Light- and electronmicroscopic distribution of taurine, an organic osmolyte, in rat renal tubule cells.

Several lines of evidence suggest that taurine acts as an organic osmolyte in the kidney. We investigated the cellular and subcellular distribution of this amino acid in rat renal tubule cells. Semi- and ultrathin sections of plastic-embedded rat kidney were incubated with an antiserum against conjugated taurine, using peroxidase-antiperoxidase and immunogold procedures, respectively. Extensive control tests confirmed the selectivity of the antiserum. Our immunocytochemical preparations revealed a highly differentiated labeling pattern. Strong labeling (judged visually or by computer-aided calculation of gold particle densities) was found in collecting duct cells throughout cortex and medulla, in proximal straight tubule cells, and in cells of the descending thin limbs of Henle's loop. Intermediate gold particle densities occurred in proximal convoluted tubule cells and intercalated cells of the collecting ducts (the gold particle in the latter being 30% of that in the collecting duct cells). The distal convoluted tubules, and thick and thin ascending limbs were almost immunonegative. It cannot be excluded that the proportion of free taurine that is retained by the fixative varies somewhat among the different cell types. Yet the highly differentiated labeling pattern that was obtained suggests that taurine is heterogeneously distributed among different populations of tubule cells, and that its level varies substantially even among cells that are exposed to the same osmotic stress.

Demonstration of cytochrome reductases in rat liver peroxisomes: biochemical and immunochemical analyses.

In this study we utilized the analytical cell fractionation approach in combination with immunoblotting techniques and immunoelectron microscopy to test for the presence of NADPH cytochrome P-450 reductase and NADH cytochrome c (b5) reductase in rat liver peroxisomes. Highly purified peroxisomes from clofibrate-treated rats exhibited both NADPH cytochrome P-450 reductase activity and NADH cytochrome c reductase activity (using cytochrome c as an electron acceptor). These activities were inhibited by the respective reductase antibodies made against the endoplasmic reticulum (ER) enzymes. Immunoblot data in combination with immunoelectron microscopy indicated that the peroxisomal NADPH cytochrome P-450 reductase is localized in the matrix of the organelle and has a subunit of molecular weight similar to that of the ER enzyme, whereas the NADH cytochrome c (b5) reductase is localized in the membranes of the peroxisomes. Again, the subunit molecular weight was similar to that of the ER enzyme. The presence of these reductases in peroxisomes further supports the role of this organelle in bile acid synthesis and cholesterol metabolism.

Capitalizing on current technology to improve the electrocardiogram data management process.

Concord Hospital was able to use an existing electrocardiogram (ECG) data management system to greatly improve efficiency regarding the managing, interpreting, viewing, and reporting of ECGs. By taking advantage of the full capabilities of the current ECG data management system, Tracemaster, we were able to design a workflow process to allow any physician to view and interpret electronically any current or previous ECGs from any computer.

A review of the automated detection of change in serial imaging studies of the brain.

Serial imaging is frequently performed on patients with diseases of the brain, to track and observe changes. Magnetic resonance imaging provides very detailed and rich information, and is therefore used frequently for this application. The data provided by MR can be so plentiful; however, that it obfuscates the information the radiologist seeks. A system which could reduce the large quantity of primitive data to a smaller and more informative subset of data, emphasizing change, would be useful. This article discusses motivating factors for the production of an automated process to this effect, and reviews the approaches of previous authors. The discussion is focused on brain tumors and multiple sclerosis, but many of the ideas are applicable to other disease processes, as well.

[The introduction of microcomputerization into the uro- and andrological fields].

Microcomputer systems are used wider and wider in medicine after their incorporation in the diagnosis and treatment of large number of diseases. Computer dispensary of patients with retained testes was used in the chair of urology in 1987 for the first time. Half of the used computer programme was written on the language DBABE. DBABE are 16 beat computers. The functional possibilities for the programme are the following: introduction of data from the first clinical examination, adding new data from the control examinations, correction of the old data, search for data according to a definite criterium, printing of informations in accordance with assigned criterium for seeking in table type, all functional possibilities for generation of informations, printed post communications. The created system for coding data allows their further mathematical processing both with practical and with scientific purposes.

[Multicenter study of obstetrico-perinatologic quality control in the Rostock district. II. Analysis of results with graphic aids].

Various graphic diagrams of obstetrical and perinatological quality assessment between 1984-1986 in the district of Rostock are demonstrated. The advantages of centralization of premature labors are visible. The prenatal diagnosis of intrauterine growth retardation is still insufficient. With an upper cesarean section rate of 10% good perinatological results are achieved. The cesarean section rate of obstetrical departments without high risk concentration is below 5%. There is a highly subjective influence in vaginal operative decision. A mean rate of prematurity may be combined with a high rate of perinatal mortality caused by an unproportional part of very low birth weight infants. The small number of data of the obstetrical basic-check-list allows different possibilities of comparing quality assessment. Different levels of the results and the reasons are better visible by means of graphs. The preliminary results of outcome control justify a general introduction. By means of further data processing we will get better mathematical and graphical interpretations.

The data processing dilemma.

With the inevitability of automation at hand, today's medical groups must face the data processing dilemma with objectivity and thoroughness. The author sketches a background of the evolution of data processing for medical groups and briefly reviews the factors involved in the decision of whether to use a service bureau or buy a computer system. The essence of this article, however, is a discussion of the development of an RFP. A detailed outline of the elements necessary to include is provided, and format, distribution, and evaluation of proposals are all addressed. In conclusion, important considerations in the final selection, negotiation, and conversion processes are presented.
