The amitochondriate eukaryote Trichomonas vaginalis contains a divergent thioredoxin-linked peroxiredoxin antioxidant system.

Trichomonas is an amitochondriate parasitic protozoon specialized for an anaerobic lifestyle. Nevertheless, it is exposed to oxygen and is able to cope with the resultant oxidative stress. In the absence of glutathione, cysteine has been thought to be the major antioxidant. We now report that the parasite contains thioredoxin reductase, which functions together with thioredoxin and thioredoxin peroxidase to detoxify potentially damaging oxidants. Thioredoxin reductase and thioredoxin also reduce cystine and so may play a role in maintaining the cellular cysteine levels. The importance of the thioredoxin system as one of the major antioxidant defense mechanisms in Trichomonas was confirmed by showing that the parasite responds to environmental changes resulting in increased oxidative stress by up-regulating thioredoxin and thioredoxin peroxidases levels. Sequence data indicate that the thioredoxin reductase of Trichomonas differs fundamentally in structure from that of its human host and thus may represent a useful drug target. The protein is generally similar to thioredoxin reductases present in other lower eukaryotes, all of which probably originated through horizontal gene transfer from a prokaryote. The phylogenetic signal in thioredoxin peroxidase is weak, but evidence from trees suggests that this gene has been subject to repeated horizontal gene transfers from different prokaryotes to different eukaryotes. The data are thus consistent with the complexity hypothesis that predicts that the evolution of simple pathways such as the thioredoxin cascade are likely to be affected by horizontal gene transfer between species.

Subsequent sexually transmitted infections among adolescent women with genital infection due to Chlamydia trachomatis, Neisseria gonorrhoeae, or Trichomonas vaginalis.

OBJECTIVE: To identify factors associated with subsequent sexually transmitted infection (STI) (within 1 year of initial infection) due to Chlamydia trachomatis, Neisseria gonorrhoeae or Trichomonas vaginalis. DESIGN: Prospective cohort study. SETTING: A sexually transmitted diseases clinic and four community-based primary care clinics for adolescents. PARTICIPANTS: Female patients (ages 15 to 19 years) with initial diagnosis of chlamydia, gonorrhea, or trichomonas. MAIN OUTCOME MEASURES: Subsequent infection by chlamydia, gonorrhea, or trichomonas. RESULTS: More than 40% of subjects were subsequently infected by at least one STI. Reinfection was common, but infections with sexually transmitted organisms other than the initial infecting organism were also common. Predictors of subsequent infection were black race, gonorrhea as the initial infection, two or more sex partners in the previous 3 months, and inconsistent condom use. CONCLUSIONS: Subsequent STI frequently follow an initial STI, but there is substantial variation in the causal organism. These data suggest the importance of comprehensive STI prevention programs for adolescents rather than organism-specific interventions.

Influence of growth conditions on RNA levels in relation to activity of core metabolic enzymes in the parasitic protists Trypanosoma brucei and Trichomonas vaginalis.

Levels of mRNAs encoding metabolic enzymes and their cellular activities were measured on continuous culture samples of the parasitic protists Trypanosoma brucei and Trichomonas vaginalis. The organisms were grown in chemostats at varying growth rates under glucose limitation or in the presence of excess glucose (EG), resulting in extensive adaptation of the cellular activities of glycolytic enzymes. rRNA and mRNA for beta-tubulin were monitored as controls. In Trypanosoma brucei levels of all RNAs showed a biphasic dependence on growth rate (= dilution rate D), with a sharp increase at higher D values. Cellular RNA levels of Trichomonas vaginalis rate-limited by glucose decreased slightly with increasing D. In EG-grown cells the opposite trend was observed. Equal levels for both carbon regimes were observed at intermediate D values. In both species the ratio between rRNA and mRNA encoding beta-tubulin was constant, independent of the carbon regime. mRNA encoding metabolic enzymes showed varying degrees of correlation with rRNA and beta-tubulin mRNA. In contrast, there was little to no correlation between mRNA levels and the activities of the enzymes they encode, even though only one of these is allosterically regulated. The data indicate that RNA levels in Trypanosoma brucei and Trichomonas vaginalis are determined by growth rate and in the latter species by the availability of the carbon and energy source. Rates of synthesis of metabolic enzymes are most likely regulated at the post-transcriptional level.

