Morphometric discrimination of females of five species of Anopheles of the subgenus Nyssorhynchus from Southern and Northwest Colombia.

The most important vectors of human Plasmodium in the neotropics belong to the subgenus Nyssorhynchus. These species are generally sympatric in terms of their geographical distributions. Some are difficult to identify based solely on examination of adult females using the available morphological keys, in these cases examination of immature stages and male genitalia is required to make correct determinations. However, in epidemiological studies it is necessary to identify the species of adult females which are found near humans, i.e. in studies of malaria transmission or evaluation of control measures. The purpose of the present study was to evaluate the discrimination of adult females of different species of Nyssorhynchus isolated mainly from Southern Colombia (department of Putumayo), using morphometric analysis. Adult females were obtained after rearing larvae collected in natural breeding places and from the progeny of females collected on humans. The morphological characteristics of the immature stages allowed the identification of four species of the subgroup Oswaldoi from Southern Colombia: Anopheles rangeli Gabaldon, Cova Garcia & Lopez, An. oswaldoi (Peryassu), An. benarrochi Gabaldon, Cova Garcia & Lopez and An. triannulatus (Neiva & Pinto). The species An. nuneztovari (Gabaldon) from the Northwest of Colombia was included for comparison. Morphometric analysis allowed differentiation of the females of all species to a confidence level approaching 90% using principal components analysis of 10 wing and leg variables, followed by canonical variate analysis of the first four principal components. We conclude that morphometrics may represent a useful taxonomic tool for this group and that its use should be further studied.

A nonparametric quantification of neural response field structures.

The response fields of higher cortical neurons are usually approximated with smooth mathematical functions for the purpose of population parameterization or theoretical modeling. We used instead two nonparametric methods (principal component analysis and independent component analysis), which provided a basis for the response field clustering. Although both methods performed satisfactorily, the principal component analysis space is more straightforward to calculate. It also gave a clear preference toward the smallest number of functional response field classes. Clustering was performed with both K-means and superparamagnetic clustering algorithms with similar results. We also show that the shapes of the eigenvectors remain consistent regardless of the response field data sets size. This finding reflects the fact that the response fields were generated by the same neural network and encode the same underlying process.

Prediction of nucleating sequences from amyloidogenic propensities of tau-related peptides.

Physical properties, including amyloid morphology, FTIR and CD spectra, enhancement of Congo red absorbance, polymerization rate, critical monomer concentration, free energy of stabilization, hydrophobicity, and the partition coefficient between soluble and amyloid states, were measured for the tau-related peptide Ac-VQIVYK amide (AcPHF6) and its single site mutants Ac-VQIVXK amide (X not equal Cys). Transmission electron microscopy showed that 15 out of the 19 peptides formed amyloid in buffer, with morphologies ranging from straight and twisted filaments to sheets and rolled sheets. Using principal component analysis (PCA), measured properties were treated in a comprehensive manner, and scores along the most significant principal components were used to define individual amino acid amyloidogenic propensities. Quantitative structure-activity modeling (QSAM) showed that residues with greater size and hydrophobicity made the largest contributions to the propensity of peptides to form amyloid. Using individual amino acid propensities, sequences within tau with high amyloid-forming potential were estimated and found to include 226VAVVR230 in the proline-rich region, 275VQIINK280 (PHF6) and 306VQIVYK311 (PHF6) within the microtubule binding region, and 392IVYK395 in the C-tail region of the protein. The results suggest that regions outside the microtubule-binding region may play important roles in tau aggregation kinetics or paired helical filament structure.

QSPR of the bioconcentration factors of non-ionic organic compounds in fish using extended topochemical atom (ETA) indices.

