[Autotransplantation of spleen tissue after traumatic spleen rupture (traumatic splenosis). A report of 4 cases].

Report on 4 cases of traumatic splenosis (autotransplantation of splenic tissues) which were diagnosed 1 to 15 years after traumatic spleen rupture in childhood or adolescence. In 3 of the 4 cases traumatic splenosis was detected casually, the other case was misinterpreted during laparoscopy as endometriosis. Histologically the autotransplants reveal functional red and white splenic pulp. This may be significant for hematologic diseases. Although clinical signs are often lacking or noncharacteristic, knowledge of traumatic splenosis is important for surgeons, internists (laparoscopy!), gynecologists, and pathologists, because splenic autotransplantants are often misinterpreted as accessory spleens, hemangiomas, endometriosis or peritoneal metastases of melanoma.

Characterization of a B cell progenitor present in neonatal bone marrow and spleen but not in adult bone marrow and spleen.

The neonatal period marks an important time in mammalian immunologic development, yet it is often ignored in studies of lymphocyte development. We identified a cell population with the phenotype heat stable Ag (HSA)low lin- CD43low that contained B cell progenitors at a high frequency in the neonatal bone marrow and spleen. Although cells with a similar phenotype can be identified in the bone marrow and spleen of adult animals, these populations showed a greatly reduced frequency of B cell progenitors. B lineage cells were detected after 7 days in culture at a frequency of 1:15 when HSAlow lin- CD43low cells from neonatal bone marrow were cultured on stromal cells and IL-7 under limiting dilution conditions. Under similar conditions, the equivalent population in adult bone marrow had a frequency of B cell progenitors that was less than 1:2000. The expression of terminal deoxynucleotidyl transferase in freshly sorted neonatal HSAlow lin- CD43low cells suggested that cells committed to the lymphocyte lineage were present in this population. These data suggested that the HSAlow lin- CD43low population of cells represents a pool of B lineage precursors that may be responsible for filling the immune compartment early in neonatal life.

Association of splenoma, peliosis and lipid histiocytosis in spleen or accessory spleen removed in 2 patients with chronic idiopathic thrombocytopenic purpura after long term treatment with steroids.

In 2 patients with a long lasting cortico-dependent chronic idiopathic thrombocytopenic purpura, splenectomy in one and ablation of an accessory spleen in the other, disclosed a complex association of splenoma, peliosis and lipidosis of the spleen, with a secondary sea blue histiocytosis in one case. The relation of this curious association with thrombocytopenia and with the long treatment with corticosteroids is discussed, with a review of the literature.

Biological activity of the spleen focus-forming virus is encoded by a molecularly cloned subgenomic fragment of spleen focus-forming virus DNA.

A biologically active subgenomic DNA fragment of a polycythemia-inducing strain of the replication-defective spleen focus-forming virus (SFFV) has been molecularly cloned. The SFFV DNA fragment includes 2.0 kilobase pairs (kbp) from the 3' end of SFFV, the long terminal repeat sequences of SFFV, and 0.4 kbp from the 5' end of SFFV. The fragment contains the previously described env-related gene of SFFV. All the properties associated with SFFV can be assigned to this SFFV DNA fragment by using a two-stage DNA transfection assay with infectious helper virus DNA. The virus recovered from the transfection assays can induce erythroblastosis, splenic foci, and polycythemia in infected mice. Fibroblast cultures transfected with the SFFV DNA fragment synthesize gp52, the known intracellular product of the env-related gene of SFFV. gp52 can also be detected in spleens from diseased mice infected with the virus recovered in the two-stage transfection. The results are consistent with the hypothesis that the env-related gene sequences of SFFV and their product gp52 are required for the initiation of SFFV-induced disease.

[Treatment of childhood anorexin with spleen deficiency by principle of invigorating spleen and benefiting qi].

