From cyclopentanone oximes to bis[1,2,3]dithiazolo-s-indacenes, cyclopenta[c][1,2]thiazine, pentathiepino-, tetrathiino-, and thienocyclopenta[1,2,3]dithiazoles as a rich source of new materials.

[Structure: see text]. The 1,5- and 1,7-s-hydrindacenedione dioximes reacted with S2Cl2 and iBu3N to give the first examples of bis[1,2,3]dithiazolo-s-indacenes; one of them was a near-infrared dye. In contrast, the silylated bicyclo[3.3.0]octan-2,6-dione dioxime reacted with S2Cl2 and Et3N to give a bicyclic 4-cyanoethylcyclopenta[1,2,3]dithiazole or, after addition of Li2S, a tricyclic 4-cyanoethyl-5,6-pentathiepinocyclopenta[1,2,3]dithiazole, also obtained from 2-cyanoethylcyclopentanone oxime, S2Cl2, and Hünig's base. In related reactions, 2-oxocyclopentylpropionate oxime gave the expected cyclopenta[1,2,3]dithiazole, in addition to an unexpected cyclopenta[c][1,2]thiazine that showed a reversible reduction wave in its CV at -0.95 V. Ethyl 2-oxocyclopentanecarboxylate oxime reacted with S2Cl2, Hünig's base, and Li2S to give a 5,6-tetrathiinocyclopenta[1,2,3]dithiazole derivative. Cyclopentathiophen-4-one oximes reacted with S2Cl2 and iBu3N to give thienocyclopenta[1,2,3]dithiazoles that showed UV-vis spectral bands that depended on the positions of the ring fusion.

Electronic structure of thiazine-indigo pigment on the basis of the crystal structure.

Thiazine-indigos are novel hydrogen-bonded pigments developed recently by Clariant that exhibit a variety of shades from yellow to red. Among these, a commercially available chlorinated derivative (THI) gives a color of vivid red, although it is only pale yellow in solution. For this reason, electronic characterization of THI has been carried out on the basis of the crystal structure. There are chains of intermolecular hydrogen bonds between the N-H group of one molecule and the O atom of the neighboring one along the b-axis, forming a two-dimensional hydrogen bond network. The direction of the transition dipole points along the long molecular axis, and the transition dipoles are arranged in "head-to-tail" fashion along the (b,-c) diagonal direction. The large bathochromic shift upon crystallization can be mostly attributed to excitonic interactions between transition dipoles arranged diagonally along the stacking axis as well as those in head-to-tail fashion on the molecular plane.

Quantitative structure-activity relationship study of N-(3-oxo-3,4-dihydro-2H-benzo[1,4]thiazine-6-carbonyl)guanidines as potent Na/H exchange inhibitors.

We have previously reported that N-(4-isopropyl-2,2-dimethyl-3-oxo-3,4-dihydro-2H-benzo[1,4]oxazine-6-car bonyl)guanidine (4b) methanesulfonate salt (KB-R9032) is a potent and highly water-soluble Na/H exchange inhibitor. In a series of studies on Na/H exchange inhibitors, we designed and synthesized N-(3-oxo-3,4-dihydro-2H-benzo[1,4]thiazine-6-carbonyl)guanidines (5) as more potent inhibitors with high water-solubility. The design strategy for 5 was based on a quantitative structure-activity relationship (QSAR) study, involving the proportional relationship between the biological activity and hydrophobicity of the ring structure of compounds 4. As expected, compounds 5 showed more potent activity than 4. It was found by using the QSAR analysis that 5 were about five-fold more potent than 4. The increase in potency of compounds 5 well agreed with our previous QSAR analysis result. The most potent derivative was the methanesulfonate salt 5d of the 4-isopropyl derivative (IC50=0.0091 microM). And in addition to the in vitro study, 5d showed significant protective activity against a rat acute myocardial infraction model.

[UV spectrophotometry in drug control. 31. Spectrophotometric behavior of drugs with dibenzo-p-thiazine chromophores].

The results of a systematic examination of the spectra of 34 drugs with dibenzo-p-thiazine chromophores in the ultraviolet and visible range were evaluated. The substances under study were subdivided into 10 different types corresponding to a different substitution in position 3 and in one case in position 5 of the phenothiazine ring system. All of these types are characterized by the bands E, K and B which are--with only a few exceptions--separated by well developed absorption minima.

Studies on annelated 1,4-benzothiazines and 1,5-benzothiazepines. XI. Synthesis and biological activity of several naphtho- and quinolino-1,4-thiazine and -1,4-thiazepine derivatives containing the imidazole ring.

Several derivatives of naphthol[1,2-b]imidazo[1',2'-d]-1,4-thiazine and -1,4-thiazepine and imidazo[1',2'-4,5]-1,4-thiazino[3,2-c]quinoline and -1,4-thiazepino[3,2-c] quinoline have been synthesized. These compounds and other imidazo[2,1-d][1,5]benzothiazepine derivatives, previously synthesized, have been tested for their possible pharmacological activities. One of these substances displayed inhibitory activity on CNS, others showed an appreciable antiinflammatory effect. None of the naphtho and quinolino derivatives showed affinity for the benzodiazepine receptor.