A divergent ADP/ATP carrier in the hydrogenosomes of Trichomonas gallinae argues for an independent origin of these organelles.

The evolution of mitochondrial ADP and ATP exchanging proteins (AACs) highlights a key event in the evolution of the eukaryotic cell, as ATP exporting carriers were indispensable in establishing the role of mitochondria as ATP-generating cellular organelles. Hydrogenosomes, i.e. ATP- and hydrogen-generating organelles of certain anaerobic unicellular eukaryotes, are believed to have evolved from the same ancestral endosymbiont that gave rise to present day mitochondria. Notably, the hydrogenosomes of the parasitic anaerobic flagellate Trichomonas seemed to be deficient in mitochondrial-type AACs. Instead, HMP 31, a different member of the mitochondrial carrier family (MCF) with a hitherto unknown function, is abundant in the hydrogenosomal membranes of Trichomonas vaginalis. Here we show that the homologous HMP 31 of closely related Trichomonas gallinae specifically transports ADP and ATP with high efficiency, as do genuine mitochondrial AACs. However, phylogenetic analysis and its resistance against bongkrekic acid (BKA, an efficient inhibitor of mitochondrial-type AACs) identify HMP 31 as a member of the mitochondrial carrier family that is distinct from all mitochondrial and hydrogenosomal AACs studied so far. Thus, our data support the hypothesis that the various hydrogenosomes evolved repeatedly and independently.

Risk factors for gonorrhoea, syphilis, and trichomonas infections among women attending family planning clinics in Nairobi, Kenya.

OBJECTIVE: To identify the risk factors for gonorrhoea, syphilis, and trichomonas infections among low risk women in Nairobi, Kenya. METHOD: In a cross-sectional study, 4,404 women attending two peri-urban family planning clinics between 1989 and 1991 were interviewed using a structured questionnaire and examined for signs of sexually transmitted disease (STD) infection. Cervical cultures for gonorrhoea, PAP smear (including microscopy for trichomonas), RPR and HIV testing were done. RESULTS: Positive cervical cultures for gonorrhoea were found in 3.2% of women, positive syphilis serology in 1.9%, and positive trichomonas microscopy in 5.2%. Genital ulcers were found in 1.9% of women. Although unmarried status and reporting more than one sex partner in the previous year were both significantly associated with each disease in the crude analysis, these associations were attenuated after controlling for each other and for other risk factors. The population attributable risks (PARs) for these factors were low (7-16%) owing to the high proportion of cases who were married and monogamous. The majority of women with microbiological evidence of infection had normal pelvic examinations. Clinical diagnostic algorithms for STDs in this population had a low sensitivity and positive predictive value. Nevertheless, a strong association between HIV seropositivity and STDs was observed. CONCLUSION: The low population attributable risks found in this study suggest that behaviour change messages directed to women, particularly if they are married have a low potential for preventing STDs. The poor performance of clinical diagnostic algorithms illustrates the desirability of testing these algorithms in a variety of populations and reinforces the need for low-cost methods of microbiologic diagnosis if populations with relatively low prevalences of these infections are to be included in programmes to diagnose and treat STDs.

Late recurrence of resistant Trichomonas vaginalis vaginitis: relapse or re-infection?

Distinguishing between re-infection and relapse of trichomonas infections is often a difficult task in the clinical setting. The chronicity of trichomonas infections and the ongoing sexual activity are two confounding factors. We present a patient with recurrent resistant vaginal trichomoniasis shortly following a sexual contact with an untreated partner after a complete response to treatment with tinidazole for nine months. We hypothesise that re-infection occurred from the asymptomatic partner who was an untreated chronic carrier of resistant trichomonas in the urogenital tract.

[The influence of antimetabolites on Trichomonas vaginalis as an experimental model].

The present study was concerned with the relationship between antimetabolites and Trichomonas vaginalis. In all of the strains (with p-nitrobenzylether of N-acetyldiiodtryrosine) tested, the growth of Trichomonas vaginalis was identical to that of the control series at concentrations up to 2 mg/ml. At higher concentrations a decrease in multiplication was noted. In the experimental series with antitrypsin, from 1 mg/ml produced an inhibiting effect upon the Trichomonas population. Further investigations on the combined inhibition test are envisaged.

Phenotypic variation of surface antigenic determinants in Trichomonas vaginalis detected by monoclonal antibodies.