UNLABELLED: 101 children of anorexia with Spleen deficiency were divided randomly into two groups, the invigorating Spleen(IS) group of 58 cases treated by modified Yi-Gong-San, and the replenishing zinc(RZ) group of 43 cases treated by 0.2% zinc sulphate syrup. One course of treatment was 3 months. RESULTS: the clinical effects of IS group were notable in promoting appetite, increasing body weight and height, eliminating polyhidrosis, preventing common cold and tracheitis etc. The total effective rate was 96.6% in IS group which was significantly better than 72.1% in the RZ group (P less than 0.01). The effect of examination showed that the levels of hair zinc after treatment were markedly increased in both groups (P less than 0.01). The average increased value of hair zinc was 62.2 +/- 9.1 (ppm) in IS group which was higher than 42.1 +/- 7.2 (ppm) in RZ group (P less than 0.05). Moreover excretion rate and Hb after treatment in the IS group were markedly higher those in the RZ group (P less than 0.01). Chemical analysis showed that the concentration of zinc in modified Yi-Gong-San was very slight (0.73 mg/pack). It indicated that arising hair zinc in IS group was not simply supply of zinc but also the treatment in appetite, digestion and absorption, with which the body was supplied enough zinc naturally by food taken.

Laparoscopic spleen-preserving pancreatic tail resection for an intrapancreatic accessory spleen mimicking a nonfunctioning endocrine tumor: report of a case.

Laparoscopic surgery is now performed for several pancreatic disorders, such as benign tumors of the pancreatic body or tail, which are a good indication for laparoscopic resection. However, the risk of pancreatic fistula after distal pancreatectomy, performed laparoscopically or by open surgery, is a topic of debate. We report the case of a 61-year-old man in whom a routine follow-up computed tomography (CT) scan showed a solid, well-defined mass, 1.5 cm in diameter, in the pancreatic tail. The mass was homogeneously enhanced from the early phase to the super-delayed phase on enhanced CT. We suspected a nonfunctioning endocrine tumor of the pancreas, and surgery was performed laparoscopically. After dissecting the pancreatic tail away from the splenic hilum and the splenic vessels, it was resected using only a linear stapler. The histological diagnosis was an intrapancreatic accessory spleen. The patient was discharged on postoperative day 14, but was readmitted 6 days later because of a pancreatic fistula, which was treated by CT-guided percutaneous drainage.

Electron exchange between Fe(II)-horse spleen ferritin and Co(III)/Mn(III) reconstituted horse spleen and Azotobacter vinelandii ferritins.

Azotobacter vinelandii bacterioferritin (AvBF) containing 800-1500 Co or Mn atoms as Co(III) and Mn(III) oxyhydroxide cores (Co-AvBF, Mn-AvBF) was synthesized by the same procedure used previously for horse spleen ferritin (HoSF). The kinetics of reduction of Co-AvBF and Mn-AvBF by ascorbic acid are first-order in each reactant. The rate constant for the reduction of Mn-AvBF (8.52 M(-1) min(-1)) is approximately 12 times larger than that for Co-AvBF (0.72 M(-1) min(-1)), which is consistent with a previous observation that Mn-HoSF is reduced approximately 10-fold faster than Co-HoSF [Zhang, B. et al. (2005) Inorg. Chem. 44, 3738-3745]. The rates of reduction of M-AvBF (M = Co and Mn) are more than twice that for the reduction of the corresponding M-HoSF. HoSF containing reduced Fe(II) cores (Fe(II)-HoSF), prepared by methyl viologen and CO, also reduces M-HoSF and M-AvBF species, with both cores remaining within ferritin, suggesting that electrons transfer through the ferritin shell. Electron transfer from Fe(II)-HoSF to Co-AvBF occurs at a rate approximately 3 times faster than that to Co-HoSF, indicating that the Co cores in AvBF are more accessible to reduction than the Co cores in HoSF. The presence of nonconductive (SiO2) or conductive (gold) surfaces known to bind ferritins enhances the rate of electron transfer. A more than approximately 4-fold increase in the apparent reaction rate is observed in the presence of gold. Although both surfaces (SiO2 and gold) enhance reaction by providing binding sites for molecular interaction, results show that ferritins with different mineral cores bound to a gold surface transfer electrons through the gold substrate so that direct contact of the reacting molecules is not required.

Right-sided spleen with partial situs inversus of the abdominal viscera: visualization of the spleen by dynamic and static 99mTc-sulfur colloid imaging and 51Cr-labeled red blood cell sequestration imaging.

In a patient with right-sided stomach and malrotation of the small bowel and cecum visualized by gastrointestinal x-ray studies using barium, a right-sided spleen was demonstrated by 99mTc-sulfur colloid and 51Cr-labeled red blood cell imaging. The sulfur colloid study also demonstrated that the liver was in its usual right upper quadrant location.

