[Effects of Radix Salviae Miltiorrhizae on the PMN-EC adhesion in vitro at the early stage of endotoxemia].

OBJECTIVE: To evaluate the effects of Radix Salviae Miltiorrhizae (RSM) on the PMN-EC adhesion in vitro at the early stage of endotoxemia, and to probe into the mechanism there in involved. METHODS: The rabbit endotoxemia model was made, and one group was treated with RSM injection (2 ml/kg) instantly and 24 h after the modeling of endotoxemia. PMN-EC adhesion rate, adhesion molecule (CD11a/CD18 and CD11b/CD18) and TNF-alpha level were measured instantly, 2 h, 4 h, 8 h, 16 h, 24 h and 48 h after endotoxemia modeling. RESULTS: It was found that after the modeling of endotoxemia, the levels of CD11a/CD18, CD11b/CD18 and PMN-EC adhesion rate instantly increased in the endotoxemia group and RSM group, they went up to the peak at 4 h in the endotoxemia group, and at 2 h, 4 h, 8 h, 16 h, 24 h and 48 h, they were markedly lower in the RSM group than in the endotoxemia group (P < 0.05). The TNF-alpha level in the endotoxemia group began to increase 2 h after the modeling and reached to the peak at 8 h, with its level still higher than normal at 48 h, while the RSM group had lower level of TNF-alpha at all time-points(P < 0.05). CONCLUSION: The data from this animal experiment indicate that Radix Salviae Miltiorrhizae can decrease PMN-EC adhesion rate and the levels of CD11a/CD18, CD11b/CD18 and TNF-alpha in case of endotoxemia so as to improve the microcirculation.

[Purification and fingerprinting development of Salvia miltiorrhiza Bunge by high-speed counter-current chromatography].

In an attempt to apply high-speed counter-current chromatography HSCCC for TCM fingerprints, the separation and purification of the Chinese medicinal plant Salvia miltiorrhiza Bunge of different localities was realized using the technique. The equipments used include a HSCCC (TBE-300) of Shenzhen Tauto Biotech containing three connected preparative coils (diameter of tube = 2.6mm, total volume = 300mL) and a 20mL sample loop and a HPLC from Shimadzu of Japan with a Ultrasphere C18 column (150 x 4.6mm ID, 5microm) and a 20microL sample loop. Salvia miltiorrhiza Bunge samples from 3 locations were separated by HSCCC in a Step-wise elution program with solvent systems A (hexane:ethanol: water = 10:5.5:4.5) and B (hexane:ethanol: water = 10:7:3) at a speed of 900 r/min and a flow-rate of 2mL/min. All the 12 peak fractions were eluted within 13 hours. The contents of each component varied greatly in different samples, which confirmed previous observation that the locations and climates have a great impact on the TCM quality and also indicated a quality control system is necessary to safeguard the quality of the herb. The retention times of the 12 peak fractions from crude extracts of the samples were collected by HPLC and the absorption spectrums of the corresponding peaks were identified. The 12 components of the three crude samples were readily distinguishable and can be used as fingerprints of S. miltiorrhiza Bunge. The relative standard deviation of the HSCCC retention times was less than 3%, which satisfies the requirement of the national standard reference index. The components 7, 8 and 11 from the standards were identified to be crypototanshinone, tanshinone I and tanshinone II A respectively. This study demonstrates that if it is possible to apply HSCCC for TCM fingerprinting, especially with samples of high viscosity and highly absorptive components. The precision and the run time of fingerprinting can be further improved if larger volume and a temperature control system is used. With these and other improvements, HSCCC is expected to play an important role in TCM development.

Compound salvia pellet, a traditional Chinese medicine, for the treatment of chronic stable angina pectoris compared with nitrates: a meta-analysis.

BACKGROUND: Compound salvia pellet (CSP), consisting of active herbal ingredients extracted from Danshen (salvia miltiorrhiza), Sanqi (panax notoginseng), and Borneol (Cinnamomum camphora), is taken most frequently by patients with angina pectoris in China. To determine the efficacy and safety of CSP for the treatment of stable angina pectoris (SAP), a meta-analysis was undertaken. MATERIAL/METHODS: An extensive search including MEDLINE, EMBASE, BA, Chinese Biomedical Database (CBM), and Chinese Cochrane Centre Controlled Trials Register from 1994 to 2004 was performed. Data were extracted independently from the included trials by two reviewers. Statistical software (RevMan 4.1) provided by the Cochrane Collaboration was applied. RESULTS: Twenty-seven randomized controlled trials (RCTs) (n=3722), regardless of language or publication status, were identified. Generally, the methodological quality of the trials, assessed by the Jadad scale, was low, except for one with 3 points. Statistical pooling of the results showed that, compared with nitrates, CSP treatment had significant effect on the improvement of angina symptoms (RR=1.13, 95%CI=[1.07, 1.20]), showed greater increased effect on the improvement of electrocardiogram (ECG) results (RR=1.39, 95%CI=[1.28, 1.50]), and the percentage of patients with adverse events was significantly decreased in the treatment of CSP in comparison with nitrates (2.4% vs. 29.7%). CONCLUSIONS: CSP has significant effect on the improvement of angina symptoms and ECG results with few adverse events. However, the methodological quality of clinical trials with CSP for SAP needs to be improved, and the outcome measures should include mortality, quality of life, and other end-points.