Bioconcentration refers to the absorption or uptake of a chemical from the media to an organism's tissues leading to greater concentration in tissues than that in the surrounding environment. Considering the importance of bioconcentration from the viewpoint of ecological safety assessment, a QSPR study was conducted based upon log BCF of 122 non-ionic organic compounds in fish using the recently introduced extended topochemical atom (ETA) indices. In deriving the models, principal component factor analysis (FA) followed by multiple linear regression (MLR), stepwise regression, partial least squares (PLS) and principal component regression analysis (PCRA) were applied as statistical tools. This was repeated with non-ETA (topological and physicochemical) descriptors and a combination set including both the ETA and non-ETA descriptors. The ETA indices suggested negative contributions of functionalities of nitro, amino and hydroxy substructures and positive contributions of branching, volume and functionality of chloro substituents. Again, the predictive ability of the developed models was compared with the previously reported models. Finally the validation of all the QSAR models was discussed based on random division, sorted log BCF data and K-means clusters for the factor scores of the original variable (ETA) matrix without the response property values. The results suggest that ETA parameters are sufficiently rich in chemical information to encode the structural features contributing to the bioconcentration of the non-ionic organic compounds in fish and thus these merit further assessment to explore their potential in QSAR/QSPR/QSTR modelling.

[A new linear neural network multi-component analysis method and its application in the analysis of VC yinqiao tablets quantitative analysis].

We measured NIR spectrum of VC yinqiao tablets with spectral instrument, analyzed the contents of acetaminophen and vitamin C in the VC yinqiao tablets with principal component analysis (PCA) and Linear Neural Network, and discussed the choice of principal component number and ANN's parameters affecting the network. To compare arithmetic performance, the authors also processed the spectral data with partial least squares and PCA-BP neural network. Compared with other two data process methods, the experiment and the result of data process showed that the PCA-linear neural network possess the best forecasting precision.

Profiling phenolic metabolites in transgenic alfalfa modified in lignin biosynthesis.

Soluble phenolics, wall-bound phenolics and soluble and core lignin were analyzed in transgenic alfalfa with genetically down-regulated O-methyltransferase genes involved in lignin biosynthesis. High performance liquid chromatography and principal component analysis were used to distinguish metabolic phenotypes of different transgenic alfalfa genotypes growing under standard greenhouse conditions. Principal component analysis of HPLC chromatograms did not resolve differences in leaf metabolite profiles between wild-type and transgenic plants of the same genetic background, although stem phenolic profiles were clearly different between wild-type and transgenic plants. However, the analytical methods clearly differentiated two non-transgenic alfalfa cultivars based on either leaf or stem profiles. Metabolic profiling provides a useful approach to monitoring the broader biochemical phenotypes of transgenic plants with altered expression of lignin pathway enzymes.

Nonlinear modeling and adaptive monitoring with fuzzy and multivariate statistical methods in biological wastewater treatment plants.

A new approach to nonlinear modeling and adaptive monitoring using fuzzy principal component regression (FPCR) is proposed and then applied to a real wastewater treatment plant (WWTP) data set. First, principal component analysis (PCA) is used to reduce the dimensionality of data and to remove collinearity. Second, the adaptive credibilistic fuzzy-c-means method is used to appropriately monitor diverse operating conditions based on the PCA score values. Then a new adaptive discrimination monitoring method is proposed to distinguish between a large process change and a simple fault. Third, a FPCR method is proposed, where the Takagi-Sugeno-Kang (TSK) fuzzy model is employed to model the relation between the PCA score values and the target output to avoid the over-fitting problem with original variables. Here, the rule bases, the centers and the widths of TSK fuzzy model are found by heuristic methods. The proposed FPCR method is applied to predict the output variable, the reduction of chemical oxygen demand in the full-scale WWTP. The result shows that it has the ability to model the nonlinear process and multiple operating conditions and is able to identify various operating regions and discriminate between a sustained fault and a simple fault (or abnormalities) occurring within the process data.

The Edinburgh cohort of HIV-positive drug users: the relationship between auditory P3 latency, cognitive function and self-rated mood.