The utility of SPECT liver-spleen imaging in the evaluation of a possible accessory spleen.

A case where an x-ray CT examination showed the existence of an unidentified perisplenic mass is presented. This mass was determined to be an accessory spleen after an evaluation with Tc-99m labeled sulfur colloid. This report describes a clinical indication for the use of SPECT imaging of a patient when a mass of uncertain significance is discovered.

Repopulation of spleens of irradiated mice after injection of spleen cell subpopulations.

Haemopoietic stem cells present in the spleen of adult mice were analysed by grafting X-irradiated animals with polystyrene-nonadherent (NABS) and polystyrene-adherent (ABS) B-enriched splenocytes from syngeneic donors. The progeny of the haemopoietic stem cells present in NABS and ABS subsets were studied with respect to size, surface markers, and response to mitogens and antigens. Ninety-six per cent of the precursors of the myeloid cell lineage (CFU-S) were present in the NABS fraction (50-fold enrichment). The presence in NABS of progenitors of functional T and B lymphocytes was also demonstrated. Twelve days after grafting with NABS, more than 80% of the recipient splenocytes were large and nonadherent granulocyte-like cells. These cells had surface similarities with NABS from normal mice, since both populations reacted with peanut agglutinin and with a rabbit anti-NABS (RAN) serum.

Murine spleen culture: homing of hemopoietic progenitor cells to spleen is not mediated by a similar mechanism to that in marrow.

We have previously shown that in long-term bone marrow cultures (LTMC) the specific recognition and binding of hemopoietic stem cells to stroma, which we call "homing," is mediated by a recognition mechanism involving a surface membrane lectin with galactosyl and mannosyl specificities. Subsequent in vivo studies in lethally irradiated mice confirmed that homing to the marrow similarly involves a galactosyl- and mannosyl-specific recognition mechanism. However, these in vivo studies suggested that homing of hemopoietic progenitor cells to spleen was based upon a different molecular recognition mechanism. In the present study splenic homing was investigated in a cell culture system composed of an adherent layer of splenic stromal cells inoculated with stroma-free stem cells from the supernate of LTMC. In this system, splenic stroma supported proliferation and differentiation of hemopoietic precursors for a few weeks. When stem cells were added to the cultures in the presence or absence of inhibitory concentrations of neoglycoprotein reagents specific for galactosyl, mannosyl, or fucosyl lectins, the pattern of production of total cells, pluripotential stem cells (CFU-S), and granulocyte-macrophage committed progenitors (CFU-GM) remained the same. These data support our in vivo observations that homing of stem cells to splenic stroma is not mediated by a surface lectin with galactosyl and mannosyl specificities as it is in bone marrow, but rather by a different molecular mechanism.

The blood circulation inside the spleen. I. The arterial blood vessels general distribution in the rat spleen.

As an attempt to investigate the different pathways followed by the blood into the spleen and to analyse their functional significance, a technique was used mainly based on the intraarterial perfusion of a Prussian blue "solution" added of some chemical mediators and vasoactive substances. Such technique provides results which may be analysed taking into account the effect of the anaesthetic used, that may influence the findings. From the anaesthetic used, the sulfuric ether and the barbital sodium produce vasoconstriction of the white pulp blood vessels, whereas the chlorpromazine-promethazine doesn't have this effect, and so the Prussian blue appears inside these vessels. The vasodilator drugs, such as succinonitrile and papaverine hydrochloride, show a general vasodilator effect on the splenic arterial system. Teh arterial vessels of the white and the red pulp, including those placed at the subcapsular areas, become enlarged; into the white pulp, either the central or the peripheral blood vessel plexus of the lymphatic follicle becomes evident. The latter readily constitutes the perifollicular and the pericolumnar plexus. The blood vessels of this plexus become permeable to the Prussian blue "solution" by the heparin sodium effect, and so the dye particles enter the marginal zone and the splenic sinuses. In addition, from the white pulp arteries arise 2 types of anastomotic arterioles which appear enlarged after succinonitrile treatment: The short anastomotic arterioles that crosses the marginal zone entering the red pulp near the white pulp; the long anastomotic arterioles which enter the red pulp and after a long course end up into or around a collector sinus. The addition of histamine dihydrochloride to the perfusion solutions shows a slight vasodilator effect mainly on the subcapsular penicillar arterioles, including the helicine arterioles. The adrenergic stimulation of the splenic blood vessels induces a generalized arterial constriction, except of the anastomotic arterioles, that becomes open; in such way, the blood pathway follows the course of the anastomotic arterioles and the collector arterioles also become constricted. The adrenergic vasoconstrictor effect is inhibited by the phenoxy-benzamine hydrochloride. The addition of acetylcholine chloride, in the dosage, used, induces a generalized arterial vessel constriction, mainly of the perifollicular plexus. This effect is inhibited by atropine sulfate which, on the other hand, produces evident enlargement of the perifollicular and pericolumnar arterial plexus.

