The rate of incorporation of the labeled precursors, acetate and propionate, into levorin components and fatty acids was studied. It was shown that within the first 12 hours of cultivation the rate of labeled precursors incorporation into levorin components A2, A3, A0 and A1 was low. In this period the acetate and propionate labeled with respect to the methyl and carboxyl groups mainly incorporated into fatty acids. A higher rate of acetate incorporation as compared to propionate at the beginning of cultivation suggested that acetate was the first link of the chain. At the beginning of the idiophase, i.e. during the period of 36 to 48 hours of cultivation, the rate of incorporation of the carboxyl and methyl groups of the labeled precursors into the levorin components increased. The rate of incorporation of the radioactive precursors into components A2 and A3 markedly increased after 72 hours of cultivation. The maximum rate of incorporation of the carboxyl and methyl groups of acetate and propionate into components A0 and A1 was observed somewhat earlier, i. e. by the 48th hour of cultivation. During the period from 72 to 96 hours of cultivation the incorporation rate of the labeled precursors for components A2 and A3 was constant and then gradually decreased. The radioactivity of components A0 and A1 was constant during the period of 48 to 72 hours of cultivation and then gradually decreased. Beginning from the 24th hour of cultivation the level of 14C-propionate incorporated into all levorin components was higher than that of 14C-acetate incorporation. After 96 hours of cultivation the rate of 2-14C-propionate incorporation into the levorin components was higher than that of 1-14C-propionate incorporation.

Deficiency of the A subunit of coagulation factor XIII causes a severe bleeding disorder requiring life long replacement therapy. The mutations causing A subunit deficiency appear to be very heterogeneous, and it is impractical to identify each mutation before genetic counselling or prenatal diagnosis can be attempted. In this study we have shown that a highly polymorphic short tandem repeat element, HUMF13A01 (AAAG)n that occurs in the 5' flanking sequence of the factor XIII A subunit gene, can be used to follow the segregation of deficiency causing mutations. We studied 6 families with factor XIII A subunit deficiency from 5 different ethnic groups. All parents were heterozygous for the repetitive element and therefore all the families were informative. The linked polymorphism was used to carry out the first prenatal diagnosis of factor XIII A subunit deficiency. The analysis of this polymorphism by the polymerase chain reaction is rapid, reliable, requires little DNA and is ideal for the genetic analysis of factor XIII A subunit deficiency.
Allele and genotype frequencies of four short tandem-repeat loci were determined in a population sample from the North of Portugal using the polymerase chain reaction (PCR). After denaturing PAGE, 6 alleles were identified for HUMTH01 (n = 419), 9 alleles for HUMVWA31A (n = 376), 12 alleles for HUMF13A1 (n = 232), and 5 alleles for HUMFES/FPS (n = 409). No deviation from Hardy-Weinberg equilibrium was found. The allele frequencies observed are similar to those of the European populations compared. The combined power of discrimination is 0.999.
In a case of disputed paternity where the alleged father was dead and the children and their mothers were available for testing, we analyzed 23 conventional hemogenetic markers and six short tandem repeat loci. We deduced the possible types of the alleged father and calculated the probability of paternity. Conventional phenotypings did not provide conclusive proof of paternity. In contrast, typing of the short tandem repeat loci gave a combined probability of paternity of 0.9986, establishing paternity. The present study demonstrates the power of short tandem repeat typing as a tool for solving such a deficiency case.
The short tandem repeat system HumCD4 was amplified by the polymerase chain reaction (PCR) on blood samples from 304 unrelated Austrian Caucasians and analyzed by horizontal, non-denaturing polyacrylamide electrophoresis. The mean exclusion chance was 0.417, the discriminating power 0.850 and the heterozygosity rate 0.628. The observed phenotype distribution is in Hardy-Weinberg equilibrium. In 100 families (200 meioses) no mutations were found. Sufficient amplification could be achieved with as little as 80 pg of high molecular weight cell-line DNA. In a degradation experiment DNA extracted from bloodstains stored for up to 28 days in a moist chamber and DNA boiled for up to 18 min could be amplified. A duplex PCR with TH01 is proposed.
We studied 404 unrelated Japanese including 202 females for trinucleotide repeat polymorphism at the AR locus (Xcen-q13) and found 27 alleles of which 25 differed in size by an integral multiple of the repeat unit. The high informativeness makes this locus suitable for forensic application.
