The effect of ACTH on the expression of Fos in rat adrenal glands was investigated immunocytochemically at both light and electron microscopic levels. An approximately 4-fold increase in the number of Fos-positive cortical cells per unit area of the adrenal cortex was found 45 min following a single injection of ACTH (i.v., 2 IU/kg b.w.), as compared with the control animals. The number of immunoreactive cells reached a maximum 90 min after ACTH injection and remained high 150 min after the injection. However, 5 h following the injection it declined significantly. At the ultrastructural level, the ACTH-induced Fos-like immunoreactivity was localized in the nuclei of cortical cells, exclusively confined to the nuclear regions associated with the euchromatin. The result that ACTH enhances Fos-like immunoreactivity suggests that ACTH is involved in c-fos induction in adrenal cortical cells. The characteristic intracellular localization of Fos-like immunoreactivity implies that Fos, once synthesized, may be rapidly translocated into the nuclei of the cells, where it participates in transcriptional regulation of genetic events.

Agouti related protein (AgRP) is a recently discovered melanocortin receptors (MCR) antagonist implicated in the control of feeding behaviour. Expression of AgRP has been shown to be localized by in situ hybridization to the arcuate nucleus and median eminence of the brain, where it acts as an antagonist to the MC3 and MC4 receptors, while in the periphery the only significant expression was located in the adrenal medulla. As AgRP is only a weak antagonist of the MC2 and MC5 receptors, which are expressed principally by adipocytes and in the adrenal cortex, the question arizes as to the function of peripheral AgRP. In this study, we investigated the expression of AgRP in the rat adrenal and suggest that it is expressed in the adrenal cortex and not as previously described in the medulla. We also show that AgRP mRNA expression is upregulated in the adrenal during fasting and in the contralateral gland following unilateral adrenalectomy but not during chronic stress. These results indicate an as yet undefined role for AgRP in the periphery and are supportive of the suggestion that a further melanocortin receptor exists.
The interactions of 2Fe-2S ferredoxins from bovine adrenal cortex, blue-green algae and leaves of three plant species with some positively and negatively charged as well as with uncharged detergents, have been studied. These interactions generally resulted in the destruction of active centers of ferredoxins represented by the 2Fe-2S cluster. The role of oxidants (oxygen and potassium ferricyanide) in cluster destruction by detergents has been established. The opposite sensitivity of plant and animal ferredoxins to anionic and nonionic detergents was revealed.
In cells of the zona fasciculata externa of the adrenal cortex of mice, the maximal value of nuclear volume is observed in evening and night time, while the mitotic peak occurs in the early part of the day. Ten day subcutaneous injection of 1.5 units of ACTH twice in 24 hr produced nuclear hypertrophy and stimulation of mitotic activity of cells of the zona fasciculata externa. The circadian periodicity of nuclear volume in mice injected with ACTH is disturbed, while the circadian rhythm of mitotic activity is retained.

The activity of succinate dehydrogenase from bull adrenal cortex was studied as affected by malonate and oxaloacetate. The both substrate analogs without preincubation (separately and in the mixture) inhibit the enzyme by the competitive type. After a 3 min oxaloacetate preincubation of the enzyme inhibition is of a mixed character. Malonate under these conditions lowers the oxaloacetate effect without changing the type of inhibition. It is supposed that the protective effect is due to a high rate of formation and decay of the enzyme-inhibitory malonate complex.
