].

The effect of unitiol treatment on the toxicity of dactinomycin, 5-fluorouracil, bleomycetin and vincristine was studied in CBA mice. Unitiol treatment was shown to decrease the acute toxicity of dactinomycin and involves a lower loss of body weight. Also, it reduced dactinomycin hemotoxicity which was assessed on the basis of leukocyte count in peripheral blood. Conversely, unitiol potentiated the acute toxic effect of 5-fluorouracil, bleomycetin and vincristine which was matched by body weight loss following treatment with cytostatic drugs. The results point to a wide range of the biological effects of unitiol.

[Effect of unithiol on the ultrastructure of the myocardium and adrenals in endotoxin shock].

Administration of unithiol to animals with endotoxin shock promotes deposition of catecholamines in chromaffin cells of the adrenals, blocks their secretion to the bloodstream and prevents the development of injuries to the cortical substance of the gland. Administration of unithiol for endotoxemia prevents damage to intracellular organelles of cardiomyocytes. However, insignificant disorders of the contractile apparatus of the cells and histohematic barriers are maintained. The capacity of unithiol to prevent the progress of destructive alterations in the heart and adrenals permits recommending it for use in combined treatment of septic conditions.