[The effect of Inj. Salviae Miltiorrhizae Co. on the retrograde axoplasmic transport in the optic nerve of rabbits with chronic IOP elevation].

With the horseradish peroxidase histochemical technique and electron microscope, the effects of 0.5% timolol, Inj. Salviae Miltiorrhizae Co. and their combination on the retrograde axoplasmic transport in the optic nerve of 230 rabbit models of chronic IOP elevation were studied. The results showed that (1) the nerve damage in chronic glaucoma was due to multiple factors; (2) the use of a drug that improved microcirculation in combination with an IOP depressor better protected the optic nerve functions than did the latter alone; and (3) Inj. Salviae Miltiorrhizae Co. improved the optic nerve axoplasmic transport under chronic IOP elevation. The mechanism of the drug in protecting the optic nerve could be through its actions of improving the local microcirculation and the tolerance of nerve tissues to anoxia.

[Prevention of radiation injury of the lungs by Salvia miltiorrhiza in mice].

The lungs and thymus of mice received X-ray irradiation on the right thorax were observed. It was found that lung injury was lighter and got recovered more rapidly in mice given Salvia miltiorrhiza than that in the control group. Microscopic thymus changes showed no difference in the two groups. The results indicated that Salvia miltiorrhiza could prevent radiation-induced pulmonary injury, but could not prevent thymus injury.

Isolation of a renal function-facilitating constituent from the Oriental drug, salviae miltiorrhizae radix.

An attempt was made to isolate the active component which exhibits an improving effect on renal function from Salviae Miltiorrhazae Radix (Chinese crude drug). Systematic isolation from aqueous extract of Salviae Miltiorrhizae Radix was carried out, and Compound 1 was found to be more effective than any of the other constituents in improving renal functional parameters; that is, a marked reduction of glomerular filtration rate following adenine ingestion was improved by administration of this substance. The renal plasma flow and renal blood flow were also increased in renal failure rats. On the basis of chemical and spectroscopic data, Compound 1 was shown to be identical with magnesium lithospermate B.

[Clinical research of compound salviae miltiorrhizae injection for severe pancreatitis].

Compound salviae miltiorrhizae injection was administered after operation for 28 cases of severe pancreatitis, and 13 cases were taken as a control group. The results showed that: (1) the difference was not obvious in the morbidity of complications between the two groups, but the mortality (3.6%) of the trial group was significantly lower than that (30.8%) of the control group (P < 0.05); (2) Hematocrit was clearly decreased from 46.1 +/- 5.2% to 33.2 +/- 3.9% in the trial one (P < 0.05), but platelet and hemoglobin showed no statistical significance. It is concluded that compound salviae miltiorrhiza injection might improve hemorheologic abnormalities of the disease, promote the recovery of the pancreatic tissue, and correct the serious complications such as adult respiratory distress syndrome etc.

Experimental study on ischemia and reperfusion injury of rat liver and effects of ligustrazine and salvia compound.

The present study was to investigate changes of free radicals in the whole rat liver, changes of ATP levels of hepatic cells, ultrastructural changes in hepatic tissue during ischemia and reperfusion in rats and the effects of ligustrazine and salvia compound. The results indicated that: 1) the free radical levels increased by 8.3-fold and 9.0-fold in the groups with 30 and 60 min ischemia after 40 min reperfusion, as compared with the group with ischemia only (P < 0.01, P < 0.01, respectively). 2) The levels of ATP returned to normal in the group with 30 min ischemia after reperfusion for 40 min and 3 d, remained low in the group with 90 min ischemia and fell again after a mild increase in the group with 60 min ischemia. 3) The hepatic and endothelial cell damage after ischemia became more severe after reperfusion, as revealed by electron microscopy. The present study also showed that ligustrazine and salvia compound have protective effects against reperfusion injury. They can be used to scavenge free radicals, improve hepatic microcirculation and alleviate hepatic and endothelial cell damage.

[Effect of radix salviae miltiorrhizae on the mitotic activity of osteoblast-like cells isolated from chicken embryo calvariae cultured in vitro].

Chicken embryo calvariae cultured in vitro, could yield 3 types of osteoblast-like cells, which differed in morphology. They were the spindle-shaped cells, globular cells, and squamous cells. Of these, the squamous cell was the chief cellular component, while both the spindle-shaped cells and the globular cells gradually transformed into squamous cells. Administration of Radix Salviae Miltiorrhizae rendered more rapid growth of the osteoblast-like cells in the early stage of culture. After 17-day of culture, when both the control group cells and the treated, group cells became quite confluent, 3H-Thymidine labelling and radioautographic study revealed a very statistically significant increase in the number of cells, bearing silver grains over the nucleus area in the treated group, as compared with the control (P < 0.001). Since osteoblasts play a pivotal role in fracture healing, administration of Radix Salviae Miltiorrhizae can exert favourable influence on bone repair through increasing the number of osteoblasts.