One hundred and six HIV-positive drug users were tested with a two-tone auditory evoked potential (AEP) task and a small battery of neuropsychological tests, to examine the relationship between the latency of the P300 component (P3) of the AEP, intellectual function, mood and drug use. Principal components analysis revealed a significant correlation between P3 latency and the first principal component (r = -0.43, P < 0.001). Varimax rotation generated three orthogonal components which we interpreted as intellectual performance, memory, and mood. Intellectual performance and self-reported mood were individually correlated with P3 latency, but memory was not (r = -0.36, P < 0.001; r = 0.23, P < 0.05; and r = -0.18, NS, respectively). Subjects with symptomatic HIV disease had a higher correlation between P3 latency and intellectual performance than subjects with asymptomatic HIV disease and, among patients with symptomatic HIV disease, poorer memory was associated with a lower CD4 count. Opiate or benzodiazepine consumption did not correlate with poor intellectual performance, memory, or self-rated mood in our sample. These results indicate that there is a relationship between AEP latency and neuropsychological measures of intellectual function, and that it is influenced by subjective mood. Surprisingly, declared current drug use has no discriminable effect on these relationships.

Mycotoxin formation in HY-320 wheat during granary storage at 15 and 19% moisture content.

Eleven-kilogram parcels of HY-320 wheat, a cultivar of the new Canada Prairie Spring class, were kept at 15 and 19% initial moisture contents (IMC) in simulated storage in a Manitoba farm granary for 60 weeks to determine biotic and abiotic changes and mycotoxin production. Ochratoxin A reached a maximum of 0.24 ppm by week 20 in the 19% IMC wheat, but was absent in the 15% IMC wheat; no other mycotoxins were detected. Temperature, moisture content, O2 and CO2 levels, fat acidity values, seed germination, microfloral incidence and abundance, and the presence of other mycotoxins were monitored. Principal component analysis of all variables showed that the first principal components accounted for 32-41% of the system variability, and contained the ochratoxin A variable. Ochratoxin A was produced in moist grain that had decreased seed germination and Alternaria activity, and high fungal activity by Penicillium and Aspergillus versicolor. Compared to other stored cereals previously studied, HY-320 wheat would be ranked in a low-risk category for mycotoxin formation, based on the ochratoxin A levels observed.

Functional and histopathologic correlation in patients with dilated cardiomyopathy: an integrated evaluation by multivariate analysis.

To correlate left ventricular function and histologic features in patients with dilated cardiomyopathy, precise indexes of hemodynamics and semiquantitative histologic data were combined for multivariate analysis. Right endomyocardial biopsy was performed at the time of cardiac catheterization. Five hemodynamic indexes were used for functional assessment: ejection fraction, ratio of end-systolic stress to volume index, end-diastolic stress, time constant (T) of left ventricular pressure fall, and end-systolic stress. Six histologic findings (disarray of myofibers, hypertrophy of myofibers, scarcity of myofibrils, nuclear changes of myofibers, vacuolization of myofibers and proliferation of collagen fibers) were graded from (-) to (4+). Each finding was assigned to category (-) or (+) according to the absence or presence of significant abnormality. Ordinary statistical analysis revealed that, although ejection fraction was lower in category (+) for proliferation of collagen fibers, ratio of end-systolic to volume index was reduced for category (+) of hypertrophy of myofibers. A significant correlation was present between hypertrophy of myofibers and proliferation of collagen fibers by Spearman rank correlation. When principal component analysis was applied to the hemodynamic data, two principal components could be extracted. Fisher's discriminant analysis could clearly differentiate two categories (-) and (+) in the semiquantitative histologic finding of proliferation of collagen fibers. The analysis indicated that contractility was reduced with elevated afterload in that category (+). Thus, proliferation of collagen fibers may play a pivotal role in deteriorating contractility in patients with dilated cardiomyopathy.

Application of multivariate analysis and artificial neural networks for the differentiation of red wines from the Canary Islands according to the island of origin.

Eleven metals (K, Na, Ca, Mg, Fe, Cu, Zn, Mn, Sr, Li, and Rb) were determined in 83 red wines from the Canary Islands. The wines presented high concentrations of Na, and the concentrations of Cu and Zn were much lower than the maximum concentrations established by the International Office of Vine and Wine (OIV). Applying principal component analysis, the dimension space was reduced to five principal components that explain 76.4% of the total variance, and the wines tend to separate on the basis of the island of production. Linear discriminant analysis (LDA) allowed a reasonable classification of wines according to the island of production. When artificial neural networks (Kohonen self-organizing maps and back-propagation feed-forward as unsupervised and supervised techniques, respectively) were applied on the matrix of data constituted by the analyzed metals, the results improved in relation to those obtained by other multivariate methods observing a differentiation of wines according to island of production.