We produced a large panel of murine monoclonal antibodies against surface determinants of Trichomonas vaginalis using the hybridoma technique. An immunoenzymatic technique (E.L.I.S.A.) was used to screen positive hybrid cells producing specific antibodies against the protozoan surface. Eleven monoclonal antibodies (Mabs) out of seventy-seven positives were further characterized. We tested antibody reactivity in order to investigate the antigenic variance among 13 different strains of Trichomonas vaginalis of different geographic origin. To elucidate the complexity of antigenic expression in Trichomonas vaginalis, further characterization of the antigenic pattern in our 13 clinical isolates was carried out by immunoblotting techniques. We demonstrate that some monoclonal antibodies react with antigens varying in molecular weight in the different strains tested. We also demonstrate the pivotal role of protozoan proteases in antigenic rearrangement.

[In vitro evaluation of the activity of butoconazole against Trichomonas vaginalis].

An in vitro study was carried out on ten first clinical isolates of Trichomonas vaginalis. Increasing levels of butaconazole were added to 150 microliters of a dilution in Roiron medium adjusted to 50,000 Trichomonas/ml. Results were read after 1, 2, 6 and 24 hours aerobic incubation at 37 degrees C. Minimum lethal concentration was 93 micrograms/microliters after one hour, 23 micrograms/microliter after six hours and 12 micrograms/microliters after 24 hours. These findings warrant in vivo studies of butoconazole against Trichomonas. This drug, which has already been proved effective in fungal infections, may prove very useful since concomitant infection of the genital tract by Trichomonas and Candida is common.

[Influence of the metabolite produced by Trichomonas vaginalis on human sperm motility in vitro].

OBJECTIVE: To investigate the effects of the metabolite produced by Trichomonas vaginalis on human sperm motility in vitro. METHODS: Trichomonas vaginalis having been cultured, the culture solution containing metabolite was obtained by removing the protozoa, then diluted into 3 kinds of concentration. Sperm was obtained from 10 healthy fertile men by masturbation and prepared by swim-up technique to produce a spermatozoon solution of high motility. Every sperm sample was divided into 4 groups (A, B, C, D). Unused culture solution was added to Group A as control, and the other 3 groups (B, C, D) were respectively incubated with the above used culture solution at 3 kinds of concentration (1.2 x 10(9)/L, 6 x 10(8)/L, 1.2 x 10(8)/L). Measurements were carried out at 30 s, 1 h, 2 h, 4 h, 6 h by CASA. RESULTS: Sperm motility decreased in both Group B and C markedly, and the effects displayed a concentration- and time-dependent manner. CONCLUSION: The metabolite of Trichomonas vaginalis can reduce human sperm motility in vitro, and may be one of the causes of infertility.

[Cloning and sequence analysis of Trichomonas vaginalis ferredoxin gene].

OBJECTIVE: To construct a recombinant plasmid containing ferredoxin gene of Trichomonas vaginalis. METHODS: Total DNA was extracted from Trichomonas vaginalis with Chelex-100 method and used as templates for PCR. Primers were designed based on the published sequence of the ferredoxin gene and used to amplify the Trichomonas vaginalis gene using PCR method. The ferredoxin gene obtained by PCR technique was directionally cloned into plasmid pMD-18T simple vector. The constructed recombinant plasmid was transferred into E. coli JM109. The transformants were screened and identified by PCR and restriction analysis. The DNA sequence of the gene was determined by Sanger's method. RESULTS: The size of amplified ferredoxin gene was 306bp. The correct recombinant plasmid was isolated and confirmed by PCR and restriction analysis. The DNA sequence of cloned gene was the same as the published sequence. CONCLUSION: The ferredoxin gene was successfully amplified and cloned into plasmid pMD-18T simple vector. The cloned ferredoxin gene could be used to produce recombinant protein and for study of its function.

[Therapeutic aspects of trichomonas infection of the female genitals].

Infection with trichomonas is a common event in patients in gynecologic practice. In patients in an outpatient department the incidence of trichomonas infection was 7%, in cases with other genital infections 16%. Especially in alcalic vaginal milieu and in Gardnerella infections higher rates of trichomonas could be observed. The recurrence rate after monotherapy was 8%.

Trichomonas infections in men.

