Control of scrotal colour in the vervet monkey.

The vervet monkey has a vivid blue scrotum which pales when the animal falls in social rank. Histology and electron microscopy showed dopa-positive melanocytes in the dermis each packed with fully melanised melanosomes. By transmitted light the scrotal skin was brown on a red background: by reflected light the colour was blue. Thus the blue colour is due to Tyndall scattering over a layer of melanin. Variation of scrotal colour was not due to changes in melanocyte number or dispersion of melanosomes. Pallor was induced by injecting fluid, and blueness could be restored by removing fluid. It is concluded that the blue-to-white colour variation is modulated by the degree of dermal hydration.

Intrauterine infections in nonhuman primates.

Gross and histologic examination of five nonhuman primate placentas revealed inflammatory processes, either of the ascending type, with chorioamnionitis and fetal vasculitis, or of the hematogenous type with villitis. These reactions were similar to those occurring in man, with known implications for perinatal outcome.

Atherogenic diet-induced modification of the subfraction distribution of high density lipoproteins in monkeys.

High density lipoproteins (HDL) were isolated from the plasma of adult male African green monkeys fed control or atherogenic diets and were then subfractionated in a density gradient by centrifugation at 50,000 rpm, 20 degrees C for 18 hours in the Beckman VTi50 vertical rotor. Material from the gradient was pooled into six subfractions for further analysis. With increasing density across the gradient, the subfractions had progressively decreasing flotation rates, molecular weights, apo A-I/apo A-II mass ratios, lipid percentages of mass, and average sizes. Apoprotein content per particle was reasonably constant compared to lipid content. The physical and chemical properties of the HDL subfractions suggested that the gradient separated three subclasses of HDL equivalent to HDL 2b, HDL 2a, and HDL3 of human beings. When values among animals were compared, the atherogenic diet shifted the relationship between total plasma cholesterol concentration and total HDL concentration from a positive to a negative correlation. The subfractions most affected were those of d less than 1.10 g/ml (HDL2b), while the 1.10 less than d less than 1.13 g/ml (HDL2a) subfractions were affected to a lesser degree; the more responsive the animal to dietary cholesterol, the lower was the mass concentrations of these subfractions. This pattern suggested that diet-induced changes in HDL2 subfraction particle concentrations may be related to the development of atherosclerosis in African green monkeys; parallel changes in particle composition were not identified.

Protection against harmful effects of a restorative procedure using an acidic cavity cleanser.

The present investigation demonstrates that direct application of a composite resin on an acid-etched, vital dentin surface is hazardous to the pulp in monkey teeth. This indicates that exposed dentin should be protected before treating the tooth with a restorative technique including acid etching. A ca(OH)2-containing base material showed promising qualities as a protective liner for this kind of restorative treatment.

Feline insular amyloid: histochemical distinction from secondary systemic amyloid.

Amyloid in islets of Langerhans from 48 domestic cats, one human, one non-human primate, and one raccoon was compared with secondary systemic amyloid from three domestic cats, one dog, one human, and one cow to determine affinity for Congo red dye after treatment of paraffin-embedded tissue sections with potassium permanganate and dilute sulfuric acid. Insular amyloid from all six species was resistant to pretreatment with potassium permanganate, i.e., affinity for Congo red was retained, whereas secondary systemic amyloid from all species was sensitive to the potassium permanganate pretreatment. Other stains did not distinguish between insular and secondary systemic amyloid. The potassium permanganate-Congo red staining procedure thus can be used to differentiate insular from secondary systemic amyloid in the cat and other species. The results also indicate that insular amyloid and secondary systemic amyloid are of different chemical composition and pathogenesis.

Semen characteristics of vervet monkeys.

Semen samples (91) from 47 vervet monkeys were collected by electroejaculation over a 2 year period. Seventy-eight of these were from 37 singly caged males of unknown fertility and 13 from 10 breeding males of known fertility. Mean values for semen characteristics of the singly caged males were: volume 0.45 ml, pH 7.8, concentration 184 x 10(6)/ml, forward progression rating 2.95 (scale 0-4), motility 55.4%, live 68% and abnormal morphology 3.5%. Mean values for semen characteristics for the breeding males were: volume 0.86 ml, pH 9.00, concentration 117.15 x 10(6)/ml, forward progression rating 3.00 (scale 0-4), motility 43.6%, live 53.3% and abnormal morphology 6%. Semen volumes in the singly caged males were lower than the volumes reported in other studies.