Analysis of mandible of newly developing strain of Japanese white rabbit (Nib:JWNS).

In the present paper, we applied analysis of the mandible to a new strain of Japanese White rabbit being developed (Nib: JWNS), at 5th and 8th generations (F5 and F8), and evaluated the process of establishment, making comparison with the parent colonies (I, K and L), and other established JW colonies (A, E, O and R). The mandibles were measured at 12 sites and the data obtained were calculated by principal component analysis and discriminant analysis. As a result of principal component analysis, the parent colonies were found to be close to one another with intermediate size of the mandible and height of condyloid and angular processes, but in F5 and F8 the mandibles were slightly shorter in height and different from those in the parent colonies. On the other hand, results of discriminant analysis revealed that mandibles were discriminated correctly 100% in colonies A, O and R, and 90% in E and I. Colonies A, O, R, E and I were therefore regarded as established strains. In the developing colonies, the discriminant rate was 70% in F5 and 80% in F8. One mandible from F5 and two from F8 were wrongly classified to the parent colony L, which was the lowest discriminant rate among the colonies examined (61.5%). The results of both statistical analyses suggest that the JWNS are almost established at 8th generation as a new strain.

Development of a reliable and valid psychosocial measure of self-perception of health in type 2 diabetes.

OBJECTIVES: The aim was to develop a reliable and valid psychosocial measure of self-perception of health in type 2 diabetes. METHODS: Item analysis and factor analyses were done in order to stabilize and establish principal components of the questionnaire. Reliability (internal consistency aspect) was established using Chronbach's alpha method. Concurrent and discriminant validities were established using conventional methods. RESULTS: Factor analysis yielded 12 significant factors (eigen value > or =1), but first six components were retained based on Scree test. These six principal varimax factors explain 49.7% of variance of psychosocial measure of self-perception of health. CONCLUSIONS: The final version after all these psychometric procedures had 27 items with six principal components. They were appropriately named as follow: (I) Positive self-feeling; (II) Sociality; (III) Attention seeking; (IV) Feel healthy; (V) Worry about health; (VI) Dependence. Total variance explained is 49.7%.

Micro-Raman spectroscopy detects individual neoplastic and normal hematopoietic cells.

Current methods for identifying neoplastic cells and discerning them from their normal counterparts are often nonspecific, slow, biologically perturbing, or a combination thereof. Here, we show that single-cell micro-Raman spectroscopy averts these shortcomings and can be used to discriminate between unfixed normal human lymphocytes and transformed Jurkat and Raji lymphocyte cell lines based on their biomolecular Raman signatures. We demonstrate that single-cell Raman spectra provide a highly reproducible biomolecular fingerprint of each cell type. Characteristic peaks, mostly due to different DNA and protein concentrations, allow for discerning normal lymphocytes from transformed lymphocytes with high confidence (p << 0.05). Spectra are also compared and analyzed by principal component analysis to demonstrate that normal and transformed cells form distinct clusters that can be defined using just two principal components. The method is shown to have a sensitivity of 98.3% for cancer detection, with 97.2% of the cells being correctly classified as belonging to the normal or transformed type. These results demonstrate the potential application of confocal micro-Raman spectroscopy as a clinical tool for single cancer cell detection based on intrinsic biomolecular signatures, therefore eliminating the need for exogenous fluorescent labeling.

Spatial and temporal features of ambient air-quality over Taiwan.

This study analyzed a time series data of daily subPSI values to determine the temporal and spatial characteristics of ambient air-quality in Taiwan by employing the Varimax rotational method. The first and second rotational principal components nearly accounted for 54 and 19% for the variation of subPSI values over Taiwan. The factor loadings between rotational principal components and subPSI values of air pollutants identified the first and second components as combustion and photochemical sources, respectively. Time series analysis of rotational component scores associated with regions showed the relationships between rotational components and subPSI values and also revealed the optimum simulation period for combustion and photochemical sources.

