The effect of exogenous -aminolaevulinate on rat liver haem and cytochromes.

The activity of delta-aminolaevulinate synthetase is generally regarded as rate-limiting for hepatic haem biosynthesis. It has been suggested that cytochrome synthesis may also be regulated by changes in delta-aminolaevulinate synthetase activity. This hypothesis was studied by injecting product, delta-aminolaevulinate, into adult rats over a 4-240h period. The concentrations of hepatic mitochondrial cytochromes a, b, c and c(1) were unchanged by treatment with delta-aminolaevulinate, allylisopropylacetamide or phenobarbital. In control animals, total microsomal haem content equalled the sum of cytochromes b(5) plus P-450. After delta-aminolaevulinate administration the total amount of microsomal haem, measured as the pyridine haemochromogen, exceeded these components, indicating the formation of a ;free' haem pool. Haem synthesis does not appear rate-limiting for hepatic cytochrome synthesis in the adult rat.

Aberration of porphyrin metabolism in iron-deficient anaemic rats.

Iron-deficient rats excreted greater amounts of 5-aminolaevulinate in urine but the output of porphobilinogen was decreased considerably. In addition, activity of the enzyme 5-aminolaevulinate dehydratase was diminished in bone marrow and liver. The results suggest that in iron deficiency a defect occurs in porphyrin synthesis at the level of porphobilinogen formation.

Mechanism and stereochemistry of the 5-aminolaevulinate synthetase reaction.

1. Two mechanisms for the biosynthesis of 5-aminolaevulinate from glycine and succinyl-CoA (3-carboxypropionyl-CoA) are considered. One of the mechanisms involves the retention of both the C-2 H atoms of glycine during the synthesis of 5-aminolaevulinate, whereas the other predicts the retention of only one of the C-2 H atoms of glycine. 2. Highly purified 5-aminolaevulinate synthetase from Rhodopseudomonas spheroides was used to show that the C-2 H atom of glycine with R configuration is specifically removed during the biosynthesis of 5-aminolaevulinate. 3. The mechanism of the condensation therefore differs from the analogous reaction of the biosynthesis of sphinganine from palmitoyl-CoA and serine, in which the C-2 H of serine is retained (Wiess, 1963).

Synthesis and degradation of mitochondrial components in hypertrophied rat heart.

The accumulation of inner mitochondrial components of rat heart was studied 1 and 3 days after constriction of the ascending aorta of rats. By 1 day after aortic constriction, the activities of three mitochondrial respiratory enzymes/mg of cardiac homogenate protein were increased; after 3 days, specific activities had levelled off or decreased. Selective accumulation of inner mitochondrial membrane components 24h after aortic constriction was further indicated by increased left ventricular cytochrome c concentration (nmol/mg of protein). By 3 days after surgery, cytochrome c concentration was significantly diminished. Low-temperature spectroscopy of isolated mitochondria showed that the ratios of cytochromes c, b and a+a(3) remained unchanged after aortic constriction, suggesting that cytochrome c was a good indicator of the response of the other mitochondrial inner-membrane cytochromes as well. The effect of cardiac hypertrophy on the turnover of cytochrome c was also examined. Cytochrome c was labelled in its haem group with delta-amino[2,3-(3)H(2)]laevulinate 3 days before aortic constriction. By 1 day after surgery the total ventricular radioactivity in cytochrome c of aortic banded animals was significantly higher than in sham-operated controls, indicating a decreased degradation rate in the former during the first postoperative day. delta-Aminolaevulinate was shown to be a particularly suitable precursor for such turnover studies, since it results in rapid pulse-labelling of cytochrome c (peak activity in 90min), is rapidly removed from the precursor pool (t((1/2))=30min) and is not reutilized.

Role of membrane-bound and free polyribosomes in the synthesis of cytochrome c in rat liver.

The functional distinction of membrane-bound and free polyribosomes for the synthesis of exportable and non-exportable proteins respectively is not so strict as was initially thought, and it was therefore decided to investigate their relative contribution to the elaboration of an internal protein integrated into a cell structure. Cytochrome c was chosen as an example of a soluble mitochondrial protein, and the incorporation of [(14)C]leucine and delta-amino[(14)C]laevulinate into the molecule was studied by using different ribosomal preparations from regenerating rat liver. A new procedure was devised for the purification of cytochrome c, based on ion-exchange chromatography combined with sodium dodecyl sulphate-polyacrylamide-gel electrophoresis. In spite of cytochrome c being a non-exportable protein, the membrane-bound polyribosomes were at least as active as the free ribosomes in the synthesis in vitro of the apoprotein and the haem moiety. The detergent-treated ribosomes could also effect the synthesis of cytochrome c, although at a lower rate. Since in liver more than two-thirds of the ribosomes are bound to the endoplasmic-reticulum membranes, it is considered that in vivo they are responsible for the synthesis of most of the cytochrome c content of the cell. This suggests that in secretory tissues the endoplasmic reticulum plays a predominant role in mitochondrial biogenesis, although free ribosomes may participate in the partial turnover of some parts of the organelle. The hypothesis on the functional specialization of the different kinds of ribosomes was therefore modified to account for their parallel intervention in the synthesis of proteins associated with membranous structures.

Purification, properties and synthesis of delta-aminolaevulinate dehydratase from Neurospora crassa.

Delta-aminolaevulinate dehydratase, the second and rate-limiting enzyme of the haem-biosynthetic pathway, was purified 300-fold from induced cultures of Neurospora crassa. The native enzyme has a mol.wt. of about 350000, whereas the salt-treated enzyme after incubation at 37 degrees C for 10 min has a mol.wt. of about 232000. The mol.wt. of the subunit is about 38000. Antibodies to the purified enzyme were raised in rabbits. By using radiolabelling and immunoprecipitation techniques it was shown that addition of iron and laevulinate to iron-deficient cultures brings about a significant increase in the synthesis of the enzyme, and protoporphyrin, the penultimate end product of the pathway, represses enzyme synthesis.