Capillary electrophoresis is a relatively new technology for the forensic analysis of STRs, and may be an excellent alternative to traditional gel-based methods. Eighty samples, obtained from six different laboratories which had previously analyzed the samples using a variety of gel-based methods, were reanalyzed by CE. It was determined that CE could obtain the same allelic designation as that reported by the comparison laboratory. Seven different loci employing two separate multiplex amplifications were used for the comparison study. One basepair resolution and average standard deviations within the range of 0.075 to 0.117 basepairs were observed.
An orthopedist had been sentenced for different offenses to two years and six months imprisonment by the Landgericht Frankfurt (superior court). One point of the ruling had been the evaluation of the use of X-ray methods without medical indication, which the Landgericht Frankfurt/Main (superior court) finally did not evaluate as bodily harm. The Bundesgerichtshof (supreme court) of Germany overruled this interpretation and referred the case back for new trial and ruling.
In the course of the investigation of a pentanucleotide repeat polymorphism at the human CD4 locus, a C-A transversion was found at the position corresponding to the 3' end of the original forward primer presented by Edwards et al. (1). In the present study, the simultaneous determination of the new sequence polymorphism and the pentanucleotide repeat polymorphism at the CD4 locus was attempted. To achieve this purpose, we adopted amplified product length polymorphism (APLP) analysis and designed some new allele-specific forward primers tagged with non-complementary nucleotides differing in length. A total of 646 DNA samples from peripheral blood of Japanese, Chinese and German populations were investigated. Although the C-A transversion was restricted to CD4*5, a new subtype allele with A and 5 repeats, designated CD4*5A, was observed at polymorphic frequencies in the three populations. The simultaneous genotyping by APLP analysis resulted in dramatically increased heterozygosity and discriminating power of the human CD4 locus.
We developed a non-isotopic method which improves the technical quality of the X-linked HUMARA locus typing process. The use of formamide and a low concentration of acrylamide increased resolution and sharpness of HUMARA alleles in silver-stained polyacrylamide gels. In addition, the construction of an allelic ladder containing amplified sequence of 9 alleles (even-numbered alleles) of the HUMARA locus, allows confident, rapid and precise assignment of discretely defined alleles. Allele and genotype frequencies for the HUMARA locus were determined in a French Canadian population sample. Observed genotype frequencies in females conformed to Hardy-Weinberg expectations. Furthermore, the HUMARA locus is highly polymorphic with 18 observed alleles and an heterozygosity value of 89.3%. Also, this locus has average powers of discrimination of 97.8% and 88.7% for testing samples of female and male origin, respectively. In the French Canadian population, the average probability of excluding a random man as the father in paternity analysis when both mother and daughter are tested for this locus is 88.0%. Together, the results indicate that the HUMARA locus provides a highly discriminatory system that is appropriate for the purposes of forensic identification and paternity testing involving a female child.
Stromelysin-3 (STR-3) is a matrix metalloproteinase with a unique pattern of expression and substrate specificity. During embryogenesis and remodeling of normal adult tissues, STR-3 is produced by stromal cells in direct contact with epithelial cells undergoing regional apoptosis and selective cell survival. STR-3 is also overexpressed by interdigitating stromal cells in primary epithelial malignancies. Although STR-3 does not degrade classic extracellular matrix components, the enzyme promotes the establishment of local tumors in nude mice by as yet undefined mechanisms. STR-3 is induced when malignant epithelial cells come into contact with surrounding stromal elements; the active stromal cell-derived 45 kDa enzyme is subsequently processed to a 35 kDa protein without enzymatic activity. We have generated MCF-7 transfectants expressing wild type or catalytically inactive 45 kDa STR-3 (STR-3wt and STR-3cat-) or secreted 35 kDa STR-3 (35 kDa STR-3sec) and evaluated their implantation and survival in nude mice. Tumors developed significantly more rapidly in animals receiving STR-3wt, rather than vector-only, STR-3cat- or 35 kDa STR-3sec transfectants. Most importantly, STR-3wt tumors had a significantly lower percentage of apoptotic cells than tumors derived from vector-only, STR-3cat- or 35 kDa STR-3sec transfectants. Taken together, these studies suggest that the active STR-3 enzyme may increase tumor take by suppressing tumor cell apoptosis and that 45 kDa to 35 kDa STR-3 processing limits STR-3 activity at the tumor/stromal interface. Because STR-3 is secreted as an active enzyme rather than a proform, subsequent 45 kDa to 35 kDa STR-3 processing may represent a novel mechanism for regulating enzymatic activity.