Trichomonas vaginalis, a common pathogen in the female genital tract, produces a characteristic clinical picture in women. Less well recognized are the manifestations of Trichomonas infestations of the male genital tract, which include urethritis and chronic prostatitis. Multiple-glass urinalysis and selective use of Trichomonas cultures may improve recognition of this organism in the family practice setting.

The influence of certain B12-antagonists upon trichomonas foetus.

The antagonistic substances used in our experiments showed various relationships to the pathogenic protozoa Trichomonas foetus. Theseinacid and L-2-Methyl-2-Aminoaethenole demonstrated comparable inhibiting influences on the Trichomonas population. Puriniribosid, 6-Mercaptopurine, 4-Mercaptopteridine and Decobald-Cobamide exercised no recognisable influence on the Trichomonas foetus.

[Effects of different fixatives for Trichomonas vaginalis in indirect fluorescent antibody test].

This paper dealt with the effects of different fixatives for Trichomonas vaginalis in indirect fluorescent antibody test. T. vaginalis fixed by formalin, methanol or alcohol displayed clear flagella. The parasites fixed by methanol and alcohol showed specific ring-like fluorescence with 1-2 bright dots. Antigen slides prepared from different Trichomonas strains, different number of parasites and anti-Trichomonas vaginalis McAb strains in IFA presented different results. Antigen slides showed the best results when using parasites at concentrations of 1.2 x 10(6) cells/ml or 2.4 x 10(5) cells/ml. Antigen preparations stored at 4 degrees C or -20 degrees C remained reactive throughout the experimental period of three months.

[Dramatic reduction of the incidence of trichomonas vaginalis infections].

In contrary to former meanings the trichomonas-vaginalis-infection doesn't play any longer an important role. Nowadays it even seems to be a rarity. So we found an infection-rate of 2.17% in 1979 (n = 4005), whereas in 1992 only in 0.19% (n = 1048). The possible causes for this pleasant development are discussed. Nevertheless a gynaecologist on principle should think of a trichomonas-vaginalis-infection not only when her/his patients show specific symptoms as to burning, itching, discharge and dyspareunia, but in every case of discharge a trichomonas-infection should be excluded-or diagnosed and treated specifically, in special cases even by culture.

Trichomoniasis of the breast diseased by fibrocystic mastopathy: pathogenic rather than saprophytic relationship (Trichomonas in fibrocystic mastopathy process).

Because of the striking similarity between histopathological pictures of chronic trichomonal cervicitis uteri, the tissue reaction after subcutaneous inoculation of Trichomonas culture in experimental animals and female breast diseased by fibrocystic mastopathy, the detection of Trichomonads was undertaken in surgically removed diseased breast parts. In 12 FCM patients, subjected to segmental breast resection, the imprint smears were prepared from dissected specimens and after supravital staining immediately examined by light microscopy. The mucous content from dilated ducts was inoculated in the culture media. The dissected tissues have been further histologically analysed by standard method. In nine out of 12 examined FCM cases the direct microscopy revealed aflagellary, pseudocystic, leucocytoid form of Trichomonads. The cultures were positive in 4 cases: in 3 patients Trichomonas tenax was identified and in the last one T vaginalis. Histopathological findings in all 12 examined cases have shown the changes characteristic for FCM. On the basis of the accumulated knowledge about pathogenic capacity of Trichomonads, it can be with great certainty claimed that these protozoa, even in their pseudocystic form are able to cause the all appearances characteristic for FCM. This first report about Trichomonas infection in the middle of FCM process gives the unexpected hope in solving of its etiology and new insight into antitrichomonal host reaction which is frequently associated with epithelial dysplasia and unrarely with precancerous lesions as earlier observed in cervix uteri.

Trichomonas tenax proteolytic activity.

In this study, proteolytic activity of Trichomonas tenax collected directly from patient's dentobacterial plaque was examined. Electrophoretic method involving polyacrylamide gels (Commassie Brilliant Blue R-250) and electrophoretic method involving gelatin-containing polyacrylamide gels, have been used to analyse Trichomonas tenax proteolytic activity. The most obvious and the fastest activities were obtained when gels were incubated in pH 4.6; followed by results of incubating in pH 5.6; while in pH 2.8 activity was less effective but still obvious. Proteolytic activities were the most effective in area of protein MW 36 kDa. Different activities of enzymes depending on pH of incubated media indicate the presence of different endopeptidases in cell lysates of protozoon Trichomonas tenax from dentobacterial plaque.