[Separation and quantitative determination of seven aqueous depsides in Salvia miltiorrhiza by HPTLC scanning].

A new analytical method for the separation and determination of seven aqueous depsides in salvia miltiorrhiza by HPTLC has been developed. The seven depsides are protocatechualdehyde, caffeic acid, methyl rosmarinate, rosmarinic acid, salvianolic acid A, B and C. Using chloroform-ethyl acetate-benzene-formic acid(2.4:2:1:0.6) as developing solvent A, protocatechualdehyde was separated; using chloroform-ethyl acetate-benzene-formic acid-methanol (1.5:2:1:1:0.1) as developing solvent B, the other six constituents were well separated. The aqueous depsides were detected at the wave lengths of lambda S = 300 nm and lambda R = 240. This method is simple, rapid, sensitive and accurate. The contents of seven depsides in several Salvia species were determined.

[Experimental study of the protective effects of astragalus and salvia miltiorrhiza bunge on glycerol induced acute renal failure in rabbits].

The astragalus and salvia miltiorrhiza bunge alcohol extracts were used in preventive treatment of glycerol induced acute renal failure (ARF) in rabbits. The experimental rabbits were divided: astragalus group (AB), salvia miltiorrhia bunge group (SM), two extracts mixture group (ABSM), and normol saline control group (C). The extracts, mixtures and normol saline were administrated before and after the induced ARF. The counts of Na, K, creatinine in urine and blood and urinary AAP, NAG were determinated during 24-14 days periodically in all 4 groups, and the renal tissues were taken from same periods for pathomorphological studies by microscopy and transmission electromicroscopy. The damage of the glycerol induced ARF was not only in the convolute tubules but also in the glomerulus. The glomerulor filtration rate reduced abruptly, and oligouria or auria developed. The study of renal functions and renal morphogy showed that the AB and ABSM groups were damaged more lesser and promptely repaired than the SM and C groups in the early stage. No glomerular sclerosis was noted in the AB, ABSM groups in later stage, but it occurred sporadically or diffusely in the SM, C groups. Therefore, astragalus is an ideal protective drug of traditional chinese medicine for ARF.

[Effect of salvia miltiorrhiza composita on superoxide dismutase and malonyldialdehyde in treating patients with non-insulin dependent diabetes mellitus (NIDDM)].

OBJECTIVE: To assess the effect of Salvia Miltiorrhiza Composita (SMCo) in treating non-insulin-dependent diabetes mellitus (NIDDM). METHODS: The superoxide dismutase (SOD) and malonyldialdehyde (MDA) levels of 20 patients with NIDDM were observed before and after treatment with Salvia Miltiorrhiza Composita and 20 patients without using SMCo were studied as controls. RESULTS: The SOD levels were significantly lower than that of normal (P < 0.05), and MDA significantly increased (P < 0.01). After treatment with SMCo, the SOD levels were significantly increased (P < 0.01), there was no difference between the treatment group and the normal group. In the control group, the SOD levels had increase tendency and MDA had decrease tendency, but there was significant difference compared with the normal (P < 0.05, P < 0.01 respectively). The total effective rate of anti-lipid peroxidation injury in the group combined with intravenous SMCo was markedly higher than in the group without SMCo therapy (90% vs 60%, P < 0.01). CONCLUSION: SMCo could resist lipid peroxidation injury.

Antimicrobial study of the resinous exudate and of diterpenoids isolated from Eupatorium salvia (Asteraceae).

The antimicrobial properties of the resinous exudate from twigs and leaves of Eupatorium salvia were tested against five gram-negative and five gram-positive bacteria. Comparison of the antimicrobial activities of 7-hydroxy-8(17)-labden-15-oic acid (salvic acid) and of its acetate, both compounds isolated from the plant, with that of the crude extract suggested that the latter ester derivative was the major active component in the exudate. These results validate the vernacular medicinal use of Eupatorium salvia as an antiseptic agent.

Chemical composition and antimicrobial action of the essential oils of Salvia desoleana and S. sclarea.

The chemical composition of the essential oils of Salvia desoleana Atzei & Picci and Salvia sclarea L. from Sardinia (Italy) was analysed by GC and GC/MS. S. desoleana oil had a high content of monoterpenic esters (linalyl acetate and alpha-terpinyl acetate) and a lower amount of the corresponding alcohols while S. sclarea oil was characterised by a higher content of alcohols and lower quantity of esters. We studied the antimicrobial activity of these oils concerning their use in pharmaceutical preparations for local application. Only weak microbiostatic inhibitory activity was seen against S. aureus, E. coli, S. epidermidis and C. albicans, but since inhibition increased progressively with contact time, better results could be obtained by using these oils in bioadhesive formulations that would also have anti-inflammatory and peripheral analgesic action at a local level, as demonstrated in experimental animals following systemic application.