Long-term effects of destruction of the endolymphatic sac in a primate species.

The endolymphatic sacs of ten vervet monkeys were surgically ablated unilaterally, and the animals were assigned predetermined survival times ranging from 6 months to 5 years. The left and right ears were prepared in anatomical continuity for light microscopic study. Four experimental ears had openings in their membranous labyrinths of unknown etiology and were not considered acceptable for correlative studies. The remaining five experimental ears showed mild cochlear endolymphatic hydrops, which was localized to the apical regions or was more severe in the apical regions than elsewhere. Four ears also showed saccular endolymphatic hydrops. The sensory and neural structures in both experimental and control ears were normal, and none of the control ears showed endolymphatic hydrops. None of the animals developed severe endolymphatic hydrops or the cochleovestibular symptoms that occur in human subjects with Meniere's disease. The results of this experiment indicate that dysfunction of the endolymphatic sac may be a contributing factor, but probably is not the sole cause of Meniere's disease.

Scanning electron microscopy of fungiform papillae on the tongue of man and monkey.

The surface topography of the fungiform papillae of homo and monkey was studied by scanning electron microscopy. Different methods of specimen preparation were tried. Satisfactory preservation was obtained with fixation in formaldehyde, or in glutaraldehyde with postfixation in OSO4, followed by critical point drying. Taste pores were found in nearly half of the fungiform papillae. Up to 3 pores, located in the dorsal surface, were observed in a single papilla. Most taste pores opened in the form of a rounded crater with a diameter of about 5-7 mum, elevated slightly above the surface of the papilla. Microvilli could be observed in large-bore pores.

Selective habituation of vestibular nystagmus by visual stimulation in the monkey.

Four monkeys were exposed to a series of unidirectional rotating visual fields, of the type known to produce circularvection in man. As in the human case, the vestibulo-ocular reflex for rotation in the direction of the optokinetic nystagmus and circularvection during the preceding visual stimulation--was markedly decreased following that visual stimulation, whereas that in the opposite direction was increased. This finding corresponds to the results of nystagmus and psychophysical circularvection experiments on man. It suggests a link between single unit studies performed under identical conditions in monkey and psychophysical investigations.

A comparative evaluation of the cognitive skills of the chimpanzee and the monkey.

A review of selected aspects of cognitive performance in apes and in monkeys suggests that only rarely can we at this time demonstrate greater cognitive competence in the ape than in the monkey. Instead it seems that the ape's cognitive style differs qualitatively from that of the monkey: not in the direction of greater capacity (using this term as a measure of continuously increasing ability), but in the way that the ape applies itself to a novel situation. From this it would follow that the greater discontinuity lies between man and apes, not between apes and monkeys.

Fluorescent anti-colonic and E. coli antibodies in ulcerative colitis.

With the indirect immunofluorescence technique using sections of colon from guenon and rat, sera from patients with ulcerative colitis yielded positive staining of goblet cells. Eight sera, thus defined positive, two negative, and five from hospital controls, were applied to agar preparations of 12 different E. coli strains, normally found in human bacterial diseases and E. coli 0 14. With E. coli 0 group antigens, 2, 8 and 14 positive fluorescence reactions were regularly found in sera where positive immunofluorescence reaction aginst colon antigen could be demonstrated, indicating that antigen(s) from colon mucosa shares antigenic determinant(s) with some E. coli bacteria or bacterial components.

Decalcification end point determination by X-rays and flame photometry to obtain superior quality pulpal sections.

Radiography and flame photometry have been compared as means of determining the end point of decalcification in relation to minimizing pulp-dentin separation in histological sections of teeth. Eighteen homologous pairs of vervet monkey incisor teeth were decalcified in a formic-citric acid mixture. At 24 hr intervals decalcification was monitored in half of the teeth by radiography and in the other half by flame photometry. When decalcification was complete as determined by the respective methods, histological specimens were prepared and separation at the pulp-dentin interface evaluated in hematoxylin and eosin stained step serial sections. The median separation was determined for the combined group and the median test applied. There was significantly less separation in the flame photometry group and within each group significantly less separation on the side where the knife cut from dentin to pulp.