An electron microscope study of the perfusion-fixed spleen. II. Fine structure of the sinus lining cells with respect to filamentous structures in monkey spleens.

In the perfusion-fixed spleen of monkey (Macaca mulatta) were studied with the electron microscope the cytoplasmatic structures of the sinus lining cells. The particular interest find the contractile filaments in this cells, which are important for the cell contraction. The connections to the smooth endoplasmatic reticulum are very strong as in the muscle fiber or muscle cell.

Influence of classic immunosuppressants on spleen colony formation in mice. 3. Concentration of colony forming units in bone marrow and spleens of cyclophosphamide-treated mice.

The concentration of colony forming units (CFU) increased nonsignificantly in bone marrow, and significantly in spleens of cyclophosphamide-treated mice, but there was no increase in the absolute number of CFU in these organs due to decreased celullarity. The serum of mice pretreated with cyclophosphamide seems to have colony-stimulating activity.

An experimental method for rapid growth of liver in spleen. The survival and proliferation of chemically induced preneoplastic hepatocytes in spleen.

Cellular suspensions (2 x 10(6) cells) of isolated preneoplastic liver cells, obtained from carcinogen-treated rats, were injected in the spleens of syngeneic rats divided into groups on the basis of no treatment, partial hepatectomy (PH), and/or feeding regimens including 2-acetylaminofluorene (AAF). Recipient rats undergoing both PH and AAF showed significantly more rapid proliferation of the preneoplastic liver cell implant, compared with other treatment groups and control. The theoretic basis for this observation, supported by a large body of data derived from hepatocarcinogenesis, is as follows: The phenotype of the donor cells has been altered by chemical carcinogens such that the liver cells develop resistance to growth-inhibiting agents such as AAF. The recipient receives PH and AAF, the former creating a strong proliferative stimulus for hepatocytes, while the latter inhibits regeneration of normal liver cells but not those resistant to the mito-inhibitory effect of AAF, ie, the carcinogen-altered donor cells. These manipulations in donors and recipients thus create a selective environment in which the implant undergoes rapid proliferation. This model of resistance induction followed by selective proliferation, built upon the principles of carcinogenesis and applied to isolated liver cell transplantation, provides an experimental basis for achieving rapid liver growth of the splenic implant.

Save the spleen--an experimental study on the effects of three tissue adhesives on deep wounds of liver and spleen.

Because opinions on the advisability of splenectomy, especially in children, have recently changed considerably, an experiment was set up to study the application of 3 hemostatics in deep injuries to the liver and to the spleen in 29 rabbits and to dogs. Of the 3 substances tested GRF-adhesive and Avitene appeared to give reasonably good results in splenic injuries, but a supplementary suturing procedure or a ligation of the vessels seemed desirable. The effect of GRF-adhesive on liver injuries was satisfactory.

An electron microscope study of the perfusion-fixed spleen. III. Fine structure of germinal centers in monkey spleens with special reference to the so-called "dendritic reticular cells".

The spleen white pulp of monkey (Macaca mulatta) was studied on electron microscopical level with special reference to cell composition of the light zone of follicular germinal or reactive centers. The presence of a special kind of the so-called "dendritic reticular cells" whose origin appear to have a bearing only on the germinal center formation was confirmed. These non-lymphoid cells, characteristic principally of a well developed Golgi complex and a conspicuous cell coat, overlying mainly the very fine ramified peripheral cytoplasmic processes, seem to support differentiation of small lymphocytes (B-cells). This specialized cell type of peculiar shape and function may be derived most probably from the rest structural reticulum cells under proper circumstances.
