Bacterial mutagenic evaluation of Luxabendazole, a new broad spectrum anthelmintic, with the Salmonella typhimurium His- and the Escherichia coli Tryp- reversion tests.
Luxabendazole is a new benzimidazole carbamate chemotherapeutic agent, which has proved to be effective against adult and immature stages of the major gastrointestinal nematodes, trematodes and cestodes. The mutagenic properties of Luxabendazole were investigated in the in vitro Ames Salmonella and E. coli tests. The product was tested at concentrations of 0.5, 5, 50, 500, 1250 and 2500 micrograms/plate in the TA1535, TA1538, TA98 and TA100 strains of Salmonella typhimurium, and 0.5, 5, 50 and 500 micrograms/plate in the WP2, WP2uvrA- and its pKM 101-containing derivative CM891 (WP2 uvrA- pKM101) strains of Escherichia coli, with and without S9 microsomal activation (post-mitochondrial liver fraction from Wistar rats pretreated with Aroclor(R)). Positive and negative controls were included in each experiment. From the present study it can be concluded that Luxabendazole, over a dose range of 0.5-2500 micrograms/plate, is unlikely to present a mutagenic hazard, as demonstrated by the Ames test.

Pharmacokinetics and anthelmintic efficacy of febantel in the racing pigeon (Columba livia).
This paper reports on the pharmacokinetics and the efficacy of febantel against Capillaria obsignata and Ascaridia columbae in the racing pigeon. Febantel was rapidly cleared from the circulation and highly metabolized. The efficacy of febantel against Capillaria and Ascaridia was studied on lightly and heavily infected pigeons. The efficacy against Ascaridia was 100% for the faecal egg count reduction (FECR) as well as for the worm reduction in all treated pigeons. After two treatments with febantel against Capillaria in heavily infected pigeons the FECR amounted to 96.7% and worm reduction was 95%. In order to effectively remove both parasites from the host, repeated treatments with febantel at a short interval could be the treatment of choice.

Effects of three anthelmintic regimes on milk yield of ewes and growth of lambs.
Forty ewes were allocated into one of four groups (n = 10) and were treated with albendazole (ALB) (3.8 mg kg(-1)) before lambing (group A), with ALB before and after lambing (group B), with moxidectin (MXD) (0.2 mg kg(-1)) before lambing (group C) or were untreated controls (group D). Counts of nematode eggs in faeces and coprocultures were carried out during the study, as well as ewes' milk yield measurements and lamb weighings. Pre-treatment mean eggs per gram (epg) counts were 640, 715, 625 and 630 for groups A, B, C and D, respectively (P > 0.05); respective counts 21 days after treatment were 5, 0, 0 and 690 epg, whilst 70 days after treatment they were 380, 145, 40 and 1120 epg. Mean lactation milk yield was 3527.5, 3893.5, 3786.4 and 3285.9 ml for groups A, B, C and D, respectively; no significant difference was evident among the four groups in milk yield collected during the suckling period, although subsequently, group B or C ewes yielded significantly more milk than controls (P < 0.05). Mean birthweight of lambs were 3.56, 3.45, 3.59 and 3.26 kg for groups A, B, C and D, respectively (P = 0.045); subsequently, lambs from treated ewes were significantly heavier than lambs from control animals (P < 0.001). We conclude that anti-parasitic treatment during the last month of pregnancy contributed to an increased birthweight of lambs of treated ewes, whilst the cumulative effect of two doses of ALB or the long persistent efficacy of MXD provided a longer protection of animals against new parasitic infections and contributed to a lactation persistence.
A method for recording the effects of anthelmintics on the movements of Ascaris lumbricoides.
Adult roundworms (Ascaris lumbricoides) obtained from pigs were suspended in modified Tyrode solution at 37 degrees in fine nylon stockings, and the movements of the stockings were recorded. This method of suspension avoided damage to the cuticle or occlusion of the alimentary tract of the worm.Piperazine salts caused gradual narcosis, the time of onset of which was related linearly to the logarithm of the concentration of drug in the bath. The method is suitable for the assay of preparations of piperazine. Tetrachlorethylene, hexylresorcinol, and oil of chenopodium all caused stimulation of movement before the worm was immobilized. Santonin caused incoordination of the movements of the anterior end of the worm; the movements of the body were apparently unaffected. Piperazine is the safest preparation available for the treatment of ascariasis.
Uncoupling activity of the anthelmintic oxyclozanide in rodents.
The uncoupling activity of oxyclozanide in warm blooded animals has been studied in whole animals, isolated tissue in vitro and on mitochondrial preparations. The onset of post mortem rigidity in mice and rats is accelerated and a contracture of striated muscle is produced. Oxyclozanide (1 muM) stimulated rat liver mitochondrial respiration and stimulated an ATP-ase activity.
The interaction between anthelmintic drugs and histamine in Ascaris suum.
1 Piperazine reduced the histamine content of Ascaris suum, yet it greatly increased the uptake of histamine from the surrounding medium, the neuromuscular structures of the nematode preferentially increasing in amount. 2 Bephenium reduced the histamine content of Ascaris and the uptake of histamine from the surrounding medium. However, the relative amount in the neuromuscular structures increased. 3 The flaccid paralysing action of piperazine may thus involve increased histamine absorption whereas the spastic paralysing action of bephenium may be independent of histamine.
