Pharmacokinetics of (Me)Arg-Lys-Pro-Trp-tert-Leu-Leu-OEt and (Me)Arg-Lys-Pro-Trp-tert-Leu-Leu administered intravenously to beagle dogs.

The pharmacokinetics of two analogues (NT-1 and NT-2) of the smallest active fragment of neurotensin were investigated in beagle dogs after intravenous (iv) administration at the doses of 0.1 and 0.5 mg/kg for NT-1 and of 0.25 and 0.5 mg/kg for NT-2. After iv administration of these two drugs, the plasma levels decreased with time with a biexponential pattern, and linear kinetic behavior was observed. The pharmacokinetic parameters (mean +/- standard error of mean) after iv administration of NT-1 at 0.1 mg/kg were as follows: the half-life of the distribution phase (t1/2 alpha) was 0.29 +/- 0.11 h, the half-life of the terminal phase (t1/2 beta) was 1.99 +/- 0.41 h, total plasma clearance (CL) was 33.2 +/- 6.3 mL/h/kg, steady-state volume of distribution (Vdss) was 57.1 +/- 2.8 mL/kg, and mean residence time (MRT) was 2.20 +/- 0.55 h. The t1/2 alpha, t1/2 beta, CL, Vdss, and MRT values after iv administration of NT-2 at 0.25 mg/kg were 0.11 +/- 0.02 h, 0.58 +/- 0.04 h, 257.3 +/- 17.2 mL/h/kg, 180.3 +/- 4.6 mL/kg, and 0.71 +/- 0.04 h, respectively. After iv administration of NT-1, NT-2 was detected in the plasma. It was confirmed from this result that NT-1 was metabolically hydrolyzed to NT-2 in the body of beagle dog.

Crystallography and magnetism of radicals with hindered hydroxyl groups: 2-(3,5-di-tert-butyl-4-hydroxyphenyl)-4,4,5,5-tetramethyl-4,5-dihydro-1H-imidazole-3-oxide-1-oxyl and 2-(3,5-di-tert-butyl-4-hydroxyphenyl)-4,4,5,5-tetramethyl-4,5-dihydro-1H-imidazole-1-oxyl.

Radicals form networks of OH... ON and OH...Me(nitroxide) interactions. In 2, a frustrated network forms with insufficient N-O units to form extended chain interactions. The magnetism of 1 fits a 1-D Heisenberg model with J/k=-25 J mol(-1), while 2 shows more complex exchange behaviour consistent with its disordered crystal lattice.

tert-Butyl 2-[2-(ethoxycarbothioylamino)-3-pyridyloxy]acetate and tert-butyl 2-(3-thioxopyrido[2,1-c][1,2,4]thiadiazol-8-yloxy)acetate.

Compounds (I), C(14)H(20)N(2)O(4)S, and (II), C(12)H(14)N(2)O(3)S(2), are two minor products of the same reaction. Both structures contain identical ester functionalities in similar orientations. Both independent molecules of (I) contain an ethoxycarbothioylamine moiety, whilst (II) possesses a novel exocyclic thione system fused with a pyridine ring.

(1R,3S,6S)-6-tert-Butyl-1-hydroxymethyl-5-methoxy-6-methyl-7-oxa-4-azaspiro[2.5]oct-4-en-8-one and dimethyl [(1S,1'R,3'S,6'S)-(6'-tert-butyl-5'-methoxy-6'-methyl-8'-oxo-7'-oxa-4'-azaspiro[2.5]oct-4'-en-1'-yl)hydroxymethyl]phosphonate.

The molecules of the title compounds, C(13)H(21)NO(4), (I), and C(15)H(26)NO(7)P, (II), are linked into chains along the c direction in (I) and along the b axis in (II) through O-H.O hydrogen bonds. The heteroatomic ring in (I) adopts a twist-boat conformation, while that in (II) has a conformation intermediate between boat and twist-boat. The P atom has a distorted tetrahedral geometry.

[Bis[(S)-2-(4-tert-butyl-4,5-dihydrooxazol-2-yl)phenyl]phenylphoshine-kappa(2)N,P](1,3-dimethyl-pi-allyl-kappa(3)C)palladium(II) hexafluorophosphate and (S)(P)-[bis[(S)-2-(4-tert-butyl-4,5-dihydrooxazol-2-yl)phenyl]phenylphosphine-kappa(2)N,P]dichloropalladium(II).

In the structures of the title compounds, [Pd(C(5)H(9))(C(32)H(37)N(2)O(2)P)]PF(6) and [PdCl(2)(C(32)H(37)N(2)O(2)P)], the bis(dihydrooxazolyl)phosphine ligand is N,P-bidentate, with S chirality on the P atom. In the allyl complex, the pi-allyl ligand ligates in a syn-syn-kappa(3)C manner.