Changes in soil microbial community structure associated with two types of genetically engineered plants analyzing by PLFA.

With the rapid expansion of GEPs(genetically engineered plants), people are more and more concerned about the ecological risks brought by their release. Assessing the effect of GEPs on soil microbial ecology is indispensable to study their ecological risks. In our study, the phospholipids fatty acid ( PLFA) method was used to analyze the microbial community of soil samples collected from fields with two types of GEPs-Bt transgenic corn and PVY ( potato virus Y) cell protein gene transgenic potato. The principal components analysis (PCA) showed all controls were on the right of related GEPs samples along the PC1 (the first principal component) axis, which means a decrease of fungi in soils with genetically engineered crop since most of PLFAs that are strongly positively correlated with PC1 represent fungi. For samples collected from Bt transgenic cornfield, the ratios of gram-positive to gram-negative bacteria were less than those of controls. For samples of transgenic potato field, these ratios were lower than those of controls when soils were collected from deep layer (20-40 cm), but were higher when soils collected from surface layer(0-20 cm). For soils collected from 0-20 cm, the ratios of fungi to bacteria for all GEPs samples were at the same level. So were such rations for all controls. Changes of soil microbial community in two types of GEPs fields were detected in our study, but the causes and more information still needs further study.

Rapid screening of the fermentation profiles of wine yeasts by Fourier transform infrared spectroscopy.

A rapid screening method for the evaluation of the major fermentation products of Saccharomyces wine yeasts was developed using Fourier transform infrared spectroscopy and principal component factor analysis. Calibration equations for the quantification of volatile acidity, glycerol, ethanol, reducing sugar and glucose concentrations in fermented Chenin blanc and synthetic musts were derived from the Fourier transform infrared spectra of small-scale fermentations. The accuracy of quantification of volatile acidity in both Chenin blanc and synthetic must was excellent, and the standard error of prediction was 0.07 g l(-1) and 0.08 g l(-1), respectively. The respective standard error of prediction in Chenin blanc and synthetic musts for ethanol was 0.32% v/v and 0.31% v/v, for glycerol was 0.38 g l(-1) and 0.32 g l(-1), for reducing sugar in Chenin blanc must was 0.56 g l(-1) and for glucose in synthetic must was 0.39 g l(-1). These values were in agreement with the accuracy obtained by the respective reference methods used for the quantification of the components. The screening method was applied to quantify the fermentation products of glycerol-overproducing hybrid yeasts and commercial wine yeasts. Principal component factor analysis of the fermentation data facilitated an overall comparison of the fermentation profiles (in terms of the components tested) of the strains. The potential of Fourier transform infrared spectroscopy as a tool to rapidly screen the fermentative properties of wine yeasts and to speed up the evaluation processes in the initial stages of yeast strain development programs is shown.

Effects of individual characteristics on healthy oral mucosa autofluorescence spectra.

Autofluorescence spectroscopy is a tool for detecting tissue alterations in vivo. In a previous study, we found spectral differences between clinically normal mucosa of different patient groups. These are possibly caused by associated patient characteristics. In the present study, we explore the influences of volunteer characteristics on healthy oral mucosa autofluorescence. Autofluorescence spectra were recorded in 96 volunteers with no clinically observable oral lesions. We applied principal components analysis to extract the relevant information. We used multivariate linear regression techniques to estimate the effect of volunteer characteristics on principal component scores. Statistically significant differences were found for all factors but age. Skin color strongly affected autofluorescence intensity. Gender differences were found in blood absorption. Alcohol consumption was associated with porphyrin-like peaks. However, all differences but those associated with skin color were of the same order of magnitude as standard deviations within categories. The effects of volunteer characteristics on autofluorescence spectra of the oral mucosa are measurable. Only the effects of skin color were large. Therefore, in lesion classification, skin color should be taken into account.
